Parasitic diseases are endemic in parts of the tropics, but there is no convincing evidence that their prevalence or incidence is increasing due to the HIV epidemic. Available scientific data on parasitic infections in patients with the Acquired Immunodeficiency Syndrome (AIDS) suggests a predominance of Pneumocystis carinii, Toxoplasma gondii and Cryptosporidium spp. For reasons which are unclear, parasitic infections such as Plasmodium falciparum, Strongyloides stercoralis and Entamoeba histolytica, where cell-mediated immune responses are also thought to be significant, do not appear to be opportunists of importance. It is being increasingly recognized that chemotherapy for parasitic diseases has a host-dependent component, although scientific data on this subject remain scanty. The management of opportunistic parasitic infections in patients infected with HIV is dogged by failures and relapses, aptly illustrating the notion of the relationship between chemotherapy and the immune response. This review discusses the immunity and chemotherapy of opportunistic parasite infections in patients infected with the Human Immunodeficiency Virus (HIV).

This paper investigates a cohort of 2187 laboratory reared Culex quinquefasciatus fed on 69 human volunteers, including 59 persons with different levels of Wuchereria bancrofti microfilariae and 10 without microfilaria. Mosquitoes were followed until death. Mosquito survival was analysed in relation to the level of microfilaria in the human and larval count in the dead mosquito. Vector mortality during the extrinsic incubation period (12 days post-engorgement) was significantly higher in mosquitoes fed on microfilaraemic volunteers (50%) than in those fed on amicrofilaraemics (29%). Both the percentage infected and the geometric mean parasite density was significantly higher among mosquitoes which died before 13 days (45% infected and 10 larvae per infected mosquito) than those surviving beyond 13 days (39% and 2·2), suggesting a parasite loss of more than 80% during the extrinsic incubation period. A large proportion (62%) of the mosquitoes that died during the early of phase of parasite development were infected (36% in low, 26% in medium and 90% in high human Mf-density). Survival analysis showed that the parasite load in mosquitoes and the human Mf-density for a given parasite load are independent risk factors of vector survival. Overall, the hazard of dying was found to be 11–15 times higher among mosquitoes fed on microfilaraemic volunteers than those fed on amicrofilaraemics. The hazard doubles for every increase of about 60–70 parasites in the vector. As a consequence of the parasite-induced reduction in vector survival, the transmission success of the parasite is reduced. The implication of the results on control/elimination of lymphatic filariasis using mass-drug administration is discussed.

Binding to the midgut microvillar surface in the sandfly Phlebotomus papatasi is a prerequisite for successful development of Leishmania major within the gut of the vector. This paper describes a method for detecting microvillar-associated proteins which act as ligands for the parasite surface glycoconjugate lipophosphoglycan (LPG). Adhesion of LPG to midgut proteins was visualized by probing midgut extracts with LPG using a Western ligand blotting technique. Procyclic L. major LPG bound to a microvillar polypeptide band of 65 kDa (estimated in the non-reduced state) and bound variably to several lower molecular weight bands, probably degradation products or subunits of the primary binding polypeptides. Specificity of binding was confirmed by co-incubating biotinylated LPG with an LPG-specific mAb which resulted in a great reduction in binding.

This paper is written with the purpose of drawing further attention to the value of the complement fixation reaction as a diagnostic test in dourine and of recommending a method of procedure and technique arrived at with an experience of 15,000 tests for dourine made at the Veterinary Research Laboratory, Lethbridge.

The identification of the regions where vector-borne diseases are transmitted is essential to study transmission patterns and to recognize future changes in environmental conditions that may potentially influence the transmission areas. SGS1, one of the lineages of Plasmodium relictum, is known to have active transmission in tropical Africa and temperate regions of Europe. Nuclear sequence data from isolates infected with SGS1 (based on merozoite surface protein 1 (MSP1) allelic diversity) have provided new insights on the distribution and transmission areas of these allelic variants. For example, MSP1 alleles transmitted in Africa differ from those transmitted in Europe, suggesting the existence of two populations of SGS1 lineages. However, no study has analysed the distribution of African and European transmitted alleles in Afro-Palearctic migratory birds. With this aim, we used a highly variable molecular marker to investigate whether juvenile house martins become infected in Europe before their first migration to Africa. We explored the MSP1 allelic diversity of P. relictum in adult and juvenile house martins. We found that juveniles were infected with SGS1 during their first weeks of life, confirming active transmission of SGS1 to house martins in Europe. Moreover, we found that all the juveniles and most of adults were infected with one European transmitted MSP1 allele, whereas two adult birds were infected with two African transmitted MSP1 alleles. These findings suggest that house martins are exposed to different strains of P. relictum in their winter and breeding quarters.

