1. The life history of Lecithodendrium chilostomum has been established; C. Lecithodendrii chilostomi penetrates a second intermediate host, the larva of Phryganea grandis, and unlike other stylet cercariae does not encyst, but feeds and grows in the host tissue as a mobile metacercaria. During pupation of the host in the following year these mobile forms migrate from the abdominal segments of the larva to the thorax, where they then encyst in the thoracic muscles in which they are also found in the imago. The largest metacercariae and the excysted worms are typical early adult Lecithodendrium chilostomum. No case of progenesis in the metacercaria was found.

2. It is estimated that the maximum swarming of the cercariae probably occurs during July, when first penetration of the intermediate host takes place. Subsequent diminished swarming and penetration proceed until November. The life of the larval trematode as a mobile metacercaria is approximately 8 months; the imagines of Phryganea grandis emerge during May and June, the infective period for the final host.

3. The metacercariae are not distributed throughout the body cavity of their larval host, but the majority are confined to the three posterior segments.

4. Limnophilus rhombicus may also serve as a second intermediate host, but the infection is very light and the metacercariae do not encyst during pupation of this host.

5. The excretory system has been determined in all stages of Lecithodendrium chilostomum; it is of the 2 (6 × 2) type. The occurrence of this type of system in other groups of cercariae is reviewed and since it is found in several widely separated families, it is suggested that its presence does not necessarily imply relationship, but is due to convergence.

6. The life histories of the following bat trematodes are indicated: Lecitho-dendrium lagena, Plagiorchis vespertilionis and Crepidostomum moeticus.

7. The life history of Dicrocoelium dendriticum is discussed.

There are three species of true parasites in the genus Aphelenchus, viz.: A. fragariae R.B., 1891, A. olesistus R.B., 1893, and A. phyllophagus n.sp. A. ormerodis R.B., 1891, is a doubtful species, possibly a young form of A. fragariae (Marcinowski, 1908).

A. fragariae causes the disease strawberry bunch; A. olesistus and phyllophagus cause leaf disease in flowering plants and ferns. The diseases are widely distributed and of considerable economic importance.

From the study of the anatomy of the three species certain points of general importance emerge: (1) The salivary glands of Aphelenchus represent the oesophageal glands of other nematode genera, which have separated from the body of the oesophagus. This separation causes the reduction in size of the posterior portion of the oesophagus, and its lack of clear demarcation from the intestine. (2) The excretory organ of Aphelenchus, and of some other nematode genera, originates from the left lateral line. (3) The vaginae are ectodermal organs distinct in origin from the gonads proper.

The life cycle of the parasitic Aphelenchi is divided into three definite stages: (A) That of residence in the definitive habitat, which for A. fragariae is in the stem and leaves of the strawberry plant, for A. olesistus and phyllophagus in the mesophyll spaces of the leaves of many plants. Nutrition and reproduction are most actively carried on in this situation, several or many generations succeeding each other, and here the pathological effects are manifest. (B) The resting stage in the soil. The worms reach the soil in fallen leaves, and the adults survive in a partially dried, dormant condition on the surface of the soil for prolonged periods. (C) The stage of immigration into the host plant. When revived by moisture the Aphelenchi may be attracted by a suitable plant, and wanders on to it; they may live as ectoparasites in the leaf axils, breeding in this situation, the larvae migrating to the definitive habitat; or they may enter the definitive habitat direct. They do not traverse the tissues of the stem during immigration. A. olesistus and phyllophagus enter the mesophyll spaces through the stomata.

It should be noted that this life cycle is that of the race, not of the individual. It comprises many generations, and in this respect resembles that of Tylenchus dipsaci and differs from that of T. tritici, which includes one generation only.

In the life of the individual Aphelenchus, embryonic development occupies not more than five days, embryonic and larval development not more than ten, and a complete generation not more than fourteen days.

High atmospheric humidity assists the migration of the worms, previous disease or injury of the host plant may do so. Some races of various species of plants are immune.

Treatment of the affected plants by chemical methods is of limited applicability. Treatment by immersion in water at 50–52° C. for five minutes is recommended by Marcinowski (1908). Prophylaxis should be based on burning of infected plants and leaves, and on the avoidance of infected surface soil in the filling of pots and boxes.

An allele-specific, one tube PCR for the sensitive and reliable detection of point mutations in Plasmodium falciparum DNA is described. Design of specific internal primers and optimization of the PCR is simple, and the procedure is robust and sensitive. Single nucleotide polymorphisms at codons 184, 1034, 1042 and 1246 of the P. falciparum multidrug resistance gene Pfmdr1, were examined in 6 laboratory isolates, to validate the technique.

