Protists are a diverse collection of eukaryotic organisms that account for a significant global infection burden. Often, the immune responses mounted against these parasites cause excessive inflammation and therefore pathology in the host. Elucidating the mechanisms of both protective and harmful immune responses is complex, and often relies of the use of animal models. In any immune response, leucocyte trafficking to the site of infection, or inflammation, is paramount, and this involves the production of chemokines, small chemotactic cytokines of approximately 8–10 kDa in size, which bind to specific chemokine receptors to induce leucocyte movement. Herein, the scientific literature investigating the role of chemokines in the propagation of immune responses against key protist infections will be reviewed, focussing on Plasmodium species, Toxoplasma gondii, Leishmania species and Cryptosporidium species. Interestingly, many studies find that chemokines can in fact, promote parasite survival in the host, by drawing in leucocytes for spread and further replication. Recent developments in drug targeting against chemokine receptors highlights the need for further understanding of the role played by these proteins and their receptors in many different diseases.

The distribution of the fluorescent ivermectin derivative (4″ 5,7 dimethyl bodipy proprionylivermectin, referred to here as bodipy ivermectin) was studied in muscle vesicle membranes from the parasite Ascaris suum. Incubation in bodipy ivermectin, at concentrations greater than 1 ng/ml, produced a dose-dependent fluorescence in the vesicles. The level of fluorescence was not depressed (but increased) by prior incubation with 1 μM unlabelled ivermectin. This observation suggested that most of the bodipy ivermectin was free in the lipid phase of the membrane and not associated with a receptor. The trypan blue quenching technique showed that the bodipy ivermectin did not cross the membrane bilayer but remained in the outer monolayer. Lateral mobility of the probe in the membrane was studied using the fluorescence recovery after photobleaching (FRAP) technique. The lateral diffusion coefficient (DL) was 14.0 x 10−10 cm2/s which was nearly three times faster than the lipid probe octadecanoyl-aminofluorescein in membranes from the same preparations; only 46% of the probe was mobile, showing that within a 1.2 μm radius the distribution of the bodipy ivermectin in the membrane was not homogeneous. These observations are used to suggest that: the site of action of livermectin is located in the outer monolayer of the membrane (possibly on the Cl− channel); and that the speed of onset of action in isolated membrane preparations may be limited by diffusion in the membrane.

Flow cytometric analysis of the binucleated protozoan parasite Giardia intestinalis gave DNA histograms with a broad Gl peak and a definable G2 + M peak with twice the DNA content of Gl. Twenty-four hour treatment with metronidazole arrested cell cycle progression of susceptible trophozoites in the G2 + M phase, but had no effect, even at toxic doses, on the DNA histogram of a line selected for resistance to metronidazole. Furazolidone was inhibitory to both stocks, causing an arrest in the S and G2 + M phases. Inhibitors of the mammalian cell cycle were also tested. Hydroxyurea, which blocks mammalian cells in Gl/S, and razoxane, which blocks in G2 + M, arrested trophozoites in the G2 + M phase whereas colchicine and gamma-irradiation had little or no effect on the cell cycle of G. intestinalis. These results suggest that the cell cycle of G. intestinalis may be controlled in a different manner from mammalian cells.

Echinostoma liei, a newly described 37-spined eehinostome from Egypt, was tested against two trematodes in paired infections in the snail host, Biomphalaria glabrata (NIH strain). E. liei, when matched with the highly predacious Brazilian echinostome Paryphostomum segregatum, was dominated by the latter's predatory rediae. Pre-existing E. liei infections were destroyed, although the P. segregatum infection was itself delayed in development by an unidentified ‘indirect antagonism’ elicited by the E. liei larvae. In concurrent exposures with the two species, or with E. liei challenge of an established P. segregatum infection, the E. liei miracidia penetrated the snail, but the infection did not become established.When E. liei and S. mansoni were paired, the former species dominated. Exposure of snails already infected with E. liei to miracidia of S. mansoni slowed growth of the latter and the schistosome sporocysts eventually disintegrated without producing cercariae. When E. liei miracidia were exposed to snails with S. mansoni infections of different ages, the echinostome became established but developed slowly. Subsequently, however, it destroyed the schistosome infection. Although E. liei is itself quickly eliminated by P. segregatum, the two species appear almost equal in anti-schistosome capacity. This can be credited in part to a deleterious or lethal effect of the E. liei-schistosome combination on the host snail, as well as to a direct anti-schistosomal effect. Greater knowledge of this host-inter-trematode balance may permit the use of trematodes to attack infected snails and their parasites in regions hyperendemic with human schistosomiasis.

