Uptake of triclabendazole by the liver fluke, Fasciola hepatica has been studied by experiments designed to block either oral uptake of drug, by use of ligatures, or trans-tegumental diffusion, by allowing the drug to bind to an excess of bovine serum albumin (BSA) in the medium. Changes to the tegumental system, musculature and gut were assessed using transmission electron microscopy. Flukes were incubated in vitro for 24 h in TCBZ.SO (15 μg/ml). Disruption to the tegument and muscle was similar in ligatured and non-ligatured flukes, suggesting that closing the oral route did not affect drug uptake. The ultrastructure of the gastrodermal cells remained unchanged. Non-ligatured flukes were also incubated for 24 h in vitro in TCBZ.SO (15 μg/ml) in the presence of red blood cells (RBCs). Oral uptake of blood was demonstrated, but gut ultrastructure remained normal, whereas the tegument was severely disrupted. In separate experiments, ligatured and non-ligatured flukes were incubated in TCBZ.SO (15 μg/ml) in the presence of BSA (30 mg/ml) for 24 h in vitro. There was a marked decrease in the degree of tegumental disruption observed compared with TCBZ.SO action alone; again, the gut remained normal. The findings support previous morphological and pharmacological studies indicating that trans-tegumental uptake of triclabendazole predominates in the liver fluke.

A pleomorphic neoplasm (PN) is described from sections of Onchocerca volvulus worms in nodules excised from Cameroonian patients. PN is confined to older, non-fecund, female worms, and those classed as moribund/dead. It is mainly composed of small, roundish, basophilic cells of diverse sizes, often forming a ‘rosette’ pattern around amorphous eosinophilic centres. The cells have a high nuclear/cytoplasmic ratio and up to 2–3 mitoses/high-power field; some become grossly enlarged, highly polymorphic and contain large, irregular blocks of chromatin. The eukaryotic PN cells first appear posteriorly in the pseudocoelom, probably from ovarian cells; they spread anteriorly, invading or compressing the uteri. Ivermectin treatment increased the prevalence PN from 3.7% of 1422 female worms in 637 patients before treatment to 17.5% of 1134 worms in 511 patients after 3 years treatment. Ivermectin at 400–800 μg/kg annually, or at 150 μg/kg or 400–800 μg/kg 3-monthly, over 3 years, did not increase the PN prevalence significantly, as compared with standard doses of 150 μg/kg annually. In other small series of African patients, PN prevalence increased in worms 2, 4, 6 and 10 months after ivermectin treatment; but there was no increase after treatment with amocarzine, albendazole or diethylcarbamazine and suramin. PN may partly account for the increased macrofilaricidal action of ivermectin on female O. volvulus in patients treated for 3 years at 3-monthly intervals.

Despite the fact that the non-human primates are our closest relatives and represent a species-rich mammalian group, little is known about their intestinal protistan parasites/commensals. Particularly, the intestinal trichomonads represent a neglected part of the fauna of the primate digestive system. We have established 30 trichomonad strains isolated from feces of 11 primate species kept in 3 Czech zoos and performed an analysis of their SSU rDNA and ITS1-5·8S rDNA-ITS2. Our results showed that intestinal trichomonads are rather common among non-human primates. Molecular phylogenetic analysis showed that the strains are unexpectedly diversified, belonging to 8 or 9 distinct species. Interestingly, the vast majority of the strains from non-human primates belonged to the genus Tetratrichomonas while no member of this genus has been found in the human intestine so far. In addition, hominoid and non-hominoid primates differed in their intestinal trichomonads. Our results suggest that captive primates possibly may be infected by intestinal trichomonads of other vertebrates such as pigs, cattle, birds, tortoises and lizards.

