Gamma-aminobutyric acid (GABA) is the most prevalent inhibitory neurotransmitter in the vertebrate brain. It can exert its influence either as GABAergic projection pathways or as local interneurons, which play an essential role in many visual functions. However, no GABAergic visual pathways have been studied in frogs so far. In the present study, GABAergic pathways in the central visual system of Rana pipiens were investigated with double-labeling techniques, combining immunocytochemistry for GABA with Rhodamine microspheres for retrograde tracing. Three GABAergic visual pathways were identified: (1) a retino-tectal projection, from retina to the contralateral optic tectum (OT); (2) an ipsilateral projection from the nucleus of the basal optic root (nBOR) to the pretectal nucleus lentiformis mesencephali (nLM); and (3) a second-order pathway from the nucleus isthmi (NI), bilaterally, to the optic tectum. These results indicate that GABA is involved in both first-order (retina to optic tectum) as well as second-order (nucleus isthmi to optic tectum) visual projections in Rana pipiens, and may play a major role in mediating visuomotor reflexs such as optokinetic nystagmus or other visually guided behaviors.

Luminance signals mediated by the magnocellular (MC) pathway play an important role in vernier tasks. MC ganglion cells show a phase advance in their responses to sinusoidal stimuli with increasing contrast due to contrast gain control mechanisms. If the phase information in MC ganglion cell responses were utilized by central mechanisms in vernier tasks, one might expect systematic errors caused by the phase advance. This systematic error may contribute to the contrast paradox phenomenon, where vernier performance deteriorates, rather than improves, when only one of the target pair increases in contrast. Vernier psychometric functions for a pair of gratings of mismatched contrast were measured to seek such misestimation. In associated electrophysiological experiments, MC and parvocellular (PC) ganglion cells' responses to similar stimuli were measured to provide a physiological reference. The psychophysical experiments show that a high-contrast grating is perceived as phase advanced in the drift direction compared to a low-contrast grating, especially at a high drift rate (8 Hz). The size of the phase advance was comparable to that seen in MC cells under similar stimulus conditions. These results are consistent with the MC pathway supporting vernier performance with achromatic gratings. The shifts in vernier psychometric functions were negligible for pairs of chromatic gratings under the conditions tested here, consistent with the lack of phase advance both in responses of PC ganglion cells and in frequency-doubled chromatic responses of MC ganglion cells.

Deuteranomalous trichromacy, which affects medium-wavelength-sensitive cones, is more common than protanomalous trichromacy, which affects long-wavelength-sensitive cones. The aim of the present work was to test the extent to which these two kinds of anomalous trichromacy affect surface-color judgments in the natural world. Simulations of 18 natural scenes under different daylight illuminants were presented on a high-resolution color monitor to 7 deuteranomalous, 7 protanomalous, and 12 normal trichromatic observers, who had to discriminate between reflectance and illuminant changes in the images. Observers' ability to judge surface color was quantified by a standard color-constancy index. Deuteranomalous trichromats performed as well as normal trichromats, but protanomalous trichromats performed more poorly than both. The results are considered in relation to the spectral coverage of cones, rod intrusion, and the characterization of anomalous trichromacy by the Rayleigh match.

How is chromatic induction affected by perceptual grouping? Chromatic induction has been studied extensively, as has grouping, but only a small number of experiments have connected them. Even fewer reports go beyond weakly controlled qualitative observations. We report here a new and substantial color shift caused by perceptual grouping: a shift in appearance due to chromatic induction in one part of the visual field occurs also in a separate region that belongs to the same group. The color appearance of a test square within various surrounds was measured by asymmetric matching. The test square was at the center of an “hourglass” structure formed by other elements in the surround. The test shifted in color appearance toward the appearance of these other elements, whose color was affected by local chromatic induction. Control experiments ruled out as explanations (1) direct chromatic induction from the other elements into the test area, and (2) the influence of background light occluded by introducing the elements forming the hourglass.

