We tested the hypothesis that 30 min of brisk walking daily would offset the increase in plasma TAG concentrations associated with substituting dietary fat for carbohydrate. Fourteen subjects (six males) aged 57 (sd 5) years underwent three 4 d trials in a counterbalanced order: (i) 4 d on a typical UK diet (40 % energy from fat, 45 % carbohydrate and 15 % protein); (ii) 4 d on an isoenergetic diet but substituting fat for carbohydrate in line with the present recommendations (30 % fat, 55 % carbohydrate and 15 % protein); (iii) 4 d on the same recommended diet with 30 min of brisk walking each day. The food provided for the first 3 d of each trial was isoenergetic with each volunteer's previously determined daily energy intake. On day 4, the subjects consumed breakfast, lunch and an early evening meal, equivalent in total to 90 % of their daily energy intake. Blood samples were collected and substrate utilisation and energy expenditure were determined in the fasted state and for 9 h postprandially. Substrate utilisation differed significantly among trials (P = 0·003); RER was higher on the recommended diet trial than during either of the other two trials (P = 0·012 and 0·021 for the UK diet and recommended diet with walking, respectively). The rise in plasma TAG concentrations over the day was steeper on the recommended diet trial than on the other two trials (trial × time interaction, P = 0·040). The increase in postprandial TAG concentrations associated with substituting dietary fat for carbohydrate was offset by 30 min of brisk walking daily.

In the present study, the protective effects of dietary Spirulina (SP) and germanium-containing Spirulina (GeSP) were compared in rats with liver injury induced by an intraperitoneal injection of d-galactosamine and lipopolysaccharide (GalN/LPS). Wistar rats were fed one of the following diets: the basal diet (GalN/LPS-CON group; n 6), the basal diet supplemented with 5 % SP or GeSP (GalN/LPS-SP and GalN/LPS-GeSP group, respectively; n 7 each). After administering these diets for 7 d, each rat was intraperitoneally injected with GalN/LPS. Increases in plasma alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were suppressed in the GalN/LPS-GeSP group (GalN/LPS-CON v. GalN/LPS-GeSP: ALT 1052 (sem 187) v. 509 (sem 88) IU/l and AST 2183 (sem 368) v. 1170 (sem 196) IU/l) following the injection of GalN/LPS. Plasma levels of interferon-γ (IFN-γ) and TNF-α in GeSP-fed rats were significantly lower when compared with those in the GalN/LPS-CON group (GalN/LPS-CON v. GalN/LPS-GeSP: IFN-γ 142·8 (sem 17·5) v. 66·8 (sem 9·7) pg/ml and TNF-α 72·3 (sem 15·4) v. 31·2 (sem 6·8) pg/ml). However, the decrease in these levels observed in the GalN/LPS-SP group was not as prominent as those observed in the GalN/LPS-GeSP group. Furthermore, the increase in liver catalase (CAT) and glutathione peroxidase (GPx) activities, as well as the level of oxidised glutathione (GSSG), was more suppressed in GeSP-fed rats (GalN/LPS-CON v. GalN/LPS-GeSP: CAT 457 (sem 47) v. 262 (sem 54) U/mg liver protein; GPx 1·30 (sem 0·11) v. 0·53 (sem 0·09) U/mg liver protein; GSSG 2·18 (sem 0·33) v. 1·31 (sem 0·24) mmol/kg liver) after the injection of GalN/LPS. These changes were more pronounced in the GalN/LPS-GeSP group than in the GalN/LPS-SP group. These results suggest that GeSP could afford a significant protective effect in the alleviation of GalN/LPS-induced hepatic damage. In addition, the results indicate that GeSP is more effective than SP.

1. Experiments were undertaken to examine the errors associated with the use of indigestible markers, the 51Cr-labelled complex of chromium ethylenediaminetetra-acetic acid (51Cr-EDTA) and 103Ru-labelled tris-(1,10-phenanthroline)-ruthenium(II) chloride (103Ru-P), to measure liquid- and solid-phase digesta kinetics in the caecum and proximal colon of sheep.

