The present study describes a novel heat-stable, water-stress-related protein with a molecular mass of 47 kDa (designated Desc47) in the entomopathogenic nematode Steinernema feltiae (IS-6). The protein was accumulated about 10-fold (from 7·84±1·85 to 74·09±4·35% relative content level [RCL]) in dehydrated clumps of infective juveniles (IJs), which had lost 34·4% of their initial water content (from 65·1±1·7% to 42·7±0·72%) in a desiccation-tolerance-inducing treatment (97% relative humidity [RH] for 3 days). The appearance of Desc47 was accompanied by trehalose accumulation (from 300 to 600 mg trehalose/g protein) during the process of inducing the IJs into a quiescent anhydrobiotic state. A second cycle of IJ dehydration did not alter the RCL of Desc47 (79·3% for the first cycle and 73·3% for the second cycle). Desc47 retained its high RCL (69·7%) in rehydrated active IJs for 3 days, reaching 51·2% of its initial RCL only after a week. No homology to other known proteins was found by mass-spectrometry electrospray-ion-trap analysis. However, of the 5 sequences obtained from the protein (ranging from 11 to 21 amino acids), the 21-amino-acid peptide NVASDAVETVGNAAGQAG(D/T)AV showed excellent homology (74% identity in 19 amino acids) to the cold-responsive protein COR14b (g6564861) from Triticum aestivum. In the Caenorhabditis elegans predicted proteome database search, the N21 yielded the first-best identity score (59% identity in 17 amino acids) to the CE-LEA homologue protein (g2353333). In plants, COR and LEA are related proteins, heat-stable, which are expressed in response to both dehydration and cold acclimation. The implication of the involvement of Desc47 and the osmoprotectant trehalose in the desiccation-tolerance mechanisms of S. feltiae is discussed.

We set out to study how anti-filarial IgG4 and IgE, which have been studied extensively in adult populations, are influenced by gender and by the degree of filarial endemicity during childhood. Development of specific IgG4 and IgE was examined in 502 children aged 3 months to 12 years, who were resident in 3 villages in South-Sulawesi with microfilaria prevalences of 6, 23 and 42%. Specific IgG4 and IgE could be detected as early as 18 months after birth, in low amounts, and increased to levels comparable to those produced by adults at the age of 3 years. A higher prevalence of anti-filarial IgG4 in boys, indicating higher filarial infection compared to girls, became apparent after the age of 7. The specific IgG4 response was strongly influenced by the degree of filarial endemicity and production of this antibody was considerably delayed in the low transmission village. With respect to IgE, it was noted that specific IgE was consistently higher in boys from infancy onwards indicating a predisposition for high IgE production in males. The influence of filarial endemicity was less profound on IgE than on IgG4. In conclusion, reactivity to filarial antigens begins early in life and is differentially influenced by gender and transmission intensity.

The ability of the nematode Anguillicola crassus to infect eel larvae (glass-eel stage) was tested. The results show that glass-eels fed on infected copepods, the natural intermediate host of the nematode, can be infected. Light microscopical examination of the infected developing swimbladder tissue revealed that the infection results in a significant thickening of the connective tissue. The basolateral labyrinth of gas gland cells is very much reduced in infected swimbladders, and the distance of gas gland cells to blood capillaries is enlarged. Critical swimming speed, defined as the speed where the larvae were no longer able to swim against the current, was similar in infected and uninfected animals. At intermediate speeds (about 60–80% of critical swimming speed) infected eels showed a slightly higher swimming activity than control animals. Resting oxygen consumption, measured as an index of metabolic activity, within the first 2 months of infection was higher in control animals, which may be due to a reduced rate of activity in infected glass-eels. By 4–5 months after the infection, however, it was significantly higher in infected animals. This may indicate that at this stage a higher activity of the animals is required to compensate for the increase in body density, but swimming performance of infected and non-infected glass-eels was not significantly different. Oxygen consumption during swimming activity, measured in a swim tunnel at 50% of maximal swimming speed, also was not affected. The results thus show that even glass-eels can be infected with A. crassus, and this probably contributes to the rapid spread of the nematode in Europe. While aerobic metabolism during swimming activity is not affected at this stage of infection, the swimbladder tissue shows severe histological changes, which most likely will impair swimbladder function.

