Freshly excysted Fasciola hepatica possess an outer glycocalyx which stimulates an antibody response in cattle infected with F. hepatica metacercariae. Sera from animals receiving either a single or a double infection were examined for levels of IgM, IgG1 and IgG2 specific for the outer glycocalyx of intact live juvenile F. hepatica. High levels of specific IgG1 were found to predominate. Using positive sera in an indirect fluorescent antibody assay, the bovine immunoglobulins labelled the outer glycocalyx in a characteristic reticulated pattern. On incubation at 37 °C this pattern rapidly broke up into discrete clumps and progressed until the entire antibody–outer glycocalyx complex was shed. The shedding phenomenon was shown to be continuous and, by using preparations of Fab fragments, was not due to the bivalent binding of the sensitizing antibody.

The behaviour of mice exposed to graded doses of Trichinella pseudospiralis larvae was monitored in an open-field situation. Ambulatory activity declined briefly during the acute phase of the infection (day 11) but regained pre-infection levels 1 week later. Animals receiving heavy doses of T. pseudospiralis exhibited a mild increase in the level of ambulatory activity during the chronic phase of the infection. The infections did not affect the level of exploratory activity. The effects of the T. pseudospiralis infections on the behaviour of the mouse host were much less pronounced than those induced by T. spiralis. It is suggested that these differences may be associated with the small size of T. pseudospiralis and the lack of capsule formation.

The cysticercoid type of larval development, involving the retraction of the scolex and the associated development of a cyst, is of widespread occurrence within cyclophyllidean cestodes, but previously its possible adaptive value has been unexplained. Scolex retraction in Hymenolepis microstoma is an active, muscular process which takes only about a minute to complete. Following its retraction the scolex develops adult structures. The cyst enclosing the scolex also differentiates further, involving the secretion of three layers of collagen fibres and the development of a myelin-like layer adjacent to the retracted scolex. The cells within the cyst, including the muscle, become condensed or are autolysed causing a decrease in thickness of the cyst and an increase in the proportion of fibrous tissue. The collagenous part of the cyst provides mechanical protection for the scolex during grinding by the teeth on entry into the mouse host. The myelin-like layer insulates the scolex from the low pH conditions of the stomach. Values of lower than pH 3·0 are lethal to newly excysted worms. The myelin-like layer loses its insulatory capacity following exposure to bile salts.

Five species of Glossina (G. morsitans, G. pallidipes, G. austeni, G. brevipalpis and G. palpalis) have ovaries containing two polytrophic ovarioles, each of which contains a single egg follicle. In all these species the right ovary is more highly developed than the left at emergence. This type of ovary is probably common to all species of Glossina.

The development of the egg follicles in G. morsitans pupae is described. It is shown that the cycle of development whereby eggs are produced alternately from right and left ovaries is established during pupal development and that some yolk deposition has occurred in follicle A (right ovary) and follicle C (left ovary) before the fly emerges from the puparium.

In G. morsitans, it is shown that the size of the egg follicles in newly emerged flies is correlated with the size (R.D.W.) of the fly, larger females having more yolk laid down in their follicles before emergence. This is probably correlated with the state of reserves in the fat body. It is also shown that there is a seasonal variation in follicle size in newly emerged flies, this variation is probably correlated with variations in temperature acting upon wild females during pregnancy and upon the pupae, as temperature is known to affect both size and fat content of wild tsetse flies.

Despite the utility of RNAi for defining gene function in Caenorhabditis elegans and early successes reported in parasitic nematodes, RNAi has proven to be stubbornly inconsistent or ineffective in the animal parasitic nematodes examined to date. Here, we summarise some of our experiences with RNAi in parasitic nematodes affecting animals and discuss the available data in the context of our own unpublished work, taking account of mode of delivery, larval activation, site of gene transcription and the presence/absence of essential RNAi pathway genes as defined by comparisons to C. elegans. We discuss future directions briefly including the evaluation of nanoparticles as a means to enhance delivery of interfering RNA to the target worm tissue.