2. First-order kinetics of markers were observed following either single injection or termination of continuous infusion. There were no differences between the half-times (T½) of marker in the caecum plus proximal colon whether calculated from marker concentration in caecal digesta or in faeces. There were also no differences in the T½ values calculated for the liquid- and solid-phase markers. When pool sizes calculated from the marker kinetics were compared with the volume of digesta present in the caecum and proximal colon at slaughter, it appeared that the 51Cr-EDTA and 103Ru-P caecal pools described the digesta contained in the entire caecum and proximal colon.

3. The flow-rates of dry matter (DM) through the caecum of sheep given 694 g lucerne (Medicago sativa) DM/d were similar whether estimated from total collection of faeces, by single injection of marker, or by the ratio, marker concentration: DM in either caecal digesta or faeces during continuous infusion of marker into either the rumen or the caecum.

4. In sheep given 553 g brome grass (Bromus inermus) DM/d the coefficient of variation of estimates of the plateau of 51Cr-EDTA marker during continuous infusion into the caecum was greater when 130 ml infusate/d were administered than with 1000 ml/d.

5. In the sheep given brome grass the lines of best fit of decline in In 51Cr-EDTA marker concentration v. time following termination of the continuous infusions described previously and following single injection of marker in 20 or 2 ml into the caecum were examined. The variation was least when 1000 ml infusate/d had been administered and was unacceptably large following a single injection of 2 ml.

6. These experiments showed that tracer techniques could provide unbiased estimates of trace kinetics in the caecum and proximal colon.

1. Metabolism of L-[1-14C]methionine, DL-[l-14C]methionine and DL-[ 1-14C]2-hydroxy-4-methylthiobutanoic acid (DL-HMB) by broiler chicks which had been fasted overnight or given a methionine-deficient diet was compared with fed (control) birds.

2. The excretion of 14C-labelled material, total 14CO2 exhaled, 14C incorporation into tissue proteins and the 14C-labelled material in perchloric-acid-soluble tissue fractions were measured 6 h after injection of the 14C-labelled materials.

3. The incorporation of 14C into tissue proteins and the relative rates of conversion of D-methionine and DL-HMB to L-methionine in tissues under different nutritional regimens were compared using protein-bound 14C:protein-free 14C values.

4. Fasted birds exhaled more 14CO2 than control birds but excreted less 14C, while methionine-deficient birds behaved very similarly to the control animals in these respects.

5. Fasted birds incorporated much less 14C into proteins of tissues other than liver and kidney from all three labelled tracers. The values for protein-bound 14C: protein-free 14C were lower in all tissues.

6. Methionine-deficient birds had similar levels of 14C in tissue proteins but lower values for protein bound 14C: protein-free 14C.

7. Examination of the values for protein-bound 14C:protein-free 14C suggest that brain and probably liver tissues from fasted and methionine-deficient birds showed improved rates of conversion of D-methionine and DL-HMB to L-methionine compared with control animals.

We have previously shown that higher intake of cruciferous vegetables is inversely associated with carotid artery intima-media thickness. To further test the hypothesis that an increased consumption of cruciferous vegetables is associated with reduced indicators of structural vascular disease in other areas of the vascular tree, we aimed to investigate the cross-sectional association between cruciferous vegetable intake and extensive calcification in the abdominal aorta. Dietary intake was assessed, using a FFQ, in 684 older women from the Calcium Intake Fracture Outcome Study. Cruciferous vegetables included cabbage, Brussels sprouts, cauliflower and broccoli. Abdominal aortic calcification (AAC) was scored using the Kauppila AAC24 scale on dual-energy X-ray absorptiometry lateral spine images and was categorised as ‘not extensive’ (0–5) or ‘extensive’ (≥6). Mean age was 74·9 (sd 2·6) years, median cruciferous vegetable intake was 28·2 (interquartile range 15·0–44·7) g/d and 128/684 (18·7 %) women had extensive AAC scores. Those with higher intakes of cruciferous vegetables (>44·6 g/d) were associated with a 46 % lower odds of having extensive AAC in comparison with those with lower intakes (<15·0 g/d) after adjustment for lifestyle, dietary and CVD risk factors (ORQ4 v. Q1 0·54, 95 % CI 0·30, 0·97, P = 0·036). Total vegetable intake and each of the other vegetable types were not related to extensive AAC (P > 0·05 for all). This study strengthens the hypothesis that higher intake of cruciferous vegetables may protect against vascular calcification.