We review the literature on parameter values relevant to the epidemiology of strongyle nematode infections of domestic sheep. Information is subdivided by parasite genus, country of origin and climate type. While field observations have been made in a large number of countries, the bulk of studies under controlled conditions have been conducted in Australia, New Zealand and the UK. For these countries, experiments and parameters are interpreted in terms of a previously published model of nematode dynamics, and are used to calculate the basic reproduction number. Average values range from less than 6 for Haemonchus contortus in New Zealand and a winter rainfall region of Australia, to more than 16 for Ostertagia circumcincta in New Zealand and the UK. Additional considerations of the effects of climate and the annual replacement of host stock show that for conditions favourable for parasite transmission this is a robust indicator of parasite epidemiology. When climate variation and annual replacement are added to the model, it is shown to reasonably describe the qualitative behaviour of an experimental data set, indicating it to be a useful tool for further investigation of some of the underlying assumptions of sheep–nematode dynamics.

Infective larvae of the dog roundworm Toxocara canis survive in the tissues of their hosts for extended periods in a state of developmental arrest, successfully evading immune destruction. This survival strategy is thought to be mediated by T. canis excretory/secretory (TES) products which downregulate or divert the immune response. We purified one of the major TES products, TES-70 and gained amino acid sequence from 4 tryptic peptides. These peptides were matched to a predicted protein from a cDNA that was isolated by expression screening a T. canis cDNA library with mouse anti-TES serum. The predicted protein (Tc-CTL-4) is similar to, but larger than, Tc-CTL-1, a 32-kDa C-type lectin secreted by T. canis larvae. Tc-CTL-4 has a signal peptide, 2 Cys-rich domains and a C-terminal calcium-dependent C-type lectin domain that shares sequence similarity with host immune cell receptors such as macrophage mannose receptor and CD23. The lectin domain was expressed in bacteria and antiserum to the purified recombinant protein was used to confirm that Tc-ctl-4 did encode the native TES-70 glycoprotein. TES-70 selectively bound to ligands on the surface of Madin–Darby Canine Kidney cells in vitro in a calcium-dependent manner, inhibitable by mammalian serum, indicating that a host glycan is the native ligand for this new parasite lectin.

The frequency distribution of first generation, Steinernema feltiae Filipjev parasitic stages was over-dispersed with the majority of hosts containing few or no parasitic stages, whilst a few hosts contained a great many. Because of high extraction efficiency, the frequency distributions of the parasitic stages and the infective stages in the soil were assumed to be directly related. To explain the frequency distribution of the parasites it was therefore necessary to account for the frequency distribution of the S. feltiae infective stages in the soil. The infective stages were spatially aggregated into 30 cm diameter patches at the site of host death. These patches were randomly distributed approximately 1 m apart. At the 1 m scale, the pooled counts of infective stages were randomly distributed. Thus, in contrast to the frequency distributions, the spatial structuring of S. feltiae changed with the spatial scale of the interaction. This dynamic spatial structuring means that the majority of samples taken would contain few or no infective stages, whilst a few soil samples would contain a great many. Thus, the spatial structuring of the infective stages generates the over-dispersed frequency distribution of the S. feltiae in the soil. Hosts, encountering infective stages from this spatial distribution will, therefore, show an over-dispersed frequency distribution of S. feltiae parasitic stages.

This detailed review of the published studies underlying ivermectin's recent registration for use in lymphatic filariasis (LF) demonstrates the drug's single-dose efficacy (over the range of 20–400μg/kg) in clearing microfilaraemia associated with both Wuchereria bancrofti and Brugia malayi infections of humans. While doses as low as 20μg/kg could effect transient microfilarial (mf) clearance, higher dosages induced greater and more sustained mf reduction. The single dose of 400μg/kg yielded maximal responses, but a number of practical considerations suggest that either 400μg/kg or 200μg/kg doses would be acceptable for use in LF control programmes. Associated safety assessments indicate that adverse events, which occur commonly following treatment of microfilaraemic individuals, develop not because of drug toxicity but because of host inflammatory responses to dying microfilariae killed by the ivermectin treatment. Ivermectin is, therefore, a highly effective and generally well tolerated microfilaricide that may soon become an essential component of many public health initiatives to interrupt transmission of lymphatic filarial infection in an effort to eliminate LF globally.

