1. Filarial worms were found in the peritoneal cavity of a monkey (Macaca speciosa) from Bengal. They were identified as Dipetalonema digitatum (Chandler, 1929).

2. The microfilariae were unsheathed and showed a well-marked nocturnal periodicity in the blood.

3. No development of the microfilariae was detected in Aëdes aegypti, A. albopictus, Anopheles quadrimaculatus or An. stephensi, but only a few mosquitoes were observed.

4. Six female worms and one male were transplanted into the peritoneal cavity of a rhesus monkey (Macaca mulatta). Ten weeks later microfilariae were found in the blood of the monkey and they persisted for at least 16 weeks.

5. Filarial worms (Dipetalonema gracile) were discovered in the peritoneal cavity of a monkey (Cebus apella) from Brazil.

6. The microfilariae were unsheathed and were found in the blood. They showed no periodicity.

7. No development of the microfilariae was detected in Aëdes aegypti, A. albopictus, Anopheles maculipennis atroparvus or An. stephensi.

8. No pathological lesions due to either of these worms were detected in the monkeys.

Unlike most bird species, individual kingfisher species (Aves: Alcedinidae) are typically parasitized by only a single genus of louse (Alcedoffula, Alcedoecus, or Emersoniella). These louse genera are typically specific to a particular kingfisher subfamily. Specifically, Alcedoecus and Emersoniella parasitize Halcyoninae, whereas Alcedoffula parasitizes Alcedininae and Cerylinae. Although Emersoniella is geographically restricted to the Indo-Pacific region, Alcedoecus and Alcedoffula are geographically widespread. We used DNA sequences from two genes, the mitochondrial COI and nuclear EF-1α genes, to infer phylogenies for the two geographically widespread genera of kingfisher lice, Alcedoffula and Alcedoecus. These phylogenies included 47 kingfisher lice sampled from 11 of the 19 currently recognized genera of kingfishers. We compared louse phylogenies to host phylogenies to reconstruct their cophylogenetic history. Two distinct clades occur within Alcedoffula, one that infests Alcedininae and a second that infests Cerylinae. All species of Alcedoecus were found only on host species of the subfamily Halcyoninae. Cophylogenetic analysis indicated that Alcedoecus, as well as the clade of Alcedoffula occurring on Alcedininae, do not show evidence of cospeciation. In contrast, the clade of Alcedoffula occurring on Cerylinae showed strong evidence of cospeciation.

Hookworms are some of the most widespread of the soil-transmitted helminths (STH) with an estimated 438.9 million people infected. Until relatively recently Ancylostoma ceylanicum was regarded as a rare cause of hookworm infection in humans, with little public health relevance. However, recent advances in molecular diagnostics have revealed a much higher prevalence of this zoonotic hookworm than previously thought, particularly in Asia. This study examined the prevalence of STH and A. ceylanicum in the municipalities of Palapag and Laoang in the Philippines utilizing real-time polymerase chain reaction (PCR) on stool samples previously collected as part of a cross-sectional survey of schistosomiasis japonica. Prevalence of hookworm in humans was high with 52.8% (n = 228/432) individuals positive for any hookworm, 34.5% (n = 149/432) infected with Necator americanus, and 29.6% (n = 128/432) with Ancylostoma spp; of these, 34 were PCR-positive for A. ceylanicum. Considering dogs, 12 (n = 33) were PCR-positive for A. ceylanicum. This is the first study to utilize molecular diagnostics to identify A. ceylanicum in the Philippines with both humans and dogs infected. Control and elimination of this zoonotic hookworm will require a multifaceted approach including chemotherapy of humans, identification of animal reservoirs, improvements in health infrastructure, and health education to help prevent infection.

