An infection of pearl oysters, Pinctada maxima, attributed to a Haplosporidium sp. by Hine and Thorne (1998) has been detected on 3 occasions and is considered to represent a serious concern to the pearling industry in Australia. The spore ornamentation of the parasite was determined by scanning electron microscopy and transmission electron microscopy. Spores of the parasite were pleomorphic, or elongated 3·5–4 μm×2·5–3·0 μm in size. Two filaments were wound around the spore and originated from 2 ‘knob-like’ posterior thickenings. Both filaments passed up one side of the spore together until just below the operculum whereupon each split and passed obliquely under the lip of the opercula lid. Each filament wrapped around the spore 4 times. The posterior thickenings seem to appear late in the development of the spore and were composed of spore wall material. A second set of branching tubular filaments composed of a different material was observed on the spore body although not on mature spores possessing a ‘knob-like’ posterior thickening. The ornamentation on the spores of the pearl oyster parasite was unique amongst described haplosporidian species where spore ornamentation is known. The parasite is named in this manuscript as Haplosporidium hinei n. sp.

The faecal egg counts of seven groups of calves exposed to infection with O. ostertagi on pastures were found to follow a similar course to those of calves experimentally infected on one occasion or of calves infected daily. They declined from an early peak according to an apparently logarithmic curve. It was concluded that faecal egg counts were largely independent of worm numbers and tended to follow a stereotyped pattern. Acute clinical ostertagiasis was as liable to lead to high egg counts as was poor thriving due to the other causes. The relevance of these findings to the epidemiology of ostertagiasis and to the use of faecal egg counts in the diagnosis of parasitic gastro-enteritis are discussed.

Metacercarial cysts of Bucephalus haimeanus occur throughout the liver of Pomatoschistus microps, greater concentrations being found around the gall bladder, blood vessels and periphery of the liver. The cyst wall is composed of three layers, an inner granular, middle vacuolated and outer nucleated layer. The inner layer consists of three zones, an inner zone of irregularly compacted, granular material; a middle densely compacted zone and an outer moderately compacted zone. Vacuoles of the middle layer vary in appearance and appear to be discharging their contents into the cyst cavity. The cells of the outer layer are flattened and elongated with elongate nuclei but no other distinct organelles. Occasionally at the outer edge of the cyst are electron-dense cells which are in the process of degeneration. Only small amounts of collagen and very few leucocytes are associated with the cysts.

The cyst wall is carbohydrate in nature containing some free aldehyde groups, galactogen and neutral mucosubstances. The inner layer appears to consist of mainly acidic mucopolysubstances and weakly acidic sulphomucins. Large amounts of protein, carbohydrate-protein complexes and DNA are also present within the cyst wall. Alkaline and acid phosphatases, ATPase, β-glucuronidase and N-acetyl-β-glucosaminidase were detected. The origin and function of the cyst wall is discussed.

The mode of appearance and assembly of cyst wall filaments on the surface of Giardia duodenalis trophozoites committed to encyst was analysed by scanning and transmission electron microscopy (SEM and TEM) and by fluorescence microscopy (FM). SEM showed a progressive appearance of fibril patches, predominantly on the anterior area of ventral and dorsal surfaces, which then spread and coalesced. By TEM, ruthenium red (RR) displayed staining in encysting cells as rodlike spots of variable diameter (3–25 nm), possibly microfibril tips with polyanionic moieties, that displayed tangential associations and random orientations over the cell membrane. In FM assays, the 1,10-phenanthroline derivative of ruthenium red (RR/oPHE) was a specific ligand for these assembling fibrils and this staining was significantly blocked by N-acetylgalactosamine (GalNac) and galactosamine (GalN). Interestingly, RR staining was lost when the cyst wall was completely assembled and thickened as observed by TEM and FM. Kinetic FM assays, in which a mAb specific for a 26 kDa Giardia cyst wall polypeptide was used concomitantly with RR/oPHE staining, showed a differential pattern for the appearance and reactivity of polypeptide and assembling GalN/GalNac-rich moieties of Giardia cyst wall.

