Here we developed the first set of nuclear microsatellite markers (nSSRs) for Stizophyllum riparium (Bignoniaceae), a widespread species of Neotropical liana. Thirty-two sets of primers were isolated from a genomic dataset obtained with an Illumina HiSeq Platform, and characterized for 57 individuals of S. riparium from three populations. Nine nSSRs were polymorphic and the number of alleles ranged from eight to 29. The unbiased expected heterozygosity per locus ranged from 0.724 to 0.952 and the polymorphic information content values ranged from 0.717 to 0.944. All nine primer pairs also amplified for two closely related species (S. inaequilaterum and S. perforatum). The new set of nuclear markers will be useful for population genetics studies of Stizophyllum as a whole.

Teosinte, the wild progenitor of maize, has immense potential for providing unique traits and is more divergent compared to inbred lines and landraces. One hundred and sixty-nine teosinte-introgressed maize backcross inbred lines were developed to widen the genetic base of maize with predomestication alleles. The population was evaluated phenotypically and genotypic data of 76 SSR markers were used to map quantitative trait loci (QTLs) governing the targeted traits. Sixty-six QTLs were detected for eight plant architect-related traits that are spread over 10 different chromosomes with phenotypic variation ranging from 2.29 to 13.97%. Maximum three stable QTLs were recorded for days to anthesis (DA) followed by two for days to silking (DS), plant height (PH) and node bearing first ear (NBE). For rest of three traits namely flag leaf length (FLL), flag leaf width (FLW) and ears per plant (E/P) only one stable QTL was detected. Among the 16 common QTLs, the marker phi328178-linked QTL governed four characters (DA, DS, FLL, FLW) simultaneously, followed by umc1622-linked (ASI, FLW, E/P), umc2341-linked (DA, DS, NBE) and phi075-linked QTLs (ASI, PH, NBE) controlling three traits each. Remaining 12 QTLs controlled two characters. Molecular association between co-localized QTLs for different traits was also validated at the phenotypic level by significant correlation estimates. For eight studied traits, 53 superior lines were identified which along with parents (teosinte and maize inbred DI-103) were grouped into 12 clusters. Therefore, lines clustered independently can be combined to accumulate desirable traits for the improvement of maize.

A total of 60 farmers' cacao accessions (Theobroma cacao L.) from Nicaragua were investigated using microsatellite markers to reveal their genetic composition and to identify potentially resistant genotypes against the black pod disease caused by Phytophthora palmivora. These accessions were compared with 21 breeders' accessions maintained locally, two Criollo accessions from Costa Rica and two accessions from Ecuador. The analyses showed a low level of differentiation among groups of farmers' accessions (FST = 0.06) and that six Nicaraguan accessions were genetically closely related to the two Criollo accessions used as a reference. In addition, seven distinct genotypes were found to have allelic composition that may indicate linkage to resistance alleles, thus being potential parental lines in future breeding programmes.

Freezing damage is a major limiting factor to Pistachio cultivation in cold climates. Assessment of pistachio genotypes to freezing stress is important for the selection of tolerance cultivars in these regions. In this study, nine genotypes belong to six Pistacia species (P. terebinthus L., P. atlantica Desf., P. atlantica subsp. kurdica P. atlantica subsp. mutica, P. khinjuk Stocks., P. vera var. Sarakhs, P. vera cv. Badami-Riz-Zarand I, P. vera cv. Badami-Riz-Zarand II) and an interspecies hybrid named UCB1 (P. atlantica × P. integrrima) were evaluated to freezing stress at temperatures of −4, −8, −12 and, −16°C in the months of December, January and February. Results showed that electrolyte leakage (EL) percentage and means of eco-physiological parameters varied among genotypes with the highest soluble carbohydrates and phenolic compounds observed in species of P. atlantica subsp. kurdica and P. vera var. Sarakhs respectively. In February, the highest calcium and potassium concentrations of shoot were observed in P. terebinths and P. vera var. Sarakhs genotypes respectively. The highest EL percentage was observed at −16°C in P. atlantica and P. khinjuk and the lowest in P. terebinthus and P. vera var. Sarakhs.