Lungworms of the genus Dictyocaulus from cattle, roe deer, and moose in Sweden were subjected to morphological and molecular analyses. The objectives of the study were to investigate whether mixed or monospecific Dictyocaulus infections occur in Swedish cattle and whether wild cervids may act as reservoirs. The morphological characters examined were thickness and shape of the buccal capsule wall (BCW) and total spicular length (TSL). Morphometry was also done on the total body length, and BCW thickness and length. In the molecular identification, we used a PCR-linked hybridization assay to probe worm DNA with species-specific oligonucleotide probes to the second internal transcribed spacer (ITS2). The results showed that the BCW shape was the most reliable morphological character for identification. Significant differences were observed in this character, but an overlap occurred between lungworms from each of the host species. With the hybridization assay, all lungworms from cattle were identified as D. viviparus, whereas those from roe deer represented a novel Dictyocaulus species demonstrating that each host had a monospecific lungworm infection. In moose, 61 (78·2%) worms belonged to the new species and 17 (21·8%) were D. eckerti. This study shows the usefulness of hybridization assay as an epidemiological tool for the specific identification of lungworms of cattle and wild cervids.

Three nucleotide data sets, one nuclear (ITS-2) and two mitochondrial (COI and l-rRNA), have been investigated in order to determine relationships among species of Strongylinae and Cyathostominae, intestinal parasites of the horse.Nucleotide sequence data reported in this paper are available in the NCBI, GenBank, accession numbers: AF263472 to AF263472 and AF268479. The data exhibited a strong mutational bias towards A and T and in the COI gene, silent sites appeared to saturate rapidly partly due to this substitution bias. Thus, the COI gene was found to be less phylogenetically informative than the l-rRNA and ITS-2 genes. Combined analysis of the l-rRNA and ITS-2 genes supported a monophyletic clade of the cyathostomes with Tridentoinfundibulum gobi, which had previously been classified as a nematode of ‘uncertain origin'. The Strongylinae grouped consistently outside the clade containing the cyathostomes and T. gobi. Molecular analysis failed to provide strong evidence for the separation of cyathostomes into classical genera, as previously defined by morphological classification.

Ascaris suum possesses a well-developed nervous system which is regulated by a number of classical neurotransmitters including acetylcholine (ACh), γ-aminobutyric acid (GABA), glutamate and serotonin. The vagina vera, the distal part of the ovijector, displays intrinsic, rhythmic activity which has been shown to be modulated by FMRFamide-related peptides (FaRPs) in vitro. Confocal scanning laser microscopy coupled with immunocytochemistry, and histochemical studies, revealed that the nerve plexus of the ovijector contains GABAergic and glutamatergic innervation. Although no distinctive cholinergic or serotoninergic innervation was apparent, cholinesterase activity was localized to discrete areas of the musculature of the vagina vera. The effects of classical transmitters on the activity of the vagina vera in vitro were examined. ACh was excitatory, stimulating a brief but powerful contraction of the vagina vera with a threshold for activity of 1 μM. Both GABA and glutamate were inhibitory, causing a cessation of contractile activity at high concentrations (> 10 μM). Although less potent than glutamate, GABA had more profound effects and induced longer-lasting paralysis of the tissue. The threshold concentrations for activity were 5 μM for glutamate and 10 μM for GABA. Serotonin had no consistent effect on the vagina vera. This study demonstrates that classical transmitters modulate the activity of the ovijector of A. suum.

