Traditionally, the family Campulidae has been associated either with the family Fasciolidae, parasites of ruminants, or the Acanthocolpidae, parasites of fishes, based on morphological similarities. Since morphology does not seem to resolve clearly the problem of the relationships of campulids, we have used the sequences of the 18S rRNA gene of the campulids Zalophotrema hepaticum, Campula oblonga and Nasitrema globicephalae, the fasciolid Fasciola hepatica, the acanthocolpid Stephanostomum baccatum and the outgroup Schistosoma mansoni to infer a phylogeny. Maximum parsimony and neighbour-joining methods were applied. Both methods indicated that campulids are closer to acanthocolpids than fasciolids. In order to confirm this relationship, we generated a second phylogeny using all the partial sequences of the 18S published for trematodes: Lobatostoma manteri, Echinostoma caproni, Calicophoron calicophorum, Tetracerasta blepta, Gyliauchen sp. and Opistorchis viverrini, plus those mentioned above, and Dicrocoelium dendriticum. The aspidogastrean L. manteri was used as the outgroup. Results were identical to the first analysis. According to this and the most recent Digenean phylogeny, which considers campulids and acanthocolpids as sister groups, we suggest that a common origin for these 2 groups would imply a host-switching process. The life-cycle of acanthocolpids includes marine gastropods as first intermediate hosts, and fishes as second intermediate and definitive hosts. In this context, the hypothesis would be that trematodes whose cycle ended in fishes were able to switch to mammalian hosts.

Young and old rediae of Parorchis acanthus Nicoll, from the digestive gland and gonad of Nucella lapillus (L.) have been described. The young redia has an anterior collar-like fold and mid-ventral birth papilla the movements of which are controlled by a special arrangement of muscles. The birth papilla is ‘solid’ but in older rediae is replaced by a pore. The pharynx is of fairly uniform size irrespective of the size of the redia and the intestine, in the young redia, is relatively muchlarger than in the older redia which facts suggest that relatively more food may be absorbed by the mouth in the young stage. The redia cavity develops as a slit in the central cell mass. As it enlarges the intestine remains close to the dorsal wall and developing embryos are carried, among the cells, towards the surface dropping into the cavity, later, to complete their development. Up to fifty-five cercariae may be present in a redia. Simple nervous and excretory systems are present.

The tegument consists of one layer; the surface bears numerous unusual branched microvilli. The syncytial protoplasm contains numerous electron dense mitochondria, vacuoles and granules. Half-desmosomes, not recorded for any other parasitic platyhelminth, occur fairly regularly on the lower limiting layer. Below the uninterrupted basement membrane are circular and longitudinal muscles and, lining the cavity, two or three layers of flattened cells containing numerous mitochondria, agranular and granular endoplasmic reticulum and Golgi complexes. Large lipid globules accumulate in the cells.

It is suggested that, in the older redia, active transport of nutrient materials through the tegument may take place.

The relationships between regional species richness and local species richness were examined in respect of helminth parasite communities in 32 species of freshwater fish in the British Isles. Fish were divided into 5 categories, for each of which the goodness of fit of the relationship to linear, exponential and power function models was tested. For all categories of fish combined, there was a significant, positive curvilinear relationship. Nested within this were two other patterns. For introduced fish, a linear model provided the best fit; for euryhaline and relict species it was impossible to determine the best model, but for the other categories the relationship was curvilinear and was best fitted by a power function model. The linear relationship found for introduced fish was interpreted as a temporary situation, reflecting the shortage of time for the communities to become saturated. It corresponded to the linear part of the curvilinear relationship of the other categories, which is believed to represent the fundamental form of the relationship for parasite communities. The communities reached a saturation level of richness, corresponding to the asymptote of the curve, which fell well below regional species richness. Explanations for local saturation are discussed, but neither community structure nor supply-side ecology can yet be preferred. It is concluded that local patterns in helminth community richness, in contrast to those in fish assemblages, are not significantly influenced by patterns on a larger, regional spatial scale and so regional species richness is not a key determinant of local species richness, nor does a knowledge of regional patterns improve predictability of local patterns.

