Unisexual females, (80–100 days post-infection) were cultured in vitro and exposed to mature male worms from either mixed or single-sex infections. A comparison was made of the uptake of [3H]thymidine (used as a marker for DNA synthesis) by isolated unisexual females or by those females exposed to males derived from either mixed or unisexual infections during in vitro culture. All males were observed to be paired with females by approximately 18 h post-culture. At 24 h post-culture females paired with males from mixed infections had an elevated uptake of [3H]thymidine compared to females that had not paired and uptake was also significantly different (P ≥ 0–001) from females exposed to males from unisexual infections. By 48 h there was no significant difference between uptake in females paired with either type of male. The frequency of uptake in the paired females was approximately double that in unstimulated females while non-paired females in the presence of males had an increase of between 35 nd 45%. In a second experiment the effect of depriving a mature egg-laying female of the paired male was investigated. These females had a continual decrease in uptake of [3H]thymidine compared to mature females maintained with male partners. The uptake of [3H]thymidine was used as a marker for DNA synthesis and the effect of the male worm on DNA synthesis in the unisexual female is discussed in view of the above results.

Glycogen synthase kinase-3 (GSK-3) is classically described as a key enzyme involved in glycogen metabolism in mammals. It has been shown to be highly conserved among several organisms, mainly in the catalytic domain region. This enzyme has already been described in Rhipicephalus (Boophilus) microplus and the ovaries of females appeared to be the major site of GSK-3 transcription. The treatment with GSK-3 specific inhibitor (alsterpaullone, bromo-indirubin-oxime 6 and indirubin-3-oxime) caused a reduction in oviposition and egg hatching in completely engorged female ticks. The effect was more pronounced in partially engorged females when alsterpaullone was administrated by artificial capillary feeding. Moreover, GSK-3 gene silencing by RNAi in partially engorged females reduced significantly both oviposition and hatching. The study of tick embryogenesis and proteins that participate in this process has been suggested as an important means for the development of novel strategies for parasite control. GSK-3 is an essential protein involved in embryonic processes and for this reason it has already been suggested as a possible antigen candidate for tick control.

Although Ixodes ricinus ticks are mainly associated with woodland, they are also present in open habitat such as pastures. The distribution of nymphal I. ricinus was monitored by drag sampling the vegetation in May–June 2003 on 61 grazed permanent pastures for cattle located in central France. After selecting explanatory variables from among a set of 155, tick abundance was modelled on the perimeter of the pasture using a negative binomial model that took into account data overdispersion. An abundant tree layer at the perimeter of the pasture associated with a high humidity before sampling greatly enhanced the average number of captured I. ricinus nymphs. The presence of apple or cherry trees around the pasture perimeter, the presence of trees or bushes at the pasture edge, woodland around the pasture and a high number of I. ricinus nymphs in the nearest woodland to the pasture were also favourable to nymph abundance in the pasture. The study highlighted that woodland vegetation associated with humidity and the presence of attractive foraging areas for tick hosts around the pasture played a key role in the abundance of I. ricinus. Finally, the results raised the question of whether and how transfer of ticks between woodland and grazed pastures occurs.

The cercaria of Trichobilharzia ocellata finds and identifies its duck host with a seriesof different behavioural phases. Dispersal and selection of the water surface as microhabitat is achieved by an intermittent swimming behaviour, which is governed by the interplay of passive dropping with forward and backward swimming movements and includes a positive phototactic and a geonegative orientation. Then the cercariae tend to cling to the water surface in an energy-saving resting position. A movement towards the duck feet as the site of entry occurs when shadows evoke forward swimming movements, which are directed away from the source of light, i.e. normally downwards. Forward swimming movements are also stimulated by touch, but only in free-swimming cercariae and not when these are in the resting position. Attachments occur only when a substrate is touched during forward swimming movements. Attachments are stimulated by warm substrates (1 °C temperature difference triggers a nearly maximal response) and by chemical components of duck-foot skin, and the readiness to attach is increased when the forward swimming movement is started by shadow stimuli.