Trichomonas vaginalis (Tv) induces host cell damage through cysteine proteinases (CPs) modulated by iron. An immunoproteomic analysis showed that trichomoniasis patient sera recognize various CPs, also some of them are present in vaginal washes (VWs). Thus, the goal of this work was to determine whether TvCP2 is expressed during infection and to assess the effect of iron on TvCP2 expression, localization and contribution to in vitro cellular damage. Western-blotting (WB) assays using TvCP2r and vaginitis patient serum samples showed that 6/9 Tv (+) but none of the Tv (−) patient sera recognized TvCP2r. WB using an anti-TvCP2r antibody and VWs from the same patients showed that in all of the Tv (+) but none of the Tv (−) VWs, the anti-TvCP2r antibody detected a 27 kDa protein band that corresponded to the mature TvCP2, which was confirmed by mass spectrometry analysis. Iron decreased the amount of TvCP2 mRNA and the protein localized on the parasite surface and cytoplasmic vesicles concomitant with the cytotoxic effect of TvCP2 on HeLa cells. Parasites pretreated with the anti-TvCP2r antibody also showed reduced levels of cytotoxicity and apoptosis induction in HeLa cell monolayers. In conclusion, these results show that TvCP2 is expressed during trichomonal infection and plays an important role in the in vitro HeLa cell cytotoxic damage under iron-restricted conditions.

1. During the period November 1936 to February 1937, eighty rabbits were examined, whilst from August 1937 to July 1938 another 446 were examined for nematodes. The following nematodes were found: Graphidium strigosum, Passalurus ambiguus, Trichostrongylus retortaeformis.

2. The incidence of the above three nematodes is given, and their seasonal incidence is discussed, with special reference to its ecological implication.

3. Evidence of host and age resistance is also demonstrated in infections with Graphidium strigosum, Passalarus ambiguus and Trichostrongylus retortaeformis.

4. The proportion of the sexes in Graphidium strigosum was found to be constant during both periods. The percentage of males in 1936–7 was 45·6 and 45·96 in 1937–8. In Passalurus ambiguus, however, the proportion of the sexes varied to a large extent.

The egg of Ixodes ricinus L. is described, and the increase in size during development is noted. It is suggested that the increase is due to the absorption of water. The egg wall consists of exo- and endochorion with a thin vitelline membrane, which becomes thicker as development proceeds and assumes a laminated appearance. The secretion layer of Gené's organ is described, and appears to function as a cement for holding the eggs together in the cluster. Chemical tests show it to be of a waxy nature.

It is deduced from experimental evidence that the egg shell is not absolutely rigid, and the weight of evidence favours the view that the shape of the egg shell is maintained by the internal pressure of the egg contents, and the elasticity of the shell.

The development processes as seen in the living egg are described. Hatching from the egg shell is brought about by a muscular force acting on a limited area at the posterior pole of the egg.

Current chemotherapeutic agents for leishmaniasis have several disadvantages interfering with the effective treatment and therefore more and better antileishmanial drugs are needed. Discovery of candidates for leishmaniasis treatment requires not only accurate and precise methodologies but also well-known biological system to measure infectivity of parasites and antileishmanial activity of the new compounds. Significant variation in the in vitro and in vivo infectivity and sensitivity to established and experimental drugs in Leishmania strains are reported. This work reports the in vitro biological behavior and antileishmanial drugs sensitivity of different green fluorescent protein transfectant Leishmanias strains. The in vitro growth kinetic and infectivity to U937 cells vary slightly in the Leishmania transfectant strains in comparison with their correspondant wild-type. However, the insertion of the pIR3(-)-eGFP may affect the sensitivity of the parasites to meglumine antimoniate (MA) and miltefosine but not to amphotericin B (AMB) and pentamidine isethionate. In consequence, AMB or pentamidine isethionate but not MA or miltefosine should be used as antileishmanial control drugs during in vitro assays of antileishmanial activity. Furthermore, is recommended to test compounds against more than one Leishmania strain in order to verify that the antileihmanial activity of these compound is similar among species.

Myxozoans are widespread and common endoparasites of fish with complex life cycles, infecting vertebrate and invertebrate hosts. There are two classes: Myxosporea and Malacosporea. To date about 2500 myxosporean species have been described. By comparison, there are only five described malacosporean species. Malacosporean development in the invertebrate hosts (freshwater bryozoans) has been relatively well studied but is poorly known in fish hosts. Our aim was to investigate the presence and development of malacosporeans infecting a diversity of fish from Brazil, Europe and the USA. We examined kidney from 256 fish belonging variously to the Salmonidae, Cyprinidae, Nemacheilidae, Esocidae, Percidae, Polyodontidae, Serrasalmidae, Cichlidae and Pimelodidae. Malacosporean infections were detected and identified by polymerase chain reaction and small subunit ribosomal DNA sequencing, and the presence of sporogonic stages was evaluated by ultrastructural examination. We found five malacosporean infections in populations of seven European fish species (brown trout, rainbow trout, white fish, dace, roach, gudgeon and stone loach). Ultrastructural analyses revealed sporogonic stages in kidney tubules of three fish species (brown trout, roach and stone loach), providing evidence that fish belonging to at least three families are true hosts. These results expand the range of fish hosts exploited by malacosporeans to complete their life cycle.

