The presence of retinol-binding protein (RBP) activity in Onchocerca cervicalis adult worms and interaction with ivermectin has been studied using high pressure size exclusion chromatography (HPSEC). Four distinct peaks of [3H]-retinol incorporation were obtained corresponding to approximate molecular weights of 150, 67, 19·7 and 4–6 kDa, the 2 smaller Mr peaks accounting for most of the binding activity. Competition for binding using non-labelled retinol at 200-fold molar excess indicated that specific binding of retinol occurred only to the 19–7 kDa fraction. Competition by ivermectin also inhibited binding of [3H]-retinol to the third peak. Following incubation with [3H]-ivermectin & peaks of similar molecular weights were also detected by HPSEC in soluble adult worm homogenate, However, in this case the 150 kDa fraction was most prominent. Both non-labelled ivermectin and non-labelled retinol at 200-fold molar excess reduced binding of [3H]-ivermectin to all & fractions. These data indicate that the putative Onchocerca RBP has an approximate molecular weight of 19·7 kDa, that retinol also binds to 3 additional fractions non-specifically, that the pattern of binding of ivermectin to adult worm material is quantitatively and qualitatively different from the binding exhibited by retinol, and that ivermectin interferes with the binding of retinol to the 19·7 kDa Onchocerca protein.

The incidence of Helminthiasis at Mombasa is herein briefly dealt with chiefly in tabular form. The tables give the result of the first 100 examinations conducted on natives of indigenous races complaining of intestinal troubles, admitted into or attending hospital.

Anthomyia pluvialis Linn. (1761) is a very common and beautiful fly, widely distributed in the old world, and easily recognisable by having characteristic black spots on the thorax and abdomen.

A mathematical model that describes the transmission dynamics of Theileria annulata is proposed that consists of 2 host components: the Hyalomma tick population and a compartmental model of T. annulata infection in the cattle population. The model was parameterized using data describing tick infestation and the infection status of cattle in Turkey from 2006 to 2008. The tick attachment rates are highly seasonal and because of the temporal separation of infectious and susceptible ticks virtually all ticks are infected by carrier cattle, so that annual peaks of disease in cattle do not impact on infection in the Hyalomma tick population. The impact of intervention measures that target the tick population both on the host and in the environment and their impact on the transmission of T. annulata were investigated. Interventions that have a limited ‘one-off’ impact and interventions that have a more permanent impact were both considered. The results from the model show the importance of targeting ticks during the period when they have left their first host as nymphs but have yet to feed on their second host.

This study aimed to set up methodology to monitor parasite-specific T-cell activation in vitro using Eimeria tenella-infected chickens. A sonicated E. tenella sporozoite protein preparation was used for the activation of chicken spleen cell cultures. Proliferation assessed by 3H-thymidin incorporation or blast transformation of T-cells assessed by immunofluorescence labelling and flow cytometry were used as read-outs for activation. Results showed that E. tenella-specific proliferation was detected in cultures of spleen cells collected in a ‘window’ between 8 and 14 days after primary infection. However, due to high variation in proliferative responses between individuals and to high background proliferation, large numbers of observations were needed to obtain significant results. Moreover, the outcome was not improved by increasing the infection dose to chickens or by depletion of T-cell receptor (TCR) γ/δ expressing cells from cultures. An E. tenella-specific blast transformation response was observed for TCRα/β expressing cells within the same ‘window’, confirming the identity of the responding cells as classic T-cells. Thus, it is possible to study the kinetics of E. tenella-specific T-cell responses in vitro. However, more in-depth phenotypic identification of the responding T-cells could improve the methodology.

Observations have been made on the frequency distribution of strongyloid infective larvae in random samples of pasture herbage collected on four occasions from a quarter-acre field grazed by five sheep carrying mixed strongyloid infections. The distribution of numbers of infective larvae of the genera Haemonchus, Trichostrongylus and Cooperia in each of the samples has been examined. The variance of each series was found to be substantially larger than its mean and, hence, the distributions were all highly overdispersed statistically. Of a number of frequency distribution models, the ‘truncated lognormal’ was found to provide the best empirical fit to the sample distributions.