Partial (DNA) sequences were collected for 2 mitochondrial loci (Srrna and Lrrna, the rrnS and rrnL rRNA genes respectively) for Schistosoma indicum group species from 4 Southeast Asian countries. The samples included 7 populations, 4 of which were previously unstudied. In 11 cases the combination of locus and population was new. The aim of the study was to provide a phylogeny based on new independent data and multiple populations (earlier studies had mostly used a common set of field samples or laboratory lines) and to examine interrelationships and phylogeography within this species group. Paraphyly of the S. indicum group was confirmed, as was the basal position of Schistosoma incognitum in the Schistosoma phylogeny. Southeast Asian Schistosoma spindale and S. incognitum populations were shown to fall into their respective con-specific cohesive groupings. Estimated divergence times for these taxa were shown to be related to Pleistocene changes in sea level and the radiation of definitive host groups. A revised phylogeographical model is proposed in the light of these findings.

Parasitic helminths are ubiquitous in most host, including human, populations. Helminths often alter the likelihood of infection and disease progression of coinfecting microparasitic pathogens (viruses, bacteria, protozoa), and there is great interest in incorporating deworming into control programmes for many major diseases (e.g. HIV, tuberculosis, malaria). However, such calls are controversial; studies show the consequences of deworming for the severity and spread of pathogens to be highly variable. Hence, the benefits of deworming, although clear for reducing the morbidity due to helminth infection per se, are unclear regarding the outcome of coinfections and comorbidities. I develop a theoretical framework to explore how helminth coinfection with other pathogens affects host mortality and pathogen spread and evolution under different interspecific parasite interactions. In all cases the outcomes of coinfection are highly context-dependent, depending on the mechanism of helminth-pathogen interaction and the quantitative level of helminth infection, with the effects of deworming potentially switching from beneficial to detrimental depending on helminth burden. Such context-dependency may explain some of the variation in the benefits of deworming seen between studies, and highlights the need for obtaining a quantitative understanding of parasite interactions across realistic helminth infection ranges. However, despite this complexity, this framework reveals predictable patterns in the effects of helminths that may aid the development of more effective, integrated management strategies to combat pathogens in this coinfected world.

Samples of Peringia ulvae were collected and measured on eight occasions, over a period of two years from an isolated pool in the saltings of the River Tamar. Growth conditions in this pool are at a maximum. Spawning occurs in the autumn and the greatest number of small specimens are found in the sieve (1 mm. mesh) in February. Maximum growth takes place during April-July. After ten months of age,.uninfected snails which have reached a length of 5¾ mm. grow only 1–1¼ mm., and gradually die off from 17 months of age onwards. Infected snails display gigantism and variation and attain dimensions of 9–10 mm. They are probably no older than uninfected specimens measuring 6¾ mm. long.

The infection rate is high and shows certain seasonal fluctuations, no doubt linked with the migrations of the wading birds which are the principal final hosts of the trematodes.

The known range of the zoonotic fox tapeworm Echinococcus multilocularis has expanded since the 1990s, and today this parasite is recorded in higher abundances throughout large parts of Europe. This phenomenon is mostly attributed to the increasing European fox populations and their invasion of urban habitats. However, these factors alone are insufficient to explain the heterogeneous distribution of the parasite in Europe. Here, we analysed the spatial interrelationship of E. multilocularis with the known distribution of seven vole species in Ticino, southern Switzerland. Among 404 necropsied foxes (1990–2006) and 79 fox faecal samples (2010–2012), E. multilocularis was consistently found in the north of the investigated area. No expansion of this endemic focus was recorded during the 22 years of the study period. This stable endemic focus is coincident with the known distribution of the vole species Microtus arvalis but not, or only partly, with the distribution of the other autochthonous vole species. Our results give evidence that this vole species plays a crucial role in the maintenance of the parasite's life cycle and that its absence could be a limiting factor for the spread of E. multilocularis in this region.

