The development of the free-living stages of Ostertagia ostertagi incubated at constant temperatures from 5 to 27 °C and in dung pats in the field was observed. The death rates were adequately described by a single exponential function and the distribution of times for development by a delayed gamma density function. The times of appearance of selected development stages such as maximum numbers of embryonated eggs and half the maximum number of infective larvae, were highly dependent on temperature. The relationships between these times and temperature were adequately described by the Arrhenius equation. The parameters of this equation and mean hourly temperatures recorded in dung pats in the field were used to predict development times in the field. A comparison of predicted and observed times showed that an initial delay accounted for a large proportion of the time required for development in dung pats, particularly the embryonation of eggs. This delay was attributed to lack of aeration associated with high moisture content of the dung pat.

Taenia saginata cysticercosis causes financial losses to the beef industry and farmers, and represents a significant source for human infection in many countries. A case-control study was conducted to identify risk factors for bovine cysticercosis on farms in Switzerland. The case group (n=119) consisted of farms with infected cattle identified at slaughter in 2005 and 2006. Infections were confirmed by morphological or molecular diagnosis. The control group (n=66) comprised randomly selected farms with cattle slaughtered in the same period but with no evidence or history of infection. In personal structured interviews with the farmers, information regarding local surroundings and farm management was collected. Logistic regression revealed the following 5 factors as being positively associated with the occurrence of bovine cysticercosis: the presence of a railway line or a car park close to areas grazed by cattle, leisure activities around these areas, use of purchased roughage and organized public activities on farms attracting visitors. This information is considered useful for government authorities to direct control strategies as well as for farmers to take measures tailored to local situations.

Parasitism in wild mammals can vary according to myriad intrinsic and extrinsic factors, many of which vary seasonally. However, seasonal variation in parasitism is rarely studied using repeated samples from known individuals. Here we used a wild population of individually recognized red deer (Cervus elaphus) on the Isle of Rum to quantify seasonality and intrinsic factors affecting gastrointestinal helminth parasitism over the course of a year. We collected 1020 non-invasive faecal samples from 328 known individuals which we then analysed for propagules of three helminth taxa: strongyle nematodes, the common liver fluke Fasciola hepatica and the tissue nematode Elaphostrongylus cervi. Zero-inflated Poisson models were used to investigate how season, age and sex were associated with parasite prevalence and count intensity, while Poisson models were used to quantify individual repeatability within and between sampling seasons. Parasite intensity and prevalence varied according to all investigated factors, with opposing seasonality, age profiles and sex biases between parasite taxa. Repeatability was moderate, decreased between seasons and varied between parasites; both F. hepatica and E. cervi showed significant between-season repeatability, while strongyle nematode counts were only repeatable within-season and showed no repeatability within individuals across the year.

This review presents a progression strategy for the discovery of new anti-parasitic drugs that uses in vitro susceptibility, time-kill and reversibility measures to define the therapeutically relevant exposure required in target tissues of animal infection models. The strategy is exemplified by the discovery of SCYX-7158 as a potential oral treatment for stage 2 (CNS) Human African Trypanosomiasis (HAT). A critique of current treatments for stage 2 HAT is included to provide context for the challenges of achieving target tissue disposition and the need for establishing pharmacokinetic–pharmacodynamic (PK–PD) measures early in the discovery paradigm. The strategy comprises 3 stages. Initially, compounds demonstrating promising in vitro activity and selectivity for the target organism over mammalian cells are advanced to in vitro metabolic stability, barrier permeability and tissue binding assays to establish that they will likely achieve and maintain therapeutic concentrations during in-life efficacy studies. Secondly, in vitro time-kill and reversibility kinetics are employed to correlate exposure (based on unbound concentrations) with in vitro activity, and to identify pharmacodynamic measures that would best predict efficacy. Lastly, this information is used to design dosing regimens for pivotal pharmacokinetic–pharmacodyamic studies in animal infection models.