Cattleya walkeriana, one of the most improved Brazilian Cattleyas, is a popular tropical orchid endemic from Brazil and currently endangered. In the present study, for the first time microsatellite markers were developed for C. walkeriana and their transferability was tested for the species C. loddigesii and C. nobilior. The markers were used for genotyping 26 C. walkeriana specimens from different growers and from different levels of improvement. The transferability was successful, with five polymorphic loci transferred to C. loddigesii and six polymorphic loci to C. nobilior. Eight loci were polymorphic, revealing a maximum of two to ten alleles per locus in C. walkeriana and two to four and two to five in C. loddigesii and C. nobilior, respectively. There was no significant linkage disequilibrium in the studied loci. For C. walkeriana, the observed and expected heterozygosities ranged from 0 to 0.963 and from 0.138 to 0.841, respectively. These markers identified polymorphisms and may be used to study the genetic diversity, gene flow or hybridization of these species.

The common dandelion (Taraxacum campylodes G.E.Haglund) is known for its medicinal properties and as a non-toxic, highly nutritious edible plant. Even though the herb is mostly collected in the wild, the food and pharmaceutical industries prefer cultivated plant material. Cultivation offers the possibility to achieve predictable yields, ensure the botanical identity of the plants and products can be certified according to organic or biodynamic standards; therefore the interest in domestication is growing. Due to the scarcity of research on dandelion cultivation in agricultural systems, the current study focused on determining the quantitative and qualitative characteristics of 13 wild populations from Latvia in an organic farming system. Populations collected at Gluda, Priekuli, Mundigciems, Marupe as well as variety TA008 had the highest dry root weight (>100 g per plant) and exceeded the root weight of variety Nouvelle more than two-fold. The highest concentration of chlorogenic acid (CHA) (0.12%) and chicoric acid (CCA) (0.15%) was found in the wild population collected in Turaida. The wild populations from Marupe and Mundigciems had a high average root weight as well as a relatively high CHA and CCA content, making them suitable for commercial growing. The present study examined the feasibility of cultivating wild dandelion populations in an organic farming system. The results revealed a high degree of variation within and between populations; therefore, dandelion wild populations can be used for domestication, breeding and selection of accessions most suitable for the pharmaceutical or food industries.

Sugarcane brown rust, caused by Puccinia melanocephala, is one of the main diseases of sugarcane in China. The identification and discovery of new resistance genes have important theoretical and practical significance for preventing outbreaks of brown rust and ensuring the sustainable production of sugarcane. To screen for polymorphic simple-sequence repeat (SSR) molecular markers for localization of brown rust resistance genes, we used two populations that are suitable for genetic linkage map construction and mapping of new resistance genes to construct resistant and susceptible genetic pools. We then screened 449 pairs of primers to identify polymorphic SSR markers in the parental lines and the resistant/susceptible genetic pools. The results showed that 25 pairs of primers directed amplification of polymorphic DNA fragments between the parents of the cross combination ‘Yuetang 03-393’ × ‘ROC 24’, and 16 pairs of primers amplified polymorphic fragments between the parents of the cross combination ‘Liucheng 03-1137’ × ‘Dezhe 93-88’. Four pairs of primers (SMC236CG, SCESSR0928, SCESSR0636 and SCESSR2551) amplified polymorphic DNA fragments between the parental lines and the resistant/susceptible genetic pools in ‘Yuetang 03-393’ × ‘ROC 24’. The results of this study will establish a solid foundation for the mapping of new brown rust resistance genes, genetic linkage map construction and the development of closely-associated molecular markers in sugarcane.

To characterize five different morphological forms of Russian olive grown in Ladakh, analysis of morphometric data of different variants was performed. Statistically significant differences among these variants were found for all the fruit and seed characters studied. Results of the study indicate presence of infra-specific differentiation within the species. Morphometric data for these variants conform to the system of nomenclature prevalent in the region for these variants. The ringmo (‘long fruited’ variety) type had the longest fruits, while bee (‘small fruited’ variety) had the smallest fruits.

Distant hybridization of cereals is often impaired by fertilization barriers. Haploid induction through intergeneric crossing is well developed in wheat but has not been successful in rice due to incompatibility issues. The present study was thus undertaken to identify fertilization barriers that hinder the compatibility of the rice cultivar Punjab Rice 121 with maize and pearl millet lines as pollinators. A total of 37,357 spikelets were pollinated, yielding 494 caryopses upon supplementation with auxins. The resultant caryopses, arising from true intergeneric crosses, lacked embryos. Imaging of the pollinated pistils at different intervals indicated that intense callose depositions block the release of generative nuclei to the ovule in these wide crosses. Rice spikelets pollinated with rice pollen (cis-generic crosses) exhibited positive indicators of fertilization reaction at the micropyle. While the cis-generic crosses initiated true caryopsis formation after 24 h, no comparative reaction was observed in the intergeneric crosses. The current survey underlines that the rice female gametophyte presents a strong pre-fertilization barrier to foreign pollen. This barrier may be modulated in the future by altering genotype and auxin combinations.