This study describes morphological changes during capsule formation of Trichinella spiralis with emphasis on nuclei within the capsule. The nuclei of the pre-existing muscle cell may become hypertrophied or degrade soon after the entrance of newborn larvae. Cytoplasm associated with these hypertrophied nuclei was basophilic in H&E staining and composed of mitochondria and rough endoplasmic reticulum. Satellite cells underwent cell division and joined to the nurse cell. These nuclei may also become hypertrophied or degrade. Cytoplasm associated with nuclei of satellite cell origin was eosinophilic in H&E staining and composed of mitochondria, polysomes, and smooth and rough endoplasmic reticulum. Eosinophilic cytoplasm (of satellite cell origin) became prevalent in the capsule as capsule formation proceeded. Infiltrated cells were observed within the capsule. Thus this study showed diversity of nuclei in the capsule of Trichinella.

Cyclosporin A (CsA) is a widely investigated experimental anti-parasitic drug whose mode of action remains unresolved. The immunosuppressive action of CsA depends on the drug binding to the intracellular receptor cyclophilin (CyP). This study investigates whether complexing of CsA with parasite CyP is equally essential for its anthelmintic action. The correlation between initial cyclosporin-induced damage in vitro and drug binding to parasite CyP has been examined. CsA and the analogues B-5-49, CsH and CsA-acetate all induced similar damage to the tapeworm Hymenolepis microstoma in vitro in incubations between 2h and 4 days. The initial foci of drug damage were the parasite surface and mitochondria in the syncytium. In a competitive binding assay only B-5-49 displaced [H3]-CsA from either crude parasite cytosolic CyP or affinity-purified CyP while CsH and CsA-acetate had no effect. These data suggest strongly that cyclosporins act on the surfaces of helminth parasites but that drug action does not involve complex formation with CyP. An alternative drug-binding site must therefore be identified which may lead to the rational design of novel anthelmintic drugs.

The intergenic spacer of the ribosomal RNA gene array from the monogenean Gyrodactylus salaris was isolated using PCR amplification. PCR products were cloned and sequenced. Three different fragments of 0·63, 1·0 and 2·62 kb, were consistently obtained. These showed homology at the 5′ and 3′ termini but differed in their overall size and intervening sequence. The 5′ end showed homology to various 28S ribosomal RNA gene sequences, suggesting that this represented the 3′ terminus of the G. salaris 28S ribosomal RNA gene. A number of features common to other eukaryotic intergenic spacers were found in the longest sequence, including A + T rich sequences, palindromic sequences and tandemly repeated elements. Two regions of 23 bp sequences arranged in non-identical tandem repeats were identified. There were 9 repeats in both regions, separated by 81 bp of non-repetitive sequence. The repeat units from the two regions shared some similarity at their 3′ ends. The G. salaris intergenic spacer sequence was examined for sequence motifs involved in the transcription of the ribosomal RNA genes in other species. Several regions with homology to transcription start sites were identified.

This review of the safety of the co-administration regimens to be used in programmes to eliminate lymphatic filariasis (albendazole+ivermectin or albendazole+diethylcarbamazine [DEC]) is based on 17 studies conducted in Sri Lanka, India, Haiti, Ghana, Tanzania, Kenya, Ecuador, the Philippines, Gabon, Papua New Guinea, and Bangladesh. The total data set comprises 90635 subject exposures and includes individuals of all ages and both genders. Results are presented for hospital-based studies, laboratory studies, active surveillance of microfilaria-positive and microfilaria-negative individuals, and passive monitoring in both community-based studies and mass treatment programmes of individuals treated with albendazole (n=1538), ivermectin (9822), DEC (576), albendazole+ivermectin (7470), albendazole+DEC (69020), or placebo (1144). The most rigorous monitoring, which includes haematological and biochemical laboratory parameters pre- and post-treatment, provides no evidence that consistent changes are induced by any treatment; the majority of abnormalities appear to be sporadic, and the addition of albendazole to either ivermectin or DEC does not increase the frequency of abnormalities. Both DEC and ivermectin show, as expected, an adverse event profile compatible with the destruction of microfilariae. The addition of albendazole to either single-drug treatment regimen does not appear to increase the frequency or intensity of events seen with these microfilaricidal drugs when used alone. Direct observations indicated that the level of adverse events, both frequency and intensity, was correlated with the level of microfilaraemia. In non microfilaraemic individuals, who form 80–90% of the ‘at risk’ populations to be treated in most national public health programmes to eliminate lymphatic filariasis (LF), the event profile with the compounds alone or in combination does not differ significantly from that of placebo. Data on the use of ivermectin+albendazole in areas either of double infection (onchocerciasis and LF), or of loiais (with or without concurrent LF) are still inadequate and further studies are needed. Additional data are also recommended for populations infected with Brugia malayi, since most data thus far derive from populations infected with Wuchereria bancrofti.