In two recent papersm Sambon and Seligmann (1907, 1908) have recorded some observations on the intracellular parasites of snakes, and have described no less than ten new species. The authors, in discussing the life histories of these parasites, have made the startling discovery that, “the life history of the haemogregarines like that of the haemoprotozoa is divided into two cycles: a schizogonic or ‘vegetative’ cycle spent in the blood of vertebrates and characterised by asexual multiplication, and a sporogonic or sexual cycle spent in the digestive organs of blood-sucking invertebrates and characterised by sexual reproduction.” The authors then go on to speak quite familiarly of young merozoites, adult schizonts, adult sporonts, and so on.

Haemonchus contortus is one of the most pathogenic parasites of small ruminants (e.g. sheep and goat). The treatment of haemonchosis is complicated because of recurrent resistance of H. contortus to common anthelmintics. The aim of this study was to compare the metabolism of the anthelmintic drug flubendazole (FLU) and the activities of selected biotransformation enzymes towards model xenobiotics in 4 different strains of H. contortus: the ISE strain (susceptible to common anthelmintics), ISE-S (resistant to ivermectin), the BR strain (resistant to benzimidazole anthelmintics) and the WR strain (resistant to all common anthelmintics). H. contortus adults were collected from the abomasums from experimentally infected lambs. The in vitro as well as ex vivo experiments were performed and analysed using HPLC with spectrofluorimetric and mass-spectrometric detection. In all H. contortus strains, 4 different FLU metabolites were detected: FLU with a reduced carbonyl group (FLU-R), glucose conjugate of FLU-R and 2 glucose conjugates of FLU. In the resistant strains, the ex vivo formation of all FLU metabolites was significantly higher than in the susceptible ISE strain. The multi-resistant WR strain formed approximately 5 times more conjugates of FLU than the susceptible ISE strain. The in vitro data also showed significant differences in FLU metabolism, in the activities of UDP-glucosyltransferase and several carbonyl-reducing enzymes between the susceptible and resistant H. contortus strains. The altered activities of certain detoxifying enzymes might protect the parasites against the toxic effect of the drugs as well as contribute to drug-resistance in these parasites.

The monogenetic trematode Discocotyle sagittata attaches itself to the gills of Salmo trutta with its long axis inclined to the long axis of the primary lamella to which it is attached, the direction of inclination being determined by the incidence of the gill-ventilating current of the host. The inclination is to some extent brought about by an asymmetrical disposition of the posterior adhesive organs, but Discocotyle appears to retain a primitive unspecialized character in that the asymmetrical disposition is facultative and not obligatory and permanent as in Axine.

The adhesive apparatus consists of four pairs of clamps and one pair of hooks. The hooks appear to be the persistent posterior hooks of the larva.

Each clamp is operated by an extrinsic muscle-tendon-fair-lead-hinged-jaws system generally similar but possibly less efficient than the corresponding system in Plectanocotyle gurnardi. Scheuring's and Sproston's interpretations of the clamp-closing mechanism of Discocotyle as a spring system are wrong.

1. Crepidostomum farionis inhabits the gall bladder, as well as the intestine and pyloric caeca, of the trout and grayling.

2. The life history of Crepidostomum farionis has been worked out and is based on the similarity of organisation of the cercaria, encysted larval trematode and the adult.

3. The first intermediate hosts in the life history of this trematode are Pisidium amnicum (Müll.) and Sphaerium corneum (L.) though the latter is unusual.

4. The second intermediate host is the larva of the mayfly, Ephemera danica (Müll.).

5. There are two generations of rediae, the first gives rise to daughter rediae, which in turn produce cercariae.

6. The rediae are characterised by the absence of ambulatory processes and a functional intestine.

7. The cercaria (n.sp.) possesses “eye spots,” stylet and gland cells (salivary?), and the excretory vesicle is tube-shaped.

8. The excretory system of the redia and the cercaria has been worked out in detail.

9. The relation of the parasites to their respective hosts is discussed. On account of the need for further observations definite conclusions are held over for a later paper.

Haemolymph free amino acids (HFAAs) in adult Tenebrio molitor varied in concentration with beetle age. Between 9 and 30 days post-emergence the concentration range was 34–94 mM with a peak occurring in 15-day-old insects. Much of this age-related variation was due to changes in the concentration of a single amino acid, proline. An analysis of concentrations of individual HFAAs in 15-day-old beetles, following infection with metacestodes of Hymenolepis diminuta, revealed that changes in female insects are more marked than those occurring in males. In the former, changes occurred in the concentration of 9 amino acids (isoleucine, leucine, arginine, serine, threonine, tyrosine, phenylalanine, proline and alanine/citrulline) whereas in the latter only 4 amino acids were affected (threonine, glycine, histidine and arginine). It is suggested that H. diminuta may interact with the endocrine system of its beetle host.

