The course of infection with a Malaysian dog strain of Ancylostoma ceylanicum was investigated in 15 inbred strains of mice, in outbred and inbred mice immunosuppressed with prednisolone, and in immuno-deficient hypothymic mice. Oral, percutaneous and subcutaneous routes of infection, in both sexes of mice, were assessed. In only one instance was a single small adult male worm found. Following oral infection, larvae migrated from the stomach to the large bowel and then a proportion of worms penetrated the perianal skin. This was followed by the appearance of larvae in the lungs. Living 3rd-stage larvae were seen in the anterior small intestine, perianal skin and lungs for the 6 weeks of the study, with peak recoveries being at 12 h, 8 days and 3 weeks, respectively. It is clear that systemic migration of larvae occurs after oral infection, and it is possible that recirculation may occur. Only a small percentage of larvae penetrated the abdominal skin after being administered percutaneously. In subcutaneous infections, a small proportion of larvae moved rapidly from the site of injection and were recovered from the lungs 2 h after infection. Most larvae, however, migrated from the injection site over the ensuing few days. Living 3rd-stage larvae were seen in the lungs and in the small intestine for the 4 weeks of observation. The strain of A. ceylanicum employed does not complete its development in mice. Nevertheless, this model offers significant potential for studying the immune responses, as well as investigating the means by which these parasites evade host defences.

Mass-scale production of Plasmodium vivax sporozoites in Anopheles stephensi was achieved using the chimpanzee (Pan troglodytes) as a source of infective blood. Membrane feeding was as successful as feeding mosquitoes directly on the animal so long as the time between drawing the blood and feeding was restricted to 45 min. Longer delays such as 2–3 h resulted in loss of infectivity in terms of oocyst production. The selected strain of A. stephensi was highly susceptible to P. vivax (Chesson strain). A strain of A. stephensi relatively refractory to P. falciparum showed no cross-refractoriness to P. vivax. Mixed infections of P. falciparum and P. vivax did not interfere with each other in their development in A. stephensi. A second normal blood meal to mosquitoes infected with P. vivax increased the yield of salivary gland sporozoites.

This paper deals with the species of the commonest genus of the American Streblidae. The descriptions of the species were made from specimens mounted on slides, in Canada balsam. Here I wish to point out again that spirit preserved specimens are unsuitable for the study of these Diptera. These specimens do not show clearly the details of the structure of some sclerotized parts, and especially the arrangement of the small setae on the thorax, which in some cases, are the only reliable characters for the separation of the species. The study of unmounted specimens of these Diptera may easily lead to incorrect identification, or to the increase of undesirable synonyms.

Horizontal gene transfer (HGT) has played an important role in the evolution of nematodes. Among candidate genes, cyanase, which is typically found only in plants, bacteria and fungi, is present in more than 35 members of the Phylum Nematoda, but absent from free-living and clade V organisms. Phylogenetic analyses showed that the cyanases of clade I organisms Trichinella spp., Trichuris spp. and Soboliphyme baturini (Subclass: Dorylaimia) represent a well-supported monophyletic clade with plant cyanases. In contrast, all cyanases found within the Subclass Chromadoria which encompasses filarioids, ascaridoids and strongyloids are homologous to those of bacteria. Western blots exhibited typical multimeric forms of the native molecule in protein extracts of Trichinella spiralis muscle larvae, where immunohistochemical staining localized the protein to the worm hypodermis and underlying muscle. Recombinant Trichinella cyanase was bioactive where gene transcription profiles support functional activity in vivo. Results suggest that: (1) independent HGT in parasitic nematodes originated from different Kingdoms; (2) cyanase acquired an active role in the biology of extant Trichinella; (3) acquisition occurred more than 400 million years ago (MYA), prior to the divergence of the Trichinellida and Dioctophymatida, and (4) early, free-living ancestors of the genus Trichinella had an association with terrestrial plants.

A stereological study was undertaken on the length, diameter and numbers of microtriches and on the structural and functional amplification factors of the adult cestodes, Hymenolepis diminuta, Hymenolepis nana, Eubothrium crassium, Proteocephalus pollanicoli, the plerocercoid of Schistocephalus solidus and the cysticercus of Taenia crassiceps. The study showed wide variations in these stereological parameters between species, stages and regionally in the same species. A comparison of the present results with previous studies has shown a large measure of agreement but some differences are evident and possible explanations of these differences are proposed.