Swimmer's itch (SI) is a painful rash caused by skin penetration by free-swimming infectious cercariae of avian schistosomes, snail-borne helminth parasites related to the causative agents of human schistosomiasis. The goal of this study was to determine if commonly collected environmental data could be used to predict daily fluctuations in SI incidence at an inland beach in northwestern Michigan. Lifeguards collected daily data over four summers, including the number of self-reported SI cases, total swimmers, water temperature, wind speed and wind direction. Mixed-effects binomial regression revealed that wind direction, wind speed and time of day were the best predictors of daily SI risk. Swimmers entering the water in the morning or on days with direct onshore wind perpendicular to the shoreline had the greatest SI risk. However, there was a negative effect of wind speed after accounting for direction, where SI risk was greatest on days with a gentle breeze originating directly offshore. These results suggest that at this beach, direct onshore winds generate a surface-water current that causes SI cercariae to aggregate in the shallow waters used by swimmers. Data are needed from additional sites to confirm whether the onshore wind is a generally important driver of SI incidence.

The structure of the various stages of Eimeria tenella grown in the chorioallantoic membranes of the chick embryo has been shown to be similar to the stages in the caecum of the bird.The effects of the anticoccidial drug, Robenidine, on the fine structure of the various stages of E. tenella, grown in the chick embryo and in the caecum of the bird, have been described. The first noticeable change occurs in the endoplasmic reticulum, including the nuclear envelope, and in the Golgi complexes of all stages. The nucleus itself, and the mitochondria, do not appear to be affected by the drug until much later, and this effect is probably due to the degeneration of the cell.It is suggested that the drug may have a direct effect upon protein metabolism of the parasite, resulting in swelling of the granular endoplasmic reticulum and Golgi complexes, or that it may have a primary effect upon some other organelle or metabolic process, and that the swelling of the endoplasmic reticulum and Golgi complexes is a secondary or tertiary phenomenon.

The population biology of the schistosome–vector snail Bulinus truncatus was studied in an irrigation system near Marrakech, Morocco using both genetic and demographic approaches. The population genetic survey was conducted in 4 sites, 2 sites being sampled on 2 separate occasions. Individuals were genotyped at 6 microsatellite loci. No variability was found at 4 loci, and the 2 other loci had less than 4 alleles. The differentiation, both spatial and temporal, among populations was extremely weak. The demographic survey was conducted using 2 capture-mark-recapture analyses in 2 separate sites, the first in 1999 and the second in 2000. The second analysis permitted the estimation of parameters based on recent methodological developments (multisite models). Although these studies provided information on several traits, we report here on dispersal only. Both analyses showed that individual dispersal is of the order of a few hundreds of metres per reproductive life, that is the scale of the whole irrigation area. Both the genetic and demographic studies indicated that this area harbours a single – or no more than a few – populations of B. truncatus. This has implications for our understanding of the coevolutionary process between snails and flukes.

The effect of continuing ingestion of eggs of A. galli on growth rates of chickens placed on different nutritional levels was studied under repeated doses of 10, 100 and 1000 infective eggs/day. With increasing amounts of protein in feed, there occurred significant weight differences between infected groups, the highest protein levels showing the longest duration in weight depression due to infection. On the other hand, assuming adequate nutrition of host, highest effects on growth rates were seen in chickens receiving daily doses of 1000 eggs daily and the lowest in chickens receiving 10 eggs daily.

The effect of repeated dose on growth rate is similar to the effect of single infections reported by previous workers except that the duration of the growth depression was much longer in the former due to pathogenic effects of inhibited larvae.

Golden hamsters infected with Spirometra erinacei plerocercoids develop a hypertriglyceridaemia characterized by an increase in very low-density lipoprotein. Acyl-CoA synthetase activity, the rate of fatty acid synthesis in the liver and triglyceride production studies showed that the activation and synthesis of fatty acid and production of triglyceride in plerocercoidinfected hamsters were not significantly different from the controls. Lipase activity in post-heparin plasma was found to be suppressed in plerocercoid-infected golden hamsters. The presence of plerocercoids also resulted in a significant reduction in serum thyroxine, but the hypertriglyceridaemia associated with plerocercoid infection was not reversed by injecting the infected hamsters with 2 μg/ day doses of L-thyroxine. The levels of serum immunoreactive insulin in plerocercoid-infected hamsters were not significantly different from the controls. We conclude that the hypertriglyceridaemia, associated with plerocercoid infection in hamsters, results predominantly from a suppression of triglyceride degradation, and that the suppression of lipase activity is probably not the result of hypothyroidism or the lack of insulin, but the result of secretion of growth hormone-like substances.