The mechanism of anthelmintic resistance against the widely used macrocyclic lactones (MLs) is still not fully understood. Pharyngeal, somatic body muscles and the ovijector have been proposed as putative sites of action as well as resistance. In the present study the effects of three avermectins and three milbemycins on adult parasitic nematodes were evaluated in vitro. The Muscle Transducer system was used to investigate the effects of MLs on muscle contraction in female Haemonchus contortus and effects on motility were measured in Ostertagia (Teladorsagia) circumcincta using the Micromotility Meter. Concentration-response curves for all substances in both systems shifted to the right in the resistant isolates. Resistance was present to ivermectin (IVM) and its components IVM B1a and IVM B1b, suggesting that both components are involved in the mode of action and resistance. No consistent patterns of potency and resistance of the substances were observed except that milbemycins generally showed lower resistance ratios (RRs) than IVM. IVM and IVM B1b were the most potent inhibitors of contraction and motility in both susceptible isolates and also showed the highest RR in both species. Low RRs for milbemycins recorded in vitro for highly resistant isolates in vivo suggest that other factors such as pharmacokinetics influence drug potency in vivo.

Metacercariae of Parvatrema timondavidi were successfully cultivated in vitro to adults which produced eggs capable of embryonation and miracidial formation. The worms were grown in NCTC 109 supplemented with 20% inactivated bovine or chicken serum with a gas phase of 8% CO2 in air at 37 or 41°C. The use of the roller tube technique produced considerable promotion of growth in the worms. Heat inactivated bovine serum had a more salutary effect than unheated serum.

A method is described for the reproducible purification of greater than 1·0 × 108 sporozoites of Eimeria spp. by centrifugation on metrizamide density-gradients. The mean contamination of the purified sporozoite fraction by sporocysts and sporocyst debris was less than 6% and the recovered sporozoites were fully viable, infective and ultrastructurally intact.

Parasites cause harm to their hosts and represent pervasive causal agents of natural selection. Understanding host proximate responses during interactions with parasites can help predict which genes and molecular pathways are targets of this selection. In the current study, we examined transcriptional changes arising from interactions between Drosophila melanogaster and their naturally occurring ectoparasitic mite, Gamasodes queenslandicus. Shifts in host transcript levels associated with behavioural avoidance revealed the involvement of genes underlying nutrient metabolism. These genetic responses were reflected in altered body lipid and glycogen levels in the flies. Mite infestation triggered a striking immune response, while male accessory gland protein transcript levels were simultaneously reduced, suggesting a trade-off between host immune responses to parasite challenge and reproduction. Comparison of transcriptional analyses during mite infestation to those during nematode and parasitoid attack identified host genes similarly expressed in flies during these interactions. Validation of the involvement of specific genes with RNA interference lines revealed candidates that may directly mediate fly–ectoparasite interactions. Our physiological and molecular characterization of the Drosophila–Gamasodes interface reveals new proximate mechanisms underlying host–parasite interactions, specifically host transcriptional shifts associated with behavioural avoidance and infestation. The results identify potential general mechanisms underlying host resistance and evolutionarily relevant trade-offs.

The present study compared the miRNA expression profiles of five Toxoplasma gondii strains, namely RH (Type I, ToxoDB10), TgXD (Type I, ToxoDB10), PRU (Type II, ToxoDB1), QHO (Type II, ToxoDB1) and TgC7 (ToxoDB9), by Solexa deep sequencing, bioinformatics analysis and real-time quantitative PCR. A total of 7, 15, 10, 12 and 10 miRNAs were found from RH, TgXD, PRU, QHO and TgC7 strains, respectively. Thirteen miRNAs were shared by three genotypes, with only one miRNA shared by all of the 5 strains and others shared by 2 or more strains. A large number of targets ranging from 1 to 185 were identified for commonly shared miRNAs and strain-specific miRNAs with complete or nearly complete complementarity. Functional prediction showed that these targets were mostly focused on catalytic activity (191 targets) and binding activity (183 targets). Nonetheless, the majority of targets and most of the miRNAs are related to the virulence or invasion proteins of different strains of T. gondii, including ROP and MIC, as well as some other proteins, such as AMA1, GRA and RHO. The present study characterized comparatively the miRNA profiles of 3 different genotypes of T. gondii, identified genotype-shared miRNAs and strain-specific miRNAs.

The effect of an efficient bactericidal solution on the hatching of Haemonchus eggs was studied and it was found that a large proportion remained viable. Based on former work, a medium was devised in which the bacteria-free eggs hatched and the larvae developed to the infective stage. These larvae were morphologically and otherwise normal, except that they were slightly smaller than faecal-grown specimens, although the size ranges overlapped. Infection tests proved that Haemonchus larvae grown under conditions of sterility were normally infective for, and produced normal adults in, a susceptible lamb.

The present paper contains the results, other than those already published, of work done in the Department of Agricultural Zoology at the Birmingham University upon the tapeworms of birds. At the commencement, a little over two years ago, the aim was to make a complete investigation of the Cestodes of British birds, both from the standpoint of anatomy and also of life-history, since, as far as I am aware, there exists no adequate description of them. It was found impossible to obtain a regular supply of any one species of tapeworm, hence the work upon one particular species could only be carried on spasmodically. The results, having to wait upon the occurrence of the material, are necessarily incomplete. Circumstances having made it necessary temporarily to postpone the work, it was considered advisable to write up and publish the results so far obtained.

