Prothymosin alpha (PTA) was detected by immunocytological and biochemical methods in oocytes at different stages of oogenesis, and in early embryos of the amphibian Bufo arenarum. In all cases PTA was detected in the nucleus and was absent from the cytoplasm. This indicates that this protein could act at the level of regulating transcription. Western blots were carried out using polyclonal antibodies with extracts of embryos at different stages of development from early fertilisation up to neural tube. With this method PTA was detected in all the samples under study.

Echinoid embryos of indirect developers comprise three tiers at the 16-cell stage: eight mesomeres, four macromeres and four micromeres. Micromeres differentiate autonomously into skeletogenic mesenchyme cells and induce adjoining macromeres to form a vegetal plate, resulting in archenteron invagination (Ransick & Davidson, 1995). It has been shown that micromeres have the potential to induce archenteron differentiation of presumptive ectoderm (Hörstadius, 1939).

Direct development, in which the larval stages are more or less abbreviated, has evolved in several phylogenetic lineages of echinoids. In most of them, the fourth cleavage of the embryos produces 16 blastomeres of almost the same size (Raff, 1987). Because of their indistinguishability, the inductive potential of vegetal-most blastomeres of direct developers remained to be studied.

The 16-cell stage embryos of the direct-developing sand dollar, Peronella japonica, are an exception and produce three tiers, the same as indirect developers. The embryos develop into two-armed pluteus-like larvae and metamorphose within 3 days after fertilisation without feeding. The micromeres of the species had been shown to differentiate into the skeletogenic mesenchyme cells (Amemiya & Arakawa, 1996). In the present study, we investigated the potential of the micromeres to induce or promote archenteron invagination in Peronella japonica.

Serine/threonine protein phosphatases expected to participate in the process of signal transduction, cell movement such as cell division and gene expression (Kinoshita et al., 1990; Healy et al., 1991; Mayer-Jaekel et al., 1993; Mumby & Walter, 1993), are classified into type 1 (PP1), type 2A (PP2A), type 2B and type 2C in mammalian cells. PP1 and PP2A are known to be strongly inhibited by okadaic acid (OA) (Tachibana et al., 1981; Bialojan Takai, 1988), a polyether fatty acid isolated from the marine sponge Halicondria okadai (Haystead et al., 1989). OA is also known to inhibit PP2A at lower concentrations than that to block PP1 in mammalian cells, but does not inhibit the activities of other phosphatase species (Ishihara et al., 1989).

The p-nitrophenyl phosphate (pNPP) splitting activity in the extract obtained from eggs of the sea urchin Hemicentrotus pulcherrimus was found to be inhibited by OA and calyculin A (CLA), potent inhibitors of PP1 and PP2A. OA-sensitive phosphatases are known to catalyse pNPP splitting (Takai & Mieskes, 1991), in the same manner as other OA-insensitive phosphatases.

Four peaks of the pNPP splitting activity were obtained by QAE-Toyopearl chromatography in the extract of sea urchin eggs. In two of these four peaks, pNPP splitting reactions were strongly inhibited by OA and CLA at quite low concentration. High sensitivities of the pNPP splitting reaction to OA and CLA in these two peaks suggest that pNPP splitting results from the reaction catalysed by PP2A. The molecular masses of proteins exhibiting OA-sensitive pNPP splitting activities in these two peaks were found to be about 160 kDa by Superdex 200HR, and were similar to that of mammalian PP2A trimeric holoenzyme. By immunoblot analyses with anti-human PP2A catalytic subunit antibody, an immunoreactive 36 kDa protein was found by SDS-PAGE in a peak of OA-sensitive pNPP splitting activity obtained by QAE-Toyopearl chromatography. Sea urchin eggs have at least two PP2A-like enzymes with similar molecular masses to that of mammalian PP2A, and one of them contains human-type catalytic subunit.

Peronella japonica, a direct developer, exhibits certain peculiar features during development, particularly heterochrony, a change in the relative timing of expression among tissues and organs. One of the important heterochronical changes in the species was found in the development of the amniotic cavity, a component of an adult rudiment. In indirect developers the amniotic cavity is formed on the left side of the larval body in the late pluteus stage. In P. japonica the organ is formed at the gastrula stage in the region located on the midline of the larval body.

