Cysticercoids of three species of Tatria have been recovered from the haemocoele of two species of damsel-fly nymphs from a lake in Mid-Wales. This is the first record of Tatria in Britain. The cysticercoids are enclosed each in a transparent capsule of host origin permanently attached to the midgut of the host. The cysticercoids of the three species are indistinguishable superficially. Specific diagnosis depending on the armature of the scolex. Tatria octacantha sp.nov. differs from the other eight species by the presence of eight apical rostellar hooks and 48 small spines on the rostellar surface. One mature adult was found in a little grebe Podiceps ruficollis ruficollis (Pallas) from the same lake and is described briefly. The cysticercoids are described in general terms and the scolex of T. octacantha in detail including the mode of operation of the rostellum and the ultrastructure of the surface layers. A layer of fibres occurs below the superficial longitudinal muscles of the rostellum providing additional support for this mobile organ. A revised key to the nine species of Tatria is included.

1. A description of four new species of gregarine found in Blaberus discoidalis and Blaberus boliviensis is given. The species have been called Protomagalhaensia granulosae, Gregarina cubensis (from B. discoidalis), Protomagalhaensia blaberae and Gregarina princisi (from B. boliviensis).

2. A discussion of the taxonomic status of the species in relation to one another and to Gregarina blattarum and Protomagalhaensia serpentula is given, and further work to elucidate this problem is suggested.

Contrary to results previously published, cortisone did not cause a rapid increase of parasitaemia in mice or hamsters infected with P. berghei, and in some cases it had an inhibitory effect. The existence of a humoral agent restricting the multiplication of the parasites in the peripheral blood was suggested.

The effect of immune inhibition on the pleomorphism of Trypanosoma brucei was studied by counting trypomastigotes and their granules on agar. A statistical analysis is presented. Busulphan 32 mg/kg, given 24 h before inoculation, caused an inhibition of the infection detectable at 72 h, but not thereafter. Immune inhibition caused by busulphan was associated with increased numbers both of agranular forms and forms containing lipid (type II) granules. After 96 h, treatment did not alter the proportion of agranular to granular forms. The remission was delayed 24 h in treated rats, the remaining trypomastigotes being more sluggish, more fragile and containing more numerous granules of increased size. We conclude that the remission is not produced by the immune reaction but by an innate process of dissolution of the ageing trypomastigotes.

1. Pipunculus semifumosus (Kowarz) is an internal parasite of a number of common British Delphacidae. The life cycle and immature stages of the parasite are described.

2. The external and internal effects of parasitism on the delphacid host are described. Reductions in size and genital aberrations are related to the stage at which the host is attacked, and are most marked in female delphacids.

1. Investigations into some of the problems associated with African East Coast fever and its vectors have been undertaken: and although these are, as yet, incomplete we consider them of sufficient interest to warrant their publication at this stage.

2. The present position regarding the distribution of certain tick species in relation to East Coast fever in Kenya is briefly reviewed; and outbreaks of what may be peculiar forms of the disease in sporadic areas are mentioned.

3. It is thought that the passage of Theileria parva through different species of ticks might throw light on some sporadic outbreaks of the disease and on the peculiar character of some of the cases encountered.

4. Transmission by nymphae and adults of Rhipicephalus appendiculatus, by adults of R. evertsi and R. simus is confirmed. Experiments with R. sanguineus, R. pulchellus and Amblyomma variegatum were negative.

5. Exposure of infective nymphae of Rhipicephalus appendiculatus to different degrees and conditions of temperature prove that the ticks remain infective after being subjected to 4–6° C., and 8–10° C. for 1, 2 and 3 weeks. Exposure to different alternating temperatures for similar periods had no effect on the virulence of the parasite. The ticks exposed to warm temperatures (35–38° C,) did not survive to the same extent as those exposed to cold temperatures. Ticks that survived for 1 week transmitted the disease.

6. Animals that ultimately recover from artificial infection may show a mild thermal reaction during the short course of the disease without showing typical clinical symptoms. In such cases diagnosis is frequently difficult, and Koch's bodies may be rare.

7. The association of Hyalomma spp. with many different diseases is briefly dealt with; and it is shown that Hyalomma impressum near planum P. Sch. is capable of transmitting East Coast fever in the nymphal and the adult stages after the larvae and nymphae respectively have fed on an infected animal. The parasite does not pass through the egg. H. impressum near planum is common in most parts of Kenya Colony.

Dark-adapted third-stage Trichonema larvae were orthokinetically stimulated by light. The response soon reached a maximum and then gradually decreased, the larvae then being inactive as long as they were kept in light. The rate of response was independent of intensity, and was a constant for any one age. Inactive larvae were activated by a mechanical stimulus, and were therefore only insensitive to light.