The free radical nitric oxide (NO), which is synthesized by nitric oxide synthase (NOS), has recently been discovered to function as a neuronal messenger. The presence of NOS was detected in the nervous system of adult Hymenolepis diminuta with NADPH-diaphorase (NADPH-d) histochemistry. The NADPH-d histochemical reaction is regarded as a selective marker for NOS in neuronal tissue. NADPH-d staining was observed in nerve fibres in the main and minor nerve cords and the transverse ring commissures, and in cell bodies in the brain commissure, along the main nerve cords, in the suckers and the rostellar sac. NADPH-d staining was also observed in the wall of the internal seminal vesicle and the genital atrium. The pattern of NADPH-d staining was compared with that of the 5-HT immunoreactive nervous elements. The NADPH-d staining reaction and the 5-HT immunoreactivity occur in separate sets of neurons. This is the first time the NADPH-d reaction has been demonstrated in the nervous system of a flatworm, indicating that NOS is present and that NO can be produced at this level of evolution.

The egg shell of Fasciola was investigated by electron microscopy and found to consist of a fine reticulum of fibrils. No evidence for concentric lamellae was discovered.

The amino acids of the shell were identified using chromatography and it was suggested that the original protein was of a simple fibrous type. An unidentified spot was thought to represent a hydrolysis product of the cross-linkage. The effect of sodium hypochlorite on the shell, was also studied.

The ‘vitelline membrane’ of the fully developed egg was found to consist of a layer of peri-vitelline material beneath which were two closely opposed unit membranes. The origin of these layers was discussed.

The permeability of the shell and vitelline membrane was studied using simple physical techniques and radiotracers. The shell was found to be freely permeable to small molecules. The vitelline membrane complex was thought to represent the main barrier to permeability but an uptake of phosphate ions amounting to 8% of total phosphate, was recorded. This may have been more apparent than real as most of the activity was probably associated with egg shells rather than contents.

The somatic muscle cells of the parasitic nematode Ascaris suum possess GABA receptors that gate chloride conductances in a similar fashion to the mammalian GABAA receptor subtype. These receptors mediate muscle relaxation and are the site of action of the anthelmintic piperazine. The properties of this receptor differ from the properties of the GABA-gated chloride receptors in the mammalian host, in particular they are not as sensitive to mammalian GABA receptor antagonists such as bicuculline and picrotoxin. Using two-electrode intracellular electrophysiological recording techniques from Ascaris muscle cells, we have tested the potency of a series of azole derivatives for their ability to block the chloride-dependent GABA response. The lead compound, SN606078, 2-(2,6-dichloro-4-trifluromethylphenyl)-4-(4,5-dicyano-lH-imidazol-2-yl)-2H-l,2,3-triazoIe, and 4 structurally related compounds reversibly blocked the conductance increase elicited by 30 μM GABA with IC50s of less than 10 μM. SN606078 (10 μM) decreased the slope of the dose-response curve for GABA, suggesting a non-competitive mechanism of action. In two-electrode voltage clamp experiments, 10μM SN606078 blocked the outward current elicited by 20 μM GABA in a voltage-dependent manner with 72 ± 2% inhibition at −20 mV and 49±6% inhibition at −40 mV. These observations indicate that SN606078 may act as an open-channel blocker of the GABA-gated chloride channel in A. suum.