The purpose of the present study was to examine the contributions made by cat posterior parietal cortex to the analyses of the relative position of objects in visual space. Two cats were trained on a landmark task in which they learned to report the position of a landmark object relative to a right or left food-reward chamber. Subsequently, three pairs of cooling loops were implanted bilaterally in contact with visuoparietal cortices forming the crown of the middle suprasylvian gyrus (MSg; architectonic area 7) and the banks of the posterior-middle suprasylvian sulcus (pMS sulcal cortex) and in contact with the ventral-posterior suprasylvian (vPS) region of visuotemporal cortex. Bilateral deactivation of pMS sulcal cortex resulted in a profound impairment for all six tested positions of the landmark, yet bilateral deactivation of neither area 7 nor vPS cortex yielded any deficits. In control tasks (visual orienting and object discrimination), there was no evidence for any degree of attentional blindness or impairment of form discrimination during bilateral deactivation of pMS cortex. Therefore, we conclude that bilateral cooling of pMS cortex, but neither area 7 nor vPS cortex, induces a specific deficit in spatial localization as examined with the landmark task. These observations have significant bearing on our understanding of visuospatial processing in cat, monkey, and human cortices.

The light-driven pupil mechanism, consisting of an assembly of mobile pigment granules inside the photoreceptor cells, has been investigated by in vivo reflection microspectrophotometry in wild type (WT) Drosophila and in the photoreceptor mutants inaC and trp. The pupillary response of a dark-adapted WT eye to a step in light is a monophasic reflectance increase reaching a plateau after ca. 15-s light adaptation. This reflectance change is due to photoreceptor pigment granules that accumulate near the tips of the rhabdomeres under light adaptation and that are withdrawn towards the periphery in the dark (Franceschini & Kirschfeld, 1976). The step response of the pupil mechanism of inaC is triphasic. Strikingly, the reflectance level at light onset is distinctly higher than that in WT, due to a partly aggregated state of the photoreceptor pigment granules near the rhabdomere tips that persists in the dark-adapted state, in line with direct calcium measurements of Peretz et al. (1994b). The step response of the pupil mechanism of inaC is slightly elevated compared to that of WT. The step response in trp is a transient, biphasic reflectance change, approximating a log normal function. This function is also a good approximation of the pulse response in WT and inaC. The intensity range of pupillary sensitivity is about 4 log unit. The range of inaC compared to that of WT is slightly (≈0.5 log unit) shifted towards lower intensities, but that in trp is strongly shifted to higher intensities (≈2.5 log unit). The results can be interpreted with the present knowledge of the primary steps in fly phototransduction and the hypothesis that the local intracellular calcium concentration determines the position of the pigment granules, and hence are in line with the notion that the pupil can be used as a qualitative Ca2+ probe.

Recent studies have demonstrated the defocus sign-dependent, bidirectional gene expression regulation of bone morphogenetic proteins, BMP2, 4 and 7 in chick RPE. In this study, we examined the effects of imposed positive (+10 D) and negative (−10 D) lenses on the gene expression of these BMPs and BMP receptors (BMPR1A, BMPR1B, BMPR2) in chick retina and choroid after monocular lens treatment for 2 or 48 h, as indicators of the roles of retinal and choroidal BMPs and receptors in postnatal eye growth regulation. In retina, although all genes were expressed, neither +10 nor −10 D lenses, worn for either 2 or 48 h, significantly altered gene expression. In contrast, treatment-related differential gene expression was detected in the choroid for both BMPs and their receptors, although interestingly, with the +10 D lens, BMP2 was up-regulated by 156.7 ± 19.7% after 2 h, while BMPR1A was down-regulated to 82.3 ± 12.5% only after 48 h. With the −10 D lens, only the gene expression of BMPR1B was significantly altered, being up-regulated by 162.3 ± 21.2% after 48 h. Untreated birds showed no difference in expression between their two eyes, for any of the genes examined. The finding that retinal gene expression for BMP2, 4, 7 and their receptors are not affected by short-term optical defocus contrasts with previous observations of sign-dependent expression changes for the same genes in the RPE. The latter changes were also larger and more consistent in direction than the choroidal gene expression changes reported here. The interrelationship between these various changes and their biological significance for eye growth regulation are yet to be elucidated.

