The flagellate to be described in this paper was found in the faeces of a native of the Bahamas who was admitted to the Seamen's Hospital, Albert Dock, in October, 1909. In the course of the examination of the perfectly fresh material it was noticed that numerous flagellates were swimming about. They were taken to be Trichomonas but on closer examination they were seen to differ markedly from these. In the character of their movements there was little to distinguish them from Trichomonas except that the undulating membrane so characteristic of this genus appeared to be absent. The body is pear shaped and there are three long flagella directed forwards from the blunt end of the body. By the lashing of these three flagella the animal is drawn along with the blunt end in advance and at the same time revolves upon its longitudinal axis. A very characteristic feature is the presence of a very large cytostome extending from the base of the three flagella towards the posterior end of the body for about one half to two thirds of its length. The movements of the animals were at first so violent that great difficulty was experienced in making out the details of the anatomy, but as the faeces cooled their activities began to abate. It was then seen that the flagella was as long or longer than the body and that the large cytostome was arranged in a slightly spiral manner: that is to say it did not run directly parallel to the longitudinal axis of the body.

Echinococcus multilocularis causes alveolar echinococcosis which is a chronic, progressive zoonotic disease that mainly affects the liver. Hepatic alveolar echinococcosis is insidious and the patients are asymptomatic most of the time. Generally, it is incidentally found on imaging studies performed for other reasons. Specific symptoms may evolve if the vascular and biliary structures of the liver are affected. Hepatic alveolar echinococcosis shows a similar pattern to malignancies in terms of radiologic and clinical features. For this reason, oncological surgical principles should be applied during the resection of hepatic alveolar echinococcosis. The gold standard surgical treatment is resection with negative surgical margin. However, in patients whose radical resection is not possible other therapeutic options include palliative resection which has no benefit to the patient, and other curative major surgical options such as ex vivo liver resection, and autotransplantation and ultimately liver transplantation. The remnant liver volume has paramount importance if resection is going to be performed. For this reason, occasionally, remnant liver volume hypertrophy is induced by employing either two-stage hepatectomy or associating liver partition and portal vein ligation for staged hepatectomy.

Species of Marshallagia are abomasal parasites in free-ranging and domesticated ungulates in temperate climatic zones throughout the world. Pervasiveness of these nematodes is significant in various parts of the world. There has been limited research in the area of Marshallagi amarshalli pathogenesis. The aim of this study was to investigate the effects of M. marshalli on the acid secretory capacity of the abomasal mucosa and the morphological changes due to parasitic migration to different parts of abomasal tissue in sheep. Ten lambs, approximately around 6 months old, were allotted to two groups of five (A and B). The sheep from group A were infected orally with a dose of 5000 third-stage larvae (L3) of M. marshalli whereas the sheep of group B were not infected. The results indicated that the development of M. marshalli in the abomasal glands of ruminants causes pathophysiological changes, which include a reduced acidity of the abomasal contents, increased abomasal pH and increased serum pepsinogen concentrations. The reduced acid secretion is explained by a replacement of functional parietal cells by undifferentiated cells. Histology changes include mucosal cell hyperplasia, loss of parietal cells and inflammatory cell infiltration, which includes numerous granulocytes and lymphocytes.

Trypanosomatid mitochondrial DNA is structured as a giant network of thousands of interlocked DNA molecules enclosed within the kinetoplast. The structure and replication mechanism of kinetoplast DNA (kDNA) is unique, thereby making it an excellent chemotherapeutic target. Alteration in the structural organization of kDNA can give rise to dyskinetoplastic (Dk) strains. In Dk cells, the kDNA is dispersed in clumps throughout the mitochondrial matrix and not organized into a network. In this work, Trypanosoma cruzi epimastigotes were treated with acriflavine, a DNA intercalating drug, which promoted a decrease in cell proliferation and induced the appearance of Dk protozoa. In treated cells, the kinetoplast lost its normal disc-shaped structure because the fibrillar arrangement was reduced to a compact, amorphous mass within the mitochondrion. Moreover, basic proteins associated with kDNA were redistributed throughout the Dk protozoal kinetoplast. We sought to understand how the disruption of the kDNA leads to the emergence of the Dk phenotype with atomic force microscopy (AFM) analysis of isolated networks. Our results demonstrate that the detachment of minicircles from the kDNA disk promotes the disassembly of the network, thereby generating Dk cells. Our data strongly suggest that acriflavine inhibits T. cruzi multiplication by interfering with kDNA replication.

