Schistosoma mansoni cercariae were incubated for 3–5 h at 37°C in various test solutions, and the rate and extent of conversion of th ecercariae to schistosomula were determined. Criteria used to identify schistosomula included: (1) loss of cercariai tail, (2) viability of organisms in saline but not in water, (3) pre-acetabular gland evacuation and (4) ability to survive in culture.

The actions of known platyhelminth FaRPs on the contractility of whole-worm preparations of the monogenean, Diclidophora merlangi have been examined in vitro for the first time. All of the peptides tested had excitatory effects on the motor activity of the worm. The order of potency for the peptides tested was: YIRFamide > GYIRFamide = RYIRFamide > GNFFRFamide = FLRFamide. However, although YIRFamide was more potent than GYIRFamide, the latter was the most efficacious on each of the motility parameters (tension, contraction amplitude and contraction frequency) examined at concentrations [ges ]0·1 μm. Serotonin, which stimulates contractility in the worm was used as a positive control. The excitatory activity of turbellarian and cestode neuropeptides on a monogenean indicates at least some structural similarities in the neuropeptide receptors of these classes of flatworm.

Fleas are insects that undergo a complete metamorphosis in the course of their development from egg to adult. The eggs laid by the mother are not, as are the eggs of lice, attached in any way to the skin, fur or feathers of the animal on which the parents are parasitic. They fall into the nest, or drop to the ground within the lair or “ run ” of the host. Hatching takes from three to ten days according to the temperature. The larvae are active, whitish maggots, eyeless and legless, which are not parasitic, but feed on organic matter in the host's bed, or in the dust that collects on the ground in its proximity.

1. Copulation between individuals of the two species O. moubata, and O. savignyi may occur, and the stimulus of coition may induce oviposition, the eggs being non-fertile.

2. There is no evidence of parthenogenesis in this species, nor do these ticks undergo ecdysis after reaching maturity.

3. An increase in temperature of 8° C. from 22° C. (a) doubles the.rate of oviposition, (b) decreases the fertility of the eggs by 30 per cent., (c) reduces the longevity of the female tick from 715 to 397 days, i.e. by 40 per cent, and (d) approximately halves the period required for metamorphosis, under laboratory conditions. Under these conditions at 37° C., reproduction is inhibited, and the longevity of the female is reduced by 80 per cent.

4. An individual may undergo from four to eight ecdyses before reaching maturity, the great majority of the males appearing after the fifth ecdysis and the females about equally after the fifth and sixth ecdyses.

5. Engorgement can take place one or two days after emergence, at any stage, the average time required being three-quarters of an hour. There is great variation in the extent of engorgement at each stage, but this is not correlated with the ecdysis period. The largest meal may be taken either before or after an individual reaches maturity.

6. Moisture has an adverse influence on the vitality of the individual, excess of moisture inhibiting growth.

7. An approximation to the stage of development attained can be made after the study of the structure of the hypostome, leg and spiracle. The larval and first four nymphal stages are fairly well differentiated, much more so than the later stages, but variation due to nutrition requires further study.

Excretions and secretions (E–S) were collected from a series of developmental stages of Litomosoides carinii maintained in vitro. Measurement of the protein content of E–S obtained from each stage indicates that the rate of production of E–S varies enormously during development of the worm. E–S was iodinated using both Iodogen and the Bolton and Hunter Reagent and was also biosynthetically labelled by incubating worms in the presence of [35S]methionine and [3H]leucine. Attempts to biosynthetically label E–S of mature worms and microfilariae with [3H]glucose were unsuccessful. Examination of radio-isotope labelled E–S by SDS–PAGE revealed that some components were sex specific and that the differences in total E–S production during development were due to the existence of both stage-specific components and components whose rate of release varied during parasite maturation. Antigenic characterization of E–S, carried out by immunoprecipitation in combination with SDS–PAGE, indicated that E–S consists of immunogenic components, a molecule which is probably a non-immunogenic parasite product, and host albumin. The implications of these findings for the construction of diagnostic tests to detect products of human filarial parasites are discussed.