Adequate dietary iodine intake in children is essential for optimal physical and neurological development. Whether lower dietary animal food and salt intake may adversely affect iodine status is under discussion. We examined the association between dietary animal:plant protein ratio with 24-h urinary iodine excretion (24-h UI, μg/d), and whether this is modified by salt intake. A 24-h UI was measured in 1959 24-h urine samples from 516 6- to 12-year-old participants of the Dortmund Nutritional and Anthropometric Longitudinally Designed Study. Parallel 3 d weighed food records were used to estimate dietary intakes. Protein sources were classified as dairy, animal and plant. A repeated-measures regression model (PROC MIXED) was used to analyse the effect of animal:plant protein ratios on 24-h UI. Animal:plant protein ratios ranged from 0·5 (95 % CI 0·4, 0·6) to 1·6 (95 % CI 1·4, 1·9) (lowest and highest quartile). After adjustment for total energy intake, main dietary iodine sources (dairy and salt intake), and further covariates, the inter-individual variation in animal:plant protein ratio was significantly associated with variation in 24-h UI. One unit higher animal:plant protein ratio predicted 6 μg/d higher 24-h UI (P= 0·002) in boys and 5 μg/d (P= 0·03) in girls. This relationship was partially mediated by a higher salt intake at higher animal:plant protein ratios. These results suggest that lower consumption of animal protein is associated with a small decline in iodine excretion, partially mediated by decreased salt intake. Because limited salt and increased intake of plant-based foods are part of a preferable healthy food pattern, effective nutrition political strategies will be required in the future to ensure appropriate iodine nutrition in adherent populations.

In a 2×2 balanced factorial experiment the biochemical effects on pigs of two dietary factors were investigated. The first factor was α-tocopherol and Se supplementation and the second factor was supplementation with α-tocopherol-stripped linseed oil. In pigs fed on diets depleted of α-tocopherol and Se, increases in concentrations of markers of lipid peroxidation (4-hydroxynonenal and hexanal) were observed. However, skeletal myopathy was only observed in those pigs fed on diets depleted of α-tocopherol and Se and supplemented with oil. In those pigs, increased lipid peroxidation was observed in heart and supraspinatus muscle. The plasma concentration of thromboxane B2 was increased in pigs fed on diets depleted of α-tocopherol and Se, suggesting an increased tendency towards platelet aggregation. However, this change was reversed in pigs depleted of α-tocopherol and Se, but supplemented with oil. This may have been a consequence of loss of arachidonic acid, the substrate for thromboxane formation, as a result of lipid peroxidation.

Male rats were fed on either a non-purified rodent diet (JS) or cholesterol-free purified diets containing wheat bran (WB) or oat bran (OB). Some animals were allowed normal access to their faeces for coprophagy (coprophagy +), while in others coprophagy was prevented by placement of a plastic cup over the anus (coprophagy −). Direct ingestion of faeces from the anus was observed in the former groups. Food intake was unaffected by diet or coprophagy status and body weight gain was unchanged with OB − and JS − but was significantly lower with WB −. Plasma cholesterol was highest with WB and equally lower with OB and JS and was unaffected by coprophagy status. Plasma triacylglycerols were highest with OB and were unaffected by coprophagy status. Caecal digesta mass was highest with JS, intermediate with OB and lowest with WB. Digesta mass was unaffected by coprophagy status with WB and JS but was higher with OB −. Digesta moisture content was lowest with WB + but highest with WB −. Digesta volatile fatty acid (VFA) concentrations were similarly lower with OB + and OB −, but were significantly lower with JS − and WB − than in the corresponding coprophagy + group. In all groups digesta butyrate concentrations were reduced by coprophagy prevention. Pools of total VFA, acetate and butyrate in the digesta were highest with JS. Pools of total VFA in digesta were highest with JS +, OB + and OB − and lowest with WB + and WB −. The propionate pool was highest with OB −, intermediate with OB +, and equally low in all other groups. The pool of butyrate was highest with JS + and lowest with OB −. Effects of oats and wheat on plasma cholesterol in the rat do not seem to be mediated through faecal re-ingestion.

