A survey was made of the helminth fauna of 657 herring gulls (474 ‘adults’ and 183 chicks) in northern Caernarvonshire and Anglesey during the period July 1962 to November 1964. A total of 31 species of helminths were identified (10 species of Trematoda, 11 of Cestoda and 10 of Nematoda), including 1 new species, four new host records and three new British records. Two species of Mallophaga were identified from the birds. In three cases (Gymnophallus deliciosus, Paricterotaenia porosa and Capillaria contorta) a significant difference was found in the burden carried by male and female birds. ‘Adult’ birds were generally found to be more heavily infected with helminths than the chicks, with the exception of the nematode Cyathostoma lari, which was found more frequently in the chicks. A new intermediate host for Capillaria contorta was noted, namely Eiseniella tetraedra.

Three species of Trematoda, two of Cestoda, and two of Nematoda were found to be cyclic (seasonal) in their time of appearance. Tetrabothrius spp. appeared most frequently in winter, whilst Anomotaenia larina was found only during the summer months. C. lari showed a primary peak of appearance in summer, with a secondary peak in winter. Capillaria contorta appeared in high numbers throughout the year.

The distribution of helminths within the birds was also examined. Gymnophallus deliciosus was generally found in the gall bladder, whilst Parorchis acanthus was found only in the rectum. Intestinal trematodes were normally found in the mid-third of the small intestine (e.g. Spelotrema excellens, Himasthla elongata). In the case of C. lingua, however, peak numbers were found in the duodenum, whilst Brachylaemus fuscatus was found most frequently in the posterior third of the small intestine. Cestodes appeared most frequently in the anterior or posterior thirds of the small intestine (e.g. Tetrabothrius spp., Anomotaenia micracantha). Hymenolepis cirrosa showed two peaks of infection along the length of the small intestine, a small one in the anterior third, a much larger one in the posterior third. Attempts were made to correlate the helminth burden found with the food eaten by the birds and the availability of intermediate hosts.

Buffalo-adapted Theileria parva causes Corridor disease in cattle. Strict control measures therefore apply to the movement of buffalo in South Africa and include mandatory testing of buffalo for the presence of T. parva. The official test is a real-time hybridization PCR assay that amplifies the V4 hypervariable region of the 18S rRNA gene of T. parva, T. sp. (buffalo) and T. sp. (bougasvlei). The effect that mixed T. parva and T. sp. (buffalo)-like infections have on accurate T. parva diagnosis was investigated in this study. In vitro mixed infection simulations indicated PCR signal suppression at 100 to 1000-fold T. sp. (buffalo) excess at low T. parva parasitaemia. Suppression of PCR signal was found in field buffalo with mixed infections. The T. parva-positive status of these cases was confirmed by selective suppression of T. sp. (buffalo) amplification using a locked nucleic acid clamp and independent assays based on the p67, p104 and Tpr genes. The incidence of mixed infections in the Corridor disease endemic region of South Africa is significant, while the prevalence in buffalo outside the endemic area is currently low. A predicted increase of T. sp. (buffalo)-like infections can affect future diagnoses where mixed infections occur, prompting the need for improvements in current diagnostics.

There is an essential requirement for highly sensitive tools that will differentiate nematode parasites of animals and plants to the species level. For studying host range, genetic variation, virulence and resistance, the availability of well defined populations is vital. Many nematode species cannot be identified with certainty using traditional morphological or morphometric techniques. This is particularly the case for the more accessible developmental stages that, depending on the particular group concerned, live as eggs and larvae in the environment or as micro-filariae that circulate in the blood or inhabit the skin. Morphological identification of these stages requires specialized expertise and is extremely time consuming. Immunological assays have their place in nematode identification but they do not discriminate between current and previous infections, an essential requirement in many epidemiological and prevalence studies. In addition to being highly sensitive, DNA-based methods of detection define present over past infection and are not dependent on the parasite stage. Many types of methodology are available for the detection and definition of nematode DNA. This paper reviews these methods citing examples that have been used with success in the laboratory as well as the field.