Adult forms of the pig whipworm, T. suis, are able to establish intimate contact with the host's tissues by the formation of a tunnel-like construction which covers a section of the thread-like oesophageal region. The extreme anterior oral end and the stouter posterior region are not covered and lie free in the lumen of the caecum. The attachment tunnel is composed of host mucosal cells and goblet cell secretion and its possible mode of construction is discussed. Host tissue not directly involved in tunnel formation appears unharmed by the presence of the parasite. T. suis is therefore not a serious pathogen under natural conditions.

It has been demonstrated that characteristic structures of the trichuroid oesophageal region, e.g. bacillary band and stichosome are metabolically active. The former penetrates the layered cuticle in the form of columnar protrusions originating in the hypodermis. Each hypodermal column represents a gland cell or bacifiary cell, which displays enzymic activity, e.g. acid phosphatase, non-specific esterase, glucose-6-phosphatase, succinate dehydrogenase, cytochrome oxidase and β-d-glucosidase. Its possible function at the host-parasite interface and in the metabolism of the whipworm is discussed. The layered cuticle is proteinaceous and enzymically inert.

Glandular activity is also displayed by the stichosome surrounding the capillary-like oesophagus. This evidence confirms earlier suggestions that the stichosome is a series of oesophageal glands.

Although the bacillary band may be involved in nutrient uptake the evidence obtained indicates that this is via the mouth which is favourably placed for the ingestion of mucoid material which was detected in the oesophagus. This may be subjected to preliminary hydrolysis by the enzymes present in the oesophagus and stichosome.

Nitric oxide (NO) has been demonstrated to be the principal effector molecule mediating intracellular killing of Leishmania, both in vitro and in vivo. We investigated the type of cell death process induced by NO for the intracellular amastigote stage of the protozoa Leishmania. Specific detection methods revealed a rapid and extensive cell death with morphological features of apoptosis in axenic amastigotes exposed to NO donors, in intracellular amastigotes inside in vitro – activated mouse macrophages and also in activated macrophages of regressive lesions in a leishmaniasis-resistant mouse model. We extended our investigations to the dog, a natural host-reservoir of Leishmania parasites, by demonstrating that co-incubation of infected macrophages with autologous lymphocytes derived from dogs immunised with purified excreted-secreted antigens of Leishmania resulted in a significant NO-mediated apoptotic cell death of intracellular amastigotes. From the biochemical point of view, NO-mediated Leishmania amastigotes apoptosis did not seem to be controlled by caspase activity as indicated by the lack of effect of cell permeable inhibitors of caspases and cysteine proteases, in contrast to specific proteasome inhibitors, such as lactacystin or calpain inhibitor I. Moreover, addition of the products of two NO molecular targets, cis-aconitase and glyceraldehyde-3-phosphate dehydrogenase, also had an inhibitory effect on the cell death induced by NO. Interestingly, activities of these two enzymes plus 6-phosphogluconate dehydrogenase, parasitic enzymes involved in both glycolysis and respiration processes, are overexpressed in amastigotes selected for their NO resistance. This review focuses on cell death of the intracellular stage of the pathogen Leishmania induced by nitrogen oxides and gives particular attention to the biochemical pathways and the molecular targets potentially involved. Questions about the role of Leishmania amastigotes NO-mediated apoptosis in the overall infection process are raised and discussed.

Mismatch in the phenology of trophically linked species as a result of climate warming has been shown to have far-reaching effects on animal communities, but implications for disease have so far received limited attention. This paper presents evidence suggestive of phenological asynchrony in a host-parasite system arising from climate change, with impacts on transmission. Diagnostic laboratory data on outbreaks of infection with the pathogenic nematode Nematodirus battus in sheep flocks in the UK were used to validate region-specific models of the effect of spring temperature on parasite transmission. The hatching of parasite eggs to produce infective larvae is driven by temperature, while the availability of susceptible hosts depends on lambing date, which is relatively insensitive to inter-annual variation in spring temperature. In southern areas and in warmer years, earlier emergence of infective larvae in spring was predicted, with decline through mortality before peak availability of susceptible lambs. Data confirmed model predictions, with fewer outbreaks recorded in those years and regions. Overlap between larval peaks and lamb availability was not reduced in northern areas, which experienced no decreases in the number of reported outbreaks. Results suggest that phenological asynchrony arising from climate warming may affect parasite transmission, with non-linear but predictable impacts on disease burden. Improved understanding of complex responses of host-parasite systems to climate change can contribute to effective adaptation of parasite control strategies.