Haematophagous insect vectors of arthropod-borne viruses (arboviruses) feed repeatedly. Consequently, they can transmit arboviruses to more than one host during the same developmental stage (intra-stadial transmission). By contrast, ixodid ticks generally feed only once at each parasitic stage (larva, nymph, and adult) and hence they have only one opportunity for tick-borne virus transmission per stadium (inter-stadial transmission). Under natural conditions, tick-infested hosts may die (from disease or other causes) before the ticks have completed their long period of engorgement. A laboratory model was used to investigate the consequences of premature host death on tick-borne virus transmission. We report intra-stadial transmission of Thogoto virus by the nymphal, male, and female ticks of Rhipicephalus appendiculatus. Tick-borne Thogoto virus infection caused viraemia and death of hamsters before the nymphal and adult ticks feeding on them had completed engorgement. The resulting partially fed ticks were allowed to continue engorgement on new, uninfected hosts (interrupted feeding). During feeding on the subsequent hosts, they transmitted the virus intra-stadially to susceptible hosts (hamsters), and to uninfected co-feeding ticks on non-susceptible hosts (guinea-pigs). Intra-stadial transmission, mediated by interrupted feeding, may help explain outbreaks of rapid and fatal tick-borne viral diseases, and the epidemiology as well as evolution of virulence, in a susceptible host population. Additionally, intra-stadial transmission provides an increased risk of tick-borne pathogen transmission to humans and domestic animals during slaughter and game hunting.

In host–parasite relationships, parasite prevalence and abundance can vary over time, potentially impacting how hosts are affected by infection. Here, the pathology, growth, condition and diet of a juvenile Cyprinus carpio cohort infected with the non-native cestode Bothriocephalus acheilognathi was measured in October 2012 (end of their first summer of life), April 2013 (end of first winter) and October 2013 (end of second summer). Pathology revealed consistent impacts, including severe compression and architectural modification of the intestine. At the end of the first summer, there was no difference in lengths and condition of the infected and uninfected fish. However, at the end of the winter period, the condition of infected fish was significantly reduced and by the end of their second summer, the infected fish were significantly smaller and remained in significantly reduced condition. Their diets were significantly different over time; infected fish consumed significantly higher proportions of food items <53 µm than uninfected individuals, a likely consequence of impaired functional traits due to infection. Thus, the sub-lethal impacts of this parasite, namely changes in histopathology, growth and trophic niche were dependent on time and/or age of the fish.

The major pathways for cysteine catabolism in Hymenolepis diminuta have been investigated. The parasite has an active cystathionine-β-synthase and, as in other tissues, this enzyme has a wide substrate specificity. However, the enzyme from H. diminuta differs significantly from the mammalian enzyme in showing a high serine sulphydrase activity and a high serine lyase activity. There was only low γ-cystathionase activity in H. diminuta and again the enzyme showed a range of substrate specificities. Cysteine aminotransferase activity was readily demonstrated in the tapeworm, but there was no evidence for 3-mercaptopyruvate sulphotransferase activity. An oxidative pathway for cysteine catabolism in H. diminuta was shown by the presence of cysteine dioxygenase and cysteine sulphinate transaminase. The properties of the helminth cysteine dioxygenase were very similar to those of rat liver. H. diminuta was able to reduce cystine to cysteine via a glutathione-cysteine transhydrogenase system.

Isozyme analysis (12 enzymes: 14 loci) was conducted on 99 isolates of Trypanosoma cruzi: 77 from Guatemala, 5 from Mexico and 17 from South American countries. Analyses of 4 population-genetic indices were undertaken to assess the possibility of genetic exchange occurring among Guatemalan isolates. The results provide evidence for a degree of genetic exchange occurring among isolates from this relatively small geographical area. Previous studies of population genetics on T. cruzi might have failed to detect this phenomenon because they tended to use isolates originating far from one another, rendering gene exchange unlikely for geographical reasons. Phylogenetic data, presented here, show considerable differences in genetic structure between Central and South American isolates, suggesting that different biological and clinical properties might be expected. For example, there are differences in clinical syndromes between Central and South America, a situation discussed further here.

A novel acidic extracellular single-stranded endonuclease was demonstrated for the first time in the excretory–secretory (E–S) products of 2 species of Trichinella. Unlike the double-stranded endonuclease reported earlier, the single-stranded molecule is divalent cation independent and is detected in both T. spiralis and T. pseudospiralis E–S products. It hydrolysed single-stranded DNA and RNA at comparable rates. The single-stranded endonuclease was sensitive to inhibition by Zn2+ and to high concentrations of NaCl. Zymographic analysis indicated that it was encoded by at least 3 peptides of Mr ∼ 50–60 kDa. The rate of hydrolysis of single-stranded targets by the E–S products was substantially higher than that of the double-stranded molecule. Due to the differences in peptide profile, divalent cation dependence, and species-specific expression, the single and double-stranded endonucleases are likely to be encoded by different proteins and may have different functions.

