In Bufo vulgaris there is a small flagellate similar to species described from insects and the Brazilian cavy. Such flagellates were formerly placed in the genus Monocercomonas but the generic name has been amended to Retortamonas Grassi 1879 by Wenyon (1926). The toad flagellate which I name R. rotunda n.sp. differs in structure from Monocercomonas bufonis Dobell 1909, which Grassé (1926) has removed to the genus Tetramastix Alexeieff 1916.

The stimulus provided to cercariae by the skin fat of their definitive hosts is not the same for Schistosoma mansoni and Austrobilharzia terrigalensis. Cercariae of the former parasite are stimulated by unsaturated fatty acids, those of the latter by cholesterol.

A reinvestigation of the penetration stimulant factors present in crude egg lecithin, which contains both cholesterol and free fatty acids as impurities, has shown that the latter are responsible for the stimulus in S. mansoni. The phospholipid fraction is inactive as is pure cholesterol. A little excitation of cercariae is produced by the monoglyceride fraction. These results fully confirm earlier findings.

Of surface active agents other than lecithin, stimulatory activity was found only in one product which possessed carboxyl groups. Of the C18 acids examined, stearic (18:0) is inactive, oleic (18:1) slightly active, linoleic (18:2) and linolenic (18:3) acids highly active. Oleic acid was shown by GC-mass spectrometry to be the most abundant acid in crude egg lecithin and is probably the main penetration stimulus present.

In South America, visceral leishmaniasis is frequently caused by Leishmania infantum and, at an unknown frequency, by Leishmania amazonensis. Therefore, mixed infections with these organisms are possible. Mixed infections might affect the clinical course, immune response, diagnosis, treatment and epidemiology of the disease. Here we describe the clinical course of mixed infections with L. amazonensis and L. infantum in a hamster model. We show that mixed infections are associated with more severe clinical disease than infection with L. amazonensis or L. infantum alone. In spleens with mixed infections, L. infantum outcompeted L. amazonensis in the tissue, but not in culture from tissue. We found increased levels of IgG in animals infected with L. infantum. Although more than 30 bands were revealed in a Western blot, the highest immunogenicity was observed with proteins having molecular masses of 95 and 90 kDa, whereas proteins with molecular masses of lower than 50 kDa were reactive frequently with serum from hamsters infected with L. amazonensis, and proteins with molecular masses of 80 and 70 kDa were reactive only with serum from hamsters infected with L. infantum. This finding has important implications regarding the biology of Leishmania and humoral immune responses to infections with these organisms.

Trilocularia acanthiaevulgaris occurs in the stomach and spiral valve of the spiny dogfish. Statistical analyses show that the stomach and spiral valve worms of male fish and the stomach worms of female fish exhibit seasonal trends in prevalence and mean worm burden, with peaks occurring in May, June and July of each year. Spiral valve worms of female fish do not show this trend. Calculation of the product-moment correlation coefficient shows that the peak in the number of stomach worms occurs approximately 1 month before that of spiral valve worms in male fish. Strobilization of the adult spiral valve worms and development of the resulting free proglottides is also seasonal, and takes place in June/July in both sexes. Seasonal trends in prevalence, mean worm burden and maturation of T. acanthiaevulgaris may be related to water temperature, and the cyclical changes in levels of reproductive hormones and thyroid activity associated with migration and mating of spiny dogfish. It is suggested that the availability of infected intermediate hosts and diet of the dogfish may also be important in explaining the seasonality of infection.

A division of labour occurs in colonies of the trematode Philophthalmus sp. within their first intermediate hosts. Two castes exist: one which reproduces and one which does not reproduce. It has been hypothesized that the benefit of the non-reproductive caste is in competitive interactions. Evidence for this from past experiments with Philophthalmus sp. colonies has been contradictory: the non-reproductive caste appears to benefit the colony in some way but not necessarily by combating interspecific competitors. The aims of this study were to consider intraspecific competition as a possible cause of the division of labour in Philophthalmus sp. colonies. Results show that mixed genotype infections occur in Philophthalmus sp. infected hosts and thus intraspecific competition is likely. Furthermore, the total number of individuals per colony is reduced in mixed genotype infections, indicating that intraspecific competition reduces colony fitness. However, the results do not indicate that the division of labour in Philophthalmus sp. plays a role in competitive interactions as the ratio of small, non-reproductive to large, reproductive individuals is unaffected by the presence of intraspecific competition. This is the first study to identify and quantify intraspecific competition in Philophthalmus sp., and to assess its selective role in this species’ division of labour.