Through the kindness of my friend Mr Clifford Dobell, F.R.S., I have come into the possession of a quantity of preserved stomach-content from the Mouse Deer of Ceylon (Tragulus meminna Milne-Edwards). As there are no descriptions, so far as I can find, of the Ciliates from Asiatic Ruminants, it seems worth while to publish a brief note on the organisms found in this material.

Third-stage (dauer) juveniles of the entomopathogenic nematode Steinernema feltiae from 3 laboratory populations were tested for infectivity to larvae of the sciarid fly Lycoriellia solani. Bioassay experiments were done to assess the effects of nematode application density, exposure time and host density on the proportion of individuals that established infection. When dauer stages were presented to a host singly, establishment rates of 28, 26 and 18% were obtained for the 3 populations, but the presence of conspecifics consistently increased the rate of establishment to 42, 33 and 21% respectively, at 20 nematodes per host. A modified binomial model was developed to show that nematode establishment was not related to application density in a linear fashion, as initial infection facilitated secondary colonization. The data suggest that 3 subpopulations of nematodes may be distinguished by their infection behaviour: a first group of individuals with the behavioural propensity to initiate infection in unparasitized insects, a second that only invaded infected hosts, and a third group of non-invaders that failed to establish in L. solani.

Human strongyloidiasis is caused by helminth Strongyloides stercoralis. It has a worldwide distribution, often neglected and cause of severe morbidity. The parasitological diagnosis is hindered by the low and irregular amount of larvae in feces. The goal of the present study was to detect IgG and IgG immune complex using conventional serum samples and saliva as alternative samples. We collected samples from 60 individuals, namely: group I composed of 30 healthy individuals; and group II composed of 30 individuals eliminating S. stercoralis larvae in feces. We calculated the area under the curve, general index of diagnostic accuracy, Kappa index and determined the correlations between different diagnostic tests. The detection of IgG levels was performed by an immunoenzymatic assay with alkaline extract of S. venezuelensis larvae as antigen. Positivity of anti-S. stercoralis IgG in serum samples from group I was 3·3%, and from group II 93·3%. The detection of immune complex indicated that group I exhibited 3·3% and group II 56·7%. In the saliva samples, IgG detection was 26·7% for group I and 43·3% for group II. Immune complex was detected in 20% of group I, and 30% of group II. IgG immune complex in conventional serum samples and saliva as alternative samples can be considered biomarkers for the diagnosis of active strongyloidiasis.

The highly pathogenic avian protozoan Leucocytozoon caulleryi infects host chicken cells, and interference by the host genome results in difficulty in obtaining protozoal DNA for genetic analysis. We used flow cytometry analysis to separate expelled L. caulleryi gametocytes from infected chicken blood and to analyse cell populations and sorting by FACS efficiency. Infected blood cells stained with SYTO-24 showed a specific area on 2-dimensional scattergrams compared to uninfected blood. The specific area was sorted, and approximately 85% of the sorted cells were identified as L. caulleryi gametocytes by microscopic observation. DNA was also extracted from the sorted fraction, and a clear increase in polymerase chain reaction (PCR) amplification of protozoal DNA was observed compared to infected blood without sorting. Host-derived DNA was also detected by PCR; however, its amplification was decreased compared to that in unsorted infected blood. This is the first report of the separation of L. caulleryi gametocytes from infected host blood using flow cytometry. This method may be applied to further genetic analyses such as studies of the dynamics of stage-specific L. caulleryi gene expression.

