American trypanosomiasis is a neglected tropical disease whose spectrum has not been quite understood, including the impact of Trypanosoma cruzi infection on the haematological parameters of different vertebrate hosts. Thus, this study was designed to compare the pattern of haematological changes induced by T. cruzi infection in order to identify possible species-specific differences among taxons. We also aimed at evaluating the use of this parameter as a tool for diagnosis during the acute phase, when symptoms are usually masked. For this purpose, we performed a systematic search on PubMed and Scopus databases to retrieve original studies published until August 2016. Thirty-one studies were selected using Prisma strategy, which were then submitted to data extraction and methodological bias analysis. Half of the studies showed that the number of erythrogram decreased in infected animals, indicating anaemia. In 68.2% of the studies, the total amount of leukogram values increased, suggesting infection. The main methodological limitations were insufficient information for T. cruzi strains identification, inoculation routes and parasitological characterization. Most of the mammalian species analysed showed the same pattern of haematological changes following T. cruzi infection, indicating that haematological parameters might direct the diagnosis of Chagas disease in the initial phase.

Neurocysticercosis (NCC) is the most common helminthic brain infection related to epilepsy. Only albendazole (ABZ) and praziquantel are used in its treatment. The development of new therapeutics has been encouraged. Taenia crassiceps cysticerci intracranial infection is the experimental model used in NCC studies. This study evaluated the histopathology of the brains of BALB/c mice experimentally infected with T. crassiceps cysticerci after the treatment with the ABZ/nitazoxanide (NTZ) combination. Thirty days after the inoculation the mice received an oral single dose of the ABZ/NTZ combination (40 mg kg−1 each). The control groups were treated with: NaCl 0.9%; ABZ or NTZ. The histopathologic evaluation of the brains was performed 24 h after treatment. The ABZ treatment induced discrete mononuclear inflammatory infiltration, meningitis, gliosis, hyperaemia and hippocampus compression; moderate ependimitis and oedema. The NTZ treatment induced accentuated inflammatory infiltration, foamy macrophages, ependimitis, choroiditis, gliosis and hyperaemia and moderate oedema. The ABZ/NTZ combination treatment induced a significant decrease in the polymorphonuclear inflammatory infiltration, ependimitis, choroiditis, gliosis, hyperaemia and ventriculomegaly in comparison with the other groups. The cysticerci showed destruction of the tegument not observed in other groups. The ABZ/NTZ combination is efficient as the parasite showed signs of destruction and lower damage to the host's tissue.

Mounting an antibody response capable of discriminating amongst and appropriately targeting different parasites is crucial in host defence. However, cross-reactive antibodies that recognize (bind to) multiple parasite species are well documented. We aimed to determine if a higher inoculating dose of one species, and thus exposure to larger amounts of antigen over a longer period of time, would fine-tune responses to that species and reduce cross-reactivity. Using the Plasmodium chabaudi chabaudi (Pcc)–Nippostrongylus brasiliensis (Nb) co-infection model in BALB/c mice, in which we previously documented cross-reactive antibodies, we manipulated the inoculating dose of Pcc across 4 orders of magnitude. We investigated antigen-specific and cross-reactive antibody responses against crude and defined recombinant antigens by enzyme linked immunosorbent assay, Western blot and antibody depletion assays. Contrary to our hypothesis that increasing exposure to Pcc would reduce cross-reactivity to Nb, we found evidence for increased avidity of a subpopulation of antibodies that recognized shared antigens. Western blot indicated proteins of apparent monomer molecular mass 28 and 98 kDa in both Nb and Pcc antigen preparations and also an Nb protein of similar size to recombinant Pcc antigen, merozoite surface protein-119. The implications of antibodies binding antigen from such phylogenetically distinct parasites are discussed.

The effect of mouse population density on the establishment, survival and fecundity of single cohorts of Heligmosomoides polygyrus was investigated. No significant relationship was found between host population density and the worm burden at post mortem. But, lower host population densities were associated with higher parasite fecundities and with higher overall mean daily faecal output. The results might be explained by a decrease in male hormone secretion or digestive disturbances associated with lower ingesta in the higher host population densities. Effects resulting from changes in immunological responses might also exist.