The organization of the cortex located immediately anterior to the second visual area (V2), i.e., the third tier visual cortex, remains controversial, especially in New World primates. In particular, there is lack of consensus regarding the exact location and extent of the lower visual quadrant representation of the third visual area V3 (or ventrolateral posterior –VLP – of a different nomenclature). Microelectrode and connectional mapping studies have revealed the existence of an upper visual quadrant representation abutting dorsal V2 anteriorly, and bordered medially and laterally by representations of the lower visual quadrant. It remains unclear whether these lower field regions are both part of a single area V3, which is split into two patches by an interposed region of upper field representation, or whether they are the lower field representations of two different areas, the dorsomedial area (DM) and area V3/VLP, respectively. To address this question, we quantitatively analyzed the patterns of corticocortical afferent connections labeled by tracer injections targeted to these two lower field regions in the dorsal aspect of the third tier cortex. We found different inter-areal connectivity patterns arising from these two regions, strongly suggesting that they belong to two different visual areas. In particular, our results indicate that the dorsal aspect of the third tier cortex consists of two distinct areas: a full area DM, representing the lower quadrant medially, and the upper quadrant laterally, and the lower quadrant representation of V3/VLP, located laterally to upper field DM. DM is predominantly connected with areas of the dorsal visual stream, and V3/VLP with areas of the ventral stream. These results prompt further functional investigations of the third tier cortex, as previous studies of this cortical territory may have pooled response properties of two very different areas into a single area V3.

Whole-cell recordings were obtained from retinal ganglion cells of the tiger salamander (Ambystoma tigrinum) in a superfused slice preparation to evaluate contributions of NMDA (N-methyl-D-aspartate) and KA/AMPA (kainate/α-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid) receptors to excitatory postsynaptic potentials (EPSPs) of retinal ganglion cells. Synaptic activation of retinal ganglion cells was achieved through the use of a brief pressure pulse of hyperosmotic Ringer (Ringer + sucrose) delivered through a microelectrode visually placed in the inner plexiform layer while whole-cell recordings were obtained from adjacent cells in the ganglion cell layer. Separation of NMDA and KA/AMPA excitatory postsynaptic currents (EPSCs) was achieved through the application of the antagonists NBQX and D-AP7, while inhibitory currents were blocked by strychnine and picrotoxin. Simple addition of the two independent EPSCs showed, most often, that the sum of the KA/AMPA and NMDA currents was less than the control response, but in some cases the sum of the two currents exceeded the magnitude of the control response. Neither result was consistent with expectations based on voltage-clamp principles and the assumption that the two currents were independent; for this reason, we considered the possibility of nonlinear interactions between KA/AMPA and NMDA receptors. Computer simulations were carried out to evaluate the summation experiments. We used both an equivalent cylinder model and a more realistic, compartmental model of a ganglion cell constrained by a passive leakage conductance, a linear KA/AMPA synaptic current, and a nonlinear NMDA current based on the well-known, voltage-sensitive Mg2+ block. Computer simulation studies suggest that the hypo- and hyper-summation of NMDA and KA/AMPA currents, observed physiologically, can be accounted for by a failure to adequately space clamp the neuron. Clamp failure leads to enhanced NMDA currents as the ion channels are relieved of the Mg2+ block; their contribution is thus exaggerated depending on the magnitude of the conductance change and the spatial location of the synaptic input.

It has been shown that the visual acuity loss experienced by the deprived eye of kittens following an early period of monocular deprivation (MD) can be alleviated rapidly following 10 days of complete darkness when imposed even as late as 14 weeks of age. To examine whether 10 days of darkness conferred benefits at any age, we measured the extent of recovery of the visual acuity of the deprived eye following the darkness imposed on adult cats that had received the same early period of MD as used in prior experiments conducted on kittens. Parallel studies conducted on different animals examined the extent to which darkness changed the magnitude of the MD-induced laminar differences of the cell soma size and immunoreactivity for the neurofilament (NF) protein in the dorsal lateral geniculate nucleus (dLGN). The results indicated that 10 days of darkness imposed at one year of age neither alleviated the acuity loss of the deprived eye induced by an earlier period of MD nor did it decrease the concurrent lamina differences of the soma size or NF loss in the dLGN.