The implications of these findings for determining the significance of the difference between two random sample estimates of the abundance of infective larvae in pasture, have been discussed. It has been shown that, because of the limit imposed by technical considerations on the maximum size of herbage samples, and because the distribution of infective larvae in pasture departs markedly from a random one, only quite large differences between the means of two random sample estimates are likely to be significant. However, because the numbers of infective larvae of the different strongyloid genera are highly correlated within sample units, considerably smaller differences between the means for different genera, within the same sample, can be detected as significant.

From a consideration of the complexity of the biological components generating populations of infective larvae on pasture, it has been concluded that simple frequency distribution models can contribute little towards an improved understanding of the population dynamics of strongyloid infective larvae in pastures grazed by sheep.

Mature asexual stages of the malaria parasite Plasmodium knowlesi synthesize proteins of Mr 180 000–225 000 that are expressed on the outer membrane of infected erythrocytes and which vary antigenically such that different parasite clones are specifically agglutinated with homologous antibody. Other non-agglutinable clones have been prepared which fail to express variant antigen on infected cells. Two agglutinable clones of different variant antigen phenotypes and a non-agglutinable clone were examined to determine the proportion of total malarial proteins represented by variant antigens. Malarial proteins were labelled with various radioactive amino acids and the sodium dodecyl sulphate—polyacrylamide gel patterns for the three clones compared by fluorography. The patterns were indistinguishable, the variant antigens being undetectable in analyses of total malarial proteins. Furthermore, these antigens were not detected by Coomassie Blue-staining of total cellular proteins after electrophoresis. Sodium dodecyl sulphate and Triton X-100 extracts of labelled cells were immunoprecipitated using a panel of sera of defined agglutination specificity. The variant antigens could not be detected in the fluorographic patterns of total malarial antigens immuno-precipitated by these sera. In contrast, after lactoperoxidase catalysed radio-iodination of intact schizont-infected cells, the 125 I-variant antigens on the cell surface were identified by demonstrating their accessibility both to antibody and to trypsin with intact cells. Thus, the variant antigens are quantitatively very minor malarial proteins that can only be detected by methods which selectively analyse the subset of proteins on the erythrocyte surface.

Most of the specimens described in the following notes were collected by the International Fishery Commission's S.S. Huxley, in the North Sea, during the years 1905–09. They were preserved in dilute formalin in sea water, or in Bles's Fluid. Owing to the length of time during which the specimens had remained in the preservative, and in some cases owing to their large size, the fixation and preservation were not in every case as perfect as might have been wished, so that many of the finer histological details could not always be determined.

The results of several experiments that included variation of the pH and concentration of the extracts and analysis of the role of enzymes separated from midgut fluid by electrophoresis indicated that hatching involved two or more proteolytic enzymes that digested the cytoplasmic layer and embryophore. Amylases from T. molitor and other sources were shown to play no part in hatching.A defined medium based on analyses of midgut fluid and containing trypsin, chymotrypsin and peptidases was shown to digest the embryophore and effect the release of hexacanths in a time comparable to that obtained with T. molitor extracts. The specificity of the enzymes attacking the cytoplasmic layer remains unknown.

Neospora caninum is an apicomplexan parasite strongly related to reproductive problems in cattle. The neosporosis control is not well established and several fronts are under development, predominantly based on immune protection, immunomodulation and chemotherapy. The use of anti-malarial drugs as therapeutic sources has, in theory, considerable potential for any apicomplexan. Drugs such as methylene blue (MB) and pyrimethamine (Pyr) represent therapeutic options for malaria; thus, their use for neosporosis should be assessed. In this work, we tested the effects of MB and Pyr on N. caninum proliferation and clearance, using LacZ-tagged tachyzoites. The drugs inhibited at nanomolar dosages and its combination demonstrated an antagonistic interaction in proliferation assays, according to the Chou and Talalay method for drug combination index. However, the drug combination significantly improved the parasite in vitro clearance. The repositioning of well-established drugs opens a short-term strategy to obtain low-cost therapeutics approaches against neosporosis.

