The study examines the rate of re-acquisition of Trichuris trichiura infection after treatment in two populations, one of mixed age and the other of children with known preintervention infection intensity. A population living in a Caribbean village was treated with mebendazole and the rate of reacquisition of infection of four age classes (2–4, 5–10, 11–30, 30+ years) monitored over a 20-month period. The reinfection rate was higher in the child age-classes indicating either that children are more exposed to infection or that adults slowly develop a partially effective acquired immunity. A cohort of children (mean age 4·5 years) was separated into 3 intensity categories on the basis of expelled worm burdens and their rate of reacquisition of infection monitored over a 12-month period. The rate of reinfection was directly and positively associated with initial infection status. This may indicate that children with low intensity infections are consistently less exposed to infection or that they have effective immune responses. The latter conclusion, however, would imply that they had acquired this immunity early in life, and so appears to contradict the suggestion that resistance is only slowly acquired by adults. Reconciling these two conclusions may require more sophisticated immunological models than have been suggested previously for geohelminthiases.

Chicory is a perennial crop that has been investigated as a forage source for outdoor-reared ruminants and pigs, and has been reported to have anthelmintic properties. Here, we investigated in vitro anthelmintic effects of forage chicory-extracts against the highly prevalent swine parasites Ascaris suum and Oesophagostomum dentatum. Methanol extracts were prepared and purified from two different cultivars of chicory (Spadona and Puna II). Marked differences were observed between the anthelmintic activity of extracts from the two cultivars. Spadona extracts had potent activity against A. suum third (L3) and fourth (L4) – stage larvae, as well as O. dentatum L4 and adults, whereas Puna II extracts had less activity against A. suum and no activity towards O. dentatum L4. Transmission-electron microscopy of A. suum L4 exposed to Spadona extracts revealed only subtle changes, perhaps indicative of a specific anthelmintic effect rather than generalized toxicity. Ultra-high liquid chromatography-mass spectrometry analysis revealed that the purified extracts were rich in sesquiterpene lactones (SL), and that the SL profile differed significantly between cultivars. This is the first report of anthelmintic activity of forage chicory towards swine nematodes. Our results indicate a significant anthelmintic effect, which may possibly be related to SL composition.

This study compared genetic diversity of Toxoplasma gondii isolates from Portugal, Austria and Israel. For this, we genotyped 90 T. gondii isolates (16 from Portugal, 67 from Austria and 7 from Israel) using 10 nested PCR-restriction length polymorphism (RFLP) genetic markers and 15 microsatellite (MS) markers. By PCR-RFLP typing, 7 isolates from Portugal chickens were identified as type II (ToxoDB #1 or #3), 4 were type III (ToxoDB #2) and the remaining 4 isolates have unique genotype pattern were designated as ToxoDB #254. One mouse virulent isolate from a bovine fetus (Bos taurus) in Portugal was type I (ToxoDB #10) at all loci and designated as TgCowPr1. All 67 isolates from Austria and 7 from Israel were type II (ToxoDB #1 or #3). By MS typing, many additional genetic variations were revealed among the type II and type III isolates. Phylogenetic analysis showed that isolates from the same geographical locations tend to cluster together, and there is little overlapping of genotypes among different locations. This study demonstrated that the MS markers can provide higher discriminatory power to reveal association of genotypes with geographical locations. Future studies of the type II strains in Europe by these MS markers will be useful to reveal transmission patterns of the parasite.

Light and transmission electron microscopy were used to study different stages of Perkinsus atlanticus (Apicomplexa) during induced zoosporulation, with fluid thioglycollate medium and seawater. Cytokinesis and nucleokinesis of different developmental stages were studied every 12 h during the incubation period of 72 h, at which time the zoospores became free. Uninucleated and flagellated zoospores present the apical complex formed by conoid, polar ring, micronemes, rhoptries and subpellicular microtubules observed at different sections. Ultrastructural details were compared with the other two species of the genus Perkinsus.