Two experiments (Exps 1 and 2) were carried out to study the effect of dietary carbohydrates on the establishment of Trichuris suis in pigs. Two experimental diets based on barley flour were used; Diet 1 was supplemented with non-fermentable carbohydrates from oat hull meal, while Diet 2 was supplemented with fermentable carbohydrates from sugar beet fibre and inulin. In Exp. 1, thirty-two pigs were allocated randomly into 4 groups. Two groups were fed Diet 1 and 2 groups were fed Diet 2. Pigs from one of each diet group were inoculated with 2000 infective T. suis eggs each and the other two groups were uninfected controls. All pigs were slaughtered 8 weeks post-inoculation (p.i.). In Exp. 2, twenty-four pigs were allocated randomly into 2 groups and fed Diet 1 or Diet 2, respectively. All the pigs were inoculated with 2000 infective T. suis eggs. Six pigs from each group were slaughtered 8 weeks p.i. and the remaining 6 pigs from each group were slaughtered 12 weeks p.i. Infections were followed by faecal egg counts and worm burdens were assessed at necropsy. Pigs fed Diet 2 had lower egg counts in both experiments; in Exp. 2 the difference was significant (P<0·05). No differences were found in worm burdens 8 weeks p.i. in both experiments, however, worms from pigs on Diet 2 were significantly shorter (P<0·0001). Pigs fed Diet 2 and slaughtered 12 weeks p.i. had significantly lower worm counts (P<0·01) compared to pigs fed Diet 1. The results indicate that fermentable carbohydrates do not affect the establishment of T. suis in naïve pigs, but result in earlier expulsion and reduced growth of the established worms. Thus, diets with highly fermentable carbohydrates may be used in the control of T. suis.

The microfibrillate component of the laminated layer of Echinococcus granulosus equinus contains, except for the zone adjacent to the germinal layer, aggregates of electron-dense bodies displaying a sub-structure of electron-lucent spheres. The tegumentary syncytial cytoplasm contains randomly distributed electron-dense granules, many occurring near the apical plasmalemma, although exocytosis was rarely seen. Granules, similar in size and sub-structure to the bodies of the aggregates, also occur in the internuncial connexions and tegumentary cytons, suggesting that they may be produced in the cytons and released into the laminated layer via the internuncial connexions and tegumentary cytoplasm. Cysts incubated for 0·5–2·5 h in serum- and non-serum-containing media showed differences from non-incubated cysts. The distal half of the syncytium contained a progressive increase in the number of granules (distal:proximal 1·9:1 at 2·5 h; cf. 1:1 in non-incubated cysts), and exocytosis of granules into the laminated layer adjacent to the cyst had occurred. This cannot be attributed wholly to serum proteins. Cysts incubated for 21 h appeared ‘normal’, suggesting re-establishment of an equilibrium. Since the matrix of the laminated layer is considered homologous to the glycocalyx of other cestodes, the possible protective role played by the granules/bodies, characteristic of Echinococcus spp., is discussed.

During autumn 2000 an extensive cross-sectional survey of the prevalence of Schistosomiasis japonicum was conducted among about 4000 villagers within 20 villages in the Anning River Valley located in the southwestern Sichuan Province. Two procedures were used to assess infection status, the Kato-Katz thick smear procedure and a miracidia hatch test. Whereas the Kato-Katz procedure provides information on both prevalence and intensity, the hatch test provides only prevalence data, albeit on a much larger volume of stool. In addition, we performed Kato-Katz smears for 15 consecutive samples on a subset of 15 individuals. The proportion of both hatch-test and Kato-Katz positive individuals in the larger cross-sectional survey was 25%. The goal of the study was to estimate both the egg and worm distributions among risk groups using both the hatch and Kato-Katz tests from the cross-sectional data and the repeated Kato-Katz smears from the longitudinal data sets. As a prelude to parameter estimation, individuals were classified into risk groups by natural village and occupation; the proportion of Kato-Katz positive subjects among the risk groups varied from 10% to 60%. We used the statistical model of de Vlas et al. (1992) and Bayesian techniques to derive both estimates of and inference about the worm and egg distribution parameters. The parameter estimates imply (1) similar eggs per gram stool (e.p.g.) per worm pair compared with earlier estimates, (2) a range of worm burdens among the risk groups and (3) estimates of risk heterogeneity within groups is sensitive to prior information on the within-person variability in egg excretion.