From a study of the organisms themselves, so far as this has been possible, and from an analysis of all accessible records relating to them, I conclude that there are four distinct species of coccidia which may parasitize man. These are: (1) Isospora hominis Rivolta, 1878 (emend.), discovered by Kjellberg in 1860. and recently investigated by Wenyon; (2) Eimeria wenyoni n.sp., a form discovered in 1915 by Wenyon; (3) Eimeria oxyspora n.sp., another new form, here described for the first time; (4) an undetermined species of Eimeria (?) which was discovered by Gubler in 1858. This last inhabits the human liver, whilst the three others probably live in the small intestine. Probably some seventy cases of infection with the Isospora have now been seen, mostly in persons who have been in the Near East; but the other parasites appear to be extremely rare, and little is known concerning their probable geographical distribution.

All these parasites are probably peculiar to man. There is no evidence that any of them is or can be parasitic in any other host. The prevailing belief that the coccidia of man are identical with those of rabbits, cats, or dogs, is therefore unfounded. Furthermore, there is as yet no good evidence to show that man may harbour any species of coccidia other than the four just enumerated. All these forms, however, require fuller investigation. They are here briefly and incompletely described from the data at present available.

There is at present no proof that the coccidia of man—with the probable exception of the species occurring in the liver—can produce a clinically recognizable pathological condition of “coccidiosis”: and as yet no method of treatment which will eradicate an infection with any species has been discovered.

We investigated the prevalence, morphological characters and molecular classifications of trematode cercariae in freshwater snails randomly collected from 59 sampling localities in Bangkok from May 2018 to March 2019. We used a crushing technique to observe the cercarial stage inside each snail body and amplified the internal transcribed spacer 2 regions of cercarial DNA using polymerase chain reaction methodology. The associated phylogenetic tree was reconstructed using Bayesian inference analyses. A total of 517 of 15 621 examined snails were infected with trematode cercariae, and the infected snails were classified into 11 species of seven families with a 3.31% overall prevalence of the infection. The Bithynia siamensis siamensis snail displayed the highest prevalence of infection (16.16%), whereas the Physella acuta snail exhibited the lowest prevalence (0.08%) of infection. Eight morphological types of cercariae were observed. The highest prevalence of infection was observed in mutabile cercaria (1.86%). Based on molecular investigations, the phylogram revealed eight cercaria types assigned to at least nine digenean trematode families, of which five belong to groups of human intestinal flukes. Although, with the exception of schistosome cercaria, trematode cercariae are not known to directly damage humans, understanding the general biology of trematode cercariae (including diversity, distribution, infection rates and host range) is important and necessary for the prevention and control of parasitic transmission that impacts aquatic cultivations, livestock farming and human health.

We established experimental metapopulations of the flour beetle, Tribolium castaneum, and its ectoparasitic mite, Acarophenax tribolii, to investigate the effects of host migration rate and local host population size on the spread of mite infections. Global prevalence across our metapopulations was less than half the observed within-patch prevalence, so that spatial structure alone afforded a great deal of protection to hosts against parasite infection. Our results showed further that migration played a determining role in occupancy, the number of patches infected within a metapopulation, while host population size played a determining role in local prevalence, the fraction of hosts infected within local patches. Local and global prevalence appeared to reach equilibrium levels on 2 different time-scales. Local host prevalence reached equilibrium values within 30 days of receiving an infected host migrant. Global prevalence increased more slowly and was clearly dependent upon occupancy, the number of host patches with at least 1 infected host, which in turn depended on the level of host migration among host patches. The effect of population size was not limited to local prevalence in patches without spatial structure but extended to sets of patches across the metapopulation. Lloyd's index of patchiness differed significantly between metapopulations with small versus large numbers of hosts. Although parasites were aggregated on hosts for both local patch sizes, they tended to aggregate to a much greater degree at the smaller host patch size. We discuss our empirical findings in light of current epidemiological theory.