The serum from rats infected intraperitoneally (i.p.) and subcutaneously (s.c.) with the tetrathyridia of Mesocestoides corti was analysed for α-amylase, alkaline phosphatase, γ-glutamyltransferase, glucose, corticosterone and total proteins. A histochemical analysis for hepatic alkaline phosphatase was also carried out. A significant decrease in activity of serum alkaline phosphatase was detected in both the i.p. and s.c. infections, whereas a large increase in hepatic alkaline phosphatase was observed in i.p. infections. A significant increase in serum cortiscosterone was found in both the i.p. and s.c. infections. No changes were detected in the other four components. The relevance of these findings is discussed with respect to the host–parasite relationship.

Dietary texture has been reported to influence parasite establishment and survival, but to what degree this relationship is modified by either the type or quantity of dietary fibre is unknown. Using a 2×4 factorial design, we explored the relationship between fibre type (soluble = pectin vs insoluble = cellulose) and fibre quantity (0, 5, 10 and 20% by dry weight) on parasitic outcomes in BALB/c mice infected with 100 Heligmosomoides polygyrus (Nematoda) larvae. Pectin, but not cellulose, exerted a significant effect on parasite egg production. Following in vitro culture of female worms, increasing levels of dietary pectin were associated with increasing release of eggs. Yet this pattern was not observed in vivo, where per capita egg production peaked at the 10% pectin concentration, but was very low in mice fed 20% pectin. Parasite establishment was elevated in mice fed 20% pectin, but was unaffected by cellulose concentration. Neither type nor quantity of fibre affected H. polygyrus survival or spatial distribution along the gastrointestinal tract. To what degree differences in parasite establishment and reproduction could be attributed to the marked effects of pectin on gut morphology (increased intestinal length, villus length, mucosa thickness and villus/crypt ratio) requires further exploration. Our data indicate that cellulose is preferable to pectin as the source of fibre for experimental diets as cellulose did not affect H. polygyrus establishment, reproduction or survival during a 4-week primary infection.

The ribosomal DNA (rDNA) unit of the liver fluke Opisthorchis viverrini has been cloned and characterized. The results demonstrated that the total length of this unit is approximately 13 kb, containing 4·2 kb of large subunit (LSU) rDNA, 2·0 kb of small subunit (SSU) rDNA, 1·0 kb of transcribed spacer DNA and 5·8 kb of non-transcribed + external transcribed spacer DNA. Examination of the non-transcribed spacer region between different rDNA units showed variation in the restriction sites rather than in the length. Judging from hybridization of the rDNA plasmid to the restriction endonuclease digest of genomic DNA, rDNA units represent 6·1% of the total genomic DNA. At the RNA level, the LSU rRNA of O. viverrini and Fasciola gigantica contained a hidden break. The molecule consisted of two fragments co-migrating with a SSU rRNA, when electrophoresis was carried out under denaturing conditions. Ribosomal RNA sequence comparison has been previously used to determine phylogenetic classification of parasitic organisms. The sequence of 381 nucleotides at the 5′ terminus of the LSU rRNA gene was determined and compared with those from species previously reported by other investigators. Phylogenetic classification of O. viverrini, as determined by rRNA gene sequence comparison, is comparable with the conventional taxonomic classification.

During the course of an investigation into the host-relationship of Ixodes hexagonus Leach a large number of birds was examined for its occurrence. These all proved negative for this species, but I. passericola Sch. was present on one robin out of 220 examined, on one starling out of eleven, on seventeen great tits out of 163, on twenty blue tits out of 540, and on four nuthatches out of fifty. A detailed report on this aspect is given later.

Studies on the susceptibility of the guinea-pig to infection with Schistosoma spindale Montgomery are reported.

Of twenty-two guinea-pigs, examined 33–153 days after exposures to 8000–50,000 cercariae of S. spindale, seventeen furnished conclusive evidence of the development of the female schistosome and two furnished inconclusive evidence. The male S. spindale developed in all the animals.