Immunohistochemical analysis of skin and draining lymph nodes of sheep repeatedly infested with the ixodid tick Hyalomma anatolicum anatolicum were studied for different antigen-presenting cells and lymphocyte subpopulations. Infiltration of neutrophils, macrophages and lymphocytes adjacent to the tick bite site were observed. Skin biopsies showed significant increases in dermal infiltration of CD8+ and γδ+ T cells at 72 h and 8 days after both primary and secondary infestation. Infiltrations of MHC-II DR/DQ decreased at 72 h after tick infestation, whereas significant increases were recorded for 8-day skin biopsies. CD1+ cellular infiltrations were observed during secondary infestations at the dermis. Decreased ratios of CD4[ratio ]CD8 T cells and MHC-II[ratio ]CD1 antigen-presenting cells were observed in both infestations compared to healthy skin biopsies. Ratios of αβ[ratio ]γδ T cells increased gradually during infestation compared to uninfested skin. The regional lymph nodes from tick-infested sheep showed an increased CD8+, γδ+ T and CD1+ cellular infiltration compared to control lymph nodes. CD4+ T cells were decreased. There were no significant changes in CD45R+ cellular infiltration either at skin lesions or regional lymph nodes.

Human serum high-density lipoprotein (HDL) is necessary and sufficient for the short-term maintenance of Plasmodium falciparum in in vitro culture. However, at high concentrations it is toxic to the parasite. A heat-labile component is apparently responsible for the stage-specific toxicity to parasites within infected erythrocytes 12–42 h after invasion, i.e. during trophozoite maturation. The effects of HDL on parasite metabolism (as determined by nucleic acid synthesis) are evident at about 30 h after invasion. Parasites treated with HDL show gross abnormalities by light and electron microscopy.

Phosphoenolpyruvate carboxykinase (PEPCK) involved in gluconeogenesis in higher vertebrates opposedly plays a significant role in glucose oxidation of the cestode parasite, Raillietina echinobothrida. Considering the importance of the enzyme in the parasite and lack of its structural details, there exists an urgent need for understanding the molecular details and development of possible modulators. Hence, in this study, PEPCK gene was obtained using rapid amplification of cDNA ends, and various biocomputational analyses were performed. Homology model of the enzyme was generated, and docking simulations were executed with its substrate, co-factor, and modulators. Computer hits were generated after structure- and ligand-based screening using Discovery Studio 4.1 software; the predicted interactions were compared with those of the existing structural information of PEPCK. In order to evaluate the docking simulation results of the modulators, PEPCK gene was cloned and the overexpressed protein was purified for kinetic studies. Enzyme kinetics and in vitro studies revealed that out of the modulators tested, tetrahydropalmatine (THP) inhibited the enzyme with lowest inhibition constant value of 93 nm. Taking the results together, we conclude that THP could be a potential inhibitor for PEPCK in the parasite.

La plupart des formes décrites ici m'ont été gracieusement données à étudier par le Professeur de l'Académic Impériale Militaire de Médecine N. A. Cholodkowski, et je saisis l'occasion de lui exprimer ma plus sincère gratitude.

Threshold density levels were investigated experimentally in rainbow trout, Oncorhynchus mykiss, for the acanthocephalan Pomphorhynchus laevis. Fifty small fish 120–150 mm long were infected with 50 cystacanths each every 4 weeks for 16 weeks, and samples of up to 10 fish examined at 1, 5, 9, 13 and 17 weeks after the original infection. The mean intestinal length was found to have increased significantly over this period, from a mean of 125 to 173 mm and the mean number of parasites recovered increased after each interval of 4 weeks, and significantly from 19·30 to 86·40 per fish over the 17 weeks. The distribution of the parasites in the alimentary tract of O. mykiss was unaltered, indicating that the favourable region for establishment had remained unchanged. The failure to detect any manifestations of competition is discussed, and the relevance of conventional approaches to experimental investigations of both intra- and interspecific competition in fish was queried, and contrasted to the situation in birds and mammals.