C-mannosylation was recently identified in the thrombospondin-related anonymous protein (TRAP) from Plasmodium falciparum salivary gland sporozoites. A candidate P. falciparum C-mannosyltransferase (PfDPY-19) was demonstrated to modify thrombospondin type 1 repeat (TSR) domains in vitro, exhibiting a different acceptor specificity than their mammalian counterparts. According to the described minimal acceptor of PfDPY19, several TSR domain-containing proteins of P. falciparum could be C-mannosylated in vivo. However, the relevance of this protein modification for the parasite viability remains unknown. In the present study, we used CRISPR/Cas9 technology to generate a PfDPY19 null mutant, demonstrating that this glycosyltransferase is not essential for the asexual blood development of the parasite. PfDPY19 gene disruption was not associated with a growth phenotype, not even under endoplasmic reticulum-stressing conditions that could impair protein folding. The data presented in this work strongly suggest that PfDPY19 is unlikely to play a critical role in the asexual blood stages of the parasite, at least under in vitro conditions.

Phosphatase activity of Leishmania spp. has been shown to deregulate the signalling pathways of the host cell. We here show that Leishmania mexicana promastigotes and amastigotes secrete proteins with phosphatase activity to the culture medium, which was higher in the Promastigote Secretion Medium (PSM) as compared with the Amastigote Secretion Medium (ASM) and was not due to cell lysis, since parasite viability was not affected by the secretion process. The biochemical characterization showed that the phosphatase activity present in PSM was higher in dephosphorylating the peptide END (pY) INASL as compared with the peptide RRA (pT)VA. In contrast, the phosphatase activity in ASM showed little dephosphorylating capacity for both peptides. Inhibition assays demonstrated that the phosphatase activity of both PSM and ASM was sensible only to protein tyrosine phosphatases inhibitors. An antibody against a protein phosphatase 2C (PP2C) of Leishmania major cross-reacted with a 44·9 kDa molecule in different cellular fractions of L. mexicana promastigotes and amastigotes, however, in PSM and ASM, the antibody recognized a protein about 70 kDa. By electron microscopy, the PP2C was localized in the flagellar pocket of amastigotes. PSM and ASM induced the production of tumor necrosis factor alpha, IL-1β, IL-12p70 and IL-10 in human macrophages.

Parasites reduce host fitness and consequently impose strong selection pressures on their hosts. It has been hypothesized that parasites are scarcer and their overall effect on hosts is weaker at higher latitudes. Although Antarctic birds have relatively low numbers of parasites, their effect on host fitness has rarely been investigated. The effect of helminth parasitism on growth rate was experimentally studied in chinstrap penguin (Pygoscelis antarctica) nestlings. In a total of 22 two-nestling broods, 1 nestling was treated with anthelminthics (for cestodes and nematodes) while its sibling was left as a control. Increased growth rate was predicted in de-wormed nestlings compared to their siblings. As expected, 15 days after treatment, the experimental nestlings had increased body mass more than their siblings. These results show a non-negligible negative effect of helminth parasites on nestling body condition that would presumably affect future survival and thus fitness, and it has been suggested there is a strong relationship between body mass and mortality in chinstrap penguins.

The malaria parasites, Plasmodium berghei yoelii and P. vinckei chabaudi, run comparable courses of infection in mice. When mice were treated with betamethasone infections with P. v. chabaudi were enhanced while those of P. b. yoelii were depressed. Antibody levels in betamethasone-treated mice infected with either P. b. yoelii or P. v. chabaudi were depressed to similar extents. The differences in parasitaemias in the two different infections are attributed to the suppression of reticulocyte production by betamethasone. P. b. yoelii preferentially invades reticulocytes and the patterns of parasitaemia follow the level of reticulocytes in the blood. P. v. chabaudi has no preference for reticulocytes and the pattern of parasitaemia is influenced by the depressed immunological response of the host.