Strains of mice poorly (B10) or non-responsive (B10.BR) to a primary infection with Trichuris muris were protected against infection by vaccination with excretory/secretory (E/S) antigen in Complete Freund's Adjuvant (CFA). Protection in these mice was slow to be expressed compared to that in good responder strains. Vaccination boosted the IgG and IgG1 antibody responses to E/S antigen and altered the antigen recognition profiles, three high molecular weight antigens (80–85, 90–95, 105–110 kDa) being recognized by antibodies in sera from vaccinated but not control mice. B10. BR mice which had experienced a patent primary infection could not be protected against challenge infections by vaccination and this was correlated with depressed levels of IgG1, but not total IgG, to E'S antigen early post-challenge compared with vaccinated infected mice which had not seen an adult primary infection. There was also lack of recognition of the three high molecular weight antigens recognized by antibodies in sera from mice infected after vaccination. It is suggested that the rapid development of high levels of IgG1 antibodies, and the recognition of the three high molecular weight antigens, may reflect events that are important in protective immunity. Immunomodulation of host immunity by T. muris may therefore be achieved, at least in part, by the suppression of specific IgG1 levels, the production of an irrelevant IgG isotype and prevention of the recognition of critical antigens.

Microfilariae belonging to the genera Brugia, Loa and Wuchereria have been stained by a wide variety of histochemical methods. The results are recorded in eight Tables.

The sheath of the microfilaria contains polyanionic material, probably polysulphate, periodate reactive mucosubstance, sulphur-protein and complex lipids. The fluid within the sheath is also polyanionic.

The cuticle of the microfilaria is stabilized by disulphide bonds possibly in a collagen but it was not possible to demonstrate collagen by staining. The cuticle also contains a periodate reactive G mucosubstance and complex lipids. The annular grooves of the cuticle also contain a periodate unreactive mucosubstance. Certain areas of the cuticle, around the excretory pore, the pores of the Schwanzgebilde, and the cephalic structures - oral ring, hook and spines - are hardened by deposition of a keratinous scleroprotein. Some reactions of the cuticle may be masked by the presence of basic protein.

The excretory vesicle contains polyanionic material but there was no clear evidence that the polyanionic material within the fluid in the sheath came from the excretory vesicle. The excretory vesicle, the anal vesicle and the subcuticular cells contain periodate reactive G mucosubstance. Most RNA staining was found in the excretory vesicle, the excretory cell, the anal vesicle, the rectal cells and the G cell.

The pharyngeal thread, the Innenkörper, the rectal cells and the anal vesicle represent the modified gut of this first stage larva. The pharyngeal thread appears to consist largely of a protein-mucosubstance complex that can be unmasked by oxidation. The Innenkörper contains a basic protein and acidic mucosubstance, possibly sialomucin, complex that can be unmasked by adjusting the electrolyte concentration of the staining media.

The relevance of some of these techniques in the analysis of possibly unique invertebrate structures is discussed briefly.

We tested the hypothesis that compositional and/or phylogenetic dissimilarity of host assemblages affect compositional and/or phylogenetic dissimilarity of parasite assemblages, to different extents depending on scale, using regional surveys of fleas parasitic on small mammals from 4 biogeographical realms. Using phylogenetic community dissimilarity metric, we calculated the compositional and phylogenetic dissimilarity components between all pairs of host and parasite communities within realms and hemispheres. We then quantified the effect of compositional or phylogenetic dissimilarity in host regional assemblages, and geographical distance between assemblages, on the compositional or phylogenetic dissimilarity of flea regional assemblages within a realm, respectively. The compositional dissimilarity in host assemblages strongly affected compositional dissimilarity in flea assemblages within all realms and within both hemispheres. However, the effect of phylogenetic dissimilarity of host assemblages on that of flea assemblages was mostly confined to the Neotropics and Nearctic, but was detected in both the Old and New World at the higher scale, possibly because of phylogenetic heterogeneity in flea and host faunas between realms. The clearer effect of the compositional rather than the phylogenetic component of host community dissimilarity on flea community dissimilarity suggests important roles for host switching and ecological fitting during the assembly history of flea communities.