The larvae needed 3 h adaptation in darkness before responding fully to a further light stimulus. Dark-adaptation required continuous darkness; larvae exposed to a flashing light responded as if in continuous light. The presence of a light-sensitive mechanism is postulated.

This study shows that ingestion of Schistocephalus solidus coracidia was related to general activity of Macrocyclops albidus copepods at the time of exposure. The lower the activity of the host, the fewer parasites it ingested. In an earlier study it was shown that large M. albidus copepods were less likely to become infected with S. solidus than small copepods, which could potentially be caused by differential ingestion of parasites. However, the current study did not show any evidence for such an effect arising through differential ingestion. Body size was not related to ingestion of parasites, but was positively correlated to activity. So, even though size did not significantly relate to ingestion of parasites, if anything, through their higher activity large copepods rather than small copepods may have ingested more parasites. This study indicates that differences in resistance to this parasite do not come about through differential ingestion of parasites. Also, an earlier study failed to show differential elimination of the parasite from the haemocoel. This leaves avoidance of penetration through the gut wall as the most plausible candidate causing large copepods to be more resistant to this parasite than small copepods.

The kinetics of serum antibody response against infection with Echinococcus multilocularis eggs was evaluated in AKR mice. The animals were infected by oral inoculation with 300 parasite eggs, and necropsied at 1, 2, 4, 6, 9, 12 and 16 weeks post-infection (p.i.), respectively. The parasite formed the laminated layer at 4 weeks p.i., the brood capsule with a massive proliferation of germinal cells at 9 weeks p.i. and protoscoleces at 16 weeks p.i. Serum antibody responses of the mice to antigen preparations from metacestodes of different stages and protoscoleces were evaluated by ELISA, immunoblotting and immunohistochemistry. In ELISA, the antibody responses began to increase at 4 weeks and became more apparent at 9 weeks p.i. and thereafter. Immunoblots using sera collected at 16 weeks p.i. showed some common bands among the 3 different antigen preparations. In addition to this, the germinal cells and brood capsules of mature metacestodes were stained strongly in an immunohistochemical study. From above, it is suggested that some antigen molecules are expressed in the parasite through these stages and stimulated host antibody responses.

Intestinal diarrheagenic polyparasitic infections are among the major public health concerns in developing countries. Here we examined stool specimens by microscopy, DNA dot blot and polymerase chain reaction (PCR) to evaluate the co-infection of four principal protozoans among amoebic dysentery cases from Northeast Indian population. The multiplex PCR confirmed Entamoeba histolytica (8·1%), Entamoeba dispar (4·8%) and mixed infection of both the parasites (3·4%) in 68 of 356 stool specimens that were positive in microscopy and/or HMe probe based DNA dot blot screening. The prevailing parasite that co-exists with E. histolytica was Giardia duodenalis (34·1%), followed by Enterocytozoon bieneusi (22·0%), Cryptosporidium parvum (14·6%) and Cyclospora cayetanensis (7·3%, P = 0·017). Symptomatic participants (odds ratio (OR) = 4·07; 95% confidence interval (CI) = 1·06, 15·68; P = 0·041), monsoon season (OR = 7·47; 95% CI = 1·40, 39·84; P = 0·046) and participants with family history of parasitic infection (OR = 4·50; 95% CI = 1·16, 17·51; P = 0·030) have significant association with overall co-infection rate. According to molecular consensus, comprehensive microscopy yielded 3·4% (12/356) false-negative and 7·6% (27/356) false-positive outcome, suggesting an improved broad-spectrum PCR-based diagnostic is required to scale down the poor sensitivity and specificity as well as implementation of integrated control strategy.

Serological antibody detection by enzyme-linked immunosorbent assay (ELISA)- and immunoblot-based methods constitutes the best indicator of human Toxocara infection. Nevertheless, the availability of serological tests, particularly western blots (WB), evaluated for sensitivity and specificity is limited. Therefore, an Anti-Toxocara-ELISA immunoglobulin g (IgG) prototype (Proto-ELISA) and an Anti-Toxocara-Westernblot (IgG) prototype (Proto-WB) were evaluated by testing 541 human sera pre-determined for Toxocara infection by an established in-house Anti-Toxocara-ELISA (IH-ELISA). To evaluate sensitivity and specificity of the newly developed ELISA and WB prototypes, results were compared to IH-ELISA and a commercial WB (Com-WB). Compared to the IH-ELISA, a sensitivity of 93.1% (229/246) and a specificity of 94.6% (279/295) of the Proto-ELISA with a Cohen's κ of 0.88 were obtained. The sensitivity of the Proto-WB was 76.7% (240/313) and specificity was 99.6% (227/228) with a Cohen's κ of 0.73 compared to those of Com-WB. A comparison to the IH-ELISA revealed 91.5% (225/246) sensitivity and 94.6% (279/295) specificity of the Proto-WB with a Cohen's κ of 0.86. Cross-reactivity was observed for some samples positive for Ascaris and Trichinella spp. in the Proto-ELISA, Proto-WB and Com-WB. Overall, the evaluated ELISA and WB prototypes showed high sensitivity and specificity, indicating high reliability of these newly developed tests.