The intestinal macroparasite communities of freshwater eels (Anguilla anguilla) captured in the south of England from Windsor (River Thames) during August 2001, and Leckford (River Test) during late June/July 2000, are reported for the first time. Parasite component communities were among the most species rich and diverse recorded from European eels. A total of 13 intestinal macroparasite species were encountered during the study, 8 from each eel host population with 3 being common to both. Acanthocephalans, nematodes and cestodes were recovered from each host population. Eels from Windsor additionally harboured Nicolla gallica (Digenea), which was also the most prevalent and abundant macroparasite species in these hosts. Each component community followed a log normal rank abundance distribution and demonstrated reduced species dominance and increased species equitability compared with previous studies. As such, the study component communities were suitable for testing the hypothesis of low infracommunity diversity in European eel hosts. Specifically, this hypothesis predicts that the intestinal macroparasite infracommunities of European eels are species-poor, displaying low density and diversity with high dominance, irrespective of component community diversity, and that this may be more pronounced in UK host populations. This hypothesis was not upheld; study findings demonstrate that higher infracommunity diversity in eel hosts is possible, and suggest that infracommunity diversity in individual eel hosts may be a simple, stochastic reflection of component community diversity.

The morphology of Paramphistomum sukumum sp.nov. from Bos indicus in the Lake Region of Tanganyika, East Africa, is described and illustrated.

A list is given showing the incidence of nine species of the family Paramphistomatidae and three species of the family Gastrothylacidae found in cattle in the Sukumaland area of the Lake Region of Tanganyika.

Factors constraining host specificity are poorly understood. Intraspecific variation in host preferences in generalist parasites may reveal which factors affect patterns of host use, and thus the evolution of specialization. Here, laboratory experiments examined genetic variation in host preferences and the effect of a refugium against infection on host use. Firstly, 6 cercarial clones of the trematode Maritrema novaezealandensis (ranging widely in heterozygosities) were exposed simultaneously to 2 alternative hosts, the amphipods Heterophoxus stephenseni and Paracalliope novizealandiae, to assess host preferences and fitness correlations with parasite heterozygosity. All clones showed a distinct preference for H. stephenseni, though the extent of this preference varied among clones. No clear association was found between heterozygosity and either parasite infection success or preference for a particular host. Secondly, cercariae were exposed to the same 2 amphipods in both the presence and absence of sand (refugium for H. stephenseni). Without sand, infection levels were significantly higher in H. stephenseni than in P. novizealandiae. With sand, H. stephenseni was able to hide, offsetting the parasite's intrinsic preferences for this host. These results demonstrate the existence of genetic variation in host preferences, as well as the effect of environmental variables on observed patterns of host use.

1. Echinobothrium brachysoma Pintner and E. affine Diesing have been found in the spiral valve of Raja clavata L. off the coast of Britain.

2. The spiral valve is lined by tubular crypts into which the scolices of the worms are thrust, the strobila, with one gravid proglottid, being free in the lumen.

3. The functional morphology of the scolex of E. brachysoma has been described. That of E. affine appears to be similar.

4. The scolex adheres, mainly, by the apical hooks which can be raised by inner and outer elevator muscles attached to their roots and closely associated with the rostellum.

5. The rostellum can be protruded by the contraction of a dorsal and a ventral protractor muscle. Movements of the rostellum elongate the scolex in a dorsoventral direction, forcing the apical hooks against the crypt wall and supporting them.

6. The peduncle hooks are not often embedded in the epithelium. Both series of hooks damage the epithelial lining of the crypt.

7. The remaining musculature, excretory and nervous systems are described.

8. The genitalia are essentially the same in both species. The number of testes varies from one proglottid to another in the same worm. In E. brachysoma the mid-ventral genital atrium is near the posterior end which affects the course of the vagina. The eggs in both species include two yolk cells which are similar to those of the Digenea and Pseudophyllidea.

Many parasite species are known to show high levels of genetic diversity, yet the consequences of this diversity for host–parasite interactions are not well understood. Variation in phenotypic traits such as growth rates and the ability to form transmission stages are raw material for natural and artificial selection to act upon with consequences for the evolution of the parasite species and disease control. In order to study genetic and phenotypic diversity amongst Theileria annulata parasites, a collection of 52 parasite clones was generated from cattle isolates and tick material recently collected in Tunisia. Genetic diversity was assessed using PCR-RFLP and monoclonal antibody markers, and genetically distinct clones selected for further study. Clones varied significantly in their growth rates in culture at 37 °C, their viability after a period of culture at 41 °C and their differentiation rates into transmission stages after culturing at 41 °C. The viability of a clone after culturing at 41 °C could not be predicted from its growth rate at 37 °C, but across clones, differentiation rates were positively correlated with growth rates at 37 °C. All 3 in vitro measures are likely to have relevance to parasite–host interactions in animals with clinical theileriosis, and should be acted on by within-host and between-host selection.