The spermiogenesis and spermatozoan morphology of the palaeacanthocephalan, P. minutus were studied by a variety of electron microscopical methods.

Spermiogenesis occurs in syncytial clusters of spermatids exhibiting synchronous development. The outstanding events of the spermiogenic process are an enormous elongation of the spermatid nucleus and the eventual breakdown of the spermatid nuclear envelope.

The mature spermatozoon is a filiform cell about 60 μm long and motile by propagated undulations. It consists of two parallel elongate components, the single flagellum and the spermatozoan body. The former has a 9 + 2 tubule organization and the latter contains glycogen granules and membrane-bound dense inclusions as well as a region of condensed chromatin of nuclear origin. No mitochondria appear to be present.

1. A study has been made of the effect of acriflavine upon Bodo caudatus. The drug has the property of producing a percentage of modified Bodos without parabasal bodies but no permanently aparabasal strains have been produced.

2. A technique was devised whereby the resistance, as judged by two different types of survival experiment, could be correlated with the percentage of modified Bodos produced when the strain is put to grow in an ascending series of concentrations of acriflavine incorporated in the culture medium.

3. The peak of the count of altered Bodos in relation to the concentration in which it occurs is found to be the index of the sensitiveness of the strain at that date. The maximum reaction as gauged by the numbers of modified Bodos produced in an untreated strain of average sensitiveness is about 70 to 80 per cent., and this occurs in so low a concentration as 1/1,000,000. 1/5,000,000 gives a count below the maximum and 1/50,000,000 produces only 1 to 4 per cent. according to the sensitiveness of the strain.

4. The mass untreated strain was originally derived from a single Bodo isolated in the autumn of 1926. This strain in 1927–28 was used as the mass untreated strain in the experiments. Its resistance was found to fluctuate from below a capacity to evolve with great difficulty in 1/50,000 to a somewhat feeble development in 1/10,000 acriflavine incorporated plates. The mass untreated strain was never able to evolve in plates containing 1/5000 acriflavine at any time.

5. Strains grown from single isolations from the untreated mass culture showed different degrees of natural resistance. The most resistant clones could develop in 1/5000 acriflavine incorporated plates, but not in 1/2000. The great importance of this natural range of resistance in estimating an acquired drug fastness is emphasized. Strains which had been made resistant could be brought to live continuously in 1/1500 acriflavine incorporated plates, but no evolution was obtained in 1/1000.

6. Evidence that drug fastness in Bodo caudatus is due to the interaction of selective inheritance and the actual modification of the quality of the Bodo by evolution in the drug is given. The modification is not apparently a mutation, it is a heritable piling up of changes in a particular direction.

7. A high resistance once acquired is retained through prolonged cultivation upon drug-free media. A gradual loss occurs, partly by a dilution through multiplication of the character impressed and partly by the survival of variants of less resistance to acriflavine, but of perfect viability in other circumstances and the competition of these within the strain. The loss of resistance is greatly accelerated in single cell cultures (clones) isolated from the resistant strains, but up to the present no treated strain has entirely lost the effect of the original exposures to the drug. The longest time elapsing between the creating of a partially resistant strain and its test for resistance above that of the untreated culture is one year.

8. The bearing of the data obtained in Bodo caudatus upon the condition found in trypanosomiasis and upon certain aspects of chemotherapy in this group is discussed.

9. Bodo caudatus being an organism without conjugation and therefore without bi-parental inheritance and consequently lacking the reorganising effect that such a periodic closing of the cycle produces, affords an example of a labile organism capable of being progressively altered within certain limits under the influence of the environment.