The ground level visual field of each eye of the leopard frog includes the entire ipsilateral 180-deg field and approximately 60 deg into the frontal contralateral field. When one eye is covered with an opaque patch, a frog responds to prey stimuli over the entire field of the other eye. Nevertheless, when one optic nerve is cut, the animal responds to prey in the ipsilateral hemifield of the connected eye, but only responds as far as about 30 deg past the frontal midline. If one optic tract is cut, the animal does not respond to prey past the frontal midline. We hypothesized that the responses past the frontal midline might be mediated by input from contralaterally projecting isthmotectal fibers. These fibers originate in the nucleus isthmi, a posterior midbrain structure. We found that when we placed an opaque patch over one eye and either ablated the ipsilateral nucleus isthmi, or cut crossing isthmotectal fibers in the optic chiasm, or blocked input to nucleus isthmi by ablating the ipsilateral tectal lobe, animals did not respond to prey stimuli past the frontal midline. We found that when we placed an opaque patch over one eye and cut crossing optic fibers in the anterior part of the optic chiasm (sparing crossing isthmotectal fibers), animals responded to prey stimuli in the nasal half of the seeing eye's contralateral frontal field. Our results suggest that contralaterally projecting isthmotectal fibers enable the frog to respond to stimuli past the frontal midline. We suggest a one-dimensional model of how nucleus isthmi influences tectal function.

We have used antibodies raised against a cytoskeletal protein, microtubule-associated protein 1A (MAP 1A), to stain adult rat retina. In cryostat and polyethylene glycol-embedded radial sections, the fiber layer, ganglion cell layer, and inner plexiform layer were highly immunoreactive, a finding that suggested that the ganglion cell somata, axons, and dendrites were recognized by these antibodies. Retrograde labeling of retinal cell somata from the superior colliculus and dorso-lateral geniculate nucleus to identify ganglion cells showed colocalization of the tracer and immunoreactive cells. Double labeling with nuclear stains revealed that many cells in the ganglion cell layer, which are likely displaced amacrine cells, were not recognized by these antibodies. Furthermore, transection of ganglion cell axons, a procedure that causes retrograde degeneration of many of the axotomized ganglion cells, led to a decrease in the number of anti-MAP 1A immunoreactive cells in retinal wholemounts. Thus, MAP 1A antibodies preferentially stain ganglion cell somata and dendrites but not amacrine cells. These antibodies should be useful ganglion cell markers.

To assess the effects of macular pigment optical density (MPOD) on isoluminant stimuli and to quantify MPOD electrophysiologically, MPOD distribution profiles were obtained in normal subjects using minimum motion and minimum flicker photometry. Isoluminance of VEP stimuli was determined using minimum flicker and tritan confusion lines were determined using a minimum distinct border criterion. Onset–offset and reversal VEPs to isoluminant red/green, blue/green, and subject-specific tritan gratings of different diameters were recorded from the same 14 subjects tested psychophysically. VEPs were additionally recorded to annular gratings. Chromatic VEP selectivity was assessed by Fourier analysis and as an index; onset negativity/(onset negativity + onset positivity). Peak MPOD varied between 0.2–0.8. Chromatic onset VEPs to all isoluminant 3-deg fields were predominantly negative. Larger blue/green and tritan stimuli elicited VEPs with additional positive, achromatic components; for 9-deg gratings, peak MPOD showed negative correlation with the power of the VEP fundamental (r = −0.70) and with the selectivity index (r = −0.83). Annular gratings elicited chromatic-specific B/G VEPs but only when isoluminance was determined for the annulus. Chromatic selectivity loss in VEPs to large B/G or Tritan gratings can be used to estimate subject-specific MPOD. An important implication is that isoluminant Tritan stimuli with short-wavelength components must be restricted in size in order to optimize koniocellular selectivity.