The female schistosomes attained maturity in 48 days or more and laid viable ova which could be detected in the liver, intestinal wall and lungs, but not in faeces.

The pathological lesions caused by the parasite consisted of thrombosis in the liver and lungs due to adult worms and pseudotubercles in the liver and nodules in the intestinal wall due to the eggs.

The guinea-pig shows varying degrees of individual susceptibility to infection with S. spindale and is an unsuitable host for the parasite, more especially for the female schistosome.

In 1911, I received from Dr Georgina Sweet of Melbourne University, two lots of Trematodes stated to have been collected respectively from the intestine of the tiger snake, Notechis scutatus, and the lungs of the copperhead snake, Denisonia superba. In 1916, Dr Burton Cleland, of the Bureau of Microbiology, Sydney, sent me two tubes containing similar parasites, obtained from the peritoneum of an unidentified snake killed on Flinders Island.

Multiple infections, those by more than one parasite in the same erythrocyte, may be adaptive for the malarial parasite as a means to ensure fertility. Alternatively they may simply be the consequence of a non-adaptive process forcing several parasites to compete for resources in one host cell. Avian hosts infected with Haemoproteus were medicated with primaquine or injected with saline solution and the density of infection and number, maturity and sex of mature multiple infections counted. Multiple infections depend on density of infection, and maturity is attained rarely and usually by gametocytes of the same sex. The role of multiple infections for fertility insurance is not supported by these results.

The Leloir pathway enzyme uridine diphosphate (UDP)-galactose 4′-epimerase from the common liver fluke Fasciola hepatica (FhGALE) was identified and characterized. The enzyme can be expressed in, and purified from, Escherichia coli. The recombinant enzyme is active: the Km (470 μm) is higher than the corresponding human enzyme (HsGALE), whereas the kcat (2·3 s−1) is substantially lower. FhGALE binds NAD+ and has  shown to be dimeric by analytical gel filtration. Like the human and yeast GALEs, FhGALE is stabilized by the substrate UDP-galactose. Molecular modelling predicted that FhGALE adopts a similar overall fold to HsGALE and that tyrosine 155 is likely to be the catalytically critical residue in the active site. In silico screening of the National Cancer Institute Developmental Therapeutics Program library identified 40 potential inhibitors of FhGALE which were tested in vitro. Of these, 6 showed concentration-dependent inhibition of FhGALE, some with nanomolar IC50 values. Two inhibitors (5-fluoroorotate and N-[(benzyloxy)carbonyl]leucyltryptophan) demonstrated selectivity for FhGALE over HsGALE. These compounds also thermally destabilized FhGALE in a concentration-dependent manner. Interestingly, the selectivity of 5-fluoroorotate was not shown by orotic acid, which differs in structure by 1 fluorine atom. These results demonstrate that, despite the structural and biochemical similarities of FhGALE and HsGALE, it is possible to discover compounds which preferentially inhibit FhGALE.

Dicyemids, poorly known parasites of benthic cephalopods, are one of the few phyla in which mitochondrial (mt) genome architecture departs from the typical ~16 kb circular metazoan genome. In addition to a putative circular genome, a series of mt minicircles that each comprises the mt encoded units (I–III) of the cytochrome c oxidase complex have been reported. Whether the structure of the mt minicircles is a consistent feature among dicyemid species is unknown. Here we analyse the complete cytochrome c oxidase subunit I (COI) minicircle molecule, containing the COI gene and an associated non-coding region (NCR), for ten dicyemid species, allowing for first time comparisons between species of minicircle architecture, NCR function and inferences of minicircle replication. Divergence in COI nucleotide sequences between dicyemid species was high (average net divergence = 31·6%) while within species diversity was lower (average net divergence = 0·2%). The NCR and putative 5′ section of the COI gene were highly divergent between dicyemid species (average net nucleotide divergence of putative 5′ COI section = 61·1%). No tRNA genes were found in the NCR, although palindrome sequences with the potential to form stem-loop structures were identified in some species, which may play a role in transcription or other biological processes.