The impact of immune sera, and peripheral blood cells (PBC) from mice immunized with Plasmodium berghei ookinetes; and of purified immunoglobulin or Fab fragments from anti-Pbs21 monoclonal antibody 13.1, upon establishment of oocyst infections in the mosquito was studied. Infections were initiated either from gametocyte-infected mice, or membrane feeders which contained either gametocytes or mature ookinetes. PBC from ookinete-immunized mice presented with non-immune serum failed to show any transmission-blocking activity. Anti-ookinete serum, intact anti-Pbs21 monoclonal antibody 13.1 or its Fab fragments, all inhibited oocyst formation significantly. When gametocyte-infected mice or gametocytes in membrane feeds were used, inhibition did not directly correlate with antibody concentration. In membrane feeders that contained ookinetes and antibody, concentration-dependent inhibition usually occurred. The efficacy of purified 13.1 IgG was dependent upon the ookinete concentration. The ookinete plasmalemma and cytoplasm were significantly disturbed after 12h in bloodmeals that contained antibody 13.1, but not in the isotype controls. These changes may have caused the observed failure of the ookinete to migrate as rapidly as the controls from the destructive environment of the bloodmeal.

A deterministic model for the demography of the free-living stages of Haemonchus contortus is described. The model consists of three compartments: the pre-infective stages, comprising the eggs and first- and second-stage larvae combined; third-stage larvae in the faeces; and third-stage larvae on the pasture. The mortality (μj) of each of these stages was a function of temperature (K, where K = (degrees Celsius + 30)/100) of the form, μj = aj exp [xj K] + bj exp [– βj K]. The minimum time to second moult (γ) and the moulting rate thereafter (σ) were also functions of temperature (In [1/γ] = c1 + c2[1/K], and σ = d1 exp [d2 K] respectively). The rate at which L3 larvae migrated out of the faeces was treated as a constant (0.165/larva/day).

Microvilli are developed on the surface of Schistosoma mansoni schistosomula during penetration of the host skin; they form rapidly but are lost approximately 90 min after penetration. Identical microvilli are also formed on schistosomula which have penetrated a mouse skin preparation in vitro, and on schistosomula prepared by mechanical separation of the tail from the body of the cercaria. The microvilli, which are limited by the trilaminate tegumental membrane of the cercaria, eventually degenerate and are cast off from the surface of the tegument. There is little change in the surface area of the schistosomulum at this time, and the formation and loss of microvilli coincides with the replacement of the cercarial tegumental membrane by the new heptalaminate membrane. It is suggested that during the cercaria/schistosomulum transformation, some intramembraneous components of the original cercarial membrane may migrate into the new heptalaminate membrane and thus be retained, while other peripheral components such as the glycocalyx are almost certainly lost.

The main pathological characteristics of hepatic fibrosis in schistosomiasis are the proliferation of hepatic stellate cells (HSCs) and the deposition of collagen type I (Col I) and collagen type III (Col III). Transforming growth factor beta-1 (TGF-β1) plays an important role in hepatic fibrosis. Paeoniflorin (PAE) has been reported to have immunoregulatory effects; however, the mechanism of its anti-hepatic fibrosis in S. japonicum has not been elucidated. In the present study, we found that mouse peritoneal macrophages (PMφs) stimulated by soluble egg antigen (SEA) of S. japonicum could secrete TGF-β1, and the TGF-β1 in the peritoneal macrophage-conditioned medium (PMCM) could induce proliferation of HSCs and secretion of Col I and III. We selected PMCM at 1:2 dilution as the optimum PMCM (OPMCM). Then we treated HSCs pre-incubated with OPMCM with PAE, and found that the inhibition of HSC proliferation or Col I and III production were closely correlated with the concentration of PAE. Further investigation found that PAE significantly decreased the Smad3 transcription and phosphorylation in HSCs stimulated by OPMCM. In conclusion, SEA plays a key role in hepatic fibrosis by inducing TGF-β1 from PMφs. PAE can exert anti-fibrogenic effects by inhibiting HSCs proliferation and down-regulating Smad3 expression and phosphorylation through TGF-β1 signalling.

