Polycercus lumbrici, previously described only by Metchnikov (1868) in Russia, has been found in Scotland in the earthworm Allolobophora terrestris.

Experimental infestation of earthworms with Polycercus lumbrici as a result of feeding them with the eggs of Paricterotaenia paradoxa shows that the proliferating cysts are the larvae of the latter as suggested by Joyeux and Baer (1939).

There is evidence that Polycercus may induce parasitic castration of the earthworm in which it occurs.

Metchnikov's description of the development of the larva is confirmed for the most part but is modified to include a retroversion of the larva in its early stages of development. The development of the scolex is shown to follow a pattern similar to that of Cysticercus fasciolaris in that the hooks are borne on a ‘prebulb’ which later invests a ‘bulb’, the former giving rise to the muscles of the walls of the double sac of the rostellum, the latter to the hook-elevator muscles.

The rostellar hooks of P. paradoxa are elevated by the action of a double layer of muscles which radiate from a conical invagination of the bulb. These muscles, in association with the muscles of the wall of the rostellum, elevate the hooks by modifying the shape of the rostellum rather than by direct action on the hooks.

The nervous system and the excretory system in the scolex of P. paradoxa are briefly described and the nature of the ‘glandular’ structure of the rostellum is discussed.

In earlier studies, proteomic characterization of splenic amastigote fractions from clinical isolates of Leishmania donovani, exhibiting significant cellular responses in cured Leishmania subjects, led to the identification of cytosolic tryparedoxin peroxidase (LdcTryP) and chaperonin-TCP20 (LdTCP20) as Th1-stimulatory proteins. Both the proteins, particularly LdTCP20 for the first time, were successfully cloned, overexpressed, purified and were found to be localized in the cytosol of purified splenic amastigotes. When evaluated against lymphocytes of cured Leishmania-infected hamsters, the purified recombinant proteins (rLdcTryP and rLdTCP20) induced their proliferations as well as nitric oxide production. Similarly, these proteins also generated Th1-type cytokines (IFN-γ/IL-12) from stimulated PBMCs of cured/endemic Leishmania patients. Further, vaccination with rLdcTryP elicited noticeable delayed-type hypersensitivity response and offered considerably good prophylactic efficacy (~78% inhibition) against L. donovani challenge in hamsters, which was well supported by the increased mRNA expression of Th1 and Th2 cytokines. However, animals vaccinated with rLdTCP20 exhibited comparatively lesser prophylactic efficacy (~55%) with inferior immunological response. The results indicate the potentiality of rLdcTryP protein, between the two, as a suitable anti-leishmanial vaccine. Since, rLdTCP20 is also an important target, for optimization, further attempts towards determination of immunodominant regions for designing fusion peptides may be taken up.

Plasma etching has been used to strip away the cortical layers of the cuticle of adult Nippostrongylus brasiliensis to reveal the struts, with their supporting fibres, which are found in the fluid-filled middle layer of the cuticle, and the basal fibre layers. The etched specimens were studied by means of scanning electron microscopy. The results support earlier work, obtained by transmission electron microscopy, on the cuticle of this nematode. Plasma etching has been shown to have potential in studying the structure of nematodes.

Although the majority of rotifers are free-living, there are many records of rotifers parasitizing both plants and animals (Thompson, 1892; Budde, 1925; Remane, 1929). The most marked development of this parasitic habit is found in the genera Balatro, Drilophaga, Zelinkiella and Albertia. In these forms, with the exception of the genus Zelinkiella, attachment to the host is performed by the mastax. Albertia vermicularis Dujardin (1838) has been recorded as a parasite of the intestine and body cavity of earthworms. This author also found specimens in slugs, but he did not indicate the host species involved in either case. This original description was rather incomplete, and it is the object of this paper to expand it by a description of specimens from Allolobophora caliginosa.

1. The suckling 10-day-old mouse harboured fewer worms than the weanling 15-day-old mouse after both 5 and 10 days' infection.

2. The adult male mouse, however, harboured significantly more worms when fed on milk than when fed on rat cake after both 5 and 10 days' infection.