Blood schizontocidal activity of 10 selected cis-fused cyclopenteno-1,2,4-trioxanes (namely Fenozan compound nos 6, 7, 11, 27, 32, 39, 44, 45, 48 and 51) have been re-investigated to establish their curative doses against the multidrug-resistant Plasmodium yoelii nigeriensis strain, which is lethal in Swiss mice. Freshly prepared formulations of these compounds prepared either in neutral groundnut (peanut) oil or in dimethyl sulfoxide (DMSO)-Tween-water, were compared for their antimalarial activity. Only 2 compounds, namely Fenozan derivatives 11 and 45, formulated in neutral groundnut oil for oral administration, showed highest activity with 100% cure rate in MDR P. yoelii nigeriensis-infected mice, while the DMSO-Tween-water formulations were inactive. Fenozan-48 produced 72·2% cure, when administered orally in groundnut oil (formulation) while its DMSO-Tween formulation was inactive. In the case of Fenozan 7, the oil and DMSO-Tween formulations produced 92·3 and 76·0% cures respectively. Fenozan derivatives nos 6, 27, 32, 39, 44 and 51 were not protective either in groundnut oil or DMSO-Tween oral formulations. The present study has applied more rigorous criteria for selection of active compounds, and has identified the 3,3-spirocyclopentane derivative Fenozan 11, and the 3,3-spirohydropyran derivative Fenozan 45, as potential blood schizontocides which can completely eliminate multidrug-resistant malaria infection in mice. Both these compounds are candidates for pre-clinical development. The present study advocates the preferred use of an oil vehicle for oral evaluation of potential antimalarial trioxanes/fenozans instead of the DMSO formulation, which gives inferior curative efficacy.

From the foregoing brief descriptions it is clear that the larva of Trichomyia urbica stands apart from other psychodid larvae not only in its mode of life, as a wood-borer, but also in its morphology. The regularly cylindrical body, devoid of dorsal plaques and long setae and the densely chitinized head give it a very different appearance from that of other psychodid larvae.

The small and heavily chitinized antennae are very different from those found in larvae of Phlebotomus, in which they are 3-segmented; Psychodes, in which they are delicate and dome-like; Sycorax, in which they are long and 4-segmented (Feuerborn, 1933); and Maruina, in which they are formed of small rods (Feuerborn, 1933).

The massive toothed mandibles, devoid of sensory processes, differ greatly from those of other genera of psychodids except Phlebotomus. Maruina, Sycorax (see Feuerborn, 1933), and Psychodes all have mandibles with large leaf-like processes and many long plumose or pectinate setae and sensory hairs or processes.

The maxillae, in all the three genera examined above, are delicate plates with ill-defined areas representing the maxillary palps, and with many delicate teeth, long setae and sensory processes arising from the body of the maxilla.

The lower lip in Trichomyia is very simple and the mentum is hardly distinguishable. In this respect it resembles Psychodes where the mentum is a simple transverse plate, and Maruina (see Feuerborn, 1933) in which it is similar except that the anterior border of the plate is fringed with long setae. In Phlebotomus the mentum is a densely chitinized plate with strong teeth on its anterior margin.

The anterior spiracles appear to be much alike in all the genera in which they are present; but in the position of the posterior spiracles Trichomyia, in which they are placed far back on the dorso-lateral region of the last segment, differs from all other genera examined except Phlebotomus, in which they occupy a similar position.

Therefore, as regards the shape of the body, nature of the cuticle, absence of long setae and of dorsal plaques, and the structure of the antennae and labrum, the larva of Trichomyia differs from all other psychodid larvae. It shares with the larva of Phlebotomus the possession of strong, simple, toothed, mandibles devoid of long setae and sensory processes. It also resembles Phlebotomus in the poorly defined epipharynx and premandibles on the labrum, and in the position of the posterior spiracles near the middle of the last segment. The weakly developed mentum of Trichomyia larvae, however, differs greatly from that of Phlebotomus larvae in which the mentum is very strongly developed as a toothed plate.