Niche apportionment models have only been applied once to parasite communities. Only the random assortment model (RA), which indicates that species abundances are independent from each other and that interspecific competition is unimportant, provided a good fit to 3 out of 6 parasite communities investigated. The generality of this result needs to be validated, however. In this study we apply 5 niche apportionment models to the parasite communities of 14 fish species from the Great Barrier Reef. We determined which model fitted the data when using either numerical abundance or biomass as an estimate of parasite abundance, and whether the fit of niche apportionment models depends on how the parasite community is defined (e.g. ecto, endoparasites or all parasites considered together). The RA model provided a good fit for the whole community of parasites in 7 fish species when using biovolume (as a surrogate of biomass) as a measure of species abundance. The RA model also fitted observed data when ecto- and endoparasites were considered separately, using abundance or biovolume, but less frequently. Variation in fish sizes among species was not associated with the probability of a model fitting the data. Total numerical abundance and biovolume of parasites were not related across host species, suggesting that they capture different aspects of abundance. Biovolume is not only a better measurement to use with niche-orientated models, it should also be the preferred descriptor to analyse parasite community structure in other contexts. Most of the biological assumptions behind the RA model, i.e. randomness in apportioning niche space, lack of interspecific competition, independence of abundance among different species, and species with variable niches in changeable environments, are in accordance with some previous findings on parasite communities. Thus, parasite communities may generally be unsaturated with species, with empty niches, and interspecific interactions may generally be unimportant in determining parasite community structure.

Intraerythrocytic granules in the blood of Sudanese fowls first seen by Balfour (1908) were named Aegyptianella pullorum by Carpano (1928).

Electron microscopy of a Rhodesian strain of A. pullorum indicated the similarity between these organisms and the Rickettsia-PLT group of organisms, also their dissimilarity to Sporozoa which we have studied. The organism was coccoid or coccobacillary, measuring up to 0·6 μm in diameter, and in longitudinal sections 1·5–3 μm. Pleomorphic and dividing forms were observed. Each organism was surrounded by two unit membranes with short internal invaginations from the inner membrane only. A layer of ribosome-like particles varying in density and depth gave the impression of an outer cortex. Irregular extensions of this layer towards the centre of the organism followed the lines of fine DNA-like filaments forming a web lattice throughout the substance of the organism. There was no well-defined nucleus as seen in Protozoa. Single as well as groups of organisms were situated in vacuoles lined by a membrane similar to the erythrocytes' own outer membrane. No intranuclear organisms were seen.

The similarity of these organisms to symbionts studied by Roshdy and Bird in both adult and immature stages of A. persicus is illustrated. From this evidence, and its apparent likeness to the tick-borne fever agent, there is good reason for including A. pullorum among the Rickettsiaceae. The position suggested for it is between the Rickettsia and PLT group of organisms, closely allied to Wolbachia, the agent of tick-borne fever (Cytoecetes sp.) and the agent of bovine heart water disease (Cowdria ruminantium).

Parasites have established numerous strategies for evading host immunity. Such tactics promote persistence and chronicity – factors that favour completion of the life cycle and transmission to vectors or the environment. This volume explores many facets of the interaction between host and parasite that will be critical to the development of meaningful vaccines capable of interrupting what has been accomplished over centuries of co-evolution.

The poultry red mite (PRM) is an obligatory haematophagous pest that causes substantial economic losses in poultry worldwide. The PRM does not live on the host but in the bird's environment and must find its host remotely. Hence, manipulating chicken odours is of interest. Several crude plant-originating volatile organic compounds (VOCs) have already been shown as repellent to Dermanyssus gallinae. We aimed to test whether these VOCs can interfere with PRM host-seeking behaviour by their oral administration to the poultry. The objectives were to determine (1) if hen odours are modified by supplemented feed ingestion and (2) if such treatment makes hens less attractive to the PRM. Chemical characterization by gas chromatography–mass spectrometry of the hen odour was conducted before and after the hens ingested the supplemented feed. The chromatograms obtained show that hen odour was substantially modified after the hens consumed it. Among the molecules recurrently detected from the supplemented hens, 26% were nearly absent in the unsupplemented hens. Behavioural choice tests to compare the effect of the modified and unmodified-host odours on the PRM show that some of the plant-originating emitted VOCs and the modified whole-hen odours were repellent to the PRM.

