The variability within schistosome populations was explored using mixed populations of cercariae from multimiracidial snail infections and individual clones of Schistosoma mansoni cercariae obtained from monomiracidial snail infections. We investigated the heterogeneity between different clones of S. mansoni with respect to infectivity and metabolism. One difference between clones of cercariae was found in the recovery of adult worms from Balb/C mice. Recovery of adult worms was greater after infections with a mixed population than with a clonal population. To investigate some biochemical features of individuals in clones or mixed populations, the uptake of [35S]methionine into individual parasites and their membrane proteins was measured. Isoelectric focusing of a soluble membrane fraction: the frozen–thawed supernatant extracted from individual clones, showed the presence of proteins of isoelectric point between 7.2 and 8.2 in all clones. These proteins were less labelled with [35S]methionine in the clones than in the mixed population. It was concluded that basic proteins are synthesized by all clones and in the mixed population but at different rates. Differences in the rate of incorporation of [35S]methionine into the surface membranes of schistosomula and adult worms derived from individual clones are reported. In addition, a direct correlation between the percentage of recovery of adult worms from mice infected with individual clones of S. mansoni and the rate of incorporation of [35S]methionine into schistosomula of these particular clones was observed. It is suggested that the high rate of metabolism shown by an individual clone may account for the enhanced survival of the cercariae derived from that clone during penetration of the skin and migration through the vertebrate host. In order to examine individuals in a population of schistosomula, from a clone or mixed population, the lysosome-specific fluorescent probe LysoTracker DND-99 was used to label the parasites and quantitative fluorescent measurements were made on individual parasites. There were significant differences between clones and a mixed population. Furthermore, the variation between individuals from a mixed population was greater than from that in any clone, just as was found in the infectivity studies. Freshly transformed schistosomula of individual clones labelled with the LysoTracker DND-99 showed less variations in the quantitative uptake of the dye within a single clone when compared to the mixed population. We conclude that for any biochemical and biological parameter, a population of cercariae consists of individuals showing a wide range of values, with a much greater range in a mixed population. This variability is likely to have great relevance for infectivity of the final host and the efficacy of drugs and the immune system.

A comparison was made between 14-day-old populations of Nematodirus battus from single infections and from those which developed concurrently with Haemonchus contortus from the day of infection and from days 5 and 10 after infection. The numbers of Nematodirus fell with decreasing length of exposure to Haemonchus and were negatively correlated with the numbers of Haemonchus. The percentage of the populations of Nematodirus in the adult stage and the mean worm lengths fell with increasing length of development with Haemonchus, the lowest values being found in infections which developed concurrently with the abomasal worm for the full 14 days (days 0–14). Mean Nematodirus worm lengths were negatively related to abomasal pH and Na+ levels in populations which were concurrent with Haemonchus for the last 9 days (days 5–14) and for the full 14 days of development studied. Nematodirus egg capacity also fell with exposure to Haemonchus, but was lower in the worm populations exposed for the last 9 days than in those exposed for 14 days. Egg capacity was related to abomasal electrolyte levels in populations which had developed concurrently with Haemonchus for the last 9 days and for the full 14 days. Significant differences between the population parameters of Nematodirus from the proximal and middle third of the small intestine were only found in the single Nematodirus infections and in those which were concurrent with Haemonchus for the last 4 days (days 10–14). The differences in the populations of Nematodirus were related to the probable changes in alimentary conditions affected by the different periods of concurrent infections of Haemonchus. It was suggested that the alimentary changes may have caused the loss of adult Nematodirus and have influenced the growth, development and reproductive capacity of the intestinal worm by acting upon the fourthand fifth-stage larvae.

1. In a mixed infection in which Haemonchus contortus, Ostertagia circumcincta and O. trifurcata predominated, the lengths were plotted against widths of eggs obtained from faeces.

2. Two-dimensional groups approximating to normal frequency distributions, showing little overlap, were observed.

3. The statistical ranges of means plus and minus twice the standard deviation for the two groups barely touched. These were, to the nearest micron, 68–84 by 41–51 for Haemonchus contortus and, for the Ostertagia group, 83–103 by 46–52.