The uniform morphology of the developmental stages of Haemogregarina species and the insufficient information supplied by the simplistic descriptions of previous authors complicates their differential diagnosis and proper species identification. In this study, we detected Haemogregarina spp. in 6 out of 22 (27·2%) examined turtles originating from Southeast Asia, Malayemys subtrijuga (n = 4), Sacalia quadriocellata (n = 1) and Platysternon megacephalum (n = 1), and compared them with the available literature data. Microscopic analysis of our isolates distinguished 2 morphological species, Haemogregarina pellegrini and one new species, being described in this paper as Haemogregarina sacaliae sp. n. Phylogenetic analyses based on 1210 bp long fragment of 18S rDNA sequences placed both haemogregarines firmly within the monophyletic Haemogregarina clade. Isolates of H. pellegrini from 2 distantly related turtle hosts, M. subtrijuga and P. megacephalum, were genetically identical. Despite the fact that numerous Haemogregarina species of turtles have been described, the incompleteness of the morphological data and relatively low host specificity provides the space for large synonymy within this taxon. Therefore, a complex approach combining microscopic analyses together with molecular-genetic methods should represent the basic standard for all taxonomic studies.

A study on the ecological distribution of alveolar Echinococcus was carried out in the Hulunbeier Pasture of Inner Mongolia, China during 1998 and 1999. Animals examined included wolves (Canis lupus), red foxes (Vulpes vulpes), sand foxes (Vulpes corsac), domestic dogs (Canis familiaris), Microtus brandti, Meriones unguiculatus, Citellus dauricus, Allactaga sibirica, Phodopus sungorus and Ochotona daurica. Three wolves were found to be infected with E. granulosus. Two sand foxes were infected with E. multilocularis. The majority of infections of alveolar echinococcus was found in M. brandti. Based on the structure of metacestodes found in the livers of naturally infected M. brandti, 3 main variants were observed. Type I had small alveolar cysts with thin cyst walls. Type II had a larger cyst with a thick cyst wall. Infection of laboratory mice with the gravid segments isolated from the naturally infected sand foxes led to the formation of mature Type I alveolar metacestodes in the lungs and Type II metacestodes in the livers of infected animals, respectively.

We have shown here that collagen type I bound efficiently to the trypomastigote surface. In addition, monoclonal and polyclonal antibodies against collagen types I and III inhibited the infection of fibroblasts by the parasite. These results suggested the presence of collagen-binding protein(s) on the parasite surface. This protein was identified from trypomastigote surface antigens using affinity chromatography on a Gelatin Ultrogel column (denatured form of collagen). These collagen-binding proteins were revealed as a low-affinity gelatin binding protein (LAG Bp) of 98 kDa, and a high-affinity binding protein (HAG Bp) of 58 and 68 kDa under non-reducing and reducing conditions respectively. In addition, HAG Bp and LAG Bp bound to collagen type I. The 58/68 kDa protein was purified to homogeneity on a wheat germ agglutinin Sepharose column. A polyclonal antibody to this glycoprotein, as well as a monoclonal antibody (McAb) 155D3 produced against the HAG Bp, immunoprecipitated two parasite surface antigens of 160 and 58 kDa under non-reducing conditions which migrated at a position of 80–85 and 68 kDa when reduced. However, only the 80–85 kDa component could be precipitated from [S] methionine-labelled trypomastigote antigens under reducing conditions. The antibodies to the 58/68 kDa glycoprotein as well as McAb 155D3 diminished the invasion of fibroblasts by parasites. Taken together these results suggest that the same receptor binds fibronectin and/or collagen and that both the 80–85 and 58/68 kDa glycoproteins form part of the same receptor. These trypomastigote surface molecules may interact with the host cell fibronectin and/or collagen during the initial phase of parasite-cell recognition.

The fecundity and longevity of Nippostrongylus brasiliensis was prolonged in mice previously infected with Nematospiroides dubius only when the former developed from the larval stage in those mice. Such worms appeared to be less immunogenic than worms which developed in mice never exposed to N. dubius.

It is proposed that prolonged fecundity and longevity resulted from an adaptation undertaken by the worms in the face of host antibodies which had been developed against the pre-existing N. dubius infection.