A study of the anterior adhesive apparatus (head organs) of Bravohollisia gussevi Lim, 1995 was carried out using light and electron microscopy. The anterior adhesive apparatus or head organs in B. gussevi comprise 6 circular openings or apertures in the antero-lateral region, associated pits lined with specialized microvillous tegument that differ from the general body tegument, a bundle of ducts, and uninucleate gland cells located lateral to the pharynx. The uninucleate glands of the anterior adhesive apparatus (head organs) comprise 2 types of cells, one kind of cell producing rod-like bodies (S1) and the other oval bodies (S2). The S1 bodies are filled with numerous, less electron-dense vesicles in an electron-dense matrix, while S2 bodies have no vesicles but contain a more homogeneous electron-dense matrix. Interlinking band-like structures were observed between S1 bodies. Similar band-like structures were found between S2 bodies. The formation of S1 bodies was followed by transmission electron microscopy. However, the formation of S2 bodies was unclear and could not be resolved. Uniciliated structures were also observed around the openings of the anterior adhesive apparatus. Each uniciliated structure is usually associated with an opening of a gland cell producing granular, electron-dense, secretory bodies, which differ from the secretions produced by the lateral gland cells of the anterior adhesive apparatus.

The two filarial Nematodes known to parasitize dromedaries in Egypt are Dipetalonema evansi and Onchocerca fasciata, of which the first is the more common. Representatives of these genera also infect man and other animals and are known to be very common in endemic areas. Doubt has always arisen in the researches of those few who worked on this subject, as to the connexion of the adult Dipetalonema evansi with the microfilariae found in the blood of Camelus dromedarius and C. bactrianus.

It has been shown that the bovine abomasal parasite, Ostertagia ostertagi, drastically modulates its microenvironment, causing epithelial cell damage, accumulation of inflammatory cells and pH changes in the stomach. The mechanisms used by the parasite to change the abomasal environment are largely unknown, but an important role has been attributed to excretory-secretory (ES) products from the parasite. In this study we have identified proteins representing a novel ES protein family, characterized by the SCP/Tpx-1/Ag5/PR-1/Sc7 protein motif. These proteins were named Oo-AL1 and Oo-AL2 (O. ostertagi ASP-like protein). Both proteins contain a signal peptide and 1 predicted N-glycosylation site. The transcript for Oo-AL1 was present from the L4 stage onwards in both male and female adult worms, whereas the Oo-AL2 transcript was hardly detectable. Western blots of somatic extracts and ES products from different developmental stages of O. ostertagi, probed with anti-Oo-AL1 antibodies, revealed Oo-AL proteins in the ES products of adult worms. An analysis of the nematode genome and EST databases indicated that these novel ES proteins are unique to O. ostertagi and its relative, Teladorsagia circumcincta, suggesting a key function in these abomasal parasites.

The detailed structure of the spiracles in the Ixodoidea has hitherto received little or no attention at the hands of zoologists; at the same time, these organs are sufficiently extraordinary to make it a matter of surprise that, so far as our knowledge of the literature goes, not one of the numerous contributors to the subject of tick anatomy has found it worth while to undertake a complete description or to publish figures to illustrate it. Batelli (1891) gives a short account of the structure of the spiracle of a tick, presumably Ixodes ricinus, with a single figure, but with this exception we have been unable to find any further information on the subject.

Intrapredatorus barri, a microsporidian parasite of the predator mosquito Culex fuscanus Wiedemann has been shown to be heterosporous. In many features this species has been reported to be similar to Amblyospora trinus, another microsporidian parasite of a different predator mosquito which was thus proposed for reassignation to the genus Intrapredatorus. In this report small subunit rDNA has been sequenced from I. barri and phylogenetic analysis of 42 microsporidia sequences has been performed.Nucleotide sequence data reported in this paper are available in the Gen Bank™ database under the accession number AY013359. The molecular data show that I. barri can not be a member of the genus Amblyospora. Furthermore, the phylogenetic analysis shows, with high confidence, that the Amblyospora spp. sequences in the present study are not monophyletic.