3. These results were discussed in relation to the effects of diet and gut length on the susceptibility of the host.

During its life-cycle, the cestode Schistocephalus solidus is parasitic in both an ectotherm (Gasterosteus aculeatus) and an endotherm (Gallus domesticus) host, and so provides an excellent model with which to study temperature adaptation in parasites. A mitochondrial fraction was prepared from the adults and plerocercoids of S. solidus and from their respective hosts; the activities of the mitochondrial adenosine triphosphatase (ATPase) were then measured over the temperature range 1–45 °C. The plerocercoids of S. solidus show evidence of immediate temperature compensation; this would provide a mechanism for withstanding the abrupt temperature change experienced during infection of the final host. Analysis of the Michaelis constant data suggests that variation of Km, a with temperature may be a major factor in this immediate temperature compensation. In response to acclimation at 5 and 19°C, plerocercoid ATPase showed inverse or paradoxical rate compensation, as did the enzyme from the fish host. Acclimation at the two temperatures had no effect on the Q10 or on the linearity of the Arrhenius plots for the plerocercoid mitochondrial ATPase and only a small effect on the Km a. Acclimation of the fish host again had only a small effect on the Km, a of the fish mitochondrial ATPase but, in contrast to the plerocercoid, there was also a significant effect on the Q10 and the Arrhenius plots. Adult S. solidus ATPase showed partial rate compensation and had a biphasic Arrhenius plot, suggesting that after infection there had been a change in the enzyme or its micro-environment. In terms of the effect of temperature on the Q10 amd Km, a and in the biphasic nature of the Arrhenius plot, the mitochondrial ATPase of adult S. solidus showed similarities with the enzyme from its bird host.

Ion channels are important target sites of anthelmintics, but little is known about those in Fasciola hepatica. In this work, we applied a planar lipid bilayer technique to characterize the properties of single ion channels in F. hepatica. Under a 200/40 mM KCl gradient, a large conductance channel of 251 pS was observed in 18% of the membranes studied. The channel was selective to K+ over Cl− with a permeability ratio of K+ to Cl− (PK/PCl) of 4·9. Open state probability (Po) of the channel was less than 0·5 and dependent on voltage (−60~+40 mV) and Ca2+ (~100 μM). The other two types of single channels observed in 11 and 5% of membranes, respectively, were a K+-permeable channel of 80 pS (PK/PCl=4·6) and a Cl−-permeable channel of 64 pS (PK/PCl=0·058). Open state probability of both channels showed little voltage dependence. The results indicate that distinct single channels of 60~251 pS are present in relative abundance and, in addition, that the planar lipid bilayer technique can be a useful tool for the study of single ion channels in F. hepatica.

In the Freetown area of Sierra Leone, the proportion of children under five years of age dying from diarrhoea-associated conditions has decreased since the 1970s from 4% to 3%. This reduction relates to children between 12 months and 5 years; deaths associated with diarrhoeal disease in infancy do not appear to have decreased significantly. Currently, 2% of children die in infancy and a further 1% before their fifth birthday from diarrhoeal diseases. Thirty-one percent of short-term growth-faltering in children under five years of age is attributed to diarrhoeal disease. Reasons for improvement in mortality may include the implementation of a National Diarrhoeal Diseases Control Programme and improved case management; some 55% of cases of acute diarrhoea in Freetown now receive oral rehydration therapy. Morbidity and mortality are influenced by the seasons. This is likely to be related to the fact that the nutritional status of children shows a significant deterioration in the early rainy season. At this time incidence of severe diarrhoea warranting hospital admission and diarrhoea mortality rates in hospital and at a Rehydration Center increased significantly. On the other hand, the presentation of dehydrating diarrhoea is greatest in the driest months of the year. Progress in the rural areas of Sierra Leone may not have been so evident although 60% of the population now have access to primary health units within 4 miles and efforts to increase host resistance have included the measles immunization programme which currently protects 60% of under-fives nation-wide. Efforts to reduce the transmission of pathogens have meant increasing access to safe water to 22% and to improved sanitation to 35% in rural areas of Sierra Leone. The sustainable prevention and control of diarrhoeal diseases in developing countries depends also upon increasing levels of adult literacy especially of women.