Immunization of chimpanzees with radiation-attenuated infective 3rd-stage larvae (L3) of Onchocerca volvulus did not induce strong protective immunity against a subsequent challenge infection; only 1 out of 4 immunized animals remained non-patent (i.e. microfilariae-negative) after challenge, and may have been protected. However, during immunization and before challenge, a broad range of adult O. volvulus-derived antigens (OvAg) and also uterus-derived OvAg were recognized by circulating antibodies; moreover, the repertory of antigens recognized increased further in subsequently patent animals after challenge, particularly in the range of Mr 12–42 kDa. In the immunized and non-patent chimpanzee, by contrast, serological recognition of uterus-derived OvAg with Mr 14 kDa and 105 kDa disappeared by 19 months post-challenge (p.c.). During immunization, Acanthocheilonema viteae L3 antigens of Mr 11–12 kDa were strongly recognized only by the non-patent animal, suggesting that recognition of these antigens may have supported resistance to the subsequent challenge infection. In immunized chimpanzees, a substantial increase in the cellular reactivity to OvAg was induced; this, however, declined by 19 months p.c. to levels similar to those seen prior to immunization. At that time, 3 out of 4 immunized animals were patently infected. The effect of exogenous cytokines on in vitro-reactivity of PBMC to OvAg was examined. Addition of exogenous IL-2 alone, IFN-γ alone, and IFN-γ in combination with IL-2, did not augment net cellular responses to OvAg by PBMC from infected and control chimpanzees. In the presence of IL-4 alone, IL-6 alone, IL-2 with IL-4, IL-2 with IL-4 and IFN-γ, or IL-2 with IL-4 and IL-6, the net cellular reactivity to OvAg increased significantly in patent chimpanzees and reached levels similar to non-patent animals. Thus, non-patent chimpanzees maintain high cellular reactivity to OvAg and in vitro cellular unresponsiveness to OvAg on the part of patent chimpanzees is reversible after addition of several cytokines which act individually or synergistically.

VmCT1 is an antimicrobial peptide (AMP) isolated from the venom of the scorpion Vaejovis mexicanus with antimicrobial, anticancer and antimalarial activities, which the rational design with Arg-substitution has yielded AMPs with higher antimicrobial activity than VmCT1. Chagas is a neglected tropical disease, becoming the development of new antichagasic agents is urgent. Thus, we aimed to evaluate the antichagasic effect of VmCT1 and three Arg-substituted analogues, as well their action mechanism. Peptides were tested against the epimastigote, trypomastigote, amastigote forms of Trypanossoma cruzi Y strain and against LLC-MK2 mammalian cells. The mechanism of action of these peptides was evaluated by means of flow cytometry and scanning electron microscopy. VmCT1 presented activity against all three forms of T. cruzi, with EC50 against trypomastigote forms of 1.37 μmol L−1 and selectivity index (SI) of 58. [Arg]3-VmCT1, [Arg]7-VmCT1 and [Arg]11-VmCT1 also showed trypanocidal effect, but [Arg]11-VmCT1 had the best effect, being able to decrease the EC50 against trypomastigote forms to 0.8 μmol L−1 and increase SI to 175. Necrosis was cell death pathway of VmCT1, as well [Arg]7-VmCT1 and [Arg]11-VmCT1, such as observed by membrane damage in flow cytometry analyses and scanning-electron-microscopy. In conclusion, [Arg]11-VmCT1 revealed promising as a candidate for new antichagasic therapeutics.

This study was designed to test the accessory function of macrophages after activation with products of Taenia multiceps coenuri. Activation was carried out by intraperitoneal injection of mice with coenurus fluid or protoscolex culture supernatant, and function was assessed by adding these macrophages in progressively increasing numbers to macrophage-depleted lymphocyte cultures transforming under the influence of plant mitogens or coenurus-fluid mitogen. In contrast to normal macrophages, which have a progressively enhancing action on the above reactions, parasite-activated macro-phages at similar concentrations were progressively inhibitory. However, low concentrations of the activated macrophages enhanced mitosis as well as, or better than, normal. Lymph node cells from injected mice showed abnormal response to macrophage-derived signals. In particular there was subnormal reaction to macrophages in the presence of coenurus mitogen. These results suggest that T. multiceps coenuri may survive in the host because of their ability to reduce effective interaction between lymphocytes and accessory cells.

Real-time PCR was used to study the duration and level of parasitaemia in mice immunized with immune-stimulating complexes (iscoms) containing recombinant NcSRS2, one of the immunodominant surface antigens of Neospora caninum. After challenge infection, blood was collected daily for 9 days. During this period the amounts of parasite DNA detected in immunized mice were significantly lower (P<0·001), and the duration of parasitaemia appeared to be shorter, than in non-immunized controls. Furthermore, the degree of parasitaemia seemed to correlate well with the amount of N. caninum DNA in the brain 3 weeks post-inoculation and with disease severity measured as changes in body weight. These results indicate that the protective immunity induced by the NcSRS2-iscoms was sufficient to reduce the level of parasitaemia, which probably reduced the number of parasites reaching the brain, and could be the reason for the reduction in brain parasite load and clinical symptoms. Furthermore, real-time PCR was found to be a sensitive means for rapid assessment of N. caninum in blood.