The ultrastructure of the body wall of the redia of Sphaeridiotrema globulus is described. The tegument, which possesses numerous microvilli, is shown to be a syncytial, cytoplasmic layer connected to underlying subtegumentary ‘cells’ by cytoplasmic processes. Between the subtegumentary ‘cells’ are located the normal parenchyma cells of the body wall, which are believed to be important in carbohydrate storage.Mitochondria, Golgi complexes, endoplasmic reticulum and beta glycogen granules are located in the tegument and subtegumentary ‘cells’. In addition, large whorled bodies and dense secretory granules appear to be formed within the subtegumentary ‘cells’ prior to their passage into the outer tegument. It is suggested that these whorled structures are contributing to the growth of the tegument. Small membrane-bound ‘vesicles’ are also seen in the tegument and some of these may be pinocytotic in nature.Following incubation, horseradish peroxidase tracer was localized within ‘vesicles’ in both the tegument, subtegumentary ‘cells’ and parenchyma cells, which indicates that the redial body wall may be important in the uptake of nutrients.Phosphatase enzymes are abundant within the tegument, particularly in association with the plasma membranes and microvilli. These enzymes, which appear to have their origin in the endoplasmic reticulum of subtegumentary cells, are believed to be associated with the uptake of nutrients through the redial tegument.

Parasitological and immunological interactions between Eimeria vermiformis or E. pragensis and Trichinella spiralis were investigated during concurrent infections in NIH, BALB/c and B10.G inbred mice. The establishment of T. spiralis was unaffected by the presence of either coccidium, but expulsion of adult worms was delayed significantly in mice infected with E. vermiformis; E. pragensis did not have this effect. Replication of E. vermiformis was enhanced in concurrent infections with T. spiralis, but that of E. pragensis was reduced. Specific immune responses to each parasite were unaffected in mice infected with T. spiralis and E. pragensis, but levels of some responses were reduced when T. spiralis and E. vermiformis were combined. Thus both in vitro antigen-induced proliferation of mesenteric lymph node cells (MLNC) and intestinal mastocytosis were lower than in singly infected mice. Mitogen (Con A) responsiveness of MLNC was not affected in mice infected with T. spiralis and E. vermiformis, and cells from these mice were capable of transferring protective immunity to the nematode in naive recipients. Injection of monoclonal antibody to interferon gamma, a major component of the cytokine response to E. vermiformis, did not prevent delay of worm expulsion in concurrent infections. The results are discussed in terms of possible interactions between the T helper cell subsets or the inflammatory components of the responses induced by each parasite.

The limpet Lepetodrilus fucensis McLean is very abundant at hydrothermal vents on the Juan de Fuca and Explorer Ridges in the northeast Pacific Ocean. This limpet is parasitized by an undescribed chitonophilid copepod throughout the limpet's range. The parasite copepodite enters the mantle cavity and attaches to the afferent branchial vein. The initial invasive stage is a vermiform endosome within the vein that develops an extensive rootlet system causing an enlargement of the afferent branchial vein. Subsequently, an ectosomal female body grows outside the vein to sizes up to 2 mm in width. Once a dwarf male attaches, egg clusters form and nauplii are released. In over 3000 limpets examined from 30 populations, prevalence averaged about 5% with localized infections in female limpets over 25%. After the establishment of limpet populations at new vents, copepod prevalence increased over the succeeding months to 3 years. Host effects were marked and included castration of both sexes and deterioration in gill condition which affected both food acquisition and the gill symbiont. There was a significantly greater parasite prevalence in larger females which likely modifies the reproductive and competitive success of local host populations.

1. Musca domestica and other non-biting flies frequenting similar feeding-grounds, are probably all liable to infection with a common flagellate. The great variability of this form is shown on comparing the flagellates in the larvae with those in the mature flies of Homalomyia, sp. and of Scatophaga lutama.

2. Infection is casual–i.e. by the mouth. In the case of the dung-flies examined, the larvae ingest faecal matter infected with herpetomonad cysts: the cysts develop into flagellates in the mid-gut, where they multiply with great rapidity: towards the close of larval life, when the larva stops feeding, they round up in the hind-gut, and are for the most part passed out as cysts. A few survive the pupal stage in a half-encysted condition, but it is probable that the infection of the adult fly is usually freshly acquired. The cycle in the fly is similar to that in the larva, and is in agreement with Patton's account of Herpetomonas muscae-domesticae. The parasite was never found in the ovaries or ova. Patton's suggestion that the degree of infection depends directly on the number of cysts ingested, is borne out by the much higher rate of infection in the larvae than in the flies: this is not surprising when we remember the complete restriction of the feeding larva to the infected area.