1. A method of trapping small mammals has been described and Apodemus sylvaticus, Clethrionomys glareolus, Microtus agrestis and Sorex araneus were captured. The method of trapping has been discussed.

2. A survey of the endoparasites found in the four host species has been produced. The two more common hosts and four of their parasites have been chosen for more detailed study.

3. The infection levels of the parasites in the two hosts have been calculated and compared with other published data.

Acetylcholinesterase (AChE) is present in all stages of the life-cycle of schistosomes and is located in muscle and on the surface of the parasite. Metrifonate is a drug that inhibits AChE. We compared the AChEs from three schistosome species (Schistosoma mansoni, Schistosoma haematobium and Schistosoma bovis) that have different susceptibilities to metrifonate in vivo. Sensitivities to AChE inhibitors were similar. The subunits of AChE were 110 kDa and 76 kDa and the dominant molecular form of AChE was a G2 form in all three species. This was the major form on the tegument while additional molecular forms were associated with the internal tissues. Differences in relative amounts of AChE activity between these species were found in the adults but not in the schistosomula. At the adult stage the major difference between species lay in the relative amounts of AChE activity in their teguments. S. haematobium teguments carried 20 times and S. bovis 6·9 times the activity present on S. mansoni teguments. These quantitative differences associate with the relative sensitivities of these species to metrifonate.

Oogenesis and fertilization in the filarial worm Dipetalonema viteae have been described. The female gametes are developed from a syncytium situated at the posterior tip of the ovary. The syncytium grows along the length of the ovary as an anucleate rachis carrying the germ cells with it. In the germinal region of the ovary the rachis is a central axial structure, but in the growth region it becomes very branched. The female gametes retain their attachment with the rachis until they have become mature primary oocytes; they acquire a surface coat whilst in the growth zone of the ovary. At fertilization the successful spermatozoon produces numerous ribosomes. Residual spermatozoa are apparently destroyed by the oocyte. The fertilized oocyte divides meiotically to yield an ovum and two polar bodies. In species destined to be unsheathed in the blood the surface coat and the original oolemma separate from the surface of the ovum. A new oolemma is produced, presumably under the stimulus of fertilization.

Paucivitellosus fragilis i sredescribed from the blenny, Salarias meleagris, found on beach-rock intertidally at Heron Island, Great Barrier Reef. The adult was also found in the mullet, Mugil cephalus and Crenimugil crenilabis at Heron Island, and Liza argentea at Brisbane.

Paucivitellosus differs from Bivesicula, Bivesiculoides and Treptodemus in having: (i) the vitellaria reduced to a single follicle on each side, (ii) two primary loops in the uterus, (iii) fully embryonated eggs in the terminal coils of the uterus, and (iv) the cirrus pouch orientated dorso-ventrally. The diagnosis of the family Bivesiculidae is emended.

The molluscan host at Heron Island is Cerithium penthusarus, in which cercariae develop in ‘fork-tailed’ rediae.

The cercaria is furcocystocercous and differs from known bivesiculid cercariae in having a protrusible glandular fold, within the mouth of the invaginated caudal chamber, by which the cercaria attaches to objects.

Evidence is presented for the view that the life-cycle is a two-host one, that there is no metacercarial stage, and that the cercaria is ingested by browsing.

Schistosome infections in humans are characterized by the development of chronic disease and high re-infection rates after treatment due to the slow development of immunity. It appears that anti-schistosome antibodies are at least partially mediating protective mechanisms. Efforts to develop a vaccine based on immunization with surface-exposed or secreted larval or worm proteins are ongoing. Schistosomes also express a large number of glycans as part of their glycoprotein and glycolipid repertoire, and antibody responses to those glycans are mounted by the infected host. This observation raises the question if glycans might also form novel vaccine targets for immune intervention in schistosomiasis. This review summarizes current knowledge of antibody responses and immunity in experimental and natural infections with Schistosoma, the expression profiles of schistosome glycans (the glycome), and antibody responses to individual antigenic glycan motifs. Future directions to study anti-glycan responses in schistosomiasis in more detail in order to address more precisely the possible role of glycans in antibody-mediated immunity are discussed.