Diagnostic PCR primers for Trichinella were constructed. Twelve pairs of primers were designed based on the sequences of random amplified polymorphic DNA, and 4 pairs of primers were designed based on the reported sequences of complementary DNA encoding excretory–secretory glycoproteins. With these primers, 31 samples of DNA from different strains of Trichinella including 5 species (Trichinella spiralis, Trichinella britovi, Trichinella nativa, Trichinella nelsoni and Trichinella pseudospiralis) and 3 phenotypes of uncertain taxonomic level (Trichinella T5, T6 and T8) were tested with PCR. Genus Trichinella can be identified by 4 different primer pairs (SB147D, SB372A, SB153, or Ts43). Trichinella spiralis can be identified by the presence of a 673 bp amplicon in PCR with the primer pair SB147B. Trichinella nelsoni can be identified using primer pair SB147F or by the presence of 673 bp and ca. 380 bp amplicon in PCR with the primer pair SB147B. Trichinella pseudospiralis can be identified by 2 primer pairs (SB147E or SB372B). Trichinella T5 can be identified by the primer pair SB147G. Trichinella T8 can be identified by its positivity by the primer pair SB147C and its negativity by the primer pair SB372C. A group of Trichinella species (T. britovi, T. nativa and Trichinella T6) can be identified by the primer pair SB372C.

During the last few decades, a considerable amount of experimental works has been carried out on the relations of insects to their environment: in certain groups such studies have advanced to an extent which renders possible tentative efforts at accurate prediction of their activities and spread. Among the arachnids, however, the experimental study of ecological relationships has been almost entirely neglected, and this despite the fact that several species, especially among the mites and ticks, are of marked economic significance. Since 1892–3, when the researches of Smith and Kilborne on piroplasmosis in cattle were published, ticks have been proved to be vectors of numerous important diseases of man and animals in many parts of the world, and much knowledge of their bionomics has been gained through observations conducted in the field and in the laboratory. The ecology of ticks under precisely controlled conditions, however, provides a practically unexplored field.

The in vivo expression and distribution of the dense granule protein GRA7 was examined in both the exoenteric (tachyzoite and bradyzoite) and enteric (coccidian) forms of Toxoplasma gondii by immunocytochemistry. There was strong staining of GRA7 in granules within all the infectious stages (tachyzoite, bradyzoite, merozoite and sporozoite). During tachyzoite development, GRA7 was secreted and was associated with the parasitophorous vacuole. In contrast, although there was staining of granules within the bradyzoites of more mature cysts, there appeared to be little staining of the tissue cyst wall or host cell. The apparent stage-specific variation in secretion of GRA7 between tachyzoites and bradyzoites was confirmed by double labelling using stage-specific markers (SAG1 and BAG1). In the enteric forms in the cat gut there was strong labelling of the PV containing early asexual and sexual stages and staining of a few granules in the apical cytoplasm of the merozoite. The positive enteric staining pattern differentiates GRA7 from the other GRA proteins (GRA1–6) which were absent in the merozoites and enteric stages. The staining pattern of GRA7 with strong staining during tachyzoite and enteric development and reduced staining in the tissue cysts is similar to that seen for NTPases. The function of GRA7 is unknown but it is unique among the dense granule proteins in being expressed in all the infectious forms of T. gondii which would point to a basic role in the vacuolar adaptations required for active parasite development.

Faecal samples from 163 captive and semi-captive individuals, 61 samples from wild individuals and 38 samples from captive groups of Bornean orangutans (Pongo pygmaeus) in Kalimantan, Indonesia, were collected during one rainy season (November 2005–May 2006) and screened for intestinal parasites using sodium acetate-acetic acid-formalin-concentration (SAFC), sedimentation, flotation, McMaster- and Baermann techniques. We aimed to identify factors influencing infection risk for specific intestinal parasites in wild orangutans and individuals living in captivity. Various genera of Protozoa (including Entamoeba, Endolimax, Iodamoeba, Balantidium, Giardia and Blastocystis), nematodes (such as Strongyloides, Trichuris, Ascaris, Enterobius, Trichostrongylus and hookworms) and one trematode (a dicrocoeliid) were identified. For the first time, the cestode Hymenolepis was detected in orangutans. Highest prevalences were found for Strongyloides (individuals 37%; groups 58%), hookworms (41%; 58%), Balantidium (40%; 61%), Entamoeba coli (29%; 53%) and a trichostrongylid (13%; 32%). In re-introduction centres, infants were at higher risk of infection with Strongyloides than adults. Infection risk for hookworms was significantly higher in wild males compared with females. In groups, the centres themselves had a significant influence on the infection risk for Balantidium. Ranging patterns of wild orangutans, overcrowding in captivity and a shift of age composition in favour of immatures seemed to be the most likely factors leading to these results.