1. Three wethers fitted with permanent rumen cannulas were given a pelleted roughage–concentrate ration containing 45% ground barley straw as roughage. The animals were given 1 kg of the diet at 08.00 h and 500 g at 17.00 h.

2. The measurements made to determine the buffering components of the rumen fluid were: rumen pH, buffering capacity value (β), total volatile fatty acids (VFA), CO2 and inorganic phosphorus concentrations. β was expressed as the quantity of titrant (m-equiv./l.) required to bring about 2 unit changes in rumen fluid pH. The pH ranges in which the β values were measured were 4–6, 5–7 and 6–8. All measurements were made at hourly intervals over a 9 h period commencing 1 h before feeding.

3. Rumen pH values were high before feeding, decreased to a minimum 2 h after feeding and then increased to approximately the original values. The concentrations of total VFA followed an inverse pattern. There was diurnal variation in the concentrations of CO2 but inorganic P levels varied very little throughout the day.

4. Significant correlations were obtained between VFA, inorganic P and CO2 levels and the β values. It would appear that the bicarbonate and phosphate buffers were not effective in regulating the rumen pH when sheep were given a roughage–concentrate ration.

1. Preschool children and schoolchildren from a rural area in the northeast of Thailand were compared with children from urban areas for prealbumin, albumin, transferrin, α1B-glycoprotein, the acute-phase reactants α1-acid glycoprotein, α1-antichymotrypsin. haptoglobin and the proteinase inhibitors α1-protease inhibitor (AlPI) as well as α1-macroglobulin (α2M). Urinary urea nitrogen: creatinine (U-C) as well as haemoglobin were also determined. Village preschool children were grouped according to their SD score for weight-for-height based on the (US) National Center for Health Statistics (1976) standard into a normal group with SD scores of ≥ −1.99 and an undernourished group with SD scores of ≤ −2.00.

2. There was no significant difference between the normal and the undernourished groups of preschool children for any of the factors measured.

3. Haemoglobin and prealbumin concentrations of preschool children were lower in the rural children than in the urban preschool children from Bangkok.

4. In rural schoolchildren haemoglobin was lower but albumin, transfemn, α1B-glycoprotein and haptoglobin were higher than in urban schoolchildren from the provincial town of Khon Kaen.

5. Serum concentrations of the proteinase inhibitors AlPI and α2M were significantly higher in the rural children than in the urban children.

6. U-C values were lower in rural schoolchildren compared with urban schoolchildren.

The effect of the weaning diet on the subsequent colonic metabolism of bran and pectin in the adult rat has been investigated. Feeding a fibre-reduced diet on its own or supplemented with bran (WB) and pectin (P) from weaning (fibre-reduced (weaning)) was compared with introducing the same diet to age-matched rats reared on a standard laboratory diet from weaning (fibre-reduced (6 weeks)). The effects of the diets on colonic metabolism were measured by wet and dry caecal contents and stool weights, caecal sac weight, and caecal and faecal short-chain fatty acids (SCFA). Final body-weights were greater for fibre-reduced (6 weeks) and fibre-reduced (6 weeks)+P groups, but not fibre-reduced (6 weeks)+WB, than those of the fibre-reduced (weaning) rats. Rats fed on fibre-reduced (6 weeks) diet had a higher total caecal SCFA content than fibre-reduced (weaning) control rats. Fibre-reduced (weaning)+P-fed rats had a threefold higher caecal concentration of both propionate and butyrate than the matched fibre-reduced (6 weeks)+P group. Fibre-reduced (weaning)+WB animals had a significantly higher butyrate caecal concentration compared with their matched fibre-reduced (6 weeks)+WB group. Fibre-reduced (weaning)+P-fed rats had a lower faecal output than the fibre-reduced (6 weeks)+P rats. There was no difference in faecal output in rats fed on either fibre-reduced (6 weeks)+ WB or fibre-reduced (weaning)+WB. The faecal concentration of SCFA was in general higher in the rats fed on fibre-reduced (weaning) alone, +P, or +WB than in those fed on fibre-reduced (6 weeks) alone, +P or +WB. Faecal output of total and individual SCFA was increased on the fibre-reduced (weaned)+WB diet compared with fibre-reduced (6 weeks)+WB-fed animals. The diet at weaning may be important in determining the pathways of caecal bacterial metabolism in the adult rat. In studying the effect of a dietary fibre on caecal metabolism and faecal output, when the diet is changed appears to be important