Since 2004 the West African countries of Burkina Faso, Mali and Niger have implemented national schistosomiasis and soil-transmitted helminthiasis control programmes with financial and technical support from the Schistosomiasis Control Initiative (SCI). In the first three years of the control programmes, nearly 13·5 million doses of praziquantel and albendazole have been administered against schistosomiasis and soil-transmitted helminthiasis with coverage rates varying between 67·0% and 93·9%. These treatments have resulted in a reduction of the prevalence and intensity of Schistosoma infection in the sentinel cohorts that were set up to monitor and evaluate the national control programmes. The challenges currently faced by these national control programmes are the ability to maintain the reduction in morbidity achieved thus far due to the mass treatment campaigns and ensuring sustainability. For reinforcement of surveillance, the establishment of a geographical information system is suggested in order to contribute towards enhanced sustainability of these programmes. Our new working hypothesis is that targeted control accompanied by periodic mass treatment campaigns (every two to three years) can contribute to maintaining the low levels of morbidity achieved thus far. The implementation of integrated neglected tropical disease control programmes in these countries will provide means to ensure the financial sustainability of control activities for the years to come.

Recombinant Schistosoma mansoni calreticulin (SmCaR) was expressed in Escherichia coli, using the glutathione S-transferase fusion protein, and its Ca2+-binding capacity was determined. Results obtained by a 45Ca2+ overlay technique showed that Ca2+-binding site(s) were present in the recombinant CaR indicating that proper folding of the protein was obtained using this system. An antiserum raised against the recombinant SmCaR showed that the native protein (Sm58) was expressed in all stages of the life-cycle from cercariae to the adult worm and in the egg. However, SmCaR seems to be a developmentally regulated protein whose expression can be used to study the post-transformational differentiation of the schistosomulum. Localization of SmCaR demonstrated that the majority of SmCaR was expressed in the epithelia of the digestive duct and in the genital organs. These results suggest that SmCaR, by regulating the Ca2+ concentration, may play an important role during cell proliferation. Finally the presence of SmCaR in miracidia and in the genital organs suggests that the antibody response directed against this protein could interfere in egg production.

The prediction that richness and diversity of helminth communities would be greater in tropical fish was tested by analysing total and intestinal helminth communities in samples of Anguilla reinhardtii taken from 10 localities in tropical and semi-tropical freshwaters in Queensland, Australia. Comparison of findings with those from A. anguilla in the UK revealed that regional species richness was higher in Queensland and that helminth communities harboured a suite of common species, virtually all eel specialists, that were responsible for the observed higher levels of similarity between component communities. All measures of community richness and diversity adopted indicated that the poorest helminth communities in A. reinhardtii were comparable with the richest ones reported from the northern temperate A. anguilla. The richest communities in A. reinhardtii were more diverse than those reported from any other species of fish, whether marine or freshwater, to date, and were comparable with the communities found in some species of aquatic birds. Both tropical conditions and age and endemism of the host, i.e. the time hypothesis, were considered as explanations for this richness but neither could be favoured. A cautious approach to generalizations based on data from temperate regions only is advocated, and the need for more studies on tropical parasite communities is strongly emphasized.

The muscular saucer-shaped haptor of the monogenean parasite Entobdella soleae is attached to the skin of its host Solea solea by means of a suction pressure generated within the sea-water-filled cavity enclosed between the cup-shaped haptor and the skin of the fish. The suction pressure is produced by the action of a pair of extrinsic muscles which are situated in the body of the parasite. Each extrinsic muscle communicates with the haptor by means of a long tendon which passes through a fair-lead in a prop-like accessory sclerite and is inserted on the end of a girder-like anterior hamulus embedded in the roof of the concavity of the haptor. The pull exerted by the muscles lifts the girders and the roof of the suction cup in which they are embedded relative to the props, thereby producing a suction pressure.