In this study, using a combined data set of SSU rDNA and gGAPDH gene sequences, we provide phylogenetic evidence that supports clustering of crocodilian trypanosomes from the Brazilian Caiman yacare (Alligatoridae) and Trypanosoma grayi, a species that circulates between African crocodiles (Crocodilydae) and tsetse flies. In a survey of trypanosomes in Caiman yacare from the Brazilian Pantanal, the prevalence of trypanosome infection was 35% as determined by microhaematocrit and haemoculture, and 9 cultures were obtained. The morphology of trypomastigotes from caiman blood and tissue imprints was compared with those described for other crocodilian trypanosomes. Differences in morphology and growth behaviour of caiman trypanosomes were corroborated by molecular polymorphism that revealed 2 genotypes. Eight isolates were ascribed to genotype Cay01 and 1 to genotype Cay02. Phylogenetic inferences based on concatenated SSU rDNA and gGAPDH sequences showed that caiman isolates are closely related to T. grayi, constituting a well-supported monophyletic assemblage (clade T. grayi). Divergence time estimates based on clade composition, and biogeographical and geological events were used to discuss the relationships between the evolutionary histories of crocodilian trypanosomes and their hosts.

The surface proteins and glycoproteins of red cells from Plasmodium berghei-infected blood have been radio-isotope labelled and compared with those of normal mouse erythrocytes using the following protein labelling probes: lactoperoxidase-catalysed radio-iodination of tyrosyl residues, periodate oxidation and NaB3H4 reduction of sialic acid and oxidation of galactosyl/N-acetylgalactosaminyl residues by galactose oxidase with subsequent NaB3H4 reduction. During P. berghei infection, new tyrosyl-labelled proteins with apparent molecular weights (Mr) of 60000, 54000, 40000 and 27500 appeared on the surface of most, if not all, red cells in the blood. Purified multinucleate cells (mostly reticulocytes) differed only in that they also had a surface protein with Mr of 83000. However, this molecule is thought to be specific to mouse reticulocytes rather than derived from parasites. In contrast to the relatively minor changes detected with radio-iodination, striking changes in glycoprotein radio-isotope labelling resulted from infection. All of the red cells in infected blood of greater than 20% parasitaemia lost their periodate-sensitive glycoprotein sialic acid. With some samples there was little change in glycoprotein labelling by the galactose oxidase method, provided neuraminidase was also added. Modification of the exocyclic hydroxyls of sialic acid is postulated to account for this. Other blood samples exhibited a dramatic loss of galactose oxidase-dependent labelling. It is suggested that these observations may relate to the excessive red cell destruction of uninfected as well as infected cells which has been inferred in many haemosporidial infections, including malaria.

A morphological study of the fine structure of the spores of Minchinia chitonis, a haplosporidian parasite of the chiton, Lepidochitona cinereus, is described. The spores contained a single nucleus, mitochondria, haplosporosomes, smooth endoplasmic reticulum, ribosomes and a large spherule (presumed Golgi apparatus). The spore wall was discontinuous at the spherule end, forming an opening covered by a lid which rested on a circumscribed flange. The flange of the spore wall and the lid were continuous in only one area which served as a hinge. The entire spore was encapsulated by epispore cytoplasm bounded by a strengthened membrane and extended to form 2 long projections, one at either end.

The effective use of biological tags in stock assessment relies on the reliable identification of the parasites concerned. This may be compromised if cryptic species are not recognized. Here we review what is known about cryptic species in aquatic hosts and its potential importance in this respect. Although strictly cryptic species may be considered as species which can be distinguished only by molecular data, we accept the far looser but more practical definition of species that cannot be readily distinguished morphologically. Cryptic species appear to have been identified most frequently as occurring in separate host species; this is heartening in that this has no significant impact on tagging studies. But cryptic species have occasionally been identified in single hosts sympatrically and are relatively common in geographically distinct populations of the same host species. Ignorance of both kinds of occurrences has the capacity to undermine the reliability of tagging analysis. We review in detail what is known of intra- and interspecific genetic variation over geographical ranges in the trematodes, based on recent molecular studies. Although the existence of cryptic species and evidence of intraspecific variability may appear daunting, we suspect that these complexities will add, and indeed have already added, to the sophistication of the information that can be derived from tagging studies.