The determination of Hyalomma sp. is discussed. In the material examined from Palestine the genital aperture of the female, the cuticle in the proximity of the stigmata and the number of ‘tunnels’ in the anterior border of the scutum are characters which facilitate diagnosis.

The junction between the gregarine trophozoite Nematocystis magna Schmidt and an epithelial cell of its earthworm host Lumbricus terrestris Muller consists of an extensive folding and meshing of the single trilaminar membrane bounding the host cell and the complex folded multilaminar limiting membrane of the trophozoite. They are separated by a structureless intercellular region of some 50 nm in width. Specialized communicating regions or permeable elements were not observed. Transport of a radioactive label across non-junctional and junctional membranes was demonstrated by using tritiated glucose. The attachment organelle of the trophozoite is shown to be capable of allowing metabolites to pass into the cell directly from the host tissue.

Some of the reindeer (Rangifer tarandus) introduced into Scotland from Northern Sweden have become infested with the deer warble fly Hypoderma diana and the deer ked Lipoptena cervi.

Evidence is presented which suggests that the larvae of H. diana proceed to their final site on the back of the host by way of the neural canal, and in this way probably caused the deaths of two reindeer in the winter of 1953–4.

A detailed analysis is presented of the frequency with which various bands of spines occurred on second-stage larvae of H. diana. An analysis of the number of chitinous rings in the two posterior spiracles on second-stage H. diana larvae showed that in any individual larva the two numbers were not independently determined.

On reindeer L. cervi produced puparia in December.

Avian malaria parasites can negatively affect many aspects of the life of the passerines. Though these parasites may strongly affect the health and thus migration patterns of the birds also during autumn, previous studies on avian malaria focused mainly on the spring migration and the breeding periods of the birds. We investigated whether the prevalence of blood parasites varies in relation to biometrical traits, body condition and arrival time in the European Robin (Erithacus rubecula) during autumn migration. We found no sex or age related differences in avian malaria prevalence and no relationship between infection status and body size or actual condition of the birds was found either. However, the timing of autumn migration differed marginally between infected and non-infected juveniles, so that parasitized individuals arrived later at the Hungarian stopover site. This is either because avian malaria infections adversely affect the migration timing or migration speed of the birds, or because later arriving individuals come from more distant populations with possibly higher blood parasite prevalence. The possible delay that parasites cause in the arrival time of the birds during autumn migration could affect the whole migratory strategy and the breeding success of the birds in the next season.

Aspects of vitellogenesis, known to be controlled by juvenile hormone, are adversely affected by Hymenolepis diminuta infection of Tenebrio molitor, in spite of circulating titres of the hormone remaining unchanged. It has therefore been proposed that juvenile hormone binding is disrupted at the tissue site level. Juvenile hormone III binding sites were located in the nuclear, microsomal and post-microsomal supernatant fractions of the follicle cells of Tenebrio molitor. When JH-III binding was quantified for both control and Hymenolepis diminuta-infected beetles, binding in the nucleus and cytosol were found to be largely unaffected. However, microsomal binding was severely disrupted; on days 3 and 6 post-infection, binding was greatly diminished, on day 9 post-infection, binding was slightly reduced and, by day 15, binding was ‘restored’ to that of control insects. Using follicle cell microsomes at day 3 post-infection, previous Scatchard analysis revealed the presence of at least two JH-III binding sites. The first is of higher affinity, Kd = 5·3 x 10·5 x 10−11 mol/mg protein and the second of lower affinity Kd = 7·7 x 10−7 M, Bmax = 9·75 x 10−11 mol/mg protein. A comparison with microsomal binding parameters of follicle cells from non-infected Tenebrio indicated that although the Bmax values were unchanged, the Kd value of the higher affinity site was increased by approximately 5-fold. These data are indicative of a parasite-induced competitive binding inhibitor.