Brachyplatystoma vaillanti is locally very common in the Amazon river, though, during my collecting tour in 1931, I examined only twenty-two examples, caught between Codajaz (nearly 1200 miles from the sea) and Gurupa. The local name of this Siluroid is Piramutāb; it is a fairly large fish (my largest specimen measuring 67 cm. in length) and possesses very long maxillary barbels (in some cases nearly as long as the body). From this fish I obtained two new species of Cestodes, one, the more numerous parasite, is a Phyllobothriid, and, so far as I am aware, this is the first occasion on which a Phyllobothriid has been described both from a fresh-water fish and from a Siluroid; and the other is a second and new species of Fuhrmann's genus Goezeella, very closely related to his G. siluri described in 1915.

The pulmonary and portal vasculature of naïve mice of the 129/Ola and CBA/Ca strains has been studied by means of the vasculature casting technique. This involves injection of pigmented vinylite resin into the arterial and venous systems, followed by digestion of the tissues with KOH. The peripheral vessels of the arterial and portal systems of CBA/Ca mice were numerous and highly branched. In contrast, casts prepared from 70–80% of naïve 129/Ola mice showed dramatic reductions in the number and extent of the peripheral vessels. In addition, such vessels appeared severely truncated. The remaining 20–30% of naïve 129/Ola mice yielded lung and liver casts that were indistinguishable from the CBA/Ca casts. Casts prepared from 129/Ola mice infected 6 weeks previously with Schistosoma mansoni cercariae showed the same segregation; faecal smears, together with observations of presence or absence of gross pathology in such mice confirmed that the vascular changes correlated with the ‘non-permissive trait’. We propose that such alterations facilitate the reportedly abnormal migration of schistosomes from the liver to the lungs in ‘non-permissive’ 129/Ola mice.

Onchocercomata with a defined worm population were analysed to elucidate the distribution of mast cells. Nodules with live females were classified according to the presence or absence of microfilariae. Immunohistochemical staining was performed using antibodies specific for mast cells or IgE. Mast cells appeared singly or in diffuse accumulations perivascularly and in inflammatory infiltrates between adult Onchocerca volvulus and in the capsular area. No mast cells were detected in cystic parts. Only few, scattered mast cells were found in the fibrous zone around the adult worm. They were increased with stronger infiltration and hence, related to the inflammatory cells. Mast cells were never localized directly at adult worms or microfilariae. A correlation of the mast cell distribution to the occurrence of eosinophils was observed regarding higher numbers of mast cells and eosinophils in nodules with microfilariae-producing females. Nodules with single males revealed higher numbers of mast cells than nodules with non-producing females, although both contained very few eosinophils. Onchocercomata with dead worms contained significantly more mast cells than those with live filariae. In conclusion, the localization and frequency of mast cells is contingent on the vitality and productivity of the worms and therefore, indirectly and directly on the release of O. volvulus antigens.

The prevalence of schistosome infections in intermediate host snails varies with snail age. The relationship between age and prevalence, the age—prevalence curve, is complex and may vary in space and time, and among parasite—host species. Field studies show that the shape of the age—prevalence curve may be seasonally variable, and that at some times there may be a decline in prevalence among older snails. This paper attempts to explain these observations in terms of the underlying epidemiological processes. A discrete-time version of Muench's catalytic model for age-dependent infection is developed. Model simulations were carried out using life-history and epidemiological parameters derived from studies of Schistosoma haematobium—Bulinus globusus in Zimbabwe. Analysis of model behaviour identifies aspects of the schistosome— snail interaction that affect the shape of the age—prevalence curve. The following features can result in a decline in prevalence among older snails. (1) A decrease in the survival rate of patent infected snails with age. (2) A decrease in the force of infection with age. (3) A high rate of loss of infection. (4) A heterogeneity in the snail population such that the probability of infection is correlated with snail fecundity. (This would occur if there existed a spatial correlation between force of infection and fecundity, or if there were a correlation between fecundity and susceptibility.) The evidence for the occurrence of these features in the field is assessed. Survival rate is related more closely to the duration of patent infection than to age per se. The evidence for age-dependent force of infection is equivocal. Significant loss-of-infection rates have yet to be demonstrated. Heterogeneities in force of infection and fecundity have been reported and, for the Zimbabwe data, this mechanism can explain seasonally in the age—prevalence curve as a function of known seasonal variation in the force of infection and snail fecundity.