In tests for chemotherapeutic activity, mice were infected with Eperythrozoon coccoides, treated with drugs for 2 days, splenectomized, and treated with drugs for a further 2 days. Blood films were examined from the second day after splenectomy, when control mice showed heavy blood infections.

Neoarsphenamine, aureomycin and terramycin were active.

Tryparsamide, Chloromycetin, alloxan and polyoxyethylene ether, D2, were slightly active.

Many drugs, listed in Table 3, were inactive.

Neoarsphenamine cleared E. coccoides from the blood very rapidly. This action was inhibited by dimercaprol (BAL).

E. coccoides closely resembles Haemobartonella muris in its sensitivity to drugs. Both micro-organisms probably have affinities with the Rickettsiae.

1. A new species of Bothriocephalus—B. pycnomerus—is described from the intestine of Ophiocephalus marulius Ham. Buch., from the Ganges and Jumna at Allahabad, India.

2. B. pycnomerus closely resembles B. histiophorus (Shipley 1901; from the marine sword-fish Histiophorus) in structure but differs chiefly in the possession of an armed scolex and its crowded indistinct segmentation.

3. Lühe's definition of the genus Bothriocephalus must be amended to include forms with armed scolices and forms in which the ventral uterine apertures are a-median and irregularly alternate.

4. Two new species of Proteocephalidae are described which possess armed muscular rostella, and for these a new genus Gangesia is created and defined. The definitions of the Order Tetraphyllidea (Lühe 1910) and of the Family Proteocephalidae (La Rue 1914) must be amended to include forms possessing muscular rostella, armed or unarmed.

5. These two new species—Gangesia wallago and G. macrones—were found in the intestines of Wallago attu Bleek and Macrones seenghala Sykes (both Siluroids) respectively, from the Ganges and Jumna at Allahabad.

6. Southwell's “Ophryocotyle bengalensis,” from Ophiocephalus striatus, Labeo rohita and Wallago attu, is probably identical with Gangesia wallago.

Knowledge regarding helminth control strategies and nematode infection of horses in Ireland is limited and only one study has been published recently. This present study was designed to investigate the current helminth control strategies followed by horse owners in Ireland. A questionnaire was formulated to collect data on general grazing, pasture management and deworming strategies including the use of fecal egg counts. Questionnaires were emailed to 700 members of the Irish Thoroughbred Breeders Association and Horse Sport Ireland. Only 78 questionnaires were returned. Respondents indicated that horses are grazed for 16–24 h day−1 during the summer and autumn (89% and 65%, respectively). Removing feces from the pasture was implemented by 37.6% of respondents. Few (22.2%) owners kept horses off pasture after worming. Overall, ivermectin and moxidectin were the most commonly administered anthelmintics in 2014 by 75% and 62% of respondents, respectively. Benzimidazole and pyrantel drugs were used by 53% and 35% of respondents, respectively. The majority of farms (81.4%) treated horses 4–5 times per year and 74.2% only estimated the weight of the horses visually. The findings of this study illustrates that many stud managers/owners do not follow best practice with regard to helminth control and more education is needed.