A total of 35 pigs aged 15 weeks old, and 21 pigs aged 8 weeks old were divided into 7 groups. Groups 1 and 2 were uninfected and challenge control groups, respectively. Groups 3 and 4 were infected weekly with 6 increasing doses of Ascaris suum eggs, and group 4 was additionally treated with pyrantel. Groups 5, 6, and 7 were immunized weekly with the 14, 42, or 97 kDa fractions from adult worms, respectively. Animals of groups 2–7 were challenged with 10 000 A. suum eggs 7 days after the last infection/immunization. Serum was sampled weekly and specific IgG1, IgG2, and IgM responses were measured. Pigs of groups 5, 6, and 7 showed high IgG1 and IgG2 responses especially against adult worms antigens, while infected groups had high IgG1 and IgM responses, especially against larva. The IgG1 responses were negatively correlated to the numbers of larvae in the lungs, and positively associated with the liver white spot numbers. There was a positive correlation between IgG2 and the numbers of white spots and lung larvae, while IgM was negatively correlated with these parasitological measures. These findings are discussed and it is suggested that acquired resistance against A. suum larvae is correlated with the induction of IgG1 and IgM, and not with IgG2, and that future vaccination protocols may focus on inducing the Th2 activity.

As has been shown previously, immunologically intact mice with patent Schistosoma mansoni infections has a significantly lower mean platelet number than intact uninfected mice (P<0·0001). However, platelet numbers in T-cell deprived mice with patent infections were not significantly different from those in uninfected T-cell deprived mice. Also, platelet counts in both the infected and uninfected T-cell deprived groups were not significantly different from those in intact uninfected mice. The S. mansoni-induced thrombocytopaenia in mice is thus seemingly immune dependent. Immunologically intact mice with chronic 12-week-old S. mansoni infections has IgG antibodies that were reactive in an ELISA-type assay wit whole fixed platelets of both mouse and human origin. In Western immunoblots the IgG antibodies from chronically-infected mice reacted in particular against mouse and human platelet antigens of 90, 37 and 30 kDa. Antisera raised from 2 rabbits, immunized respectively with mouse and human platelet antigens, cross-reacted with antigens of the larval, adult worm and egg stages of S. mansoni. These results support the hypothesis that an anti-platelet antibody response may be the cause of the thrombocytopaenia observed in mice with patent schistosome infections.

Cystic echinococcosis (CE) is a zoonotic disease caused by a complex of species known as Echinococcus granulosus sensu lato. CE is endemic in Argentina, Chile, Peru, Uruguay and the South part of Brazil. In contrast, little is known regarding the presence of CE in Bolivia. In this study, 35 cysts isolated from livestock (mostly from the Department of La Paz) and 3 from humans (La Paz, Oruro and Potosi) were genetically characterized analysing the sequence of the cox1 gene (1609 bp). In total, 30 cysts (from La Paz, Cochabamba and Beni) were characterized as E. granulosus sensu stricto (3 fertile and 4 non-fertile cysts from sheep, 8 fertile and 12 non-fertile cysts from cattle and 3 fertile cysts from humans). A detailed analysis of the cox1 haplotypes of E. granulosus s.s. is included. Echinococcus ortleppi (G5) was found in 5 fertile cysts from cattle (from La Paz and Cochabamba). Echinococcus intermedius (G7) was identified in 3 fertile cysts from pigs (from Santa Cruz). Additionally, E. granulosus s.s. was detected in 4 dog faecal samples, while E. ortleppi was present in other two dog faecal samples. The implications of these preliminary results in the future implementation of control measures are discussed.

Vulvar phenotypes and the rate of development of eggs over a range of temperatures were used to compare Haemonchus contortus populations in New York State and Ohio. These parameters indicated that the westernmost boundary of the subspecies Haemonchus contortus cayugensis is the Chautauqua valley in New York. The Haemonchus ecotype in Ohio had a vulvar phenotype formula similar to that described for the subspecies Haemonchus contortus contortus. The relationship between slope and intercept of the regression of rate of egg hatch on temperature was different for morphs within ecotypes as well as between the New York and Ohio ecotypes. Linguiform-A appeared to be the cold-adapted morph in both ecotypes. Smooth was the warm-adapted morph in New York with linguiform-B filling that niche in Ohio.