Angiostrongylus vasorum is a cardiovascular nematode increasingly found in dogs and foxes in endemic foci throughout Europe. The present study evaluates ELISAs for detection of circulating antigens and specific antibodies against A. vasorum in foxes. Blood and worm burdens (WBs) from carcasses of 215 Swiss wild red foxes (Vulpes vulpes) and from 75 farmed foxes of different age groups experimentally inoculated once or repeatedly with infective doses of 50, 100 or 200 third-stage larvae were obtained. Antigen detection in the naturally infected Swiss foxes had 91·2% sensitivity and 89·4% specificity, whereas the corresponding figures for antibody detection were 42·2 and 92·0%. The experimentally infected foxes became positive for circulating antigens 5–10 weeks post-inoculation (wpi) and remained highly positive up to 22 wpi, irrespectively of further challenge inoculation. The antibody responses in the same foxes were highly variable: high optical density (OD) values were reached 5–7 wpi in all animals, followed by a decrease in over half of the animals despite accumulating and consequently high WBs resulting in persistent infections. After each challenge, a slight increase of OD values was observed 7 weeks later. We hypothesize that infected foxes develop a variable and non-protective immunity. Such parasite tolerance allows long-term survival of A. vasorum in the animals, and may explain why the parasite appears to spread rapidly within a fox population, an epidemiological dynamic that is evident in many parts of Europe where A. vasorum has been found over the last decades.

Parasites that primarily infect white-tailed deer (Odocoileus virginianus), such as liver flukes (Fascioloides magna) and meningeal worm (Parelaphostrongylus tenuis), can cause morbidity and mortality when incidentally infecting moose (Alces alces). Ecological factors are expected to influence spatial variation in infection risk by affecting the survival of free-living life stages outside the host and the abundance of intermediate gastropod hosts. Here, we investigate how ecology influenced the fine-scale distribution of these parasites in deer in Voyageurs National Park, Minnesota. Deer pellet groups (N = 295) were sampled for the presence of P. tenuis larvae and F. magna eggs. We found that deer were significantly more likely to be infected with P. tenuis in habitats with less upland deciduous forest and more upland mixed conifer forest and shrub, a pattern that mirrored microhabitat differences in gastropod abundances. Deer were also more likely to be infected with F. magna in areas with more marshland, specifically rooted-floating aquatic marshes (RFAMs). The environment played a larger role than deer density in determining spatial patterns of infection for both parasites, highlighting the importance of considering ecological factors on all stages of a parasite's life cycle in order to understand its occurrence within the definitive host.

1. Certain flocks in the Aberystwyth Area having suffered considerably from Liver Rot, a Survey of the Agricultural Zoology was carried out between October, 1913, and October, 1916, special attention being given to the above disease and the bionomics of the host snail L. truncatula.

2. The Area comprised some 250 sq. miles of North Cardiganshire, etc., including Plynlymon, and falls into six natural divisions.

3. The presence of the disease has caused a number of local modifications in farm practice, designed to prevent, or mitigate losses. The disease was found to be on the wane within the Area, though endemic in certain places.

4. L. truncatula and L. peregra are the only abundant local species of the Genus. Considerable differences occur in the distribution of these two species. L. peregra was only studied in order to better understand the habits of L. truncatula. This latter species occurs in shallow water floored with Diatomaceous clayey mud, and is absent from soft mud, and peat. There is a marked coincidence between the distribution of clay and L. truncatula, and L. truncatula and Liver Rot.

5. The growth of the snails takes place throughout the year unless interrupted by drought or severe frost. Ova are deposited in March–April and hatch in May. The resulting snails, when they become adults, measure about 5 mm. in length. They in turn deposit ova during June–July. Drought usually limits further progress until the late Autumn, the snail being often very scarce during August–September.