Soil moisture deficit is the major constraint for sesame crop production during its main rainfed and summer cultivation seasons. In summer cultivation, the crop frequently gets exposed to soil moisture deficit at various crop growth stages. Therefore, it is essential to identify the traits along with promising genotypes adapted to soil moisture deficit. A set of 35 sesame genotypes with checks was used to quantify the variation in morpho-physiological, yield, and quality traits under irrigated (WW) and deficit soil moisture stress (WS) conditions in the summer seasons of 2021 and 2022. The analysis of variance revealed the presence of high variability among the genotypes for various measured traits. The mean performance indicated that WS negatively affects the growth, development, yield and quality traits. Moreover, the correlation, path analysis and D2 analysis studies suggested that the traits, viz. leaf area (LA), total dry matter (TDM), canopy temperature (CT), number of branches per plant (NBP) and number of seeds per capsule (NSC) were significantly associated with seed yield under both the conditions. Quality traits like palmitic acid and oleic acid correlated positively with seed yield, particularly under WS. Furthermore, the genotypes with lower canopy temperatures were found to be better seed yielders under WS. In addition, mean performance and cluster analysis suggested that the genotypes: IC- 205776, JSCDT-112, JCSDT-26, IC-205610, and IC-204300, secured higher seed yield along with superior agronomical traits and net photosynthetic rate. These selected genotypes were most promising and could be used in future sesame crop improvement programmes.

Diversity in agronomically important traits of wild emmer wheat, Triticum dicoccoides, the progenitor of most cultivated wheats, was examined in l60 genotypes representing 16 populations derived from a wide ecogeographical spectrum of climate, soil and water availability in Israel and Turkey. The analysis included 14 quantitative economic characters including heading date, culm length, biomass, grain yield and its components, and grain protein percentage.

The results led to two major conclusions: (i) agronomically important traits varied genetically: ecogeographic diversity in these traits is dramatically displayed when populations are grown in a relatively standardized environment; and (ii) genetic diversity is revealed not only between, but also within populations. Thus, genotypes excelling in some of these characters were found in several populations.

Valtrate is the major active component of iridoids in Valeriana jatamansi plants, and has been widely used for the treatment of various diseases, giving rise to rapidly increasing market demands. The gene VjG10H from V. jatamansi encoding geraniol 10-hydroxylase (G10H) was introduced into V. jatamansi hairy roots and a transgenic hairy root culture system of V. jatamansi with good valtrate production ability was successfully established for the first time. The valtrate content of VjG10H-transformed lines ranged from 8.12 to 10.77 mg/g, with an average valtrate content (9.52 mg/g) of all the four G lines being higher than the non-transgenic hairy root line. Overexpression of VjG10H significantly improved valtrate production, indicating that G10H played an important role in stimulating valtrate accumulation. MeJA treatment stimulated valtrate accumulation in all of the VjG10H overexpression recombinant lines compared to untreated cultures, indicating that methyl jasmonate treatment was another effective regulation target for metabolic engineering of valtrate biosynthesis in V. jatamansi. Thus, transgenic hairy root technology coupled with elicitor treatments may be a promising strategy to increase valtrate yield in V. jatamansi.

DNA fingerprinting using microsatellites is a useful aid in cultivar identification, but has rarely been applied to garden plants. Eleven microsatellite markers were developed for the valuable garden plant Hesperantha coccinea (Schizostylis coccinea), and used to determine relatedness of accessions. Several accessions, described as separate cultivars, appeared to have identical genotypes. Among the 53 accessions tested, there were 34 unique multilocus genotypes. The level of polymorphism detected in the cultivars was high, with on average seven alleles per locus and an average expected heterozygosity of 0.72 across loci. It is clear from the genotypes that a large proportion of the cultivars are closely related to each other. The resulting markers can now be used to generate a complete database of all known cultivars of the species and to detect essentially derived cultivars. As an extension of this study, the markers identified here could also inform us about the genetic diversity in wild populations.