3. It is important to use some reliable cytological stain such as iron-haematoxylin as a control to Romanowsky stains where possible, seeing that very different results are sometimes given by the two methods.

4. The apparent double flagellum is produced in the course of longitudinal division. The new flagellum grows up alongside the old, and is not merely split off from it: this is best seen in the flagellate from the larva, where the body of the organism usually divides at an earlier period than in the fly. Study of the living flagellate is necessary to a clear understanding of the process of division.

5. I have seen no conjugation in the living herpetomonads. Occasionally flagellates were met with in Giemsa preparations devoid of a tropho-nucleus. Such individuals might be regarded as male gametes; it is more likely that they are simply degenerate forms. Sufficient consideration is not given to the possibility of degeneration in richly-nourished, rapidly multiplying protozoa, such as the trypanosomes and their allies.

6. In encystment the flagellum is not cast off bodily, but is drawn down into the cell by the kineto-nucleus, which moves to a position either alongside of, or posterior to the tropho-nucleus. In this way apparent Crithidia or even trypanosome forms are produced, but there is no hint of an undulating membrane. Small blunt “trypanosomes” were also produced occasionally by adhesion of the flagellum to the body-wall in the “stickiness” resulting from confinement under a cover-glass: the efforts of the flagellum to free itself raised up an undulating membrane, and produced a very deceptive appearance.

7. The early stages of encystment could be induced by keeping the flagellates under waxed-down cover-slips, where they would continue to live for 30—40 hours. Similar results were got by transferring flagellates to agar-agar, or by subjecting the larval host to starvation for a day or two.

8. From what I have seen, I do not agree with Chatton and Alilaire's suggestion to divide the genus Herpetomonas into Leptomonas and Herpetomonas proper. I do not think that these authors are justified in regarding the double flagellum and the presence of a long rhizoplast as generic characters.

Objectives. Among parasites, Taylor's power law identifies a tight relationship in aggregation of macroparasite infection intensity with few exceptions; notably, the nematode family Oxyuridae tends to have higher than expected aggregation. Oxyuridae infect a wide range of mammalian hosts and have a unique reproductive strategy that involves conventional horizontal transmission, as well as re-infection of an already infected host. We asked the question, do the unique aspects of pinworm life-history explain an exception to the widely observed patterns of aggregation of parasite populations? Methods. We empirically examined the differences among Oxyuridae (genus: Syphacia) compared with other helminth (genus: Heligmosomoides) parasite aggregations in 2 rodent hosts with similar ecology: the yellow-necked mouse (Apodemus flavicollis) from Trento, Italy and the white-footed mouse (Peromyscus leucopus) from Pennsylvania, USA. To investigate the effects of pinworm life-history characteristics on generating aggregation, we present a stochastic model that explores aggregation under a range of host-self-infection, parasite death, and transmission scenarios. Results. Oxyuridae parasites had consistently greater aggregation compared to other nematodes regardless of host or parasite species identity, and pinworm aggregation exceeded the range of macroparasite aggregation described previously. Conclusions. Our simulations demonstrate that host-self-infection, on its own, is sufficient to generate aggregation values greater than the predicted values.

A comprehensive account is given of the evolution of registration guidelines and safety testing procedures for microbial insecticidal agents. Particular emphasis is given to the use of viruses for pest control and the various guidelines developed to assess their possible hazards. The likely gains and risks associated with using viruses are discussed. Several meetings have been held in the last 10 years to assess the hazards of virus insecticides. Some of these meetings have produced recommendations, some have developed guidelines for safety testing. These meetings are reviewed. The various guidelines developed for safety testing are critically evaluated and the UK Registration Criteria for Biological Agents used as Pesticides are reproduced in full. Examples of viruses that have been safety tested and registered for use are given and the criteria used are described. Conclusions are drawn on the merits of the guidelines presently available and the likely future development of safety testing schemes is considered.