The transmission success of Onchocerca volvulus is thought to be influenced by a variety of regulatory or density-dependent processes that act at various points in the two-host life-cycle. This paper examines one component of the life-cycle, namely, the ingestion of microfilariae by the simuliid vector, to assess the relationship between intake of larvae and the density of parasites in the skin of the human host. Analysis is based on data from three areas in which onchocerciasis is endemic and includes published information as well as new data collected in field studies. The three areas are: Guatemala (Simulium ochraceum sl.), West and Central Africa (savanna members of the S. damnosum complex), and South Venezuela (S. guianense). The data record experimental studies of parasite uptake by flies captured in the field and fed to repletion on locally infected subjects who harboured varying intensities of dermal microfilarial infection. Regression analyses of log transformed counts of parasite burdens ingested by the flies plotted against log transformed counts of microfilariae per mg of skin revealed little evidence for saturation in parasite uptake by the flies as the intensity in the human host increased. There was a positive and highly significant rank correlation between both variables for the three blackfly species. In an alternative analysis a model was fitted to data on prevalence of flies with ingested microfilariae (mff) versus dermal mean intensities. The model assumed an overdispersed distribution of the number of mff/fly and a given functional relationship between intake and skin load. The results of both approaches were consistent. It is concluded that parasite ingestiossssn by the vector host is not strongly density dependent in the three geographical areas and ranges of dermal loads examined. It therefore appears that this transmission process is of reduced importance as a regulatory mechanism in the dynamics of parasite population growth.

A deterministic model of the population biology of the non-infective free-living phase of Ostertagia ostertagi is described and tested. A comparison of field and laboratory studies indicates that the pre-infective larvae of O. ostertagi spend about twice as long in this phase in the field as in laboratory culture (13 days and 7 days respectively at 15 °C), and that the subsequent transition (i.e. second moult) to the infective phase also proceeds at a much slower rate (about 10 times less at 15 °C) in the field. Because of this discrepancy, estimates of model parameters are based entirely on measurements from field studies. A two-parameter model of pre-infective development in the field is derived. It describes egg and larval development as a direct non-linear function of air temperature. We also show that the mortality of the eggs and pre.infective larvae increases with temperature (although this relationship probably also subsumes a dependence on faecal moisture content). The model is shown to be a good mimic of the population dynamics of the non-infective free-living phase when tested against an independent data set, and is the basis of a larger model of the epidemiology of ostertagiasis.

Phosphorylcholine (PC), an immunodominant component of the cell wall of certain bacteria, fungi and nematodes, is known to induce low anti-PC antibody levels during natural infection by Trichinella spiralis. This article reports a study in which spleen cells from BCF1 mice infected with Trichinella sp. larvae were found to produce large numbers of direct haemolytic plaques in response to PC conjugated to sheep red blood cells (SRBC) after muscle-encysted larvae had been killed by treatment with mebendazole. Inhibition of the response by PC-chloride, immunodiffusion and immunoelectrophoretic studies with the anti-PC IgA (TEPC-15) and anti-idiotype T15 serum assays showed the plaque-forming cell (PFC) response to be specific for PC. The absence of haemolytic plaques when unconjugated SRBC or TNP-SRBC were used as indicator cells ruled out involvement of a polyclonal response. Greatest anti-PC PFC response was found to be associated with a microsomal fraction designated FCpl, a particulate fraction behaving as a thymus-dependent antigen. The FCpl fractions from all four strains of Trichinella employed induced anti-PC PFC responses when injected into mice. These results suggest that FCpl is a suitable antigen for use in detailed studies of immune responses to Trichinella and related parasites.