Schistosomiasis is a significant cause of human morbidity and mortality. We performed a genome-wide transcriptional survey of liver biopsies obtained from Chinese patients with chronic schistosomiasis only, or chronic schistosomiasis with a current or past history of viral hepatitis B. Both disease groups were compared with patients with no prior history or indicators of any liver disease. Analysis showed in the main, downregulation in gene expression, particularly those involved in signal transduction via EIF2 signalling and mTOR signalling, as were genes associated with cellular remodelling. Focusing on immune associated pathways, genes were generally downregulated. However, a set of three genes associated with granulocytes, MMP7, CLDN7, CXCL6 were upregulated. Differential gene profiles unique to schistosomiasis included the gene Granulin which was decreased despite being generally considered a marker for liver disease, and IGBP2 which is associated with increased liver size, and was the most upregulated gene in schistosomiasis only patients, all of which presented with hepatomegaly. The unique features of gene expression, in conjunction with previous reports in the murine model of the cellular composition of granulomas, granuloma formation and recovery, provide an increased understanding of the molecular immunopathology and general physiological processes underlying hepatic schistosomiasis.

The CXCR3 chemokine CXCL10 or IFN-γ inducible protein 10 (IP-10) has been identified as an important biomarker of cerebral malaria (CM) mortality in children. Studies in mouse malaria infection models have shown that CXCL10 blockade alleviates brain intravascular inflammation and protects infected mice from CM. Despite the key role that CXCL10 plays in the development of CM, the leucocytic sources of CXCL10 in response to human malaria are not known. Here we investigated CXCL10 responses to Plasmodium falciparum in peripheral blood mononuclear cells (PBMCs). We found that PBMCs from malaria-unexposed donors produce CXCL10 in response to P. falciparum and that this response is IFN-γ-dependent. Moreover, CD14+ monocytes were identified as the main leucocytic sources of CXCL10 in peripheral blood, suggesting an important role for innate immune responses in the activation of this pathway involved in the development of symptomatic malaria.

Biochemical studies of benzimidazole derivatives are important to determine their mode of action and activity against parasites. The lack of antihelminthic alternatives to treat parasitic infections and albendazole resistance cases make the search for new antiparasitary drugs of utmost importance. The 6-chloro-5-(1-naphthyloxy)-2-(trifluoromethyl)-1H-benzimidazole (RCB20) is a benzimidazole derivative with promising effect. This study evaluated the effect of different concentrations of RCB20 in the alternative energetic pathway of in vitro Taenia crassiceps cysticerci. The parasites were in vitro exposed to 6·5 and 13 µ m of RCB20 and albendazole sulfoxide (ABZSO). The quantification of acetate, acetoacetate, β-hydroxybutyrate, fumarate and propionate was performed by high-performance liquid chromatography. The quantification of urea, creatinine and total proteins was performed by spectrophotometry. The increase in β-hydroxybutyrate reflects the enhancement of the fatty acid oxidation in the treated groups. Volatile fatty acids secretion, acetate and propionate, was increased in the treated groups. The secretion mechanisms of the treated parasites were impaired due to organic acids increased concentrations in the cysticerci. It is possible to conclude that the metabolic effect on alternative energetic pathways is slightly increased in the parasites treated with RCB20 than the ones treated with ABZSO.

Autophagy process in Toxoplasma gondii plays a vital role in regulating parasite survival or death. Thus, once having an understanding of certain effects of autophagy on the transformation of tachyzoite to bradyzoite this will allow us to elucidate the function of autophagy during parasite development. Herein, we used three TgAtg proteins involved in Atg8 conjugation system, TgAtg3, TgAtg7 and TgAtg8 to evaluate the autophagy level in tachyzoite and bradyzoite of Toxoplasma in vitro based on Pru TgAtg7-HA transgenic strains. We showed that both TgAtg3 and TgAtg8 were expressed at a significantly lower level in bradyzoites than in tachyzoites. Importantly, the number of parasites containing fluorescence-labelled TgAtg8 puncta was significantly reduced in bradyzoites than in tachyzoites, suggesting that autophagy is downregulated in Toxoplasma bradyzoite in vitro. Moreover, after treatment with drugs, bradyzoite-specific gene BAG1 levels decreased significantly in rapamycin-treated bradyzoites and increased significantly in 3-MA-treated bradyzoites in comparison with control bradyzoites, indicating that Toxoplasma autophagy is involved in the transformation of tachyzoite to bradyzoite in vitro. Together, it is suggested that autophagy may serve as a potential strategy to regulate the transformation.