Angiostrongylus cantonensis has been found in Florida, USA, from the panhandle in the north to Miami and surrounding areas in the southern parts of the state, in both definitive and intermediate hosts in a limited studies completed in 2015. Additional studies have identified this parasite in a variety of intermediate hosts, both native and non-native gastropod species, with new host species recorded. Many areas in Florida with higher A. cantonensis prevalence were those with a high human population density, which suggests it is a matter of time before human infections occur in Florida. Case reports in the state currently involve non-human primates and include a gibbon and orangutan in Miami. Here, we report the current status of A. cantonensis in the state, as well as the infection in a capuchin monkey and presumptive infection in a red ruffed lemur in Gainesville, Florida.

This study investigated the genetic and antigenic diversity of Theileria parva in cattle from the Eastern and Southern zones of Tanzania. Thirty-nine (62%) positive samples were genotyped using 14 mini- and microsatellite markers with coverage of all four T. parva chromosomes. Wright's F index (F ST = 0 × 094) indicated a high level of panmixis. Linkage equilibrium was observed in the two zones studied, suggesting existence of a panmyctic population. In addition, sequence analysis of CD8+ T-cell target antigen genes Tp1 revealed a single protein sequence in all samples analysed, which is also present in the T. parva Muguga strain, which is a component of the FAO1 vaccine. All Tp2 epitope sequences were identical to those in the T. parva Muguga strain, except for one variant of a Tp2 epitope, which is found in T. parva Kiambu 5 strain, also a component the FAO1 vaccine. Neighbour joining tree of the nucleotide sequences of Tp2 showed clustering according to geographical origin. Our results show low genetic and antigenic diversity of T. parva within the populations analysed. This has very important implications for the development of sustainable control measures for T. parva in Eastern and Southern zones of Tanzania, where East Coast fever is endemic.

The essential oil (EO) of Thymus capitatus, seven fractions (F1–F7) obtained from silica gel chromatography, and several pure EO components were evaluated with respect to in vitro activities against Echinococcus multilocularis metacestodes and germinal layer (GL) cells. Attempts to evaluate physical damage in metacestodes by phosphoglucose isomerase (PGI) assay failed because EO and F1–F7 interfered with the PGI-activity measurements. A metacestode viability assay based on Alamar Blue, as well as transmission electron microscopy, demonstrated that exposure to EO, F2 and F4 impaired metacestode viability. F2 and F4 exhibited higher toxicity against metacestodes than against mammalian cells, whereas EO was as toxic to mammalian cells as to the parasite. However, none of these fractions exhibited notable activity against isolated E. multilocularis GL cells. Analysis by gas chromatography-mass spectrometry showed that carvacrol was the major component of the EO (82.4%), as well as of the fractions F3 (94.4%), F4 (98.1%) and F5 (90.7%). Other major components of EO were β-caryophyllene, limonene, thymol and eugenol. However, exposure of metacestodes to these components was ineffective. Thus, fractions F2 and F4 of T. capitatus EO contain potent anti-echinococcal compounds, but the activities of these two fractions are most likely based on synergistic effects between several major and minor constituents.

Between 2008 and 2011, the head of 150 Euthynnus alletteratus (Osteichthyes: Scombridae) caught inshore off the southeastern Iberian coast (western Mediterranean Sea) were examined for parasites. Two monogeneans, four didymozoid trematodes and four copepods were found. Parasite abundance showed a positive relationship with the annual sea surface temperature, except for Pseudocycnus appendiculatus, but negative with the sea depth (Capsala manteri, Neonematobothrium cf. kawakawa and Caligus bonito). Prevalences and mean abundances differed significantly among sampling areas, except for C. manteri, Oesophagocystis sp. 2 and Ceratocolax euthynni, and sampling years (Melanocystis cf. kawakawa, N.cf. kawakawa, P. appendiculatus and Unicolax collateralis). Results indicate that the parasite abundances of E. alletteratus in the western Mediterranean Sea depend mainly on regional environmental variables, which can show interannual variations. The presence of pelagic parasites, i.e. didymozoids and P. appendiculatus, could indicate that E. alletteratus migrates between inshore and offshore pelagic domains. The different parasite faunas reported in E. alletteratus populations from the western Atlantic Ocean and the Mediterranean Sea appear to point out the geographical host isolation. These results suggest that E. alletteratus inhabiting the western Mediterranean Sea performs inshore-offshore small-scale migrations, and not transoceanic migrations between the western Atlantic Ocean and Mediterranean Sea.