In the present study, the ability of partial embryos isolated from 8- or 16-cell stage embryos of P. japonica to differentiate an amniotic cavity was investigated to assess the regulative potential of a direct developer.

The embryos were dissected at 8-cell stage with a glass needle to obtain half embryos. Some of the half embryos were further divided into four blastomeres to obtain mesomere pairs. Each half embryo and blastomere that did not form micromeres but divided equally during the next cleavage was identified as an animal cap and presumptive mesomere pair. Isolated animal caps and mesomere pairs were cultured, and differentiation of the amniotic cavity was examined at 24 and 48 h after fertilisation, when the organ in the normal embryos had already completed differentiation.

Cell division and motility are generated by the cytoskeletal structures contained in networks of actin filaments and microtubules. Among the Ca2+-activated protein kinases, protein kinase C (PKC) can be activated with tumor promoter, 12-O-tetradecanoyl phorbol-13-acetate (TPA) (Kikkawa et al., 1983), which induces alterations in the cytoskeleton of cultured cells. To study the regulation of the cytoskeleton by PKC, we investigated the formation of actin filaments, microtubules, changes in cell shape and Ca2+ signalling after treatment with TPA in unfertilised eggs of the sea urchin Hemicentrotus pulcherrimus.

Formation of actin filaments and microtubles was induced in the eggs by the TPA treatment. When the eggs were treated with an inactive TPA, 4α-PMA, these cytoskeletal changes did not occur. Moreover, elongation of actin filaments was inhibited by the Ca2+ chelating agent BAPTA, but microtubule organisation was unaffected by intracellular Ca2+ chelation in the TPA-treated egg. TPA is known to induce alkalisation in sea urchin eggs (Swann & Whitaker, 1985) and when the eggs are treated with TPA in Na+-free seawater, the rise in intracellular pH (pHi) is depressed. However, elongation of actin filaments and microtubules occurred under these conditions. An increase in pHi is considered to be essential for triggering the bundle of actin filaments (Begg et al., 1982) and microtubule assembly (Schatten et al., 1992). These results show that actin and microtubule assemblies require activation of PKC, and that the formation of actin and microtubules may be related to the type of PKC isoform: Ca2+-dependent or -independent. Moreover, this mechanism is essentially independent of cytoplasmic alkalisation.

Protein phosphorylation is highly coupled with sperm motility activation in several animal species. The micro-tubule based flagellar motor protein, dynein, is a candidate for a phosphoprotein related to sperm activation in many animal species (Morisawa & Hayashi, 1985; Hayashi et al., 1987; Dey & Brokaw, 1991; Stephens & Prior, 1992; Inaba et al., 1998, 1999). Sperm motility of the ascidians Ciona intestinalis and C. savignyi is activated by a factor derived from unfertilised eggs named sperm activating and attracting factor (SAAF). SAAF elevates the intracellular cyclic AMP (cAMP) level by a mechanism dependent on membrane hyperpolarisation and extracellular Ca2+ (Yoshida et al., 1994; Izumi et al., 1999). Experiments using demembranated Ciona sperm showed that cAMP is required prior to ATP for the activation of axonemal movement (Opreska & Brokaw, 1983; Morisawa et al., 1984; Brokaw, 1985; Dey & Brokaw, 1991; Chaudhry et al., 1995) and that many sperm flagellar proteins including dynein light chain are phosphorylated during incubation of demembranated sperm with ATP and cAMP (Dey & Brokaw, 1991). However, there is no evidence of which proteins are phosphorylated during the SAAF-dependent activation of Ciona sperm motility.