An in vitro dialysis method was employed to determine the effect on the Fe and Zn absorption of the type (beef, pork and soyabean) and the amount (10 and 30 g/kg) of protein present. In addition, the effects of low- and high-molecular-weight (LMW and HMW respectively) digestion products were investigated. After in vitro digestion and dialysis a lower percentage of N, Fe and Zn was found in the LMW fractions from beef, pork and soyabean proteins when the protein level was increased from 10 to 30 g/kg; the higher level of protein being associated with a lower percentage of hydrolysed protein. The highest percentage levels of intrinsic Fe were always found in the HMW fractions, independent of the type and the level of proteins studied, while in the case of Zn, both HMW and LMW fractions gave similar values. An interaction was found between inorganic Zn and non-haem-Fe. The addition of inorganic Zn (10 μg/ml) caused a significant decrease in the in vitro availability of Fe from soyabean protein, while it did not affect the dialysability of intrinsic Fe from beef and pork proteins. Our results showed that the type and the level of the protein had a positive effect on the dialysability of extrinsic Fe. We postulate that the effect of a protein on the absorption of extrinsic Fe could be accounted for by free amino acids and/or small peptides released during the digestion process and also by the undigested or partially-digested HMW fractions of hydrolysed proteins which could play a fundamental role in the availability of this essential element.

We questioned whether polyamines coming from the diet or produced by intestinal microflora or by intracellular metabolism influence intestinal functions. Therefore, we compared pathogen-free rats and germ-free rats receiving a diet with low polyamine content and either treated or not treated with difluoromethylornithine (DFMO) and/or methylglyoxal bis (guanylhydrazone) (MGBG). Wet weight, protein content, DNA content, sucrase (EC3.2.1.48), maltase (EC 3.2.1.20) and lactase (EC 3.2.1.23) specific activities, amounts of putrescine, spennidine and spemine were measured in the mucosa of the proximal and distal intestine. Body weight was also determined. Rats without microflora had a higher specific activity of maltase and higher amounts of spermidine and spermine but lower lactase specific activity than pathogen-free animals; the low-polyamine diet given to gem-free rats had little effect on the functional variables measured (decrease of maltase and lactase specific activities) and did not modify the amounts of polyamines. DFMO and/or MGBG administered to germ-free rats receiving a low-polyamine diet induced modifications of most of the variables studied. Body weight and wet weight of proximal and distal intestine decreased, disaccharidase specific activities decreased, and amounts of polyamines changed according to the inhibitor used. Thus, our results showed that the deprivation of polyamine supply from microflora or from the diet failed, under our experimental conditions, to affect the intestinal properties analysed but exogenous and endogenous polyamine restriction altered general properties of the organism as well as intestinal functions.