The study aims to evaluate the antiprotozoal activities of 20 plant metabolites on Trypanosoma cruzi and Leishmania amazonensis amastigotes. Compounds 1–20 were obtained and identified by using chromatographic and spectroscopic techniques. The antiparasitic assays were performed on the intracellular form of T. cruzi and L. amazonensis using human leukaemic THP-1 cells as the host. The mechanism of action of the most active compounds was explored in silico by molecular docking using T. cruzi trypanothione reductase (TR) as a target, whereas the in vitro studies were performed by enzymatic assay using T. cruzi recombinant TR. In addition, the mitochondrial membrane potential was evaluated by flow cytometry. Two flavonoids, one triterpene and three acetogenins showed from high to moderate trypanocidal activities with IC50 values ranging 3.6–37.2 µm while three of the metabolites were moderately leishmanicidal. The molecular docking study revealed interactions between TR and the most trypanocidal compounds 1 (abyssinone IV) and 2 (atalantoflavone). In contrast, both showed no effect on TR in vitro. For the mitochondrial membrane potential assay, atalantoflavone (2) displayed a dose-dependent depolarization. On the basis of the aforementioned results, this compound's structure could be chemically explored in order to develop more potent trypanocidal derivatives.

The free-living larval stages of Bunostomum trigonocephalum are better adapted to low temperatures than high temperatures. Most survived at 10°C.

In tap water, 1st-stage larvae survived for 11 days at 0°C, 17 days at 5°C, 20 days at 10°C, 15 days at 15°C, 18 days at 20°C, 15 days at 25°C, 9 days at 30°C, 7 days at 34°C, 4 days at 35°C, 16 h at 40°C, 40 min at 45°C and 20 min at 50°C.

In tap water infective larvae survived for 40 days at 0°C, 70 days at 5°C, 100 days at 10 and 15°C. 65 days at 20°C, 48 days at 25°C, 38 days at 30°C, 14 days at 34°C, 6 days at 35°C, 5 days at 40°C, 3 h at 45°C and 105 min at 50°C.

In tap water at room temperature infective larvae survived for 26 days in January, 20 days in February, 21 days in March, 13 days in April, 10 days in May, 6 days in June, 9 days in July and August, 11 days in September, 12 days in October, 14 days in November and 17 days in December.

There is great heterogeneity in parasite communities among hosts, understanding the nature and drivers of such variations is still a great scientific quest. Here, we analyse the variation in parasite communities by addressing the following questions: (i) What is the beta-diversity component (nestedness or turnover) that most contributes to beta diversity in parasite communities among anuran species? (ii) Does the beta diversity of parasite communities follow a non-random pattern? (iii) Is the dissimilarity in composition of parasite communities related to the phylogenetic or functional dissimilarity among hosts? We found that turnover in parasite assemblages was the main component of beta diversity, but the variation observed both in the total beta diversity and in its components did not differ from the respective null models. The dissimilarity among parasite communities was not related to the phylogenetic species variability or functional dissimilarity among anuran species for most localities. In short, our findings may indicate a process of resource tracking by the parasite species, in which the resource may not necessarily be conserved phylogenetically in their hosts.

Camelids (llama, alpaca, vicunãs, guanacos) are important for the economy of South America and Eimeria infections are an important cause of mortality in camelids. Of the six species of Eimeria in camelids, Eimeria macusaniensis, considered the most pathogenic, is distinctive; its oocysts are the largest among all Eimeria species in animals, its prepatent period is more than 1 month, and its oocysts have been found in mummies from prehistoric times. Although, E. macusaniensis gametogonic stages are found associated with enteritis in naturally infected camelids, the schizogonic stages are unknown and clinical disease has been reported in some camelids with no oocysts in feces. Described herein are morphological details of gametogonic development and oocyst formation of E. macusaniensis in a naturally infected llama (Lama lama), solely infected with this parasite. Microgamonts, macrogamonts and oocysts were located in large (up to 300 µm diameter) parasitophorous vacuoles of enterocytes in the ileum. Schizonts were not found. Review of previous reports suggests that multinucleated microgamonts have been mistaken for schizonts. Gametogonic development described in the present study can serve as a guide for differential diagnosis of Eimeria species in the histological sections of intestines.