Primary spermatocytes originating from prepubertal mouse testes were electrofused to metaphase II (MII)-stage oocytes, enucleated either by the conventional micromanipulation method or by chemical treatment with etoposide and cycloheximide. These experiments were followed by assessment of morphological changes in transferred nuclei using light microscopy, by chromosomal analyses and by screening of hybrids for the presence or absence of DNA synthesis using anti-bromodeoxyuridine antibody and immunofluorescence staining of the hybrids. The results show differences between the two types of ooplasts in susceptibility to activation stimuli. However, when activated, both types of ooplasts gave rise to hybrids of similar morphology. From 35.3% to 63% of activated hybrids originating from chemically or microsurgically enucleated oocytes, respectively, contained one large pronucleus in cytoplasm, 62% or 31.6% hybrids from those two groups, respectively, possessed two smaller pronuclei and a few contained three or four pronuclei. No DNA synthesis was detected in any hybrid containing one or more pronuclei. The chromosome spreads of hybrids with premature chromosome condensation (PCC) morphology (before activation) show that most of the hybrids had a diploid (2n) number of chromosomes. The nature and regularity of the cell division cycle in the hybrids are discussed.

The ultrastructure of in vivo-produced ovine embryos, at the morula, early blastocyst and late blastocyst stages, was evaluated using transmission electron microscopy. Embryonic cells were characterized by the presence of intact intercellular junctions, numerous mitochondria, smooth endoplasmic reticulum cisternae and light vesicles. Polyribosomes, rough endoplasmic reticulum cisternae, secondary lysosomes, Golgi complexes and lipid droplets were also observed in the cytoplasm. The nucleus was well defined and organized, with an intact envelope rich in nuclear pore complexes, and one or more reticular nucleoli. Microvilli were present in external blastomeres of morulae and became more abundant in trophectoderm cells of early and late blastocysts. Light vesicles seemed to be associated with small cisternae of Golgi and endoplasmic reticulum origin. These cisternae fused and created light vesicles with engulfed heterogeneous cytosolic structures, small cisternae and vesicles. Their labile membrane enabled them to rapidly coalesce into medium-sized vesicles that began to engulf mitochondria and lipid droplets, forming giant vacuoles mostly filled with fat. Incomplete matured secretory vesicles were observed to exocytose into the perivitelline space of morulae, whereas fully matured secretory vesicles appeared only in trophectoderm cells, being exocytosed into the blastocoelic cavity. These observations suggested that these endoplasmic-/Golgi-derived vesicles behave as active autophagic organelles presenting probably a maturation process from compact morulae to blastocyst.

Our study aimed to establish the response of Salminus franciscanus to hypophysation and describe the main morphological events of its embryonic process. Wild fish were captured in São Francisco River and selected broodstock (females: 66.4 ± 11.1 cm and 4.04 ± 2.32 kg; males: 58.3 ± 10.2 cm and 3.62 ± 1.12 kg) were kept at 26.1 ± 0.6°C for induction of final maturation/gamete release via the hypophysation technique. In females, two doses (0.8 and 5.6 mg/kg body weight) of crude pituitary extract of common carp (Cyprinus carpio) were administered with a 14 h interval. For males, a single dose (2.7 mg/kg body weight) of crude pituitary extract was applied at the same time as the females’ second dose. Oocytes and sperm were manually stripped 8 h after a females’ second hormonal dose. Fertilization was carried out using the dry method. Eggs were kept in funnel-type 60 L incubators at 24.3 ± 0.3°C and were analyzed and photographed every 10 min. After hormonal induction, 60% of females and 100% of males reacted positively and no broodstock mortality was recorded. The females released an average of 385.2 ± 78.4 g of oocytes and the fertilization rate observed was 50.4 ± 12.3%. The blastopore closure occurred at 7.5 h, somite formation at 12 h and hatching at 20 h post-fertilization. In general, the results of this study improve the understanding of the reproductive biology of dourado and confirm its potential for fish farming in the neotropical region.

This study aimed to investigate the ability of disulphide-less crotamine (dLCr) to complex DNA and to evaluate whether the DNA–dLCr complex is capable of improving transfection in bovine embryos. Three experiments were performed to: (i) evaluate the formation and stability of the DNA–dLCr complex; (ii) assess the dLCr embryotoxicity by exposure of bovine embryos to dLCr; and (iii) assess the efficiency of bovine embryo transfection after microinjection of the DNA–dLCr complex or green fluorescent protein (GFP) plasmid alone (control). DNA complexation by dLCr after 30 min of incubation at 1:100 and 1:50 proportions presented higher efficiency (P < 0.05) than the two controls: native crotamine (NCr) 1:10 and lipofectamine. There was no difference between DNA–dLCr 1:25 and the controls. The DNA–dLCr complexation was evaluated at different proportions and times. In all, at least half of maximum complexation was achieved within the initial 30 min. No embryotoxicity of dLCr was verified after exposure of in vitro fertilized embryos to different concentrations of the peptide. The effectiveness of dLCr to improve exogenous gene expression was evaluated by microinjection of the DNA–dLCr complex into in vitro fertilized zygotes, followed by verification of both embryo development and GFP expression. From embryos microinjected with DNA only, 4.6% and 2.8% expressed the GFP transgene at day 5 and day 7, respectively. The DNA–dLCr complex did not increase the number of GFP-positive embryos. In conclusion, dLCr forms a complex with DNA and its application in in vitro culture is possible. However, the dLCr peptide sequence should be redesigned to improve GFP expression.