Lower prenatal exposure to n-3 PUFA relative to n-6 PUFA has been hypothesised to influence allergy development, but evidence remains largely inconsistent. In the Dutch Maastricht Essential Fatty Acid Birth (MEFAB) (n 293) and Greek RHEA Mother–Child (n 213) cohorts, we investigated whether cord blood phospholipid PUFA concentrations are associated with symptoms of wheeze, asthma, rhinitis and eczema at the age of 6–7 years. Information on allergy-related phenotypes was collected using validated questionnaires. We estimated relative risks (RR) and 95 % CI for associations of PUFA with child outcomes using multivariable generalised linear regression models. In pooled analyses, higher concentration of the n-3 long-chain EPA and DHA and a higher total n-3:n-6 PUFA ratio were associated with lower risk of current wheeze (RR 0·61; 95 % CI 0·45, 0·82 per sd increase in EPA+DHA and 0·54; 95 % CI 0·39, 0·75 per unit increase in the n-3:n-6 ratio) and reduced asthma risk (RR 0·50; 95 % CI 0·31, 0·79 for EPA+DHA and 0·43; 95 % CI 0·26, 0·70 for the n-3:n-6 ratio). No associations were observed for other allergy-related phenotypes. The results were similar across cohorts. In conclusion, higher EPA and DHA concentrations and a higher n-3:n-6 fatty acid ratio at birth were associated with lower risk of child wheeze and asthma. Our findings suggest that dietary interventions resulting in a marked increase in the n-3:n-6 PUFA ratio, and mainly in n-3 long-chain PUFA intake in late gestation, may reduce the risk of asthma symptoms in mid-childhood.

Absorption from wheat intrinsically and extrinsically labelled with 67Zn and extrinsically labelled with 65Zn was measured from 67Zn faecal excretion and 6sZn whole-body retention in rats. There were significant differences between the extrinsically- and intrinsically-labelled 67Zn (P < 0·001), but not between the extrinsically-labelled 65Zn and intrinsically-labelled 67Zn. The effect of chicken meat on the absorption of Zn from intrinsically-labelled wheat was also studied in the rat. Mean Zn absorption from wheat and chicken meat fed separately was 18·5 and 68·2% respectively, and from a mixture of the two containing the same level of Zn was 50·1%. The apparent absorption of Zn from the composite meal was significantly higher than predicted from the results of the foods on their own (P < 0·001).

There is a relationship between the fatty acid profile in skeletal muscle phospholipids and peripheral resistance to insulin in adults, but similar data have not been reported in infancy and childhood. The objective of this study was to investigate the fatty acid composition of skeletal muscle and adipose tissue across the paediatric age range. The fatty acid profile of skeletal muscle phospholipids and adipose tissue triacylglycerols was analysed in ninety-three healthy Spanish infants and children distributed into four groups: group 1 (0 to <2 years, n 10); group 2 (2 to <5 years, n 41); group 3 (5 to <10 years, n 24); group 4 (10 to 15 years, n 18). In skeletal muscle phospholipids, oleic acid (18: 1n-9cis) content decreased significantly whereas that of linoleic (18: 2n-6) acid increased significantly with age (P for trend <0·01). In adipose tissue, the contents of triacylglycerol and linoleic acid increased significantly across the paediatric age range (P for trend <0·01), whereas dihomo-γ-linolenic (20: 3n-6) and arachidonic (20: 4n-6) showed significant differences between groups. The variations in fatty acid composition observed with age indicated an imbalance in dietary n-3/n-6 long-chain PUFA.

We aimed to examine the contribution of blood lipids to the association between BMI and blood pressure (BP) in children with overweight and obesity. Data were collected in elementary and high schools of Chaoyang District, Beijing, China in 2012. Participants’ weight, height, BP and fasting plasma lipid profile were measured by standard protocols. Mediation analysis was used to examine the mediation role of blood lipids on the relation between BMI and BP, with age included as a covariate. We found that in boys 8·29 % (mediation effect=0·106, P=0·012) of the association between BMI and systolic BP was mediated through TAG. TAG mediated 12·53 % (mediation effect=0·093, P=0·018) and LDL-cholesterol mediated 7·75 % (mediation effect=0·57, P=0·046) of the association between BMI and diastolic BP was mediated by TAG and LDL-cholesterol, respectively. However, blood lipids did not show the mediation effect in girls. Our findings suggested that there was a sex difference in the contribution of blood lipids to the association between BMI and BP. Controlling TAG or LDL-cholesterol may be beneficial for reducing the risk of the BMI-related high BP in overweight boys; however, this outcome is not the case when controlling TAG or LDL-cholesterol in girls. This study may provide clues to explore the underlying mechanism of the association between obesity and hypertension.