We used color contrast adaptation to examine the chromatic and contrast selectivity of central color mechanisms. Adaptation to a field whose color varies along a single axis of color space induces a selective loss in sensitivity to the adapting axis. The resulting changes in color appearance are consistent with mechanisms formed by different linear combinations of the cone signals. We asked whether the visual system could also adjust to higher-order variations in the adapting stimulus, by adapting observers to interleaved variations along both the L versus M and the S versus LM cardinal axes. The perceived hue of test stimuli was then measured with an asymmetric matching task. Frequency analysis of the hue shifts revealed weak but systematic hue rotations away from each cardinal axis and toward the diagonal intermediate axes. Such shifts could arise if the adapted channels include mechanisms with narrow chromatic selectivity, as some physiological recordings suggest, but could also reflect how adaptation alters the contrast response function. In either case they imply the presence of more than two mechanisms within the chromatic plane. In a second set of measurements, we adapted to either the L versus M or the S versus LM axis alone and tested whether the changes in hue could be accounted for by changes in relative contrast along the two axes. For high contrasts the hue biases are larger than the contrast changes predict. This dissociation implies that the contrast and hue changes are not carried by a common underlying signal, and could arise if the contrast along a single color direction is encoded by more than one mechanism with different contrast sensitivities or if different subsets of channels encode contrast and hue. Such variations in contrast sensitivity are also consistent with physiological recordings of cortical neurons.

During locomotion, observers respond to objects in the environment that may represent obstacles to avoid or landmarks for navigation. Although much is known about how visual cortical neurons respond to stimulus objects moving against a blank background, nothing is known about their responses when objects are embedded in optic flow fields (the patterns of motion seen during locomotion). We recorded from cells in the lateral suprasylvian visual area (LS) of the cat, an area probably analogous to area MT. In our first experiments, optic flow simulations mimicked the view of a cat trotting across a plain covered with small balls; a black bar lying on the balls served as a target object. In subsequent experiments, optic flow simulations were composed of natural elements, with target objects representing bushes, rocks, and variants of these. Cells did not respond to the target bar in the presence of optic flow backgrounds, although they did respond to it in the absence of a background. However, 273/423 cells responded to at least one of the taller, naturalistic objects embedded in optic flow simulations. These responses might represent a form of image segmentation, in that cells detected objects against a complex background. Surprisingly, the responsiveness of cells to objects in optic flow fields was not correlated with preferred direction as measured with a moving bar or whole-field texture. Because the direction of object motion was determined solely by receptive-field location, it often differed considerably from a cell's preferred direction. About a quarter of the cells responded well to objects in optic flow movies but more weakly or not at all to bars moving in the same direction as the object, suggesting that the optic flow background modified or suppressed direction selectivity.

Previous studies report that background luminance flicker, which is asynchronous with signal averaging, reduces the amplitude and increases the latency of the pattern-onset visual evoked potential (VEP). This effect has been attributed to saturation of the magnocellular (m-) pathway by the flicker stimulus. In the current study, we evaluate this hypothesis and further characterize this effect. We found that flicker had similar effects on the pattern-onset and pattern-reversal VEP, suggesting that the reversal and onset responses have similar generators. Chromatic flicker decreased latency of the chromatic VEP whereas luminance flicker increased peak latency to luminance targets. This result indicates that luminance flicker saturates a rapidly conducting m-pathway whereas chromatic flicker saturates a more slowly conducting parvocellular (p-) pathway. Finally, evoked potentials to chromatic and luminance stimuli were recorded from 34 electrodes over the scalp in the presence of static and asynchronously modulated backgrounds. An equivalent dipole model was used to assess occipital, parietal, and temporal lobe components of the surface response topography. Results showed that chromatic flicker reduced activity to a greater extent in the ventral visual pathway whereas luminance flicker reduced activity to a greater extent in the dorsal visual pathway to parietal lobe. We conclude that the VEP to isoluminant color and luminance stimuli contains both m- and p-pathway components. Asynchronous flicker can be used to selectively reduce the contribution of these pathways to the surface recorded VEP. Our results provide evidence of parallel pathways in the human visual system, with a dorsal luminance channel projecting predominantly to the posterior parietal lobe and a ventral color channel projecting predominantly to inferior temporal lobe.