Some members of the ABC-transporter superfamily, such as P-glycoprotein and the multidrug resistance associated protein, may confer resistance to the avermectin subclass of macrocyclic lactones. The aim of this study was to examine the presence of ABC transporters in both sea lice (Lepeophtheirus salmonis) and its Atlantic salmon host (Salmo salar) using monoclonal antibodies (C219 and JSB-1, with high selectivity for P-gp) and a new polyclonal antibody (SL0525) generated against a putative sea louse ABC transporter. The antibody raised to SL0525 did not react with rat P-gp, suggesting that an ABC transporter, not necessarily P-gp, was isolated. C219 was the only antibody to localize P-gp in all 3 salmon tissues (intestine, kidney and liver). American lobster (Homarus americanus) was used as a reference crustacean for L. salmonis immunostaining reactions and showed positive staining in the hepatopancreatic and intestinal tissues with all 3 antibodies. The L. salmonis showed positive staining in the intestinal epithelial lining with all antibodies. This report represents the first documented evidence for the expression of ABC transporters in L. salmonis, its Atlantic salmon host, and the American lobster.

L-Amino acid esters, such as L-Leu-OMe, kill Leishmania amazonensis amastigotes by a mechanism which appears to involve ester hydrolysis by cysteine proteinases located in the parasite megasomes. We have examined the killing of isolated amastigotes by L-dipeptide esters and derived some structure-activity correlations. Toxicity of the compounds for the parasites was measured by a tetrazolium (MTT) reduction assay. The results show that active dipeptide esters contained at least I hydrophobic amino acid (Leu, Ile, Val, Phe or Trp). The activity of homodipeptide methyl esters depended on the nature of the amino acid, as indicated by the following series: Phe-Phe-OMe < Val-Val-OMe < Leu-Leu-OMe < Trp-Trp-OMe < Ile-Ile-OMe. The nature of the amino acids in Leu-X-OMe and X-Leu-OMe was relatively unimportant when X was Phe, Trp or Val. However, when X was Ala or Gly, Leu-X-OMe was several-fold more active than X-Leu-OMe.

To evaluate their protective potential, excretions and secretions of adult female Angiostrongylus cantonensis were prepared from an in vitro culture fluid and injected together with Freund's complete and incomplete adjuvants into rats and mice. Under appropriate conditions, judging from reduced mortality rates and/or prolonged survival periods, these animals were protected against a lethal challenge infection with 3rd-stage larvae. Although both ‘immunized’ and unimmunized mice died following such a lethal infection, the survival period for the ‘immunized’ group was significantly longer than that of unimmunized controls (40·8 compared with 26·7 days). On the other hand, the mortality rate of ‘immunized’ rats was reduced to 25% as compared with 100% in unimmunized controls. There was also evidence of adverse effects on parasite development in these ‘immunized’ rats. The number of young adult worms recovered from the brain 21 days after challenge infection and both the number and size of mature male and female worms in the lungs 60 days after infection were significantly reduced in the ‘immunized’ rats. Moreover, the reproductive activity of female worms was depressed, as evident from the output of 1st-stage larvae in the faeces and the number of eggs released during maintenance in vitro. The quantity of proteins released during in vitro maintenance of adult worms in a chemically defined serum-free medium was 0·2–0·5 μ/worm/day. The concentrated culture fluid contained at least 7 different antigenic components when analysed with hyperimmune rabbit antiserum to female excretions and secretions (FES) antigen. Polyacrylamidegelelectrophoresis in sodium dodecylsulphate showed the presence of 3 major and at least 12 minor protein components with molecular weights varying from 24000 to 115000 daltons. Different lines of evidence presented in this study show immunological cross-reactivity among antigens from different developmental stages.