This report documents the presence of an active thymidine kinase (TK) system within Mesocestoides vogae tetrathyridia as quantified by tritiated thymidine ([3H]-TdR) incorporation using liquid scintillation counting. A 100-fold increase in [3H]-TdR incorporation was observed at 37 °C when compared with its incorporation at 0 °C. Thymidine's competitive analogue, BrdU, competed for sites within newly replicated DNA. Immunohistochemical trials performed here using antibodies against BrdU identified cells that have entered and passed through S-phase. Positively stained nuclei were most numerous at the anterior tip of tetrathyridia especially within the ganglia, lesser numbers of these cells occurred along the growing commissure and amongst surface tegumental cytons suggesting that stem cells do not exist in one region but are found throughout the entire body. As M. vogae has no internal organ systems the major sites for cell proliferation are those exhibiting maximal cell recruitment and undergoing tissue repair. These results show that it is possible to monitor changes in the cell recruitment pattern within this cestode. Thus use of BrdU and immunohistochemistry demonstrates how spatial arrangement and cellular reorganization can be successfully traced within M. vogae.

It recently became clear that adult eo- and palaeacanthocephalans parasitizing fish can bioconcentrate several heavy metals to significantly higher concentrations than the tissues of their definitive hosts. Following this discovery the lead accumulation of the archiacanthocephalan Moniliformis moniliformis was investigated using experimentally infected male Wistar rats of the CD-M-strain. The worms were allowed to grow up for 4 weeks post-infection followed by a 3 weeks oral lead exposure of the rats. After the exposure period the rats were killed and the metal levels were determined in muscle, liver, intestine and kidney of the rats as well as in different organs of female and male acanthocephalans. Lead concentrations were found to be highest in female M. moniliformis followed by the kidneys of the rats. Male worms contained approximately the same lead concentration as the hosts' kidneys. Lead analysis of the worms' organs revealed the highest lead concentration in the eggs of female acanthocephalans, followed by the cement gland of male Worms. Whilst the lead burden of the presoma was higher than that detected in the kidneys of the rats, the lead content of the metasoma was even lower than in the kidneys. A lead uptake of M. moniliformis from the intestinal lumen of the host became apparent as the faeces of infected rats contained significantly less lead compared to the uninfected conspecifics. Thus, this study reveals that lead accumulation also occurs in archiacanthocephalans parasitizing mammals. But the degree of metal bioconcentration is considerably lower compared to eo- and palaeacanthocephalans in fish. Anyway, due to a lack of adequate sentinel species in terrestrial biotopes the host–parasite system rat– M. moniliformis appears to be a useful and promising bioindication system especially in urban ecosystems in temperate regions.

Previous studies have indicated that between 60 and 80% of a population of entomopathogenic nematodes do not infect their insect hosts at any one period in time. Two hypotheses explain this behaviour: the first that there is a subpopulation of non-infectious nematodes and the second that the non-infectious group is created by inhibitory cues derived from infected insects. Through an experimental approach with the Galleria mellonella–Steinernema feltiae system we show that both mechanisms operate together. When conditions for infection were optimized, the sum of individual infection behaviours was similar to the number infecting as a population, implying observed infection rates are driven by intrinsic mechanisms. In addition, there was evidence that an infected host released a chemical cue into the environment which inhibited subsequent levels of infection. This degree of inhibition was independent of the number of infecting nematodes. Both these mechanisms are dynamic, so the observed proportion of infectious nematodes depended heavily on the time of exposure. The implications of these findings for both the design of laboratory trials and the use of entomopathogenic nematodes in biological control are discussed.