The parasitic nematode Strongyloides stercoralis, has several alternative developmental pathways. Upon exiting the host (humans, other primates and dogs) in faeces, 1st-stage larvae (L1) can enter the direct pathway, in which they moult twice to reach the infective 3rd-stage. Alternatively, if they enter the indirect pathway, they moult 4 times and become free-living adults. The choice of route depends, in part, on environmental cues. In this investigation it was shown that at temperatures below 34 °C the larvae enter the indirect pathway and develop to free-living adulthood. Conversely, at temperatures approaching body temperature (34 °C and above), that are unfavorable for the survival of free-living stages, larvae develop directly to infectivity. The time-period within the L1's development during which temperature influenced the choice of the pathway depended on the temperature, but, at any given temperature, occurred approximately in the middle of the time-span spent in the L1 stage, which varied inversely with temperature. This critical period was associated with the time-interval in which the number of cells in the genital primordium began to increase, thus providing a morphological marker for the pathway decision in individual worms. Sensing the environment is the function of the amphidial neurons, and therefore we examined the role of individual amphidial neurons in controlling entry into the direct pathway to infectivity. The temperature-sensitive developmental switch is controlled by the neuron pair ALD (which also controls thermotaxis), as seen by the loss of control when these neurons are ablated. Thus, in S. stercoralis a single amphidial neuron pair controls both developmental and behavioural functions.

Balamuthia mandrillaris, a soil amoeba, is the causative agent of Balamuthia granulomatous amoebic encephalitis, a life-threatening brain infection. This amoeba is acquired from contaminated soil and may enter the host through cutaneous lesions or through nasal passages, migrating to the lungs or brain. During invasion, B. mandrillaris has access to components of the extracellular matrix (ECM) of the host. Therefore, we investigated the interaction of B. mandrillaris with 3 ECM glycoproteins (collagen-I, fibronectin and laminin-1) that are encountered in host connective tissues and at the basal lamina. Using optical microscopy, amoeba association on ECM-coated surfaces was examined. Binding of amoebae on laminin was greater than that on collagen or fibronectin. Laminin-adhered B. mandrillaris exhibited elongated and spread forms, distinctive from those observed for amoebae on a plastic surface. Collagen and fibronectin-adhered B. mandrillaris presented elongated shapes with cellular expansions. Binding to collagen, fibronectin, or laminin was inhibited when amoebae were pre-treated with sialic acid. Treatment with galactose resulted in diminished binding of amoebae on laminin, while mannose increased binding in all coating conditions tested. Dependence of divalent cations on amoeba binding was demonstrated for laminin-amoeba interaction. Collectively, the results indicate that B. mandrillaris recognizes specific glycoproteins of the mammalian extracellular matrix.

Entomopathogenic nematodes of the genus Heterorhabditis are morphologically conservative, consequently the majority of isolates remain unassigned at the species level. When a Heterorhabditis infective juvenile infects an insect host, it develops into a protandrous hermaphrodite female. These first-generation females give rise to a second generation which contains both males and females. Because of this complex life-cycle and also because of uncertainty as to whether second-generation females are amphimictic, cross-breeding studies to facilitate species determination have not been carried out previously. We demonstrate here that second-generation Heterorhabditis females are amphimictic. Because of this finding, we have been able to develop a successful cross-breeding technique for the purposes of species determination in Heterorhabditis. Interstrain crosses using second-generation males and females from the appropriate strains have been successfully set up in Xenorhabdus luminescens-treated G. mellonella cadavers and also on agar plates. Using the techniques described here we confirm that H. bacteriophora (Brecon strain), H. megidis and H. zealandica are distinct biological species, we note that the H. bacteriophora group contains at least 2 species and we provide evidence for the existence of a new Irish species of Heterorhabditis.