We describe for the first time the cytogenetic characteristics of mouse ‘embryos’ obtained by oocyte fusion (oocyte fusion products; OFP). Our results indicate that, after fusion, meiosis II is resumed correctly, with extrusion of two haploid polar bodies, and that metaphase synchronisation of the two haploid sets and chromosome segregation during the first cleavage are also normal.

To investigate the roles of lncRNA deleted in lymphocytic leukaemia 1 (DLEU1) on migration and invasion of human trophoblast cells. Human chorionic trophoblast cell line HTR8/SVneo was cultured and transfected using lncRNA DLEU1 small interfering RNA. Real-time quantitative polymerase chain reaction was used to detect lncRNA DLEU1 expression. The activity of migration regulatory protein CDC42 was detected by western blot. The downstream miRNA targets of lncRNA and mRNAs targeted by corresponding miRNAs were respectively predicted using bioinformatics analyses. Compared with the control group, the expression of lncRNA DLEU1 in the small interfering RNA group was significantly decreased (P < 0.05). There was no significant change in cell proliferation capacity for transfected cells (lncRNA DLEU1 siRNA-1, P = 0.537; lncRNA DLEU1 siRNA-2, P = 0.384), but cell migration (lncRNA DLEU1 siRNA-1, P = 0.025; lncRNA DLEU1 siRNA-2, P = 0.019) and invasion (lncRNA DLEU1 siRNA-1, P = 0.0327; lncRNA DLEU1 siRNA-2, P = 0.021) was significantly reduced. CDC42 activity in the lncRNA DLEU1 knockdown group decreased and the phosphorylation of cofilin increased. Therefore, downregulation of lncRNA DLEU1 suppressed the migration and invasion of human trophoblast cells.

The aim of our study was to investigate the influence of vitrification on developmental rate and quality (total number of cells, number of blastomeres in inner cell mass (ICM) area, apoptotic index and embryo diameter) of transgenic (carrying an endogenous–hFVIII or exogenous–enhanced green fluorescent protein (EGFP) gene) rabbit embryos. EGFP-positive rabbit embryos were produced under in vitro conditions by the microinjection of foreign genes into the pronucleus of fertilized eggs. The transgenic rabbit embryos with the hFVIII gene were produced by mating homozygous transgenic rabbits and flushing at the single-cell stage. Developmental rate of vitrified/thawed transgenic embryos that reached hatching blastocyst stage (68.00% and 69.00%) and differed significantly (p < 0.001) from those in control embryos (100.00%). Significant difference (p < 0.05) was found in total cell counts between control (117.00 ± 36.00) and vitrified (141.00 ± 34.80) hFVIII-positive embryos. The higher proportion of ICM cells (32.00%) and greatest embryo diameter (130.85 ± 10.90) were found in the control group compared with the transgenic. Ratio of apoptotic cells was significantly higher (p < 0.01) in the control group (2.50%) and vitrified EGFP-positive embryos (2.90%) compared with the vitrified, hFVIII-positive group of embryos (0.70%). Our results demonstrate that neither gene microinjection itself, nor exogenous (EGFP) and endogenous (hFVIII) gene expression interferes with developmental rate and quality of rabbit embryos. However, a combination of microinjection and vitrification significantly decreases (p < 0.001) the survival rate of rabbit embryos.