Monogenean communities of fish have generally been considered non-interactive as negative interspecific interactions have rarely been reported. Most of the earlier studies on monogenean communities, however, have been conducted not only in systems with relatively low parasite abundances but, more importantly, at study scales where microhabitat-level interactions between the parasites are easily overlooked. We examined the communities of 3 abundant Dactylogyrus (Monogenea) species on the gills of crucian carp (Carassius carassius) by analysing the interactions at the scale of individual gill filaments, where interactions between the species, if any, should most likely take place. Contrary to our expectations, we did not find evidence for competitive exclusion between the species, which suggests that monogenean communities are non-interactive even in high parasite abundances. At the species level, individual parasites were highly aggregated within the filaments, essentially showing a strong tendency to occur at either end of a filament. This, together with the result of differences in the distribution of juvenile parasites within the filaments compared to adults, suggests that these parasites are able to actively seek out their conspecifics in small-scale microhabitats during maturation, which again could enhance their mate-finding.

Oogonia of Meloidogyne javanica are radially arranged around a central rachis to which they are attached by cytoplasmic bridges. As the oocytes mature the rachis disappears and the oocytes pass through the oviduct in tandem. The oviduct-spermatotheca valve is constructed of two rows of tightly packed cells of which there are four per row. The nuclei of these cells are large and contain balloon-shaped cytoplasmic invaginations. The spermatotheca is characterized by microtubules which extend to its lumen and by invaginations of plasmalemma. Cells of the distal uterine region contain large intracytoplasmic spaces bordered by endoplasmic reticulum whereas proximal uterine cells have dense cytoplasm and large areas of compact endoplasmic reticulum. Egg-shell formation begins in the spermatotheca with the modification of the oolemma to form the vitelline layer. The chitinous layer begins in the distal portion of the uterus and appears to originate from the egg. Proline-containing protein is incorporated into the chitinous and lipid layers as the egg passes through the mid-region of the uterus and formation of the lipid layer in this region completes egg development.

Although early reports on HIV and malaria in co-infected subjects indicated little apparent interaction between the two infections, more recent investigations have found evidence for HIV increasing the risk from malaria. Conversely, increased viral load in susceptible cells occurs in malaria-infected people. However, the overall pattern of results is still somewhat confusing and contradictory. While morbidity from malaria may be greater in HIV-positive patients and in several reports the mortality risk is also higher, major increases in blood-stage parasitaemias that one might expect are not generally observed. The results of surveys are summarized and discussed in the context of what is known of malaria and HIV immunology in the light of recent data from humans as well as animal models.

The cellular reactions to the migration of challenge schistosomula of Schistosoma mansoni through the cheek pouch skin in Chinese hamsters immunized 20 weeks previously via the abdomen has been investigated at the ultrastructural level. The composition of the leucocytic infiltrate into the dermis and epidermis changed with time after infection. At 24 and 48 h the infiltrate was composed mainly of neutrophils and mononuclear cells. At 72 h fewer neutrophils but more eosinophils were present. The interaction of the leucocytes with the challenge schistosomula is described. Degranulation of eosinophils at the cell-parasite interface has been observed. Despite the fact that approximately 30% of 48 h schistosomula and 50% of 72 h schistosomula examined had adherent leucocytes only a few of the parasites exhibited signs of damage, consisting of tegumental vacuolation and changes in the electron opacity of the tegumental cytoplasm. There was no strong evidence of increased attrition of challenge schistosomula in the cheek pouch skin relative to the primary infection, although some dead challenge parasites were observed. It is concluded that a substantial proportion of the attrition of challenge infections in the Chinese hamster occurs at a post-dermal location.

The descriptions by Bassett-Smith (1898–9), Thompson and Scott (1903) and Wilson (1906) constitute practically all the work which has been done on the interesting group of Copepoda parasitic on fishes along the Ceylonese coast. To this short recorded list the present author has added five new species (1932–4). In a short series of papers, of which this is the first, it is proposed to describe a number of species, some of which are new forms, collected since those dates.