Passive transfer experiments showed that 76% of the resistance induced in CBA/Ca mice by exposure to radiation-attenuated cercariae of Schistosoma mansoni could be transferred to naive recipients by administration of donor serum. The level of protection achieved depended on the volume of serum administered and immunity was demonstrated most consistently with serum harvested from thrice vaccinated donors. The serum was twice as effective when given to the recipients at the time of cercarial challenge as compared to administration 5 days after challenge, a result which indicates that serum-dependent challenge elimination is probably accomplished in the cutaneous tissues. This view was confirmed by the observation that passive protection could be ablated by administration of a monoclonal antibody which we have shown elsewhere to deplete the cutaneous inflammatory reaction to cercarial penetration. Histopathological studies revealed that vaccine serum induced subdermal inflammatory reactions in challenged recipient mice which were identical both in induration and kinetics to those seen in conventionally vaccinated individuals; on days 4 and 5 post-challenge, the reactions comprised 60% mononuclear cells and 40% eosinophils. Challenge larvae, which had transformed from skin-stage to lung-stage parasites, became trapped within such reactions and eventually showed generalized vacuolation consistent with the onset of damage. Some foci were seen to contain degenerated leucocytes, free eosinophil granules and debris which is thought to represent the remnants of dead parasites. Small focal reactions were identified on occasion in naive challenged mice and in recipients of normal mouse serum, but these reactions comprised predominantly mononuclear cells and were rarely seen to encompass challenge parasites. These data show that serum-transferred resistance in the vaccinated CBA/Ca mouse model involves the induction of a cutaneous inflammatory response in the recipients.

Theileria annulata (Ta)-infected leucocytes are able to disseminate in scid mice. The dose of virulent parasites of the Ta-Ode line required to achieve quantifiable dissemination was found to be 2×10 cells given i.p. Dissemination was higher on day 11 post-inoculation than on day 18. The attenuated Ta-Ode cells were found to disseminate very poorly compared to their virulent progenitors, which correlates with a marked reduction in matrix metalloproteinase (MMP) expression. A daily i.p. injection of mice with BB94, a synthetic inhibitor of MMPs, almost completely ablated dissemination compared to controls. This provides strong evidence that metastasis of Theileria annulata macroschizont-infected host cells is mediated by host MMPs induced by the parasite. This has important implications for explaining a number of pathological features of tropical theileriosis in cattle.

The infective larvae of S. ratti had a high Qo2 (9·8 μl/mg dry weight/h at 20 °C). Their R.Q. was 0·74 indicating that lipid was the main metabolite.

As the infective larvae aged there was an exponential decrease in their infectivity and a corresponding decrease with age in the Qo2 rate of activity and rate of substrate utilization. The R.Q. of fresh and aged larvae was the same showing that there was no change in the substrate being utilized.

The infective larvae of S. ratti contained 25% lipid 3% glycogen and 0·25% free sugars (dry weight). The free sugars consisted of glucose and trehalose in approximately equal amounts. Both lipid and glycogen decreased as the larvae aged.

Analyses showed all the normal lipid components to be present, the major lipid fraction detected was free fatty acid. As the larvae aged there was a big decrease in the amounts of triglyceride and free fatty acid. There was, however, no difference in the amounts of cholesterol or in the phospholipid composition of fresh and aged larvae.

The fatty-acid composition of the larvae was analysed; 68% of the total fatty acids were unsaturated and the major fatty acid was C18:1. As the larvae aged, the fatty acids were not all catabolized at the same rate; the acid C18:1 was the most heavily catabolized decreasing by 75% in 8 days, whilst C20:1 decreased by only half this amount in the same time.

The distribution of lipid and glycogen was investigated histochemically. Lipid droplets were confined to the posterior two-thirds of the infective larvae and occurred in the lateral lines, non-contractile parts of the muscles and in the cells of the intestine. As the larvae aged the lipid in the body wall and lateral lines disappeared first. Dark granules, possibly excretory products, accumulated in old larvae. Glycogen was found in the lateral lines throughout the length of the larva.