The group of haemosporidian parasites is of general interest to basic and applied science, since several species infect mammals, leading to malaria and associated disease symptoms. Although the great majority of haemosporidian parasites appear in bird hosts, as in the case of Leucocytozoon buteonis, there is little genomic information about genetic aspects of their co-evolution with hosts. Consequently, there is a high need for parasite-enrichment strategies enabling further analyses of the genomes, namely without exposure to DNA-intercalating dyes. Here, we used flow cytometry without an additional labelling step to enrich L. buteonis from infected buzzard blood. A specific, defined area of two-dimensional scattergramms was sorted and the fraction was further analysed. The successful enrichment of L. buteonis in the sorted fraction was demonstrated by Giemsa-staining and qPCR revealing a clear increase of parasite-specific genes, while host-specific genes were significantly decreased. This is the first report describing a labelling-free enrichment approach of L. buteonis from infected buzzard blood. The enrichment of parasites presented here is free of nucleic acid-intercalating dyes which may interfere with fluorescence-based methods or subsequent sequencing approaches.

The localization of adult Schistosoma mansoni originating from Guadeloupe (West Indies) was analysed in the natural host Rattus rattus in experimental and natural infections. A transfer of schistosomes from the porto-mesenteric system to the lungs occurs between the 4th and the 20th weeks post-infection, with a peak between 4 and 8 weeks; it should be noted that the worms start laying eggs at 4 weeks post-infection. In both experimentally and naturally infected R. rattus the relocation of schistosomes in the lungs is correlated with the total worm burden. Between 6 and 12 weeks post-infection some adult worms and numerous eggs are trapped in the liver. A high mortality in the worm population coincides with the migration of the parasites from the porto-mesenteric veins to the lungs. In re-infected R. rattus a relationship appears between the presence of schistosomes in the lungs and resistance to reinfection. It is concluded that, as already postulated previously in mice, resistance to re-infection in the natural host R. rattus is at least partly correlated with the hepatic pathology caused by the schistosomes of the primary infection.

Mermithid nematodes (Nematoda: Mermithida: Mermithidae) parasitize a wide range of both terrestrial and aquatic invertebrate hosts, yet are recorded in bumble bees (Insecta: Hymenoptera: Apidae: Bombus) only six times historically. Little is known about the specific identity of these parasites. In a single-season nationwide survey of internal parasites of 3646 bumble bees, we encountered six additional instances of mermithid parasitism in four bumble bee species and genetically characterized them using two regions of 18S to identify the specific host–parasite relationships. Three samples from the northeastern USA are morphologically and genetically identified as Mermis nigrescens, whereas three specimens collected from a single agricultural locality in the southeast USA fell into a clade with currently undescribed species. Nucleotide sequences of the V2–V6 region of 18S from the southeastern specimens were 2.6–3.0% divergent from one another, and 2.2–4.0% dissimilar to the nearest matches to available data. The dearth of available data prohibits positive identification of this parasite and its affinity for specific bumble bee hosts. By doubling the records of mermithid parasitism of bumble bee hosts and providing genetic data, this work will inform future investigations of this rare phenomenon.

Evolutionary theory predicts that the sex ratio of Plasmodium gametocytes will be determined by the number of gametes produced per male gametocyte (male fecundity), parasite clonal diversity and any factor that reduces male gametes' ability to find and combine with female gametes. Despite the importance of male gametocyte fecundity for sex ratio theory as applied to malaria parasites, few data are available on gamete production by male gametocytes. In this study, exflagellating gametes, a measure of male fecundity, were counted for 866 gametocytes from 26 natural infections of the lizard malaria parasite, Plasmodium mexicanum. The maximum male fecundity observed was 8, but most gametocytes produced 2–3 gametes, a value consistent with the typical sex ratio observed for P. mexicanum. Male gametocytes in infections with higher gametocytaemia had lower fecundity. Male fecundity was not correlated with gametocyte size, but differed among infections, suggesting genetic variation for fecundity. Fecundity and sex ratio were correlated (more female gametocytes with higher fecundity) as predicted by theory. Results agree with evolutionary theory, but also suggest a possible tradeoff between production time and fecundity, which could explain the low fecundity of this species, the variation among infections, and the correlation with gametocytaemia.