A new flagellate, Proteromonas hemidactyli sp.nov., is described from the large intestine of the lizard, Hemidactylus brooki, in Hyderabad, India. It is characterized by a long, slender and fusiform body, two similar but subequal flagella, a kidney-shaped sub-blepharoplastic body, a ribbon-like paranuclear body and a vesicular nucleus. A parabasal body is absent.

An important element in most approaches to the subject of parasitism is the consideration of environment. Parasites are set apart within animal ecology because they experience two environments, one the ‘external’ conditions and the other created by the living body of the host. As in any ecological system, external environmental conditions have a major influence on life history parameters: these conditions may be experienced directly by ‘off-host’ stages of a parasite or, to a greater or lesser extent, indirectly through the body of the host. However, uniquely in parasitic associations, the internal (host) environment has a dual influence on the physiological conditions encountered by parasites: first, the host buffers the external conditions (by homeostatic mechanisms moderating environmental fluctuations, by behavioural responses selecting appropriate habitat conditions, etc.), but second, the host creates a suite of hostile factors associated with immune defence. This living, reactive environment has no parallels elsewhere in free-living animal ecology: it has the characteristic of reacting specifically to kill the organisms within its boundaries.

The ultrastructure of the pseudohaptoral squamodiscs of the monogenean Diplectanum aequans is described. The most prominent feature of the squamodisc is the presence of epidermally-embedded spines which are covered by the outer epidermal membrane. These spines, which are ‘hollow’, are composed of a moderately electron-dense material with denser fibrils embedded within it. The squamodisc epidermis is similar in appearance to the haptor epidermis, with the exception of the epidermis ‘inside’ the spines. Here the epidermis contains free ribosomes and stacks of alternating electron-lucent/electron-dense, membrane-bound sacs. The arrangement of the musculature of the spines is described. The free, prehensile, edges of the squamodiscs contain numerous radially orientated nerves, some of which are assumed to be mechano-receptors.

The foundation of an Institute for Research in Parasitology in Cambridge is an achievement worthy of being recorded in some detail since it affords a recognition of the value of a branch of biological science that cannot but gratify all who have gained an insight into the deep significance of the many problems which parasitology presents when viewed from the standpoint of pure science.

Dinobdella ferox is the most reported and studied leech species parasitizing mammalian mucus membranes. However, our knowledge of D. ferox during parasitizing and its effects on hosts has remained limited. To investigate the behaviour of D. ferox in the parasitic period and the subsequent host symptoms, the author was infested voluntarily with three juvenile leeches under regularly-conducted examinations of complete blood count (CBC) and nasal endoscopy (NE). After the first head protrusion of the parasitizing leech at the host nostril, the occurrence of bleeding-related symptoms decreased and ceased completely weeks before the end of the parasitic period. Negligible impacts of the parasitizing leech on the host were shown by CBCs, and a parasitizing leech was able to hide from NE despite being so large it at times protrudes out of the host nostril. Finally, parasitizing D. ferox were coaxed to emerge at the nostril by water only after their first head protrusion, indicating that non-invasive removal of parasitizing D. ferox before head protrusion might be difficult. Through the voluntary infestation, the full documentation of behaviours of D. ferox in the parasitic period and host symptoms shown in this study will promote better diagnoses and treatments of nasal leech infestation.

Mathematical modelling provides an effective way to challenge conventional wisdom about parasite evolution and investigate why parasites ‘do what they do’ within the host. Models can reveal when intuition cannot explain observed patterns, when more complicated biology must be considered, and when experimental and statistical methods are likely to mislead. We describe how models of within-host infection dynamics can refine experimental design, and focus on the case study of malaria to highlight how integration between models and data can guide understanding of parasite fitness in three areas: (1) the adaptive significance of chronic infections; (2) the potential for tradeoffs between virulence and transmission; and (3) the implications of within-vector dynamics. We emphasize that models are often useful when they highlight unexpected patterns in parasite evolution, revealing instead why intuition yields the wrong answer and what combination of theory and data are needed to advance understanding.