The Trypanosoma cruzi cyclophilin gene family comprises 15 paralogues whose nominal masses vary from 19 to 110 kDa, namely TcCyP19, TcCyP20, TcCyP21, TcCyP22, TcCyP24, TcCyP25, TcCyP26, TcCyP28, TcCyP29, TcCyP30, TcCyP34, TcCyP35, TcCyP40, TcCyP42 and TcCyP110. Under the conditions used, only some of the T. cruzi cyclophilin paralogue products could be isolated by affinity chromatography. The 15 paralogues were aligned with 495 cyclophilins from diverse organisms. Analyses of clusters formed by the T. cruzi cyclophilins with others encoded in various genomes revealed that 8 of them (TcCyP19, TcCyP21, TcCyP22, TcCyP24, TcCyP35, TcCyP40, TcCyP42 and TcCyP110) have orthologues in many different genomes whereas the other 7 display less-defined patterns of their sequence attributes and their classification to a specific group of cyclophilin's orthologues remains uncertain. Seven epimastigote cDNA clones encoding cyclophilin isoforms were further studied. These genes were found dispersed throughout the genome of the parasite. Amastigote and trypomastigote mRNAs encoding these 7 genes were also detected. We isolated 4 cyclosporin A-binding proteins in T. cruzi epimastigote extracts, which were identified by mass spectrometry as TcCyP19, TcCyP22, TcCyP28 and TcCyP40. Cyclosporin A-binding to these cyclophilins might be of importance to the mechanism of action of Cyclosporin A and its non-immunosuppressive analogues, whose trypanocidal effects were previously reported, and therefore, of potential interest in the chemotherapy of Chagas' disease.

The incidence and prevalence of coccidia have been studied in populations of free-living red kangaroos, Megaleia rufa, and grey kangaroos, Macropus canguru, in four geographically distinct areas in Australia.

Two new species of Eimeria—E. wilcanniensis and E. toganmainensis—are described from M. rufa, and five—E. cunnamullensis, E. gungahlinensis, E. purchasei, E. kogoni, and E. hestermani—from M. canguru.

The incidence of coccidia ranged from 6 to 14% in populations of M. rufa and from 26 to 70% in M. canguru. The susceptibility of the animals to coccidia was analysed in relation to the tibia length. Red kangaroos with tibia approximately 360 mm. long and corresponding to an age of about 12 months were found to be most susceptible. No clinical cases of coccidiosis were seen in the field.

Self-fertilization (or selfing), defined as the fusion of male and female reproductive cells originating from the same individual, is the most extreme case of inbreeding. Although most hermaphroditic organisms are in principle able to self-fertilize, this reproductive strategy is commonly associated with a major disadvantage: inbreeding depression. Deleterious effects due to the loss of genetic diversity have been documented in numerous organisms including parasites. Here we studied the effects of inbreeding depression on the offspring of the progenetic trematode Coitocaecum parvum. The parasite can use 2 alternative life-history strategies: either it matures early, via progenesis, and produces eggs by selfing in its second intermediate host, or it waits and reproduces by out-crossing in its definitive host. We measured various key parameters of parasite fitness (i.e. hatching and multiplication rates, infectivity, survival) in offspring produced by both selfing and out-crossing. Altogether, we found no significant difference in the fitness of offspring from progenetic (selfing) and adult (out-crossing) parents. In addition, we found no evidence that either strategy (progenesis or the normal three-host cycle) is heritable, i.e. the strategy adopted by offspring is independent of that used by their parents. Although it is unclear why both reproductive strategies are maintained in C. parvum populations, our conclusion is that producing eggs by selfing has few, if any, negative effects on parasite offspring. Inbreeding depression is unlikely to be a factor acting on the maintenance of the normal three-host life cycle, and thus out-crossing, in C. parvum populations.

Parasites with complex life cycles have developed numerous and very diverse adaptations to increase the likelihood of completing this cycle. For example, some parasites can abbreviate their life cycles by skipping the definitive host and reproducing inside their intermediate host. The resulting shorter life cycle is clearly advantageous when definitive hosts are absent or rare. In species where life-cycle abbreviation is facultative, this strategy should be adopted in response to seasonally variable environmental conditions. The hermaphroditic trematode Coitocaecum parvum is able to mature precociously (progenesis), and produce eggs by selfing while still inside its amphipod second intermediate host. Several environmental factors such as fish definitive host density and water temperature are known to influence the life-history strategy adopted by laboratory raised C. parvum. Here we document the seasonal variation of environmental parameters and its association with the proportion of progenetic individuals in a parasite population in its natural environment. We found obvious seasonal patterns in both water temperature and C. parvum host densities. However, despite being temporally variable, the proportion of progenetic C. parvum individuals was not correlated with any single parameter. The results show that C. parvum life-history strategy is not as flexible as previously thought. It is possible that the parasite's natural environment contains so many layers of heterogeneity that C. parvum does not possess the ability to adjust its life-history strategy to accurately match the current conditions.