A simulation model for effects of Eimeria acervulina infection on technical and economic characteristics in broiler production is presented. The model describes development over time of the growth depression, feed intake reduction, and decrease in feed efficiency associated with infection. The model also shows a phase of compensatory growth during which earlier negative effects are counterbalanced, at least partly. Major assumptions made were: infection with E. acervulina occurs in each flock; production is affected in each flock; compensatory growth takes place because immunity develops and cell regeneration occurs. The results show that the pattern of development of the production characteristics during a flock cycle depends on the initial contamination level. Both a high and low initial contamination level results in a lower average daily gain, a worse feed to gain ratio, and a reduced net revenue compared to an intermediate contamination level.

Cysts of a Sarcocystis species were found in large numbers in skeletal muscle of an otter (Lutra lutra) which was raised in Norway and died in captivity in Sweden. This is the first report of Sarcocystis infection in the otter. The sarcocysts were 0·3–2·3 mm long and 0·06–0·25 mm wide. As judged by light microscopy the sarcocyst walls were thin (<3 μm) with a serrated surface but without visible projections. By transmission electron microscopy, the sarcocyst wall measured 0·6–1·8 μm and had minute undulations covering the entire sarcocyst surface giving the wall a wavy appearance. Septa were indistinct. The sarcocysts contained few metrocytes and numerous bradyzoites. Sarcocysts were not found in 69 other otters subjected to necropsy in Sweden.

During five months spent in Colombo (April–August 1944) a large number of small mammals and birds were examined for larval Trombiculinae.

From November 1944 to February 1945 similar investigations were carried out on the Maldive Islands. The number of species obtained included three species of the genus Gahrliepia Oudemans, 1912 (not included in this paper), Trombicula deliensis Walch, 1923, Trombicula acuscutellaris Walch, 1923, Neoschöngastia indica Hirst, 1915, Walchia turmalis Gater, 1932 (a second species of Walchia not included in this paper), a new species of Schöngastia herein named ‘maldiviensis’ n.sp., and Womersleyia minuta n.g., n.sp.

Snails, Elimia semicarinata, infected with Proterometra macrostoma were collected monthly in 1990 and 1991 from North Elkhorn Creek near Lexington, Kentucky, and kept on a 12:12 h light–dark cycle for 2 weeks. Cercariae emerging from snails were classified into 8 strains (I–VIII) based on differences in number and distribution of spined and spineless papillae on the tail. Cercariae also had unique patterns of emergence, swimming behaviour and infectivity in 4 species of sunfish. Of 513 infected snails collected in May, 339 had pure infections with the strain frequencies (% of 339): I, 46·6; II, 7·7; III, 12·1; IV, 8·8; V, 0·6; VI, 2·7; VII, 11·8; VIII, 9·7. In the multiple infections, 159 snails shed 2 strains, 14 shed 3, and 1 snail shed 4 strains simultaneously. A comparison of sunfish and parasite populations in Kentucky, Ohio and Michigan indicated that strain frequency in P. macrostoma is regulated by the species composition of the sunfish population.

In 1892 Louis Léger described three species of gregarine from the alimentary tract of the larva of Tipula: Actinocephalus tipulae, Clepsidrina longa, and Eirmocystis ventricosa2.

The immunogenicity of larval Trichuris muris in Schofield strain albino mice has been studied using infections abbreviated by chemotherapy. When mice were exposed to an initial infection lasting from 7 to 14 days their immunity to reinfection increased progressively, but the immunity stimulated by abbreviated infections was less than that stimulated by uninterrupted immunizing infections which last for approximately 16–18 days. The immune response of mice to T. muris appears to be stimulated by, and effective against, larvae of all ages and does not show stage specificity.

Echinococcus multilocularis is the causative agent of alveolar echinococcosis (AE), a potentially fatal zoonotic disease. Large parts of Turkey are considered as endemic for E. multilocularis. The aim of this study was to determine the occurrence of metacestode of E. multilocularis in wild rodents in Erzurum, an endemic region for human AE in Turkey. During the sampling period, a total of 498 rodents were trapped in twenty counties of Erzurum Province. Suspected lesions were observed on the livers of 48 rodents, and then partial fragment of mitochondrial 12S rRNA gene was PCR-amplified. Five liver samples exhibited E. multilocularis infection. The prevalence of E. multilocularis for Microtus spp. was 1·3%. All of the infected rodents had fertile metacestodes. Infected rodents were morphologically and molecularly analysed and were confirmed to be Microtus irani by the mitochondrial cytochrome b gene sequence analysis. This is the first report of the presence of E. multilocularis in rodent intermediate hosts in Turkey. Our findings of infected M. irani with protoscoleces show that this rodent can act as suitable intermediate host for E. multilocularis’ life cycle in Turkey. However, there was a complete lack of data on the infection of carnivores from the country. An extensive survey is recommended to determine the prevalence of E. multilocularis in definitive hosts in this endemic region.