Trypanosomes are blood-borne parasites that can cause severe disease in both humans and animals, yet little is known of the pathogenicity and life-cycles of trypanosomes in native Australian mammals. Trypanosoma copemani is known to be infective to a variety of Australian marsupials and has recently been shown to be potentially zoonotic as it is resistant to normal human serum. In the present study, in vivo and in vitro examination of blood and cultures from Australian marsupials was conducted using light microscopy, immunofluorescence, scanning electron microscopy and fluorescence in situ hybridization. Promastigote, sphaeromastigote and amastigote life-cycle stages were detected in vivo and in vitro. Novel trypanosome-like stages were also detected both in vivo and in vitro representing an oval stage, an extremely thin stage, an adherent stage and a tiny round stage. The tiny round and adherent stages appeared to adhere to erythrocytes causing potential haematological damage with clinical effects similar to haemolytic anaemia. The present study shows for the first time that trypomastigotes are not the only life-cycle stages circulating within the blood stream of trypanosome infected Australian native marsupials and provides insights into possible pathogenic mechanisms of this potentially zoonotic trypanosome species.

Temperature is expected to modulate the responses of organisms to stress. Here, we aimed to assess the influence of temperature on the interaction between parasitism and fungicide contamination. Specifically, using the cladoceran Daphnia as a model system, we explored the isolated and interactive effects of parasite challenge (yeast Metschnikowia bicuspidata) and exposure to fungicides (copper sulphate and tebuconazole) at two temperatures (17 and 20 °C), in a fully factorial design. Confirming a previous study, M. bicuspidata infection and copper exposure caused independent effects on Daphnia life history, whereas infection was permanently suppressed with tebuconazole exposure. Here, we show that higher temperature generally increased the virulence of the parasite, with the hosts developing signs of infection earlier, reproducing less and dying at an earlier age. These effects were consistent across copper concentrations, whereas the joint effects of temperature (which enhanced the difference between non-infected and infected hosts) and the anti-parasitic action of tebuconazole resulted in a more pronounced parasite × tebuconazole interaction at the higher temperature. Thus, besides independently influencing parasite and contaminant effects, the temperature can act as a modulator of interactions between pollution and disease.

Echinococcus multilocularis is the causative agent of alveolar echinococcosis (AE), one of the most threatening zoonoses in Eurasia. Human AE is widespread in the Erzurum region of Turkey, but the situation of the disease in intermediate and definitive hosts is unknown. A Eurasian lynx (Lynx lynx) was killed in a traffic accident in the north of Erzurum, and was taken to our laboratory. Sedimentation and counting technique (SCT), DNA isolation and polymerase chain reaction (PCR) analysis were performed. The SCT results showed that the lynx was infected with E. multilocularis with a medium (745 worms) worm burden. The DNA of adult worms obtained from the lynx was analyzed with a species-specific PCR, and the worms were confirmed to be E. multilocularis by 12S rRNA gene sequence analysis. This is the first report of E. multilocularis from Eurasian lynx in Turkey.

Interest in the long-term low temperature preservation of malaria parasites was initiated by Coggeshall's (1939) demonstration that the intra-erythrocytic stages could survive storage at −76°C for 70 days. Numerous studies have subsequently been published in which a variety of techniques have been used. In addition, several reviews have included sections on the cryopreservation of malaria, the most recent being by James (1980), Leef, Hollingdale, & Beaudoin (1981), Nguyen-Dinh, Chemangey-Masaba, Spencer, Campbell, Chin & Collins (1981) and a WHO memorandum (1981). Most of these reports and reviews have dealt with the problems of cryopreserving malaria parasites from a parasitological viewpoint. In an attempt to provide a cryobiologically orientated review of value both to cryobiologists and practicing parasitologists the published technical reports have been summarized in chronological order in Tables 1 and 2, and the important cryobiological parameters are discussed.

During the eighteen months spent with the League of Nations International Commission on Human Trypanosomiasis, I had a unique opportunity for studying the transmissibility of the human trypanosomes by Glossina palpalis.

Malaria is endemic over a wide area of the earth's surface. Although popularly associated with tropical or subtropical countries, it occurs as far north as the south of Sweden and Lake Ladoga in Russia and as far south as Bechuanaland, Swaziland and Natal in South Africa and the Argentine in South America. Though most frequent in low-lying districts it may be found at altitudes as great as 6000–9000 ft.