A specific enzyme-linked immunosorbent assay, based on a 48 kDa metallo-proteinase isolated from the frontal glands of a porocephalid pentastomid (Porocephalus crotali) in rat intermediate hosts, is described. SDS–PAGE analysis of whole nymphal homogenate, followed by Western blotting and probing with immune rat serum, revealed that only 2 protein bands label consistently; these are the 48 kDa metallo-proteinase and a 150 kDa metallo-proteinase from the body fluid, but only the former labels strongly. Furthermore this response is detected earlier and persists much longer. These characteristics uniquely qualify the 48 kDa metallo-proteinase as a diagnostic antigen. We review the literature concerning the related porocephalid Linguatula serrata from the nasal cavities of domestic dogs, which can zoonotically infect man principally through the ingestion of eggs. We conclude that the high prevalences in dogs in certain areas, most notably the Middle East, must inevitably signal significant human infections. Our ELISA can be readily adapted for the serodiagnosis of linguatulosis and this should establish whether our prediction is true.

The aim of the present study was to compare the helminth infra and component communities of wild and stocked brown trout in Lough Feeagh, in the west of Ireland, and also to examine the establishment and development of helminth communities in stocked brown trout. Fish were sampled in May, August and November 1997 and 1998 and an additional sample of wild brown trout was examined in April 1997. In total 217 wild trout and 122 stocked trout were examined. The acanthocephalans Acanthocephalus clavula and Pomphorhynchus laevis were the first parasite species to infect stocked trout in May 1997. In May 1998 both acanthocephalan species along with the trout specialists Eubothrium crassum and Crepidostomum farionis were the first to infect trout. Mean species richness values for stocked trout increased from May to November, in 1997 and 1998. For wild trout, mean species richness values increased from April to November, in 1997 and decreased from May to November in 1998. The parasite communities of wild trout were richer than those of stocked trout in May of both years. In August the parasite communities recorded for wild and stocked trout were similar in terms of the number of species present but differed in terms of structure, and in November the parasite communities of stocked trout were richer than those of wild trout.

Spermatogenesis and sperm maturation in Nematospiroides dubius were studied using electron microscopy. The testis is telegonic and germ cells in the zones of mitosis, growth and meiosis are connected by a central anucleate mass of cytoplasm, the rachis. The early part of spermatogenesis is dominated by the synthesis and growth of membrane-bound vesicles called membranous organelles, which originate from RER-associated Golgi bodies. Following meiosis the spermatids separate from the rachis and their chroinatin, which is no longer bounded by a nuclear envelope, condenses into an arrow-head shape and is extruded to form a tail-like structure. After insemination spermatozoa undergo a profound change called activation. The cytoplasmic region which was previously long and cylindrical becomes spherical and the membranous organelles which lined its perimeter fuse with the plasma membrane and become confined to the posterior hemisphere of the sperm, close to the nuclear tail. The anterior half of the sperm is devoid of organelles but contains many filaments organized into clumps and chains; this region being responsible for amoeboid locomotion of the sperm.

The effect of the microtubule inhibitors coichicine (1×10−3 M) and tubulozole-C(1×10−6 M) on the ultrastructure of adult Fasciola hepatica has been determined in vitro by transmission electron microscopy (TEM), using both intact flukes and tissue-slice material. With colchicine treatment, the apical membrane of the tegument became increasingly convoluted and blebbed, while accumulations of T1 secretory bodies occurred in the basal region of the syncytium, leading to progressively fewer secretory bodies in the syncytium. In the tegumental cells there were distinct accumulations of T1 secretory bodies around the Golgi complexes, which remained active for up to 12 h incubation. Tubulozole-treated flukes showed more severe effects, with initial accumulations of secretory bodies, both at the tegumental apex and base. This was followed in the later time-periods by the sloughing of the tegumental syncytium. In the underlying tegumental cells, the granular endoplasmic reticulum (GER) cisternae were swollen and disrupted, becoming concentrated around the nucleus. The Golgi complexes were dispersed to the periphery of the cells and gradually disappeared from the cytoplasm. After treatment with both drugs, the cell population in the vitelline follicles was altered, with an abnormally large proportion of stem cells and relatively few intermediate type I cells. The nurse cell cytoplasm became fragmented and was no longer in contact with the vitelline cells, while the shell globule clusters within the intermediate type 2 and mature cells were loosely packed. In the mature vitelline cells, ‘yolk’ globules and glycogen deposits became fewer than normal and lipid droplets were observed. The results are discussed in relation to the different modes of action of the two drugs and potential significance of this to anthelmintic (benzimidazole) therapy.