Damage to the surface membrane of adult Schistosoma mansoni, and the activity of the excretory system, as shown by resorufin fluorescence, was observed following treatment with praziquantel and incubation with other molecules. Praziquantel treatment induced damage to the surface membrane as measured by the use of a variety of fluorescent compounds. The excretory system of the male worm was inhibited immediately after praziquantel treatment, but fully recovered after culture for 2 h following removal of praziquantel. The excretory system of the female, observed to be minimally active in untreated worm pairs, was often greatly activated in paired females, as shown by intense resorufin labelling, after praziquantel treatment, and this continued during recovery of the male excretory system. In experiments with normal worm pairs, the female could be activated by inhibiting the metabolic rate of the pair by a cooling procedure. The effects on the excretory system of changes in culture conditions (such as changes in pH, concentrations of bacterial lipopolysaccharide, cytokines, reactive oxygen species, compounds which remove cholesterol, such as beta-methyl cyclodextrin, and damaging basic poly-L-lysine) were also assessed. It is concluded that the extensive excretory system of the adult worm is responsive to drug treatment and to certain changes in environmental conditions. Its activity seems to be strongly linked to the integrity of the surface membrane.

1. Eggs and larvae of Trichostrongylus retortaeformis were used.

2. The rate of hatching of eggs was shown to be mainly related to temperature. From November to March, when maximum temperatures were below 50° F., there was no hatching. When maximum temperatures of 50–55° F. occurred eggs hatched on or before the fifteenth day, but never during the first 8 days. Eggs hatched in 8 days or less when maximum temperatures of 60–80° F. occurred.

3. When the rate of evaporation in the air was high, eggs still hatched and reached the infective stage, the grass blades reducing the rate of loss of moisture from the faecal pellet. Laboratory experiments show that eggs may not develop to the infective stage if the faecal pellets are on a grassless portion of the pasture. This is most likely to occur when the rate of evaporation is high and the temperature low.

4. Hatching may be delayed by cold conditions, but some eggs remain viable for long periods and they hatch when the temperature rises. Eggs passed by the host in the autumn can survive a cold winter and hatch in the spring, but eggs passed during the coldest period die.

5. During periods when the maximum temperature never exceeded 55° F., little or no migration of larvae occurred. When temperatures rose above 55° F. the number of larvae migrating increased; but rise of temperature was associated with increase in the rate of evaporation. High rates of evaporation reduced the number of larvae migrating on the grass blades.

6. Some infective larvae died soon after exposure on grass plots, but a small number survived long periods. In cold weather some larvae were still alive after 20 weeks. A high death-rate occurred in warm weather. A large proportion of the larvae died during periods in which the rate of evaporation was high; in one of these periods 95% of the larvae were dead at the end of 4 weeks' exposure.

7. The number of larvae on grass blades of a pasture was shown to be dependent, at any time, upon the climate at that time, and upon past conditions which had influenced hatching and survival:

The present findings apply to a particular area, the four northern counties of England, and must be viewed within the limits of the data presented.

Only one tick, Ixodes ricinus L., the so-called sheep tick, infests sheep and cattle in northern England. I. canisuga Johnston, alone or with I. ricinus, has been found on sheep dogs and foxes.

The distribution of the sheep tick in this region is markedly discontinuous, the total infested areas including only about one-fifth of the hilly country, to which the tick is almost exclusively confined.

In north-west Northumberland, tick distribution shows a significant degree of correlation with certain factors inherent in or influenced by the surface geology. Broadly speaking, where surface geology lends itself to relatively good natural drainage and soil, the grazing is relatively good and ticks are absent; and where surface geology results in relatively poor natural drainage and/or soil, the grazing is relatively poor (‘rough’) and ticks are present.

No consistent correlations between soil characteristics—i.e. pH, available phosphates and potash, soil textures, mechanical analysis and soil depth—and tick distribution have been found. Nor is there a consistent correlation- between quality of natural drainage and tick distribution. The interaction of soil and natural drainage factors, however, profoundly influences the character of the vegetation layer.