Leishmania infantum immunoelectrophoretic antigen 24 (AG 24), a visceral leishmaniasis associated immunodominant antigen, has been characterized with a monospecific antiserum by combining SDS–PAGE, immunoblotting, metabolic labelling, radio-immunoprecipitation and in vitro poly A+ mRNA translation. AG 24 appeared to correspond to a multi-antigen family of 6–9 members ranging from 20 to 31 kDa and proteinic by nature with no post-translational modifications. A similar banding pattern was recognized by infection sera. AG 24 was not found exposed on the cell surface.

The larvae and puparia of Limnophora exurda are to be found in tufts of wet moss or liverworts in or near small waterfalls.

The larvae are carnivorous and exhibit the characteristic morphological characters of carnivorous anthomyid larvae.

Cattle were inoculated with dilutions of Theileria parva (Muguga) stabilate without concurrent antibiotic treatment. The dilutions were selected to induce severe East Coast fever infections, which in some cases would lead to death. Serum samples were collected and the complement fixation (CF), indirect haemagglutination (IHA) and indirect immunofluorescence (IFA) tests were used to detect T. parva serum antibodies. Sera were fractionated on Sephadex G200 to determine the activities of specific immunoglobulin fractions. Sequential production of IgM antibody followed by 7SIg antibody was demonstrated by the IHA and CF tests. All IFA activity was restricted to the 7SIg fraction. Serologic reactions in the IHA test were detected as early as 4 days after inoculation of the stabilate, CF activity after 8 days and IFA activity after 21 days.

Chagas disease (CD) is caused by the intracellular protozoan parasite Trypanosoma cruzi and affects more than 10 million people in poor areas of Latin America. There is an urgent need for alternative drugs with better safety, broader efficacy, lower costs and shorter time of administration. Thus the biological activity of viniconazole, a chloroaryl-substituted imidazole was investigated using in vitro and in vivo screening models of T. cruzi infection. Ultrastructural findings demonstrated that the most frequent cellular damage was associated with plasma membrane (blebs and shedding events), Golgi (swelling aspects) and the appearance of large numbers of vacuoles suggesting an autophagic process. Our data demonstrated that although this compound is effective against bloodstream and intracellular forms (16 and 24 μ m, respectively) in vitro, it does not present in vivo efficacy. Due to the urgent need for novel agents against T. cruzi, the screening of natural and synthetic products must be further supported with the aim of finding more selective and affordable drugs for CD.

The fox tapeworm Echinococcus multilocularis is typically perpetuated in a cycle with red foxes as definitive hosts and various rodent species as intermediate hosts. In this study, foxes were baited with a highly efficient drug against cestodes (praziquantel) in 5 blocks of 1 km2. Voles, Arvicola terrestris, the most abundant intermediate host species, were trapped in the 5 baited blocks and in 5 non-baited control blocks. Baiting the foxes reduced the prevalence of E. multilocularis in fox faecal samples in the baited blocks, but voles trapped in the two blocks did not differ in their infection rates. However, voles from the baited blocks had significantly smaller spleen masses and were more likely to be infested with mites than those from the control blocks, possibly reflecting different immunological activities. Our study suggests that the environmental contamination with E. multilocularis eggs, and perhaps those of other tapeworms, influences the immune system of the intermediate host species A. terrestris in the wild.

Experiments to study the effect of extrinsic factors on the copulation of normal adult Nippostrongylus brasiliensis are described.Worms copulated when in contact with the rat small intestine, whether it was ligatured, maintained inside or outside the peritoneal cavity, slit open or its lumen obstructed, provided that the blood supply remained intact and the temperature was within a few degrees of normal. Copulation did not occur in the colon, pleural cavity, peritoneal cavity, pregnant or non-pregnant uterus of the live rat or under a variety of in vitro conditions.These results indicate the importance of extrinsic factors for successful copulation since all the worms used in the experiments were intrinsically ready for immediate copulation in their normal environment.Worms observed during in vitro experiments were more vigorous and erratic in their movements than worms in their normal environment. The possible significance of this observation is discussed both in the context of in vitro reproduction experiments and in vitro experiments to study the physiology and biochemistry of the worm.