Five species of parasitic copepods are described based on the material collected during cruise 16 of R/V Anton Bruun. They are: from the gill chambers of macrourids, Chondracanthodes deflexus Wilson, Lateracanthus quadripedis Kabata and Gusev and Brachiella annulata Markevitch; from the anal fin of a macrourid, Nudiclavella galapagoense gen.nov., sp.nov.; and from the gill filaments of brotulids, Naobranchia maxima sp.nov. All fishes were caught with FVSL (free vehicle set lines) in the deep sea off the Galápagos Islands, ranging from 1710 to 3330 m.

Homologues of c-fos and c-jun from total DNA of Taenia crassiceps and Taenia solium were cloned and sequenced. The amino acid alignment analysis revealed that c-fos DNAs from T. crassiceps and T. solium were highly homologous (96%), and both have high homology compared to several mammalian c-fos proteins (93% to mouse, 96% to rat and 86% to human). The c-jun protein alignment showed higher homology (T. crassiceps and T. solium have 98%), when compared with mouse, rat and human, being 92%, 98% and 93% respectively. RT–PCR amplification of the parasite's total RNA, showed that T. crassiceps expressed both AP-1 complex genes, while T. solium only expressed c-fos. Southern blot hybridization analysis confirmed the true origin of each amplified gene. AP-1 transcription gene expression is regulated by oestradiol in the same fashion as their mammalian counterparts only in T. crassiceps. To study if AP-1 genes are involved in a physiological function of the cyst, reproduction was studied in vitro. Oestradiol treatment stimulated reproduction in T. crassiceps but not in T. solium cysticerci. This is the first report of the detection and functionality of AP-1 transcription factor genes in any species of helminth parasite.

We asked if and how feeding performance of fleas on an auxiliary host is affected by the phylogenetic distance between this host and the principal host of a flea. We investigated the feeding of 2 flea species, Parapulex chephrenis and Xenopsylla ramesis, on a principal (Acomys cahirinus and Meriones crassus, respectively) and 8 auxiliary host species. We predicted that fleas would perform better (higher proportion of fleas would feed and take larger bloodmeals) on (a) a principal rather than an auxiliary host and (b) auxiliary hosts phylogenetically closer to a principal host. Although feeding performance of fleas differed among different hosts, we found that: (1) fleas did not always perform better on a principal host than on an auxiliary host; and (2) flea performance on an auxiliary host was not negatively correlated with phylogenetic distance of this host from the principal host. In some cases, fleas fed better on hosts that were phylogenetically distant from their principal host. We concluded that variation in flea feeding performance among host species results from interplay between (a) inherent species-specific host defence abilities, (b) inherent species-specific flea abilities to withstand host defences and (c) evolutionary tightness of association between a particular host species and a particular flea species.

While working on parasites of fish for the New York State Conservation Department during the summer of 1928 it was possible to secure experimental evidence on the life-cycle of Proteocephalus pinguis La Rue 1911. The adult parasite lives in the digestive tract of Esox lucius Linnaeus and E. reticulatus Le Sueur where it ranges in length between 50 and 90 mm. Since La Rue (1914) gives a complete description of the adult it is not necessary to go further into the morphology of this form. Adult tapeworms for the experiments were taken from E. lucius from Lake Erie and Ellicott Creek, near Buffalo, New York, and from E. reticulatus from Barrett and China Ponds near Carmel, New York.

The interaction between the 2 nodular worm species in the pig, Oesophagostomum dentatum (O.d.) and Oesophagostomum quadrispinulatum (O.q.), was studied by comparing the development and distribution of the species following single or mixed infections. The faecal egg excretion levels were assessed at regular intervals from week 3 post-inoculation, and indicated a strong negative impact of the introduction of O.q. on the continued egg excretion of O.d. All pigs were killed 9 weeks after the first inoculations to determine the composition and location of the worm burdens in the large intestine. O.q. was found more anteriorly located in the intestine than O.d., thus confirming previous descriptions. When both species were present, the distribution of O.d. was moved further posteriorly and was more spread out than in single-species infections. There appeared to be no adverse effect of O.d. on the establishment and fecundity of O.q. However, the worm recoveries corroborated the egg excretion observations, namely reduced worm burdens of O.d. if O.q. was introduced, or if O.q. was already present. It is uncertain whether this effect is caused by differences in host reaction against the two species, or whether a more specific competition occurs between the two nodular worm species in pigs.