The relationship between faecal nematode egg count and the number of adult Telordasagia circumcincta was examined in 508 naturally infected lambs at 6–7 months of age. The relationship was found to be convex and was empirically described by a gamma-type function of the form y = anb e−cn, where n is the number of adult nematodes present and y is the number of nematode eggs per gramme (epg) of faeces. This equation predicted that the peak expected egg count (277 epg) would occur at a worm burden of 2167 adult worms. The absence of a linear relationship between egg counts and worm numbers is consistent with the existence of severe density-dependent constraints on the fecundity of T. circumcincta.

We recently showed that, in our Trichinella spiralis rat model, first exposure, but not re-exposure to infective-stage larvae evoked heat shock responses in 4 test organs. Our work, however, failed to implicate either early complete clearance of challenge muscle larvae (ML), or rapid elimination of newborn larvae (NBL) in the phenomenon noted in reinfected rats. This study clarifies that issue using 2 established facts in T. spiralis biology and anti-T. spiralis immunology. That is, adult worms injure gut cells and immune destruction of NBL requires release of material also toxic to host cells. To approach the above problem we analysed relevant and irrelevant rat organs for increased heat shock protein (HSP) production at 1, 7, 14, 20 and 27 p.i. during first and second infections. Organs examined were intestines, mesenteric lymph nodes (MLN), heart and lungs. Using densitometric analyses of immunoblots, increased HSP expression was detected on day 7 in intestines from both primary and secondary-infected rats albeit that the change in the latter was just short of significant. Interestingly, MLN only exhibited increased HSP levels in the reinfected rat model with increased HSP levels persisting for 1 week. A lasting shock response was detected in reinfected rats; in contrast, first exposure resulted in shock responses being evident in lungs at either day 7 or day 14, only. These findings suggest that (i) in immune rats, a few challenge ML develop into adults, produce NBLwhich are trapped within MLN, and (ii) that anti-T. spiralis and/or anti-NBL immunity is associated with an, as yet, uncomprehended stress to host's heart tissues.

DEC-fortified salt has been used successfully as a principal public health tool to eliminate lymphatic filariasis (LF) in China and, less extensively, in several other countries. Studies from 1967 to the present conducted in Brazil, Japan, Tanzania, India, China, and Taiwan involving administration of DEC salt for 18 days to 1 year, have shown this intervention to be effective for both bancroftian and brugian filariasis, as measured by reductions in both microfilarial density and positivity, and in some studies through reduction in mosquito positivity rates as well. Furthermore, studies suggest specific advantages from using DEC salt, including lack of side effects, particularly for bancroftian filariasis, and ability to reduce prevalence below 1% when used in conjunction with standard regimens of DEC tablets. However, use of DEC salt as a control tool suffers from a concern that health authorities might find it difficult to manage a programme involving a commodity such as salt. In the past decade, the very successful global efforts to eliminate iodine deficiency through universal salt iodization have demonstrated that partnership with the salt industry can be both successful and effective as a public health tool. Use of DEC salt can be most successfully implemented in areas in which (a) there is adequate governmental support for its use and for elimination of filariasis, (b) filariasis-endemic areas are clearly defined, (c) political leaders, health officials and the salt industry agree that DEC salt is an appropriate intervention, (d) the salt industry is well-organized and has known distribution patterns, (e) a successful national salt iodization effort exists, (f) a monitoring system exists that ensures adequacy of salt iodine content during production and that can also measure household coverage, and (g) measurement of impact on transmission of LF with the new antigen or filarial DNA detection methods can be established. There are advantages and disadvantages of using DEC-fortified salt compared with other interventions for LF elimination programmes, but rather than being considered as a ‘competing’ intervention, DEC salt should be seen as an additional option. Indeed, it is likely that many countries will derive maximal benefit from the synergistic effects of combining different intervention strategies in their national programmes to eliminate lymphatic filariasis.