6. Desiccation speedily kills L. truncatula. Local environmental conditions—rainfall, aspect, soil, vegetation, etc.—play a very important part in respect to its survival. Rainfall and its distribution through the year is the most important factor governing the increase, spread, and infectivity of the snail.

7. The ova masses dry rapidly and then resemble a hard inconspicuous scale. They resume their former size and shape when moisture returns, and many appear to be able to hatch subsequently, though the amount of desiccation they will withstand is not yet determined. In spite of prolonged droughts, L. truncatula re-appears, unless ditches, etc., have been cleaned out.

8. Roadside ditches are very frequently inhabited by infected snails and the drainage from them may be the cause of cases of rot.

9. Cercariae of D. hepaticum were obtained from L. truncatula in large numbers practically throughout the year; but not from snails measuring less than 4·5 mm. in length. Seventy cercariae were given off naturally, within 48 hours, from one specimen measuring 7 mm. in length. Heavily parasitized snails are adversely affected. A few cercariae, resembling those of D. hepaticum were obtained once from L. peregra.

Trials were carried out to study the humoral immune response of blackflies to filariae following infection using the intrathoracic injection technique. An induced 66 kDa protein was abundant in the haemolymph of the European species Simulium ornatum following infection with bovine Onchocerca lienalis. This protein was apparently at higher concentrations in the haemolymph of sham-inoculated flies, i.e. flies that received sterile medium without the parasites. A molecule of the same size was also observed in the haemolymph of infected S. damnosum s.l. following infection with human O. volvulus or bovine O. ochengi. However, the level of this protein was lower in blackflies injected with microfilariae of bovine O. dukei. Unlike O. volvulus and O. ochengi this species is not transmitted by S. damnosum s.l. under natural conditions. No such reaction was observed if the African blackflies had received a sham inoculation. Feeding experiments with wild-caught nulliparous S. damnosum sl. on Onchocerca-infected cattle supported the results of the injection trials. The 66 kDa protein could only be found in the haemolymph of specimens infected via a blood meal. This 66 kDa molecule was identified as phenoloxidase. It appeared in the haemolymph due to the activation of the prophenoloxidase system following the filarial infection and we hypothesize that it may be sequestered by the parasites, as part of a non-self recognition system.

The development of a new species, Bacillidium vesiculoformis n. sp. (Microspora, Mrazekiidae), is described from the freshwater oligochaete Nais simplex (Oligochaeta, Naididae). Initial stages of parasite development consist of a monokaryotic merogony within a haemocyte of the intestinal blood sinus. The resulting hypertrophied haemocyte is attached to the chloragocytes of the sinus by fine cytoplasmic extensions with the sinus around the cell becoming greatly enlarged. The meronts within the haemocyte form diplokaryotic sporonts that undergo sporogenesis directly within the cytoplasm of the host cell. The infected cell becomes packed with spores and developmental stages, causing it dramatically to increase in size, eventually rupturing the oligochaete and cell. Sporogony appears to be disporoblastic. Released spores were observed to have an adhesive quality. Transmission studies conducted with mature spores failed to transmit the parasite horizontally although vertical transmission was observed. Phylogenetic analysis of the parasite demonstrated that B. vesiculoformis clustered with microsporidian parasites of bryozoa and two other microsporidians, Janacekia debaiseuxi and an unidentified Bacillidium sp.

Experiments were carried out to show the different hygroscopic properties of certain foods supplied to flea larvae. These show that larvae of Ceratophyllus wickhami require about 15 to 28 per cent, of water in their food. Dried blood, albumen (blood) and material from the bottom of a squirrel's nest were used.

Newly hatched flea larvae were fed on different substances at varying humidities and a constant temperature. A relative humidity of 80 per cent. gave the best results.

Dried blood is a convenient food for breeding flea larvae in the laboratory, and may be used with or without a mixing medium, such as sand. Albumen is a more simple food on which a few larvae will survive, but at lower humdities than dried blood.