An electron microscope was used to investigate the ultrastructure of the tendon, which was found to consist largely of unbranched banded fibrils which differed from the collagen fibrils of vertebrate tendon in their diameter and periodicity.

Three new species of Isospora from Australian lizards are described, viz. Isospora amphiboluri from Amphibolurus barbatus (Agamidae), I. egerniae from Egernia whitii (Scincidae), and I. ablephari from Ablepharus boutonii (Scincidae) and Heteronota bineoi (Gekkonidae). A further Isospora sp. is mentioned from Phyllurus cornutus (Gekkonidae). A table of comparative data of lacertilian Isospora is given, and also a host check-list.

The body wall of the daughter sporocyst of Cercaria bucephalopsis haimaena Lacaze-Duthiers, 1854, is shown by the electron-microscope to consist of an external syncytial tegument, lying on a basement membrane, and an internal cellular subtegument, which surrounds a body cavity containing developing cercariae.

The syncytial tegument has areas of dense cytoplasm alternating with sparse reticulate cytoplasm. The dense cytoplasm contains nuclei, a few mitochondria and secretory products which probably include a neutral mucopolysaccharide. This may be the precursor of the acid mucopolysaccharide in the sparse reticulate cytoplasm and on the surface of the daughter sporocyst.

Nutrients are probably absorbed through the outer plasma membrane of the tegument, pass through the sparse reticulate cytoplasm in the middle region of the tegument, the inner plasma membrane and basement membrane into the subtegument.

The ultrastructure of the tegument of adult and larval Digenea are compared.

T cells are central to immunity in malaria. CD4+ helper T cells favour the generation of high-affinity antibodies that are effective against blood stages and they are necessary to establish immunological memory. The intrahepatic stage of infection can be eliminated by specific CD8+ cytotoxic T cells (CTL). Cytokines secreted by CD4+ T cells may also contribute to liver stage immunity. Evolution has selected varied mechanisms in pathogens to avoid recognition by T cells. T cells recognize foreign epitopes as complexes with host major histocompatibility (MHC) molecules. Thus, a simple form of evasion is to mutate amino acid residues which allow binding to an MHC allele. Recently, more sophisticated forms of polymorphic evasion have been described. In altered peptide ligand (APL) antagonism, the concurrent presentation of particular closely related epitope variants can prevent memory T cell effector functions such as cytotoxicity, lymphokine production and proliferation. In immune interference, the effect of the concurrent presentation of such related epitope variants can go a step further and prevent the induction of memory T cells from naive precursors. The analysis of immune responses to a protein of P. falciparum, the circumsporozoite protein (CSP), indicates that the malaria parasite may utilize these evasion strategies.

In contrast to the growing knowledge accumulated on plant resistance to pathogens, mechanisms of parasite resistance largely remain to be elucidated in animal species. In the present study we investigated mechanisms underlying resistance/susceptibility in the snail–trematode system Biomphalaria glabrata–Echinostoma caproni. In particular, we compared the effect of the parasite excretory–secretory (E–S) products on the defence functions of haemocytes from 2 susceptible and 2 resistant snail strains. In vitro experiments showed that E. caproni E–S products inhibit adhesion and phagocytosis of haemocytes from susceptible snails. A partial biochemical characterization also suggested that the interfering factor(s) is (are) heat-labile glycosylated polypeptides of molecular mass between 10 and 30 kDa. Interestingly, haemocytes from resistant snails remained unaffected by the parasite E–S products, suggesting that a constitutive difference results in their refractoriness to the parasite's immunosuppressive factor(s).

A delayed gamma distribution satisfactorily described the distribution of times to hatching of Ostertagia circumcincta eggs incubated in 0·1% saline at temperatures between 6 and 20 °C. Below 6 °C hatching of eggs was extremely variable. The relationship between rates of development and temperature within the range 10 to 20 °C was more closely described by a non-linear function than a linear one. The non-linear function was incorporated into a temperature summation equation which satisfactorily predicted the hatching of eggs incubated under conditions of alternating temperatures.