The development of resistance by the Houghton strain of Eimeria tenella (H) to monensin has been studied by serially passaging the strain in groups of 30 birds, each inoculated with 106 oocysts and given 100 p.p.m. of drug in the food. After 16 passages a line partially resistant to this concentration of monensin was produced. Passage of the line in birds medicated with 200 or 300 p.p.m. of monensin did not result in the acquisition of a greater degree of resistance. A further reduction in sensitivity to 100 p.p.m. monensin was obtained, however, when the line was serially passaged in 15 groups of 6 birds each given 3 × 106 oocysts. Five passages in the absence of monensin resulted in restoration of sensitivity, suggesting that the resistance developed was not stable. The pathogenic effects of the line that had been passaged in chickens given 300 p.p.m. of monensin were less than those of the parent line.

Trypanosomosis is a major cause of mortality for dogs in Nigeria and treatment with diminazene aceturate has steadily become less effective, either as a result of low quality of the locally available diminazene preparations or of drug resistance. To investigate these alternatives, samples of locally obtained drugs were analysed for diminazene aceturate content and a strain of Trypanosoma brucei brucei was isolated from a diminazene-refractory dog in Nsukka, south-eastern Nigeria, and used to infect albino rats. The quality of diminazene aceturate-based preparations was variable, with two preparations containing less than 95% of the stated active compound. Rats infected with T. brucei isolated from the dog were treated 7 and 10 days after infection either with 7 mg/kg diminazene aceturate (intraperitoneally, once) or with 4 mg/kg pentamidine isethionate (intramuscularly, 7 consecutive days). Relapse rates were 100% for both trypanocides in the groups of rat treated 10 days post-infection, and 83% and 50% of rats treated 7 days after infection relapsed to diminazene aceturate and pentamidine isethionate, respectively. Careful consideration of physiological parameters showed that pentamidine was only marginally superior to diminazene aceturate as applied in this study. It was concluded that dogs in Nigeria are infected with genuinely diminazene aceturate-resistant trypanosomes that appear to be cross-resistant to pentamidine isethionate.

There has been much difference of opinion as to the specificity of Strongyloides in different hosts, therefore an opportunity has been made use of to compare representatives of this genus, in various stages of their life histories, from monkeys, cats and man, and to compare these with the published characteristics of other forms.

The reduction in worm burden obtained by treatment of Schistosoma mansoni with praziquantel and oxamniquine was greater in mice with heavy infections than in relatively lightly infected animals. The reduction in worm burden achieved by each drug correlated with the size of the pre-treatment worm burden (r2 = 0·82 and 0·81 for praziquantel and oxamniquine, respectively). Intensity of infection did not affect the degree of tegumental damage and drug-induced antigen exposure on worms recovered soon after treatment with praziquantel. However, praziquantel-treated worms from mice with heavy infections had significantly more murine antibody attached to the treated-worm surface than worms from praziquantel-treated lightly infected mice. Heavily infected mice had greater levels of circulating anti-worm antibodies than lighter infected mice. The correlation between infection intensity and cure rates achieved by praziquantel and oxamniquine may thus be a reflection of the higher litres of relevant antibody in heavily infected mice mediating death of drug-treated worms.

1. The life-history of Phaenoserphus viator is described.

Four larval instars are found, endoparasitic in the larvae of Pterostichus niger. At thee nd of the last larval instar the parasites, which may number as many as 45 in a single host, emerge, and while still attached, pupate without spinning a cocoon.

Adults may appear in August or September.

The effect of the parasite in inhibiting metamorphosis of the host is discussed.

2. The first observed larva is atracheate and incompletely segmented at first and is of the polypod type bearing paired prolegs on the body segments.

Subsequent instars are apodate.

The tracheal system develops progressively in the several instars, but only becomes functional in the final stage.

3. The anatomy of the larva is briefly described with the exception of the musculature.

Tracheal development is described. Gas only appears in the tracheae after the development of the tracheole cells puts the tracheae into communication with the body wall and other organs.

In the circulatory system an important accessory organ is the neural sinus, formed by the enclosure of the ventral nerve cord beneath a connective tissue curtain.