The fluid layer within the cuticle of the nematode Nippostrongylus brasiliensis contains haemoglobin. The absorbance spectra of the haemoglobins extracted from whole nematodes were measured spectrophoto-metrically and the spectra of cuticular haemoglobin in individual living nematodes was obtained by microdensitometry. Adult female nematodes were confined under cover-slips in oxygenated saline and measurements of optical density of a small area of the cuticular haemoglobin were taken at 10 sec intervals. The optical density decreased over a period of 4 min at 543 nm and increased at 553 nm indicating deoxygenation of the cuticular haemoglobin; irrigation with fresh saline restored the haemoglobin to its oxygenated state. This demonstrates that cuticular haemoglobin loads and unloads oxygen in the living animal. Isoelectric focusing showed that mature nematodes have haemoglobins which are isoelectric around pH 7·0 and at pH 9·8. Isolated cuticular fluid contained the haemoglobin which was isoelectric at pH 9·8, but no other haemoglobin, and immature adults, which do not have cuticular haemoglobin, contained only those haemoglobins which were isoelectric at pH 7·0. Sodium dodecyl sulphate polyacrylamide gel electrophoresis and column chromatography showed that the molecular weight of the cuticular haemoglobin sub-unit is 16·9–17·5 × 103 Daltons and that the haemoglobin exists primarily as a tetrameric molecule with a molecular weight of 65–75 × 103 Daltons. The presence of cuticular haemoglobin may allow adult Nippostrongylus to exploit oxygen-deficient areas within the environment of the rat's intestine where it would otherwise be unable to survive for prolonged periods.

Parasites sometimes expand their host range and cause new disease aetiologies. Genetic changes can then occur due to host-specific adaptive alterations, particularly when parasites cross between evolutionarily distant hosts. Characterizing genetic variation in Cryptosporidium from humans and other animals may have important implications for understanding disease dynamics and transmission. We analyse sequences from four loci (gp60, HSP-70, COWP and actin) representing multiple Cryptosporidium species reported in humans. We predicted low genetic diversity in species that present unusual human infections due to founder events and bottlenecks. High genetic diversity was observed in isolates from humans of Cryptosporidium meleagridis, Cryptosporidium cuniculus, Cryptosporidium hominis and Cryptosporidium parvum. A deviation of expected values of neutrality using Tajima's D was observed in C. cuniculus and C. meleagridis. The high genetic diversity in C. meleagridis and C. cuniculus did not match our expectations but deviations from neutrality indicate a recent decrease in genetic variability through a population bottleneck after an expansion event. Cryptosporidium hominis was also found with a significant Tajima's D positive value likely caused by recent population expansion of unusual genotypes in humans. These insights indicate that changes in genetic diversity can help us to understand host-parasite adaptation and evolution.

We have previously reported that the mechanisms of neuromyopathic damage induced by Trypanosoma cruzi are mediated by T cells and are parasite-strain dependent. In the present study our aim was to determine whether the humoral response against muscle and nervous system is also parasite-strain dependent. Of the sera from mice infected with RA and CA-I T. cruzi strains, 93% reacted against antigens of the nervous system (sciatic nerve, spinal cord and brain). No differences in the ability to recognize heart antigens were found between RA (48%) and CA-I (63%) antisera. Reactivity against skeletal muscle was only relevant in anti-CA-I sera at 270 days post-infection. Each of the antisera assayed in Western blots developed a particular antigenic pattern, but 3 antigens in the nervous system of molecular weight 48, 60 and 70 kDa were detected by 42, 28 and 23% of the sera, respectively. On the other hand, deposits of IgG were observed at the interstitial matrix in sciatic nerve and as endomisial and sarcolemmal patterns in skeletal muscle by IFAT for both RA and CA-I antisera. Absorption of sera with parasite antigens did not abolish the autoreactivity. Our results suggest that major serum autoreactivity from T. cruzi-infected mice is not parasite-strain dependent and does not arise from molecular mimicry.