In 1945 Sproston stated that an alternation of sexual phases takes place in Kuhnia scombri, a monogenean of Scomber scombrus, but an examination of egg-laying, the state of the sexual organs and the presence of sperms in this parasite at various times of the year have shown that there is no evidence for the existence of such a phenomenon. Sproston's observations are examined with particular reference to problems in measuring the length of the parasite and to the unreliability of using the presence or absence of eggs in the uterus as an index of femaleness.

Adults of the porocephalid pentastomid Porocephalus crotali infect the lung of rattlesnake definitive hosts and larvae develop in rat intermediate hosts. In the latter, nymphs encyst within a variety of tissue sites (commonly abdominal fat bodies and lungs) and each becomes the focus of an eosinophilic granuloma. From an early stage in infections, granulomas become increasingly infiltrated by mast cells which, using conventional histology and paired immunofluorescence against mast cell proteases, appear to be exclusively of the mucosal phenotype. Mucosal mast cells are concentrated along the dorsal region of the parasite and in a plug of tissue containing degenerating cuticles within independent granulomas, which is located between its head and tail. ELISAs against the rat mast cell proteases I and II (RMCP I and II), extracted from abdominal fat, lung, spleen, liver and kidney granulomas at various intervals post-infection, reveal a substantially elevated concentration of RMCP II in all lesions. In fat, concentrations increase up to about 100 days post-infection, at which time moulting ceases and inflammatory responses subside. RMCP II was scarcely detectable in matched control tissues. Unlike infections with certain nematode parasites, where enteric mucosal mast cells secrete RMCP II systemically, concentrations of RMCP II in the serum of infected rats were significantly reduced when compared with age-matched uninfected controls. These results confirm that P. crotali can selectively recruit mucosal mast cells to a variety of tissue sites, most of which are non-mucosal. Possible mechanisms are discussed.

Leishmaniasis are diseases caused by parasites of the genus Leishmania and transmitted to humans by the bite of infected insects of the subfamily Phlebotominae. Current drug therapy shows high toxicity and severe adverse effects. Recently, two oligopeptidases (OPBs) were identified in Leishmania amazonensis, namely oligopeptidase B (OPB) and oligopeptidase B2 (OPB2). These OPBs could be ideal targets, since both enzymes are expressed in all parasite lifecycle and were not identified in human. This work aimed to identify possible dual inhibitors of OPB and OPB2 from L. amazonensis. The three-dimensional structures of both enzymes were built by comparative modelling and used to perform a virtual screening of ZINC database by DOCK Blaster server. It is the first time that OPB models from L. amazonensis are used to virtual screening approach. Four hundred compounds were identified as possible inhibitors to each enzyme. The top scored compounds were submitted to refinement by AutoDock program. The best results suggest that compounds interact with important residues, as Tyr490, Glu612 and Arg655 (OPB numbers). The identified compounds showed better results than antipain and drugs currently used against leishmaniasis when ADMET in silico were performed. These compounds could be explored in order to find dual inhibitors of OPB and OPB2 from L. amazonensis.

LACK (Leishmania analogue of the receptor kinase C) is a conserved protein in the protozoan of the genus Leishmania, which is associated with the immunopathogenesis and susceptibility of BALB/c mice to Leishmania major infection. We previously demonstrated that intranasal immunization with a plasmid DNA encoding the p36/LACK leishmanial antigen (pCI-neo-LACK) followed by challenge 7 days after a booster dose effectively protects BALB/c mice against both cutaneous and visceral leishmaniasis. In the present study, the correlation between systemic mRNA expression after nasal DNA uptake, and the duration of protective immunity was addressed. LACK mRNA transcripts were detected in the spleen, brain, cervical lymph nodes and popliteal lymph nodes as early as 7 days, lasting 3 months after vaccination with pCI-neo-LACK. The kinetics of transcript expression correlated with enhanced cutaneous hypersensitivity against parasite antigens. Leishmania chagasi infection at 7 days or 3 months, but not 6 months after vaccination resulted in significantly lower parasite loads as compared with non-vaccinated controls. Protection also correlated with enhanced spleen cell responsiveness to parasite antigens leading to increased IFN- γ and IL-4 and decreased IL-10 production. Together, these data demonstrate that the protection conferred by the intranasal DNA vaccine lasts at least 3 months and is associated with expression of vaccine mRNA in peripheral organs.