Cells of the cat's perigeniculate nucleus (PGN), part of the visual sector of the thalamic reticular nucleus (TRN), provide GABAergic inhibition to the A and A1 layers of the dorsal lateral geniculate nucleus (LGNd) and, therefore, may control information flow from the retina to the cortex. Previous electrophysiological experiments suggested that the PGN may be subdivided on the basis of ocular dominance thus reflecting the afferent and efferent projections with lamina A and A1 of the LGNd. The present study utilized the ability of wheat germ agglutinin-horseradish peroxidase (WGA-HRP) to be transported transneuronally following intraocular injections in four cats to examine whether there is any anatomical evidence for eye specific layers within the PGN. Sections were processed with tetramethylbenzidine. Light WGA-HRP transneuronal labeling of LGNd collaterals and somata were seen in the PGN and very light labeling (but not somata) was seen in the TRN. Neither the cells of the PGN projecting to the LGNd nor the LGNd relay collaterals within the PGN were clearly organized into nonoverlapping laminae related to the eye specific layers of the LGNd. However, parts of the PGN immediately adjacent to the LGNd appear devoid of connections with lamina A1 thus creating a thin monocular segment for the contralateral eye.

Based on clinical findings, diabetic retinopathy (DR) has traditionally been defined as a retinal microvasculopathy. Retinal neuronal dysfunction is now recognized as an early event in the diabetic retina before development of overt DR. While detrimental effects of diabetes on the survival and function of inner retinal cells, such as retinal ganglion cells and amacrine cells, are widely recognized, evidence that photoreceptors in the outer retina undergo early alterations in diabetes has emerged more recently. We review data from preclinical and clinical studies demonstrating a conserved reduction of electrophysiological function in diabetic retinas, as well as evidence for photoreceptor loss. Complementing in vivo studies, we discuss the ex vivo electroretinography technique as a useful method to investigate photoreceptor function in isolated retinas from diabetic animal models. Finally, we consider the possibility that early photoreceptor pathology contributes to the progression of DR, and discuss possible mechanisms of photoreceptor damage in the diabetic retina, such as enhanced production of reactive oxygen species and other inflammatory factors whose detrimental effects may be augmented by phototransduction.

Amodal completion is the most common form of visual completion occurring when portions of an object are hidden, due to their occlusion behind another object (Michotte, 1951). Just as a shape is completed amodally behind another occluding shape, so is a color behind another occluding color or behind a lighting: a bright light reflected by a three-dimensional object. Four possible phenomenal combinations related to the amodal completion of color are shown: amodal or modal coloration or discoloration. Purposes of four experiments were: (1) to demonstrate the amodal completion of color under different stimulus conditions and under chromatic and achromatic conditions and (2) to extract the general principles ruling the amodal completion of color: “which, among many, is the color that completes amodally?” and, consequently, “which is the region of an object that determines its color?” The results showed the effectiveness of the amodal completion of color and that chromatic and achromatic conditions reveal different results. Four general principles of the amodal completion of color, useful to understand the more general problem of phenomenal organization of color, are suggested.

Amacrine cells are a diverse set of local circuit neurons of the inner retina, and they all release either GABA or glycine, amino acid neurotransmitters that are generally inhibitory. But some types of amacrine cells have another function besides inhibiting other neurons. One glycinergic amacrine cell, the Aii type, excites a subset of bipolar cells via extensive gap junctions while inhibiting others at chemical synapses. Many types of GABAergic amacrine cells also release monoamines, acetylcholine, or neuropeptides. There is now good evidence that another type of amacrine cell releases glycine at some of its synapses and releases the excitatory amino acid glutamate at others. The glutamatergic synapses are made onto a subset of retinal ganglion cells and amacrine cells and have the asymmetric postsynaptic densities characteristic of central excitatory synapses. The glycinergic synapses are made onto other types of ganglion cells and have the symmetric postsynaptic densities characteristic of central inhibitory synapses. These amacrine cells, which contain vesicular glutamate transporter 3, will be the focus of this brief review.