Pretreatment of zona-free Chinese hamster (CH) oocytes with three kinds of lectin – concanavalin A (Con-A), phytohaemagglutinin-P (PHA) and wheat germ agglutinin (WGA) – was attempted in order to improve penetration by golden hamster (GH) spermatozoa in vitro. Con-A had no significant effect on penetration at 2 μg/ml, adequately facilitated oocyte-sperm fusion at 4 μg/ml, and caused excessive sperm binding and resultant severe polyspermy at 10 μg/ml. Neither PHA nor WGA had positive effects on sperm penetration at any concentrations (2–10 μg/ml) examined. Using the Con-A (4 μg/ml)pretreatment, high rates of interspecific fertilisation and subsequent chromosome analysis of hybrid 1-cell zygotes were achieved. Among 258 CH oocytes used, 212 (82.2%) were fertilised and 153 (72.2% of fertilised ova) developed to the first cleavage metaphase. Eventually, 132 CH-derived chromosome complements and 153 GH-derived ones were successfully karyoanalysed. Incidences of aneuploidy and structural anomaly were 3.1% and 2.3% in CH complements, and 1.4% and 6.5% in GH complements, respectively. These incidences were not significantly different from those obtained by intraspecific in vivo fertilisation, suggesting that our interspecific in vitro fertilisation system does not cause chromosome aberrations.

Zygotic genome activation (ZGA) is a critical event in early embryonic development, and thousands of genes are involved in this delicate and sophisticated biological process. To date, however, only a handful of these genes have revealed their core functions in this special process, and therefore the roles of other genes still remain unclear. In the present study, we used previously published transcriptome profiling to identify potential key genes (candidate genes) in minor ZGA and major ZGA in both human and mouse specimens, and further identified the conserved genes across species. Our results showed that 887 and 760 genes, respectively, were thought to be specific to human and mouse in major ZGA, and the other 135 genes were considered to be orthologous genes. Moreover, the conserved genes were most enriched in rRNA processing in the nucleus and cytosol, ribonucleoprotein complex biogenesis, ribonucleoprotein complex assembly and ribosome large subunit biogenesis. The findings of this first comprehensive identification and characterization of candidate genes in minor and major ZGA provide relevant insights for future studies on ZGA.

The global transition towards diets high in calories has contributed to 2.1 billion people becoming overweight, or obese, which damages male reproduction and harms offspring. Recently, more and more studies have shown that paternal exposure to stress closely affects the health of offspring in an intergenerational and transgenerational way. SET Domain Containing 2 (SETD2), a key epigenetic gene, is highly conserved among species, is a crucial methyltransferase for converting histone 3 lysine 36 dimethylation (H3K36me2) into histone 3 lysine 36 trimethylation (H3K36me3), and plays an important regulator in the response to stress. In this study, we compared patterns of SETD2 expression and the H3K36me3 pattern in pre-implantation embryos derived from normal or obese mice induced by high diet. The results showed that SETD2 mRNA was significantly higher in the high-fat diet (HFD) group than the control diet (CD) group at the 2-cell, 4-cell, 8-cell, and 16-cell stages, and at the morula and blastocyst stages. The relative levels of H3K36me3 in the HFD group at the 2-cell, 4-cell, 8-cell, 16-cell, morula stage, and blastocyst stage were significantly higher than in the CD group. These results indicated that dietary changes in parental generation (F0) male mice fed a HFD were traceable in SETD2/H3K36me3 in embryos, and that a paternal high-fat diet brings about adverse effects for offspring that might be related to SETD2/H3K36me3, which throws new light on the effect of paternal obesity on offspring from an epigenetic perspective.

We have recently reported that multiple aster formation after in vitro fertilization (IVF) was one of the factors that negatively affected the developmental competence of vitrified-warmed bovine matured oocytes, and that short-term culture of the post-warm oocytes with an inhibitor of Rho-associated coiled-coil kinase (ROCK) suppressed the multiple aster formation and improved the blastocyst yield. The present study was conducted to investigate whether increased multiple aster formation following IVF was involved in impaired developmental competence of stored ovary-derived bovine oocytes. Oocytes retrieved from 1-day stored ovaries had lower developmental potential to day 8 blastocysts when compared with those from fresh ovaries (37 versus 63%). Immunostaining of α-tubulin 10 h post-IVF revealed that a higher incidence of multiple aster formation occurred in oocytes retrieved from stored ovaries than from fresh ovaries (31 versus 15%). Treatment of post-in vitro maturated (post-IVM) oocytes with ROCK inhibitor for 2 h significantly suppressed the incidence of multiple aster formation (10 versus 32% in the control group). However, the suppression effect of ROCK inhibitor on multiple aster formation in IVM/IVF oocytes did not improve blastocyst yield from stored ovary-derived oocytes (41 versus 37% in the control group). These results suggested that the higher incidence of multiple aster formation by bovine ovary storage was not responsible for the decreased developmental competence of IVF oocytes.