Hookworms feed on blood, utilizing haemoglobin for nutrition, growth and reproduction. The haemoglobin digestion cascade has been partially elucidated, but the process immediately preceding this event, haemolysis, has received considerably less attention. We have cloned and expressed Ancylostoma caninum mRNAs encoding 2 proteins belonging to the saposin-like protein (SAPLIP) family, termed Ac-slp-1 and Ac-slp-2. The open reading frames of SLP-1 and SLP-2 were used to identify expressed sequence tags encoding SAPLIPs from the 4 major clades of animal parasitic nematodes. Both Ac-slp-1 and slp-2 mRNAs were shown to be expressed in all life stages assessed, with slp-1 predominantly being expressed in third-stage larvae (L3) before and after activation with dog serum. Recombinant SLP-1 and SLP-2 were expressed in insect cells and used to raise specific antisera in mice. These antisera were used as probes in fluorescence microscopy to localize the anatomic expression sites of both proteins to small, punctate organelles or vesicles within the intestinal cells of adult worms; weak staining was detected on the microvillar brush border of the intestine. Using transmission electron microscopy, both proteins were localized to similar vesicles in the intestinal cells of the L3. Recombinant proteins contained C-terminal purification tags that potentially precluded dimerization and possibly interfered with the subsequent detection of haemolytic activity. Their expression in the gut of the L3 and adult stages suggests a role for these hookworm SAPLIPs in the lysis of host cells during tissue migration and/or feeding.

Toxoplasma gondii, an obligatory intracellular protozoan parasite, is one of the causative agents of ovine abortion, as reported in many countries. Different techniques are being used to detect this pathogen in infected ovine fetuses. One of the most sensitive and specific diagnostic techniques is Nested-PCR amplification of the B1 target gene of the organism. In total, 200 brain samples of aborted ovine fetuses and maternal sera submitted from different parts of Khorasan Razavi province, Iran were investigated to track the role of Toxoplasma gondii in ovine abortion by a slightly modified Nested-PCR and IFAT assays, respectively. Among all samples, 27 (13·5%) were PCR-positive and 31 (15·5%) were IFAT-positive and the Toxoplasma-induced abortion prevalence calculated was 8·8% to18·2% with 95% confidence interval. Results show that high levels of congenital transmission may occur in 27/31(87%) of pregnancies with an excellent logical agreement (ĸ=0·9) between 2 different tests. According to the results of this study, the Nested-PCR employed in this investigation could be recommended as an applied routine test for the routine examination and confirmation of Toxoplasma gondii-induced ovine abortion.

Sprague–Dawley rats were infected by intraperitoneal injection with either the cysticerci of Taenia crassiceps or the tetrathyridia of Mesocestoides corti. Both species of metacestodes reproduce asexually in the peritoneal cavity. The effect of T. crassiceps upon the rats was to cause chronic inflammation of the peritoneum, fibrous adhesions between the visceral organs and hyperplasia of the gut mesenteries which became extended into a large sac-like structure. Dead parasites surrounded by white calcareous material were found free in the peritoneum and attached to the inner body wall and viscera. The histopathology of the hyperplastic mesenteries and fibrous membrane is described. The tetrathyridia of M. corti were found free in the peritoneal cavity as well as in the liver (mainly the left liver-lobe), gut mesenteries and in an outgrowth of the connective tissue associated with the pancreas. The connective tissue outgrowth comprised 2 layers; an inner reticular layer and a thickened marginal layer. Metacestodes were found in collagen—fibrin capsules within the connective tissue. A layer of mucopolysaccharide material was interposed between the parasite and the capsule wall. A possible method by which the parasites gain entry into the connective tissue is described. The connective tissues of rats harbouring either M. corti or T. crassiceps had increased numbers of mast cells compared to connective tissues from control rats. Younger mast cells containing biogenic amines predominated in infected tissues whereas older heparin-containing cells dominated control tissues.