Defences against parasites are characterized by inducible, amplifiable responses, often with a memory component. Inducible defences with similar properties are common in a variety of other types of interactions, for example many aquatic invertebrates produce inducible structural defences against their predators and competitors. Most inducible defences have the following properties: (1) a threshold of activation; (2) an amplification of response with increasing stimulus; (3) a memory component. Specificity, amplification and memory are the basis for defining a defence as ‘immune’ (Klein, 1982), and these properties are present in both the vertebrate and invertebrate internal defence responses to pathogens. Invertebrates differ in the absence of immunoglobulins and therefore in reduced specificity. Although the reduced specificity of invertebrate internal defence systems is often viewed as proof of their ‘primitiveness’, the differences in defence systems of vertebrates and invertebrates may be more related to their respective selection regimes than to phylogeny. The syngeneic recognition system of vertebrates functions to recognize small departures from self, such as would arise from neoplasia. Are vertebrates under more intense selection from neoplasia, perhaps due to a greater incidence of hormonal imbalance or hypersensitivity reactions? The invertebrate internal defence systems are all less discriminating than the vertebrate, but there are marked differences in degree of discrimination depending on whether the group is colonial or not. Even the phyla of colonial animals with quite simple body plans, the sponges and cnidarians, have a more discriminating recognition system than the phyla of solitary animals with more complex body plans, such as the molluscs and arthropods. The primary effectors of all invertebrate responses to parasites are encapsulation and phagocytosis, although in some phyla there are specific antibacterial proteins that can also be induced. A primary effector of the vertebrate immune system is also based on phagocytosis, but is mediated largely by immunoglobulins. Although memory is a widespread property of inducible defence systems, the mechanisms, underlying memory components are unknown except in the vertebrate immune response. The function and adaptiveness of many of these accelerated or amplified second-set responses remain unclear, although it is tempting to conclude that a memory component is a beneficial adaptation to multiple, intermittent stimuli. From this review I conclude that the conditions that favour the evolution of inducible defences include: (1) an intermittent, unpredictable selective regime; (2) selection for a ‘camouflaged’ defence, to slow parasite counter-adaptation; (3) reliable cues and (4) high costs of defence.

Previous studies have reported nematodes of the Spirocercidae family in the stomach nodules of red foxes (Vulpes vulpes) described as Spirocerca sp. or Spirocerca lupi (Rudolphi, 1819). We characterized spirurid worms collected from red foxes and compared them to S. lupi from domestic dogs by morphometric and phylogenetic analyses. Nematodes from red foxes differed from S. lupi by the presence of six triangular teeth-like buccal capsule structures, which are absent in the latter. Additionally, in female worms from red foxes, the distance of the vulva opening to the anterior end and the ratio of the glandular-to-muscular oesophagus lengths were larger than those of S. lupi (P < 0.006). In males, the lengths of the whole oesophagus and glandular part, the ratio of the glandular-to-muscular oesophagus and the comparison of the oesophagus to the total body length were smaller in S. lupi (all P < 0.044). Phylogenetic analyses revealed that S. lupi and the red foxes spirurid represent monophyletic sister groups with pairwise nucleotide distances of 9.2 and 0.2% in the cytochrome oxidase 1 and 18S genes, respectively. Based on these comparisons, the nematodes from red foxes were considered to belong to a separate species, for which the name Spirocerca vulpis sp. nov. is proposed.