The aim of this study was to elucidate the regulatory responses and the long-term effect of praziquantel treatment in chronically Schistosoma bovis-infected West African Dwarf goats. Forty-two goats were used and the design comprised a primary infection followed by treatment at week 13, challenge infection at week 36 and termination at week 52. Dependent variables included clinico-pathological data, worm numbers, faecal and tissue egg counts, and gross pathology of the liver. The results showed that primary infections remained suppressed for up to 52 weeks and, although challenge infections imposed on 36-week-old primary infections established fully, the impairment of their egg production capacity provided protection against clinico-pathological consequences measured by body weight and haemoglobin levels. The study also confirmed a high efficacy (97·7%) of praziquantel for treatment of S. bovis infection in goats and showed that anthelminthic removal of primary infections does not interfere with the ability of the goat to elicit a marked resistance to a subsequent challenge infection. Although treated goats had more fibrous scarring of livers than untreated goats, no negative effects of liver lesions were reflected in weight gains of treated goats. This study provides strong evidence for the beneficial effects of anthelminthic treatment of young domestic stock as an element of treatment and preventive programmes.

In this study, we have produced a monoclonal antibody (MoAb) against 16 kDa antigen of Paramphistomum gracile (16 kDaAgPg), and developed an accurate sandwich enzyme-linked immunosorbent assay (sandwich ELISA) for the detection of circulating 16 kDaAg in the serum and fecal samples from cattle naturally infected with P. gracile. MoAb 1D10 was immobilized on a microtitre plate, and the antigen in the samples was captured and detected with biotinylated rabbit anti-16 kDaAgPg antibody. The lower detection limit of sandwich ELISA was 3·5 pg mL−1, and no cross-reaction with other parasite antigens was evaluated. The reliability of the assay was examined using the serum and fecal samples from cattle naturally infected with P. gracile, Fasciola gigantica, Moniezia benedeni, Trichuris sp., Strongyloides sp., strongylids and non-infected animals. The sandwich ELISA showed the sensitivity, specificity and accuracy at 98·33, 100 and 99·55% (serum samples), and 96·67, 100 and 99·09% (fecal samples). Therefore, this detection method is a rapid and excellent potential assay for the accurate diagnosis of paramphistomosis.

In order to assess the prevalence of the cattle filaria Onchocerca ochengi in onchocerciasis vectors (Simulium damnosum s.l.) in North Cameroon, we searched for a means to morphologically identify its developing larvae, which closely resemble those of O. volvulus. To this end microfilariae of the 2 Onchocerca species were isolated from slaughter cattle in Ngaoundéré and injected into neonate Simulium species. Whereas the early developmental stages (sausage stage, L2 and pre-infective larva) were indistinguishable, the infective larvae (L3) of O. ochengi were longer (median: 740 μm), more slender (diameter = 19·3 μm = 2·6% of body length) and had a relatively shorter tail (4·9% of body length) than those of O. volvulus (680 μm, 20·5 μm, 3·0% and 5·8% respectively). The tail of O. ochengi L3 was thick and rounded, whereas it was slightly tapering in O. volvulus L3. O. ochengi L3 produced by feeding flies on infected cattle in a different area in North Cameroon (Sora Mboum) showed the same features as intrathoracically produced O. ochengi L3 from Ngaoundéré, but were even longer (785 μm). On the basis of the differences in length, relative diameter, length of the tail and shape of the tail, a simple key for the separation of O. volvulus and O. ochengi L3 was elaborated, and 248 L3 found in wild S. damnosum s.l. were separated into ‘O. ochengi’ (160 L3) and ‘O. volvulus’ (88 L3) following this key. Sequential dot blot hybridization of each of the 248 larvae with a DNA probe which reacts with O. ochengi and O. volvulus but not with other Onchocerca species (pOo5/1) and with an O. volvulus-specific DNA probe (pOv12) revealed that the morphological identification had been correct in 86–91% of the cases. Only a small proportion (6–9%) of the dot blots did not react with either probe. Since this proportion was equal in experiments using experimentally produced L3 and in experiments using wild L3, the non-hybridization was certainly due to a loss of L3 during washing of the filters and not due to the presence of other unknown L3 species resembling O. volvulus and O. ochengi. Our study shows that in Cameroon it is possible to identify O. volvulus and O. ochengi infective larvae during routine fly dissections by morphology alone.