The effects of a liposomal preparation of lipophilic immunomodulator β-D-GlcNstearoyl-(1-4)-norMurNAc-L-Abu-D-isoGln (N-L18-norAbu-GMDP) were investigated on resistance to Cryptosporidium parvum infection in neonatal kids. The liposomal preparation was administered subcutaneously or intranasally/orally (i.n./p.o.) twice at doses of 100 μg, 200 μg, or 1000 μg per kid pre-infection challenge. The treatment schemes were (i) 72 and 24 h pre-infection challenge, (ii) 24 h pre-infection challenge and 24 h post-infection challenge (oral inoculation with 1×107 oocysts of C. parvum in 5 ml of PBS). Administration of liposomal N-L18-norAbu-GMDP by i.n./p.o. route at the cumulative dose of 2000 μg per kid 72 and 24 h pre-infection challenge, lead to substantially increased clearance of coccidian parasites from various parts of the intestine. On the basis of histological examination, the distribution of cryptosporidia in the intestine and the severity of the infection, treated kids were classified on day 5 as having a strong reduction in infection in comparison to the control group (P<0·05). No cryptosporidia were found on the mucosal surface of treated kids by day 10, while the intestines of the control kids were still infected. All doses and routes of administration were judged effective with respect to suppression of cryptosporidia infections.

The snail Bulinus nasutus 1214 possesses a potent haemagglutinin (end-point titre with human erythrocytes, 2−18) in its cell-free haemolymph which also binds to the miracidia (but not other larvae) of the incompatible parasite Schistosoma margrebowiei. We have purified a protein possessing this haemagglutinating property from the plasma of this snail. The native Mr of this protein was estimated by SDS polyacrylamide gel electrophoresis to be 210 kDa; under denaturing conditions in a 7.5% PAGE gel it ran as a major band of 135 kDa. Proteins of similar Mr were also found in the haemolymph of 16 other Bulinus spp. (the major intermediate hosts of human and veterinary schistosomiasis in Africa) although the plasma of none of these agglutinated human erythrocytes. Nonetheless, Cleveland mapping of the Mr 135 kDa bands from these different Bulinus spp. revealed 4 identical major peptide fragments (30, 28, 19 and 16 kDa) in each, thus demonstrating a similarity in the primary structure of these plasma proteins. Antisera from Balb/C mice immunized with the 135 kDa polypeptide from Bulinus truncatus 1521 cross-reacted in Western blots with the 135 kDa band of other members of the same truncatus/tropicus species complex but not with species from the africanus or forskalii species groups.

The anterior adhesive mechanism was studied for Merizocotyle icopae (Monogenea: Monocotylidae). Adult anterior apertures can open and close. In addition, duct endings terminating within the apertures are everted or retracted depending on the stage of attachment. Adhesive in adults is synthesized from all 3 secretory types (rod-shaped, small and large spheroidal bodies) found within anterior apertures. All exit together and undergo mixing to produce the adhesive matrix, a process that depletes duct contents. A greater number of ducts carrying rod-shaped bodies is depleted than ducts containing spheroidal bodies which changes the ratio of secretory types present on detachment. Detachment involves elongation of duct endings and secretion of additional matrix as the worm pulls away from the substrate. The change in secretory type ratio putatively modifies the properties of the secreted matrix enabling detachment. Only after detachment do ducts refill. During attachment, individual secretory bodies undergo morphological changes. The larval and adult adhesive matrix differs. Anterior adhesive in oncomiracidia does not show fibres with banding whereas banded fibres comprise a large part of adult adhesive. The data suggest that this is the result of adult spheroidal secretions modifying the way in which the adult adhesive matrix forms.

In February 1928 I obtained from a ♂ Scyllium canicula, taken at Plymouth, a ♀ Lernaeopoda scyllicola bearing a younger and better preserved male than any I had hitherto observed. It is the first time in this genus that a male has been found anywhere other than on the female's shoulder, namely on one of the abdominal appendages, and the male was probably alive at the moment of fixation. The heads of the sexes point in opposite directions so that in Fig. 1 the abdominal appendage is “upside down,” and the genital process of the male is near a vulva of the female which is however fertilised since it bears egg-strings. I am now fortunate in having the kind services of Miss E. C. Humphreys to illustrate this male which is not shrunk into its carapace and desire that Figs 1. and 2 shall supersede my figures of 1918. The near second maxilla has been omitted to show the method of attachment of the male.