Flea larvae will eat a great variety of other foods, including squirrel faeces, ground-up cockroach and dead adult fleas, but these do not provide a complete diet.

It does not seem necessary for the larvae to have dried blood from the host of their parents, rabbit or human blood being quite as satisfactory.

Due to the low informative value of available morphological characters, cytogenetic and molecular methods, based on rDNA sequencing, were used to characterize adult and larval stages of Phyllodistomum spp. Species studied have 18 chromosomes with comparable absolute and relative lengths. Conventional Giemsa staining and karyometric analysis revealed clear differences in chromosome morphology of larval Phyllodistomum spp. infecting two bivalve host species, Sphaerium corneum and Pisidium amnicum. However, karyotypes of adult P. folium from three-spined sticklebacks and larval stages from S. corneum appear almost identical both with respect to the relative lengths and centromeric indices of the corresponding chromosome pairs. The entire internal transcribed spacer (ITS) region (ITS-1, 5.8S and ITS-2) and the D1-D3 region of 28S gene were sequenced and compared. Again, sufficient differences were observed between larval Phyllodistomum spp., while adult P. folium and larvae from S. corneum showed a high level of similarity. So, both cytogenetic and molecular data support the suggestion that they represent developmental stages of the same species. The results were compared with published data obtained by cytogenetic and molecular studies on the other Phyllodistomum species. Differences revealed in karyotype and rDNA sequences leads to the conclusion that the cercariaeum of P. folium sensu Sinitsin, 1905 could not be regarded as the larva of adult P. folium from three-spined stickleback.

In the course of the examination of the contents of the paunch and rumen of 70 Ruminants a Ciliate of an interesting type has been met with on eight occasions. It is surprising that a new form should be found in this situation as a number of workers seem to have devoted no little time to researches on the fauna of the ruminant stomach. One cannot think that this animal is to be found only in England, and we are driven to the conclusion that it must have been overlooked up to the present. As the organism is relatively small for a Ciliate and of a rather transparent appearance this is understandable, especially because it is rarely abundantly present. Although it has been found eight times, yet in a coverslip preparation where there will be several specimens of the other Ciliates in every field of the microscope, there may not be more than one or two specimens of this new form.

Primers and DNA probes designed for use in the specific detection of the paramyxean parasites Marteilia sydneyi and Marteilia refringens were tested for their potential to cross-react with closely related species in Polymerase Chain Reaction (PCR) and in situ hybridization. PCR primers and a DNA probe designed within the ITS1 rRNA of M. sydneyi were specific for M. sydneyi when compared with related species of Marteilia and Marteilioides. PCR primers designed within the 18S rRNA of M. refringens were specific in the detection of this species in PCR while a DNA probe (named Smart 2) designed on the same gene cross-reacted with M. sydneyi in tissue sections of Saccostrea glomerata as well as Marteilioides sp. infecting Striostrea mytiloides. Though not species specific, the Smart 2 probe provided a stronger signal in detection of all stages of M. sydneyi than the ITS1 probe. The ITS probe is proposed for use as a confirmatory diagnostic tool for M. sydneyi.

This paper investigated the intestinal distribution and fecundity of 2 species of Diplostomum parasites, D. spathaceum and D. pseudospathaceum, in 2 species of definitive hosts, herring gull (Larus argentatus) and common gull (L. canus), using both empirical field data and experimental infections. At the level of individual hosts, the parasite species occupied different parts within the intestine, but the fecundity of the worms, measured as the number of eggs in the uterus, did not differ between the parasite species except in wild common gulls. Interestingly, egg numbers in individual hosts were positively correlated between the parasite species suggesting that some birds provided better resources for the parasite species. At the host population level, fecundity of the worms did not differ between the host species or between adult birds and chicks. Both parasite species were also aggregated to the same host individuals and it is likely that aggregation is transferred to gulls from fish intermediate hosts. Individual differences in suitability and parasite numbers between hosts provide important grounds and implications for epidemiological model-based parasite prevention strategies.