The prevalence of vector-borne parasites such as haemosporidian species is influenced by several environmental factors. While the negative effects of parasitism on hosts are well documented, these can also be amplified by interactions with environmental stressors, many of which are anthropogenic. Yet, we know little about the possible effects of anthropogenic perturbations on parasite prevalence. The goals of this study were to assess the prevalence and environmental determinants of haemosporidian parasites in a declining population of Tree swallows (Tachycineta bicolor) living in an agricultural landscape in southern Québec, Canada. Overall, a low prevalence and a moderate lineage diversity were identified in both adults and nestlings, confirming that transmission can occur during the breeding period. Anthropic areas, extensive cultures (hayfields and pastures) and forest cover within 500 km of nest boxes, as well as daily temperature fluctuations, were all related to infection by haemosporidian parasites. These findings suggest that anthropogenic alterations of landscape composition can modulate the prevalence of haemosporidian parasites in Tree swallows. Our results represent a baseline for future comparative studies assessing haemosporidian parasite prevalence in human-modified landscapes.

Theileria-free waterbuck (Kobus defassa) born in captivity were successfully infected with Theileria parva sporozoites derived from ticks infected by feeding on African buffalo (Syncerus caffer). All waterbuck underwent mild infections with the development of sporadic schizont and piroplasm parasitosis when inoculated with sporozoite doses lethal to cattle. A carrier state of T. parva was demonstrated by feeding clean R. appendiculatus nymphs on two of these infected waterbuck. Tick batches from these waterbuck on 2 of 5 occasions transmitted lethal Theileria infections to cattle. In a separate experiment, waterbuck cells were infected and transformed in vitro by T. parva sporozoites derived from buffalo but not by cattle-derived T. parva (Muguga) sporozoites. Waterbuck cells infected in vitro with T. parva isolated from buffalo were inoculated into autologous waterbuck but no infections developed. Theileria parva isolates generated in this study from various sources were characterized using anti-T. parva schizont monoclonal antibodies (MAbs), and it was found that buffalo-derived and waterbuck-passaged isolates had different profiles. Species-specific synthetic oligonucleotide probes, restriction fragment length polymorphism (RFLP) analysis with cloned T. parva DNA probes, and DNA sequence analysis of the p67 sporozoite antigen gene confirmed that the waterbuck-passaged parasite was T. parva. The Tpr repetitive probe hybridization patterns from the waterbuck-passaged parasites were different from the other samples tested. The ribosomal genotype of the waterbuck-passaged T. parva was similar to that of cattle-derived T. parva Muguga. Analyses with both probes and MAbs suggested that a minor parasite population present within the T. parva 7014 buffalo- derived stock had been selected during waterbuck passage. A variable region of the p67 sporozoite antigen gene of the waterbuck-passaged T. parva was similar to that of cattle-derived T. parva stocks and different from that of buffalo- derived parasites. Based on these results, methods were suggested to confirm and quantitate the involvement of waterbuck in the epidemiology of cattle theileriosis.

The control of gastrointestinal nematodes among ruminants maintained in zoological parks remains difficult due to infective stages develop in the soil. For the purpose to improve the possibilities of the control of gastrointestinal nematodes (genera Trichostrongylus, Nematodirus, Chabertia and Haemonchus) affecting wild captive bovidae ruminants belonging to the subfamilies Antilopinae, Caprinae, Bovinae and Reduncinae, commercial pelleted feed enriched with a blend of 104–105 spores of both filamentous fungi Mucor circinelloides + Duddingtonia flagrans per kg meal was provided for a period of 3.5 years. All animals were dewormed at the beginning of the trial and also when exceeding a cut-off point of 300 eggs per gram of feces (EPG). The anthelmintic efficacy ranged between 96 and 100%. The need for repeating the administration of parasiticide treatment disappeared at the 24th month of study in the Antilopinae individuals, and at the 8th month in the Caprinae, Bovinae and Reduncinae. No side-effects were observed on the skin or in the digestive, respiratory or reproductive system. It was concluded that this strategy provides a sustainable tool for preventing the contamination of paddocks where captive ruminants are maintained, decreasing the risk of infection by gastrointestinal nematodes and consequently the need of frequent deworming.