Changes in the amount and distribution of acid and alkaline phosphatase, non-specific esterase, glycogen, lipid and acid mucopolysaccharide in the caecal wall and liver of turkey poults infected with Histomonas meleagridis have been studied histochemically. A microdensitometer was used to measure changes in activity and distribution of acid phosphatase and non-specific esterase in the caecal mucosa. During the course of the infection there is a marked reduction in activity and distribution of acid phosphatase and non-specific esterase but little change in the amounts and distribution of alkaline phosphatase, glycogen, lipid and acid mucopolysaccharide in the wall of the main part of the caecum. Similar, but smaller, changes occurred in the wall of the neck region of the caecum. In the liver most changes occurred in the immediate vicinity of the parasites. Initially, there was a reduction in the amount of glycogen in the parasitic lesions but later in the infection there was a marked loss of glycogen in all regions of the liver. Changes in the caecum are apparently brought about by the parasite prior to and after invasion of the caecal tissues; changes in the liver occur after it has been invaded.

The first rigorous proof applicable to the migration pathway of an infective juvenile macroparasite inside its host is presented. Third-stage larvae of a homogonic strain of Strongyloides ratti applied in exact doses of less than 20 to the skin of the flank of young rats were recovered 16–40 h later in the naso-frontal part of the head. The peak proportion of the dose recovered between 20 and 25 h in this site had a mean value of 0·316 ± 0·021 in 48 animals. In 40 other rats infected simultaneously the mean proportion of the dose that reached the small intestine was at least 0·837+0·013. Proof resides in verification of the inequality > 1. With appropriate statistical tests the excess of the sum of the means of these two proportions over unity is shown to have a probability of occurring by chance of 1 in 3·5 × 106. Thus it is effectively certain that the naso-frontal portion of the head is part of at least one pathway taken by this parasite on its way from the skin to the intestine of its host. By suitable protection of the infection site it was confirmed that migration to the head was achieved by an internal route and not as a result of grooming. Larvae were recovered from the cranium in the same rats over the period 15–40 h, but the peak proportion of the dose occurred at 20 h, and < in this location. Whether the cranium is also part of the pathway is therefore still undecided. The significance of this novel analysis in the general context of in-host migration of infective stages is discussed and it is concluded, following its application to data sets from other authors, that the only cases in which proof can be demonstrated are the anterior skull of the rat for S. ratti (present data) and the lung of the same host for Nippostrongylus brasiliensis (Twohy, 1956).

An analysis of the course of infection of mixed oral infections of 12 cystacanths of Moniliformis dubius in 174 male and 179 female Wistar rats has been undertaken.

There was a marked decline in the average recovery rate of worms of both sexes from hosts of both sexes during the course of the infection.

Female worms from both male and female rats showed, on average, a greater power of survival than male worms from the third period (10–13 weeks) onwards.

Male rats were found to retain, on average, a greater number of worms of both sexes than female rats.

The suggestion that there may be a limit to the number of niches available to helminth species in the intestine of Anguilla anguilla was investigated by examining the frequency distributions of the number of helminth species per eel and the relationships between maximum and mean infracommunity richness and component community richness in 1 locality over 17 years and in 64 localities throughout Ireland and England. The maximum number of species per eel did not exceed 4 in the 1 locality, or 3 in the 64 localities. In both the single and the several localities, the relationship between maximum and mean infracommunity richness and component community richness was curvilinear and best described by a power or polynomial function. This was interpreted to mean that infracommunity richness became increasingly independent of component community richness, and that infracommunities were saturated at values well below the higher level of helminth richness immediately available for colonization i.e. component community richness. It is argued that these findings cannot be explained by supply-side ecology, pool exhaustion or transmission rates, but only by infracommunity processes acting to impose a fixed limit to the number of species in an infracommunity. Most infracommunities are species poor, and the limiting factors will only operate as species richness rises to determine a maximum. Acceptance of a limit to the number of niches available also resolves the apparent inconsistency between the occurrence and importance of interspecific competition and the nature of isolationist communities.