Trypanosoma cruzi, the agent of Chagas disease is associated with a very high clinical and epidemiological pleomorphism. This might be better understood through studies on the evolutionary history of the parasite. We explored here the value of antigen genes for the understanding of the evolution within T. cruzi. We selected 11 genes and 12 loci associated with different functions and considered to be involved in host-parasite interaction (cell adhesion, infection, molecular mimicry). The polymorphism of the respective genes in a sample representative of the diversity of T. cruzi was screened by PCR-RFLP and evolutionary relationships were inferred by phenetic analysis. Our results support the classification of T. cruzi in 2 major lineages and 6 discrete typing units (DTUs). The topology of the PCR-RFLP tree was the one that better fitted with the epidemiological features of the different DTUs: (i) lineage I, being encountered in sylvatic as well as domestic transmission cycles, (ii) IIa/c being associated with a sylvatic transmission cycle and (iii) IIb/d/e being associated with a domestic transmission cycle. Our study also supported the hypothesis that the evolutionary history of T. cruzi has been shaped by a series of hybridization events in the framework of a predominant clonal evolution pattern.

The anatomy of the oncomiracidium of Capsala martinieri, a monogenean skin parasite of the sun fish Mola mola, is described and compared with that of the oncomiracidium of Entobdella soleae, a skin parasite of the common sole Solea solea.

The oncomiracidium of C. martinieri is characterized by the occurrence of an extensive bilaterally symmetrical and constant pattern of body pigment of unknown function.

Expression of metalloprotease activities during metacyclogenesis of a series of strains and clones of Trypanosoma cruzi was investigated using SDS-polyacrylamide gel electrophoresis with a range of different co-polymerized proteolytic substrates. A complex pattern of metalloprotease expression, with considerable quantitative and qualitative variability between strains and life-cycle stages, was apparent. This is in contrast to previously published data, and data presented in this study, which demonstrate a high degree of conservation of expression of cysteine proteases in different strains and clones of T. cruzi. All the metalloprotease activities identified partitioned into the detergent phase of a Triton X-114 extract, suggesting that they are membrane-bound. Developmental regulation of expression during metacyclogenesis, either in terms of quantity, or in terms of difference in relative amounts of different isoforms, was apparent for all isolates studied except CL14. However, a clearly metacyclic-specific/metacyclic-enriched metalloprotease was detected only in T. cruzi Dm28c and 383, and our results demonstrate that a metacyclic-specific metalloprotease common to all isolates of the parasite could not be detected, at least at this level of analysis.

The properties of 4 fluorescent lipid compounds in the surface membrane of adult male Schistosoma mansoni worms were examined by fluorescent microscopy and fluorescent recovery after photobleaching (FRAP). The data suggest that the probes N-(4,4-difluoro-5,7-dimethyl-4-bora-3a,4a-diaza-s-indacene-3-pentanoyl) sphingosine (BODIPY FL ceramide) and PKH2 pass through the outer membrane and enter structures in or below the membrane. In contrast 5-(N-octadecanoyl)aminofluorescein (AF18) and N-(4,4-difluoro-5,7-dimethyl-4-bora-3a,4a-diaza-s-indacene-3-pentanoyl) sphingosylphosphocholine (BODIPY FL sphingomyelin) insert into the outer monolayer. The DL values of these latter 2 compounds, 8·83 ± 2·35 × 10−9 cm2 s−1 and 2·76 ± 0·53 × 10−9cm2 s−1, respectively, suggest that they enter different domains. Furthermore, it was observed that both BODIPY FL ceramide and BODIPY FL sphingomyelin entered particular structures in or under the surface membrane. The possible nature of these particles is discussed.

1. Careful search has failed to disclose the presence of flame cells in the excretory system of the adult liver fluke.

2. Fat globules are produced in the walls of the excretory system, and are freed into the lumen, and thus to the exterior.

3. These globules do not contain appreciable quantities of cholesterol, cholesterides or lipins, but may contain some unsaturated fatty material.

4. The aqueous medium in which the globules float is either neutral or slightly alkaline.

5. The distribution of reduced ascorbic acid has been investigated by the silver nitrate technique. This indicates that the main concentration occurs in the smaller vessels of the excretory system, and in the submuscular tissues of the body wall.