In the four northern counties, the same plant dominants occur on tick-infested and tick-free hill lands. Where grazing is rough ticks are usually present irrespective of whether the dominant plant is one of the rough grasses (Nardus, Molinia, Agrostis, Aira), bracken or heather; where grazing is relatively good (i.e. not rough), ticks are invariably absent.

Observations on a smaller scale show that the thicker, i.e. rougher, the vegetation layer, the denser the tick population and also the thicker the vegetation layer, the thicker the basal mat. On five plots it has been shown that there is a consistent positive correlation between mat thickness and tick population density: the thicker the mat,’ the denser the tick population irrespective of whether bracken or grass is the dominant. Thus the chief controlling factor in tick distribution is the physical character of the vegetation layer.

Macroparasites are almost always aggregated across their host populations, hence the Negative Binomial Distribution (NBD) with its exponent parameter k is widely used for modelling, quantifying or analysing parasite distributions. However, many studies have pointed out some drawbacks in the use of the NBD, with respect to the sensitivity of k to the mean number of parasites per host or the under-representation of the heavily infected hosts in the estimate of k. In this study, we compare the fit of the NBD with 4 other widely used distributions on observed parasitic gastrointestinal nematode distributions in their sheep host populations (11 datasets). Distributions were fitted to observed data using maximum likelihood estimator and the best fits were selected using the Akaike's Information Criterion (AIC). A simulation study was also conducted in order to assess the possible bias in parameter estimations especially in the case of small sample sizes. We found that the NBD is seldom the best fit for gastrointestinal nematode distributions. The Weibull distribution was clearly more appropriate over a very wide range of degrees of aggregation, mainly because it was more flexible in fitting the heavily infected hosts. Moreover, the Weibull distribution estimates are less sensitive to sample size. Thus, when possible, we suggest to carefully check on observed data if the NBD is appropriate before conducting any further analysis on parasite distributions.

Parasite life-cycles present intriguing model systems to study divergence in resource use and ecology between parasite taxa. In ecologically similar taxa, consistent selective forces may lead to convergence of life-history traits, but resource overlap and similarity of life-cycles may also promote divergence between the taxa in (1) use of host species or (2) specific niche within a host. We studied the life-history characteristics of 2 sympatric species of Diplostomum parasites, D. spathaceum and D. gasterostei, concentrating particularly on differences in intermediate host use and characteristics of the infective stages between the species. This group of trematodes is a notoriously difficult challenge for morphological taxonomy and therefore any information on the ecology of these species can also be helpful in resolving their taxonomy. We observed that these species indeed had diverged as they used mainly different snail and fish species as intermediate hosts and in controlled experiments infected different regions of the eye (lens and vitreous body) of a novel fish host. Interestingly, cercarial characteristics (activity and life-span) were similar between the species and the species were difficult to separate at the cercarial stage unless one observes their swimming behaviour. The release of cercaria from the snail hosts was higher in D. spathaceum, but when cercarial numbers were proportioned to the volume of the snail host, the production was higher in D. gasterostei suggesting differences in the rate of snail host exploitation between the parasite species. These results corroborate the prediction that closely related parasite taxa which are competing for the same resources should have diverged in their life-history characteristics and host use.

Nitric oxide (NO) production has been suggested to play a role as effector molecule in the control of the malarial infections. However, the roles of this molecule are debated. To assess whether blood-stage parasite killing is NO dependent, we investigated the course of blood-stage Plasmodium chabaudi chabaudi (Pcc) infections in inducible nitric oxide synthase (iNOS)-deficient mice. Parasitaemia, haematological alterations, and survival were not affected by the lack of iNOS. To exclude a role of NO produced by other NOS, controls included NO suppression by oral administration of aminoguanidine (AG), a NOS inhibitor. As in iNOS-deficient mice, no difference in the parasitaemia course, survival and haematological values was observed after AG treatment. Our results indicate that NO production is not required for protection against malaria in our murine experimental model. However, C57BL/6 mice treated with AG lost their resistance to Pcc infections, suggesting that the requirement for NO production for parasite killing in murine blood-stage malaria might be strain dependent.