The members of the Aegilops genus serve as a vast pool of allele discovery for wheat improvement in abiotic and biotic stress responses. Aegilops mutica Boiss (Amblyopyrym muticum (Boiss) Eig) is an unexplored candidate with significant potential. Even though it has been used in cytogenetics applications within the last century, natural population diversity and allele discovery have been neglected. As an endemic species for Anatolia and the lower Caucasian region, it has an unexplored population structure. Here, seventy-five genotypes from five different newly collected populations from central Anatolia were evaluated with 29 polymorphic SSR loci. Significant diversity within (83%) and between (17%) the populations was obtained. Three of the populations were clearly separated, while two had some level of the mixture. Relatively easy cross-species hybridization and introgressions make Ae. mutica a good candidate for novel allele discovery and pre-breeding. Here, for the first time, representative natural populations of Ae. mutica were compared and population structures were revealed with SSR markers which may clear the misty vision that geneticists might have regarding Ae. mutica. This could be exploited in genetic resource conservation and breeding programs and maybe a point for further studies.

Gamma ray-induced mutations have been widely used to improve existing crop germplasm and create novel genetic variation. In the current study, a multi-year experiment was carried out to induce and isolate mutants with desirable agro-morphological traits and improved grain hardness through evaluation of induced mutant populations generated in soft-textured wheat variety HPW 89 irradiated with gamma ray dose of 250, 300 and 350 Gy. Mutagen sensitivity studies revealed a higher frequency of biological damage and seedling mortality for doses beyond 300 Gy in the M1 generation. However, the mutagenic treatments in the M2–3 populations significantly altered the magnitude of the biometrical traits. Results from the variability and association studies among traits showed that biological yield per plant, 1000-grain weight, spike length, grains per spike and plant height may be prioritized for higher genetic gain and could be used as selection criteria parameters. Multivariate analysis indicated induction of heterogeneity among mutant populations. Overall, 250–300 Gy doses were found ideal for a successful wheat mutation programme and 293 agro-morphologically superior wheat mutants were identified, out of which 108 had semi-hard grain texture based on single kernel characterization system. Among these, nine mutants were found to have the highest grain hardness index due to induced changes in one or both puroindoline genes. Hence, these mutants identified for several traits along with harder grain texture will serve as important genetic resource in future wheat-breeding programmes.

This study was designed to understand how drought stress impact on the physiological and molecular status of genetically diverse Lolium perenne L. (perennial ryegrass) populations. The sensitivities of the 36 perennial ryegrass populations were checked against PEG-6000-simulated drought stress by analysing physiological status. Afterward, drought-related gene expressions were evaluated under normal and stressed conditions. The drought stress significantly up-regulated the drought-related genes in P6, P10, P15 and P30, while no difference was observed in drought-sensitive ones. The results collectively showed that four populations (P6, P10, P15, P30) are tolerant to drought stress and activate drought-related genes. Genetic similarities of the populations were also monitored using microsatellite (SSR) markers. The ten SSR primers detected 38 alleles. Surprisingly, the drought-tolerant populations were classified into different groups by principal component (PCA) and cluster analysis, showing that they are genetically different. Moreover, four unique alleles were identified by two SSR primers (P-07, and P-08) in P6, P10 and P15. In addition, the drought-tolerant populations (P6, P10, P15, P30) can be used to develop new perennial ryegrass cultivars for areas having drought problems.

Ancho (width) chile peppers have economic, social, culinary and cultural importance in Mexico and worldwide. This chile type considers divergent subtypes that altogether have not been analysed and therefore their morphological diversity has not been systematically described. The objectives were to describe the morphological diversity of ancho pepper landraces from Mexico, to identify groups of similarity and to define the traits with the higher contribution to the total variation. Eighty-six landraces of ancho chile peppers (red, ‘mulatos’, ‘miahuatecos’, ‘cristalinos’ and ‘huacle’), collected in six states of Mexico, and two commercial controls were evaluated in two localities, in a simple randomized complete block experimental design. We recorded 76 morphological traits. Statistical analysis included a combined ANOVA, Pearson's correlation coefficient, discriminant analysis, principal components and clusters. The morphological diversity in ancho chile peppers was mainly made up of fruit width, fruit wall thickness, stem diameter, corolla length, seed weight per fruit, plant height, stem length and pubescence. We defined four groups, which made it possible to differentiate ancho chile peppers of Puebla and the huacle chile pepper of Oaxaca from populations collected in the north and ‘Bajío’ (midland) parts of Mexico. Ancho chile peppers of Mexico showed wide morphological differences according to the type of chile pepper and seed collection regions. The traits that contributed the greatest morphological diversity were fruit width, fruit wall thickness, stem diameter, corolla length, seed weight per fruit, plant height, stem length and pubescence.