The follicle-stimulating hormone (FSH) and its receptor regulate the quantity and quality of spermatozoa production. Several studies have analyzed the effect of single nucleotide polymorphisms (SNPs) in exon 10 of the FSH receptor (FSHR) on basic semen parameters without yet reaching a firm consensus. The aim of this study was to evaluate the effect of p.Thr307Ala and p.Asn680Ser polymorphisms in exon 10 of the FSHR gene, in infertile men, on intracytoplasmic sperm injection (ICSI) outcomes. This study was conducted between March 2019 and February 2020 on infertile couples undergoing ICSI at Al Hadi Laboratory and Medical Center, Lebanon. Couples with severe infertility factors that may impair gametogenesis/embryogenesis (e.g. advanced maternal age, premature ovarian failure, underwent gonadotoxic treatments, etc.) were excluded from the study. Semen and blood samples were collected from infertile men on the day of oocyte collection. Infertile men (n = 173) were screened for FSHR variants using polymerase chain reaction-restriction fragment length polymorphism. Moreover, fertilization rates, embryo quality, and pregnancy outcomes were evaluated. Higher sperm concentrations were found in the p.Thr307Ala group than the p.Thr307Thr (P < 0.01) and p.Ala307Ala (P < 0.05) groups. Furthermore, fertilization rate was significantly lower in the p.Ala307Ala genotype than in the p.Thr307Thr genotype (P < 0.05). We showed that FSHR variants in infertile men undergoing ICSI could affect sperm concentration, motility, and fertilization rates. Therefore, it will be important to confirm these results in further studies using a larger sample size.

Vertebrate pigmentation provides an ideal system for studying the intersections between evolution, genetics, and developmental biology. Teleost fish, with their accessible developmental stages and intense and diverse colours produced by chromatophores, are an ideal group for study. We set out to test whether Betta splendens is a good model organism for studying the evolution and development of diverse pigmentation. Our results demonstrate that B. splendens can be bred to produce large numbers of offspring with easily visualized pigment cells. Depending on the colour of the parents, there was variation in larval pigmentation patterns both within and between breeding events. In juveniles the developing adult pigmentation patterns showed even greater variation. These results suggest that B. splendens has great potential as a model organism for pigmentation studies.

It has been reported that the number of cells per embryo increases from the cleavage stage to the pluteus stage. Also, it has been reported that the number of cells per embryo from the early gastrula stage to the mid-gastrula stage increases very slightly (Mizoguchi, 1999). A detailed analysis of cell proliferation during this period would thus seem to be necessary.

On the other hand, Roccheri et al. (1997) reported spontaneous apoptosis at the early pluteus stage, especially in the regions of arm and intestine. However, it is unknown whether apoptosis occurs before the pluteus stage.

Using the Tumor, Neuro and/or Cardio TACS in situ apoptosis detection kit (terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labelling: TUNEL method, Trevigen, USA), the Comet assay kit (Trevigen, USA) and DAPI (4,6-diamidino-2-phenylinodole dihydrochloride) staining, we investigated the distribution of apoptosis-like cells in sea urchins during early embryogenesis to clarify the relationship between cell death and morphogenesis.

The embryos of the sea urchin Hemicentrotus pulcherrimus were used in the present study. Three methods of detection of apoptosis signals in the embryos were performed: (1) defects in chromosomes and punctured nuclear envelopes of cells were detected by DAPI staining; (2) apoptosis-like cells were stained a brown colour by the TUNEL method; (3) some clear tails which detected the fragmentation of DNA were found by the Comet assay. These signals indicate apoptosis-like cells.