The imaginal discs of the hypodermis are briefly described, these being clearly defined in the head, thorax, and posterior abdominal segments.

The nervous system consists of a brain, suboesophageal ganglion and 11 ventral ganglia, the most posterior being tripartite. This system is connected with the sympathetic, by nerves passing from the cerebral commissures to a frontal ganglion which lies above the oesophagus and behind the labrum.

To address whether a fish host is involved in the life cycles of malacosporeans of the genus Buddenbrockia, cohabitation experiments with different bryozoan and fish species were conducted. Samples were analysed by malacosporean-specific PCR, partial sequencing of the 18S rDNA, and light and electron microscopy. Co-habitation challenges with bryozoans resulted in malacosporean infections detected mainly in the kidney of common carp (Cyprinus carpio) and minnow (Phoxinus phoxinus). Sequences of the minnow parasite and of worm-like Buddenbrockia stages in Plumatella repens were identical and showed 99·5% similarity to Buddenbrockia plumatellae and 96·3% similarity to the sequence obtained from carp. One sample, comprising 4–5 zooids of statoblast-raised bryozoans cohabitated with infected carp was PCR-positive, but no overt infection could be observed in the remaining colony. Light and electron-microscopy of kidney samples of infected minnows revealed single cells within kidney tubules, whereas in carp, sporogonic stages were found in kidney tubules. Phylogenetic analysis of the Buddenbrockia spp. known to date placed the carp-infecting species at the base of the B. plumatellae clade, but low posterior probability makes this node questionable. The present study showed that Buddenbrockia spp. were able to infect cyprinid fish, showing stages in kidney-tubules strikingly similar to those of T. bryosalmonae.

Experiments are described on the inactivation by gamma-rays, X-rays, and alpha-rays of the viruses of tomato bushy stunt, tobacco necrosis, tobacco ringspot, tobacco mosaic and potato virus X. Within the errors of the experiment the inactivation curves appear to be exponential, and the inactivation doses increase in the order gamma-rays, X-rays, of wave-length 1·5 A., X-rays of wave-length 8·3 A., and alpha-rays.

A theory is given explaining these results and correlating the inactivation dose with the virus size. Estimates of the sizes of the viruses obtained from the radiation experiments he within the range of the sizes given by other methods, but are somewhat lower than the most probable sizes. Possible explanations of the discrepancy which are discussed are (a) the virus particle is not the molecule, in the sense of the smallest infective unit, or (b) certain structural changes in the virus molecule produced by the radiation may still leave it infective. Some of these may perhaps show themselves as mutations.

The in vitro absorption of glucose and l-methionine in the intestine of broiler chickens was measured 7, 14 and 21 days post-inoculation (p.i.) with sporulated oocysts of Eimeria acervulina, E. mivati, E. maxima or E. brunetti. The small intestine of each bird was divided into 8 regions of equal length and absorption was measured on 3 tissue disks of equal size from each region. The absorption rate of each substrate with each coccidial species was measured based on (1) an equal area from each region, (2) an equal weight from each region, (3) the total absorption in each region and (4) the total potential absorption in the intestine. Comparisons of absorption rate of equal areas in each intestinal region demonstrated that infected birds at 7 days p.i. absorbed significantly less substrate per unit area in the regions of maximum infection than uninfected controls. Malabsorption was less apparent when the weight of the region was used as the unit of measurement. Compensatory absorption was seen in some uninfected regions with E. acervulina. The total potential intestinal absorption at 7 days p.i. was reduced with E. mivati, E. maxima and E. brunetti but not with E. acervulina. At 14 days p.i., total l-methionine and glucose absorption in some regions of the intestine was significantly increased with E. acervulina but not with E. mivati, E. maxima or E. brunetti. No absorption differences were seen at 21 days p.i. with any species.

Iron-59 (as a ferric chloride solution or as a component of haemoglobin) was introduced into the lumen of the gut of Fasciola hepatica. It was detected by autoradiography in high concentrations in the intestinal epithelium, the cuticular epithelium, the myoblasts and the cells forming the walls of the excretory tubules. Carbon-14, introduced as l-phenylalanine-C-14, was concentrated in the same tissues, with the exception of the intestinal epithelium. These findings are discussed in relation to the function of the respective tissues.