Transmission of parasites may be mediated by their habitat, consisting of abiotic and biotic components. I investigated the effect of 2 important habitat components in intertidal ecosystems, tidal level (abiotic) and density of upstream hosts (biotic), on the transmission of trematode cercariae to cockle (Cerastoderma edule) hosts. A field survey showed no general trend in metacercarial loads of cockles regarding tidal level but species-dependent reactions. Parasites originating from Littorina littorea (Himasthla elongata, Renicola roscovita) showed highest infection levels in the low intertidal while parasites originating from Hydrobia ulvae (H. continua, H. interrupta) showed highest infection levels in the mid-intertidal. This reflected the density of upstream hosts at both tidal levels and positive relationships between the density of upstream hosts and metacercarial load in cockles suggested the biotic habitat component to be the dominant factor in transmission. This was confirmed by a field experiment, manipulating tidal level and the density of infected upstream snail hosts. While tidal level had no significant effect on the number of metacercariae of H. elongata acquired by cockles, the effect of upstream host density was strong. In conclusion, although tidal level usually is a very important abiotic habitat component in intertidal ecosystems leading to conspicuous zonation patterns in free-living organisms, it seems of minor importance for trematode transmission. In contrast, the biotic component upstream host density is suggested to be the dominant predictor for trematode transmission to second intermediate hosts. Assessing the relative importance of abiotic and biotic habitat components in transmission is vital for the understanding of transmission processes in the field.

Two natural human interferon alpha preparations, (nHuIFN-μ [Cantell]) and (Nhuifn-μ [ISI]), were used for the oral treatment of cattle experimentally infected with Theileria parva parva. In the first experiment, 8 Friesian bulls were inoculated with a 1 in 10 dilution of a sporozoite stabilate of T. p. parva (Marikebuni) stock. Four of the cattle were treated daily with 1 international unit/kg body weight (i.u./kg bwt) of nHuIFN-μ (Cantell) from day –2 to day 8 p.i. None of the 4 calves given IFN developed clinical theileriosis, but 3 of the 4 control calves died of theileriosis while the fourth had a mild infection. Three of 4 treated calves and the 1 surviving control calf developed a detectable antibody response to T. p. parva schizont antigen but, on challenged with a 10-fold higher dose of stabilate, the surviving control animal and only 1 of the 4 treated calves proved to be immune. In a second experiment, 4 groups of 4 calves were inoculated with the same stabilate dilution. Three treatment groups were given either 1 i.u. nHuIFN-μ (Cantell), 1 i.u. nHuIFN-μ (ISI), or 10 i.u. nHuIFN-μ (ISI)/kg bwt from day –2 to day 8 p.i. once daily and the fourth group were controls. Clinical theileriosis occurred in 2 controls, 2 calves given 10 i.u. nHuINF-μ (ISI), 1 calf given 1 i.u. nHuIFN-μ (ISI) and no calves given 1 i.u. nHuIFN-μ (Cantell)/kg bwt. Of these, 2, 1, 0 and 0 cattle died in the respective groups. All the surviving cattle proved to be immune on homologous challenge with 10-fold higher dose of stabilate except the 2 cattle which did not develop high antibody responses. A third experiment using an undiluted challenge of T. p. parva (Muguga) sporozoite stabilate (10α) on 8 steers. Four steers were treated with 1 i.u. nHuIFN-α (Cantell)/kg bwt and 4 were controls. All calves developed acute theileriosis and the experiment was terminated. Cells of the C2 lymphoblastoid cell line, infected with T. p. parva (Muguga) schizonts, were cultured in vitro with various concentrations (0-01-100 i.u./ml) of nHuIFN-a (Cantell). The IFN appeared to have no effect on host cell or parasite developmental variables when compared to untreated control cultures.