The recent development of analytical models of trypanosomiasis has increased the general applicability of models to the strategic control of the disease. An analysis of detailed data on tsetse abundance and infection rates and trypanosome prevalence in village-based trypanotolerant cattle over 4 years in The Gambia showed that seasonal patterns of abundance in Glossina morsitans-infested areas were consistent, and that the rates of trypanosome infection remained relatively unchanging. However, there were two distinct seasonal trypanosome prevalence patterns in cattle, with peaks occurring either in May/June/July or November/December. The peaks of trypanosome prevalence therefore occurred either 4 months before or after the times of peak challenge from G. morsitans, not 1 or 2 months after as predicted by analytical models. In G. palpalis-infested areas there was little seasonal variation in abundance or trypanosome infections, but peak trypanosome prevalence still occurred mostly in June/July. Despite the incongruity between the months of peak prevalence and challenge, the overall annual prevalence rates and tsetse challenge indices showed a significant linear relationship. It is concluded that existing analytical models need to be refined to take into account trypanotolerance and the various influences on the expression of this trait.

Only 10 species of Nycteribiidae have been recorded previously from the Ethiopian Region and 3 further species from Madagascar. The writer recently described the new genus Dipseliopoda with three species, a new species of Eucampsipoda from Africa and another from Madagascar, (Theodor, 1955), 3 new species of Basilia and a new species of Tripselia from Africa (Theodor, 1956).

Heterogeneities in parasite infection among conspecific hosts often manifest as sex- or size-biased infections, which are typically attributed to differential host susceptibility and exposure. Since parasite fitness is often tied to host quality, host preference by parasites is likely to be under strong selection. We test the hypothesis that host preference is sufficient to generate variability in infection rate among conspecifics. Specifically, we ask whether the mite Macrocheles muscaedomesticae is able to discriminate between Drosophila hydei hosts of different sex and size, while explicitly accounting for the potential confounding effects of these two factors. Our results indicate a preference for female hosts, but this preference appears to be driven by size and not sex per se. When differences in body size were controlled for, the sex-biased infection disappeared, while mites presented with the choice of two female flies of disparate sizes were more likely to select the larger host. Across the distribution of fly body weight in this study, mites preferentially attached to flies of intermediate size. This study provides evidence that mite choice for certain host types can play an important role in parasite transmission, even in the absence of differential susceptibility or exposure among hosts.

Microsporidia are intracellular parasites, frequently infecting the planktonic crustacean Daphnia. Questioning the ability to detect and identify microsporidia with conventional microscopic techniques, we applied molecular methods in order to investigate the distribution and co-infection patterns of this parasite among 8 communities of the Daphnia longispina hybrid complex. Eight microsporidian taxa were detected, including 3 that previously had not been characterized genetically. Microsporidian communities from nearby lakes were found to be more similar to each other, apparently due to short distance dispersal via secondary hosts. Moreover, we detected seasonal (but not interannual) changes in microsporidian community structure. With some microsporidia being host-specific, these changes might have resulted from seasonal changes in host taxon and clonal composition. The 2 dominant and closely related parasite species were found mainly in single infections, whereas another pair of related microsporidians was found predominantly in co-infections; suggesting species-level differences in the ability to colonize infected hosts. By applying molecular methods, we were not only able to unambiguously identify parasite taxa but also to reveal multiple infections that otherwise would have remained undetected. Given the increased level of accuracy and sensitivity, we highly recommend molecular approaches in future parasite surveys of Daphnia infections.