This study furthers understanding of cestode infections in a marine environment through time and space by following seasonal fluctuations in infection parameters of three cestode species (Gilquinia squali, Trilocularia gracilis and Phyllobothrium squali) parasitizing spiny dogfish (Squalus acanthias) in the northwest Atlantic and comparing them to work previously published from the northeast Atlantic on T. gracilis. For each cestode species, host size, season and presence of the other cestode species were analysed using generalized linear models to determine if they were good predictors of prevalence and intensity. Infection parameters differed across season for the three cestode species. However, within T. gracilis seasonal trends were found to be remarkably similar on both sides of the Atlantic, differing only in a somewhat delayed decline in prevalence in the northwest Atlantic. The differences seen in infection measures across cestode species likely reflect the unique life history strategies of different parasite species. While general trends appear to be maintained across disparate localities, variation seen is likely due to differences in accessibility to intermediate hosts and host diet across sites. The knowledge gained from understanding cestode infections in the vast ocean environment allows us to speculate about the factors driving fluctuations in parasite infections in elasmobranchs.

Pathogens are increasingly implicated in amphibian declines but less is known about parasites and the role they play. We focused on a genus of nematodes (Rhabdias) that is widespread in amphibians and examined their genetic diversity, abundance (prevalence and intensity), and impact in a common toad (Rhinella horribilis) in Panama. Our molecular data show that toads were infected by at least four lineages of Rhabdias, most likely Rhabdias pseudosphaerocephala, and multiple lineages were present in the same geographic locality, the same host and even the same lung. Mean prevalence of infection per site was 63% and mean intensity of infection was 31 worms. There was a significant effect of host size on infection status in the wild: larger toads were more likely to be infected than were smaller conspecifics. Our experimental infections showed that toadlets that were penetrated by many infective Rhabdias larvae grew less than those who were penetrated by few larvae. Exposure to Rhabdias reduced toadlet locomotor performance (both sustained speed and endurance) but did not influence toadlet survival. The effects of Rhabdias infection on their host appear to be primarily sublethal, however, dose-dependent reduction in growth and an overall impaired locomotor performance still represents a significant reduction in host fitness.

Since the discovery of Perkinsus marinus as the cause of dermo disease in Crassostrea virginica, salinity and temperature have been identified as the main environmental drivers of parasite prevalence. However, little is known about how these variables affect the movement of the parasite from host to water column. In order to elucidate how environmental factors can influence the abundance of this parasite in the water column, we conducted a series of experiments testing the effects of time of day, temperature and salinity on the release of P. marinus cells from infected oysters. We found that P. marinus cells were released on a diurnal cycle, with most cells released during the hottest and brightest period of the day (12:00–18:00). Temperature also had a strong and immediate effect on the number of cells released, but salinity did not, only influencing the intensity of infection over the course of several months. Taken together, our results demonstrate that (1) the number of parasites in the water column fluctuates according to a diurnal cycle, (2) temperature and salinity act on different timescales to influence parasite abundance, and (3) live infected oysters may substantially contribute to the abundance of transmissive parasites in the water column under particular environmental conditions.

Angiostrongylus cantonensis (A. cantonensis) is the most common cause of parasitic eosinophilic meningitis worldwide. By using an animal model of BALB/c mice infected with A. cantonensis, previous studies indicated that the anthelmintic drug, albendazole, could kill A. cantonensis larvae and prevent further infection. However, the dead larvae will induce severe immune responses targeting at brain tissues. To alleviate the detrimental effects caused by the dead larvae, we administered curcumin, a traditional anti-inflammatory agent, as a complementary treatment in addition to albendazole therapy, to determine whether curcumin could be beneficial for treatment. The results showed that although curcumin treatment alone did not reduce worm number, combined treatment by albendazole and curcumin helped to reduce eosinophil count in the cerebrospinal fluid, better than using albendazole alone. This alleviating effect did not affect albendazole treatment alone, since histological analysis showed similar worm eradication with or without addition of curcumin. Nevertheless, curcumin treatment alone and combined albendazole-curcumin treatment did not inhibit MMP-9 expression in the brain tissue. In conclusion, curcumin, when used as a complementary treatment to albendazole, could help to alleviate eosinophilic meningitis through suppression of eosinophil count in the cerebrospinal fluid.