Activated oncospheres of Taenia ovis were incubated for 24 h or 48 h at 37.5°C in Medium 858 alone or in Medium 858 enriched with 20% lamb serum or foetal calf serum. Antigens contained in the supernatant medium from these incubations were concentrated, emulsified in Freund's incomplete adjuvant and injected intramuscularly into lambs. These lambs developed a very high level of immunity to subsequent challenge infection with T. ovis eggs.

Sexually reproducing pathogens such as Cyclospora cayetanensis often produce genetically heterogeneous infections where the number of unique sequence types detected at any given locus varies depending on which locus is sequenced. The genotypes assigned to these infections quickly become complex when additional loci are analysed. This genetic heterogeneity confounds the utility of traditional sequence-typing and phylogenetic approaches for aiding epidemiological trace-back, and requires new methods to address this complexity. Here, we describe an ensemble of two similarity-based classification algorithms, including a Bayesian and heuristic component that infer the relatedness of C. cayetanensis infections. The ensemble requires a set of haplotypes as input and assigns arbitrary distances to specimen pairs reflecting their most likely relationships. The approach was applied to data generated from a test cohort of 88 human fecal specimens containing C. cayetanensis, including 30 from patients whose infections were associated with epidemiologically defined outbreak clusters of cyclosporiasis. The ensemble assigned specimens to plausible clusters of genetically related infections despite their complex haplotype composition. These relationships were corroborated by a significant number of epidemiological linkages (P < 0.0001) suggesting the ensemble's utility for aiding epidemiological trace-back investigations of cyclosporiasis.

The comparative development of Echinococcus multilocularis was studied in its definitive hosts, the fox, dog, cat and raccoon dog, beyond the pre-patent period to 90 days post-infection. All host species, apart from cats were susceptible to infection and capable of supporting substantial worm burdens. Although worms in cats matured and produced thick-shelled eggs, their overall development was retarded compared to that in other species in which the parasite matured rapidly producing large populations of gravid worms. E. multilocularis matured rapidly in foxes and raccoon dogs and this was sustained in raccoon dogs but not in foxes in which maturation of worms declined during the later stages of infection, in contrast to that in both raccoon dogs and dogs. These populations were sustained for longer in raccoon dogs and dogs compared to foxes. Cats would appear to have only a minor role in the maintenance of E. multilocularis in endemic areas, and infections in cats may be of minimal public health significance. In contrast, foxes, dogs and the recently recognized definitive host the raccoon dog, are all capable of playing significant roles in the epidemiology of alveolar echinococcosis. This study also demonstrated that the developmental processes of growth, segmentation, proglottization and maturation in adult Echinococcus are independent and can be influenced by environmental factors thus confirming earlier in vitro observations.

The numerical relationships between a population of Lepeophtheirus pectoralis and its host population (plaice) have been described. There was no significant difference between infection levels in male and female plaice. In most of the monthly samples (75%) there was no significant relationship between host length and either the incidence or intensity of infection. The samples in which significant positive correlations occurred were usually taken within the period November to April. It is probable that the immigration of smaller fish, with lower levels of infection, from shallower water produces this effect.

The parasite population was found to have an overdispersed distribution (variance > mean) within its habitat, the host population. The frequency distributions of numbers of parasites per fish were calculated and the negative binomial distribution was fitted to these. The fit was good in 22 of the 24 samples and the value of k, the exponent, was found to be directly related to the mean density of the parasite population. Iwao's (1970) method of dispersion analysis indicated that the observed negative binomial was generated by a model of randomly distributed clumps or colonies of individuals. A possible mechanism of producing this pattern was proposed based on evidence from studies of mortality in the parasite population (which indicated that all-or-none type mortality operates on the clumps at certain periods during the year), the infection process (which demonstrated that a single wave of infection produces an overdispersed spatial distribution) and the examination of parasites from individual fish (which displayed a clumping of parasites at certain stages rather than an even spread over the range of possible stages). The mechanism proposed is that the adaptations for host location of the infective copepodid stage tend to produce aggregations of these larvae in favourable areas of a heterogeneous habitat. Fish moving randomly into these areas will tend to be infected by more than one larva (a clump of larvae) at any one time when available for infection. The fish can be regarded as equivalent to random samples of the free larval population and the aggregated distribution of the parasite population on its hosts is therefore directly related to the aggregated distribution of the free-swimming infective larvae.