While group formation provides antipredatory defences, increases foraging efficiency and mating opportunities, it can be counterintuitive by promoting disease transmission amongst social hosts. Upon introduction of a pathogen, uninfected individuals often modify their social preferences to reduce infection risk. Infected hosts also exhibit behavioural changes, for example, removing themselves from a group to prevent an epidemic. Conversely, here we show how Trinidadian guppies infected with a directly transmitted ectoparasite, Gyrodactylus turnbulli, significantly increase their contact rates with uninfected conspecifics. As uninfected fish never perform this behaviour, this is suggestive of a parasite-mediated behavioural response of infected hosts, presumably to offload their parasites. In the early stages of infection, however, such behavioural modifications are ineffective in alleviating parasite burdens. Additionally, we show that fish exposed to G. turnbulli infections for a second time, spent less time associating than those exposed to parasites for the first time. We speculate that individuals build and retain an infection cue repertoire, enabling them to rapidly recognize and avoid infectious conspecifics. This study highlights the importance of considering host behavioural modifications when investigating disease transmission dynamics.

It is generally assumed that the digenean human liver fluke, Fasciola hepatica, gained entry to South America during the 15th century upon arrival of Europeans and their livestock. Nonetheless in Patagonia, Argentina, digenean eggs similar to F. hepatica have been observed in deer coprolites dating back to 2300 years B.P. The main objective of our present study was to identify and characterize these eggs using an ancient DNA (aDNA) study. Eggs were isolated and used for aDNA extraction, amplification and sequencing of partial regions from the cytochrome c oxidase subunit 1 and the nicotinamide adenine dinucleotide dehydrogenase subunit 1 mitochondrial genes. Also, phylogenetic trees were constructed using Bayesian and maximum likelihood. Our results confirm the presence of F. hepatica in South America from at least 2300 years B.P. This is the first report and the first aDNA study of this trematode in South America prior to the arrival of the European cattle in the 15th century. The present work contributes to the study of phylogenetic and palaeobiogeographical aspects of F. hepatica and its settlement across America.

Strongyloides venezuelensis is a parasitic nematode that infects rodents. Although Strongyloides species described to date are known to exhibit parthenogenetic reproduction in the parasitic stage of their life cycle and sexual reproduction in the free-living stage, we did not observe any free-living males in S. venezuelensis in our strain, suggesting that the nematode is likely to depend on parthenogenetic reproduction. We confirmed by cytological analysis that S. venezuelensis produces eggs by parthenogenesis during the parasitic stage of its life cycle. Phylogenetic analysis using nearly the full length of 18S and D3 region of 28S ribosomal RNA gene suggested that S. venezuelensis is distantly related to another rodent parasite, namely Strongyloides ratti, but more closely related to a ruminant parasite, Strongyloides papillosus. Karyotype analysis revealed S. venezuelensis reproduces with mitotic parthenogenesis, and has the same number of chromosomes as S. papillosus (2n = 4), but differs from S. ratti (2n = 6) in this regard. These results, taken together, suggest that S. venezuelensis evolved its parasitism for rodents independently from S. ratti and, therefore, is likely to have a different reproductive strategy.

The central nervous system (CNS) has been recognized as an immunologically specialized microenvironment, where immune surveillance takes a distinctive character, and where delicate neuronal networks are sustained by anti-inflammatory factors that maintain local homeostasis. However, when a foreign agent such as a parasite establishes in the CNS, a set of immune defences is mounted and several immune molecules are released to promote an array of responses, which ultimately would control the infection and associated damage. Instead, a host–parasite relationship is established, in the context of which a close biochemical coevolution and communication at all organization levels between two complex organisms have developed. The ability of the parasite to establish in its host is associated with several evasion mechanisms to the immune response and its capacity for exploiting host-derived molecules. In this context, the CNS is deeply involved in modulating immune functions, either protective or pathogenic, and possibly in parasitic activity as well, via interactions with evolutionarily conserved molecules such as growth factors, neuropeptides and hormones. This review presents available evidence on some examples of CNS parasitic infections inducing different morbi-mortality grades in low- or middle-income countries, to illustrate how the CNS microenvironment affect pathogen establishment, growth, survival and reproduction in immunocompetent hosts. A better understanding of the influence of the CNS microenvironment on neuroinfections may provide relevant insights into the mechanisms underlying these pathologies.