Parasites are known to affect the predatory behaviour or diet of their hosts. In relation to biological invasions, parasites may significantly influence the invasiveness of the host population and/or mediate the relationships between the invader and the invaded community. Dikerogammarus villosus, a recently introduced species, has had a major impact in European rivers. Notably, its high position in trophic web and high predatory behaviour, have both facilitated its invasive success, and affected other macroinvertebrate taxa in colonized habitats. The intracellular parasite Cucumispora dikerogammari, specific to D. villosus, has successfully dispersed together with this amphipod. Data presented here have shown that D. villosus infected by this parasite have a reduced predatory behaviour compared with healthy individuals, and are much more active suggesting that the co-invasive parasite may diminish the predatory pressure of D. villosus on newly colonized communities.

Parasite ecologists are often interested in the repeatability of patterns in parasite communities in space and/or time, because of implications for the dynamics of host-parasite interactions. Field studies usually examine temporal and spatial variation in isolation or limit themselves to a small number of host populations. Here, we studied the macroparasite communities of 12 populations of three-spined stickleback, Gasterosteus aculeatus L., on North Uist, Scotland, separated by small geographical distances, during the breeding season in 2 consecutive years (2007 and 2008) to determine: (1) the extent of spatial variation in macroparasite communities, (2) whether this variation is consistent across years, and (3) whether habitat characteristics can explain differences in macroparasite community composition among populations. We found substantial variation in parasite communities among populations. Generally, measures of parasite community composition were higher in 2008 than in 2007, but this effect of year was consistent across populations, such that the relative differences in these measures among populations changed little between years. These data suggest that there is short-term stability in the spatial variation in macroparasite communities of North Uist sticklebacks. However, none of the 5 habitat characteristics measured explained spatial variation in any measure of parasite community composition.

Recent work has shown that mice with subclinical parasitic infections suffer impaired spatial learning and memory, as assayed in an open-field water maze. Although the mechanism underlying this effect is not clear, the phenomenon has been reported following infection with both a protozoan parasite (Eimeria vermiformis) and a gastrointestinal nematode (Heligmosomoides polygyrus). In a variety of experiments, we examined the effects of a different gastrointestinal nematode, Strongyloides ratti, on the ability of rats and mice to learn a spatial or a discrimination task. Animals were tested at various stages post-infection, with different levels of infection, using different lines of S. ratti and with varying experimental protocols. All animals learned the tasks, but we found no evidence of an effect of S. ratti infection on learning or memory. Even rats infected with approximately 5000 S. ratti larvae, a dose which has an impact on rat body size, showed no deficit in learning ability. Various reasons for the conflict between our results and those previously reported for E. vermiformis and H. polygyrus are discussed. Our results show that impaired learning and memory following parasitic infection is not a ubiquitous or at least easily replicated phenomenon.

A herd of 20 cattle was introduced on a ranch in Kilifi, Coast Province of Kenya, where they were in contact with Glossina austeni for 6 months. In total, 65 trypanosome isolates were made from these animals. Examination of the isolates revealed that 61 were Trypanosoma congolense and 4 were T. theileri. Out of the 61 T. congolense isolates, 55 were successfully passaged and cloned in mice to provide trypanosome populations for further analyses. The stocks and their clones were inoculated into goats on which teneral G. morsitans centralis were later fed in order to provide metacyclics for use in serodeme analysis. Identification of serodemes was carried out by indirect immunofluorescence and neutralization using antimetacyclic hyperimmune sera prepared in mice against metacyclics of cloned trypanosome populations. So far 4 serodemes have been identified in 8 stocks and 7 clones. Each of the 9 stocks contained a mixture of at least 2 of the 4 serodemes identified. Furthermore, stocks isolated sequentially from individual animals contained the same serodemes despite repeated treatment with a curative dose (6 mg/kg body weight) of Berenil between isolations. From the latter finding, it can be inferred that the 4 serodemes were present on the ranch throughout the study period.