The Plasmodium falciparum population in Asar village, eastern Sudan, where malaria transmission is markedly seasonal, was monitored monthly over a period of 15 months. A cohort of infected patients was treated and then followed monthly throughout the dry season until the next transmission season. Parasitaemia detected by microscopy among the cohort reduced dramatically following treatment, but remained sporadic during the dry season, and reappeared following the onset of the next wet season. However between 40 and 50% of the cohort retained a persisting parasitaemia detectable by PCR throughout the dry season. These parasites were genetically complex, consisting of multiple clones with a large repertoire of alleles of the studied genes. While the number of clones per host dropped significantly following treatment of acute cases during the transmission season, drug treated people nevertheless maintained an average of one clone throughout the dry season. Allele frequencies of MSP-1, MSP-2 and GLURP showed slight, statistically insignificant, fluctuations between the dry and wet seasons. A higher frequency of inbreeding was estimated among the parasites that survived the dry season compared to the wet season.

This paper describes a study of 194 children (aged 9–13) from a mountain village in Ecuador who were infected with one or more species of intestinal helminth or protozoan parasite. In addition to parasite load, the assessment consisted of a battery of psychological and neuropsychological tests, an EEG examination, measures of iodine level, presence of goitre and level of nutrition. We found that, in general, parasite infection, as measured at the baseline level, was not associated with cognitive impairment. The intensity of infection with A. lumbricoides, however, was correlated with the level of verbal ability and with inhibition-control aspects of cognitive behaviour. Multivariate analysis with level of nutrition, EEG status and parasite burden showed a consistent main effect of the degree of nutrition on neuropsychological performance, particularly the language, problem solving and inhibition-control dimensions.

The systematics and life-cycles of the helminths of a local population of Gammarus lacustris in a eutrophic lake near Edmonton, Alberta, is reported as part of a larger study of the composition and seasonal dynamics of the helminth fauna of gammarids.

A total of 12 species of helminths, including eight cestodes, one nematode and three acanthocephalans, were recovered. Of these, eleven were new host records, ten were assigned to an intermediate host species for the first time, and one, Hymenolepis albertensis sp.nov., was described for the first time.

Adults of all twelve helminths were raised in experimentally infested birds and the life-cycles of five species (Lateriporus clerci, L. skrjabini, Hymenolepis albertensis sp.nov., Fimbriaria fasciolaris and Polymorphus marilis) were completed in the laboratory. The larvae are described, and the developmental period in the gammarids, prepatent period and life span of the adults are given for many of the helminths.

The rate of development of the cysticercoids of Lateriporus skrjabini was shown to be directly related to the size of the gammarid and inversely related to the intensity of infestation.

The proboscis-hook formula was not a good diagnostic character for the separation of the three acanthocephalans, Polymorphus contortus, P. marilis and P. paradoxus; however, the size of the largest hook and the structure of the cystacanth body-wall were good diagnostic characters.

Among six parasitised larve of Stegomya scutellaris Walker, collected by Dr W. A. Lamborn in the Federated Malay States, Five Contained a ciliate which I have described(Parasitology, XIII. p.216)under the name of Lambornella stegomyiae n.g., n.sp., while the remaining larva harboured a new parasitic fungus which forms the subject of this communication. The larva had been fixed and was preserved in formaldehyde solution (10 per cent.) and was labelled: “Larva of Stegomyia scutellaris Walker. Infestation with protozoal parasite and a luxuriant growth of Vorticella. Kajang,” by Dr Lamborn. When the larva was examined under a low power the surface of its body was found to be extensively covered with tufts of Vorticella, whilst its interior harboured an enormous number of parasites which in the gills and the posterior segments were packed in solid masses completely filling these parts (Fig. 1). As seen by transparency the parasites are oval in shape, 37·5 to 57 μ long and 20 to 30 μ in diameter, surrounded by a more or less thick yellowish wall. In their external appearance, size, and position in the host, they are so surprisingly similar to Lambornella that it was at first quite natural to take them for the resting stages or cysts of this ciliate.

1. Three new species of Myxosoma (Sporozoa, Cnidosporidia, Myxosporidia) are described from freshwater fish of the genus Tilapia in East Africa.

2. Two of these, M. heterospora and M. brachyspora spp.nov., inhabit the abdominal viscera: the former was found in 41 of 43 fish examined, while the latter was less common. The third species, M. homeospora sp.nov., produces small oval or circular ‘cysts’ in the subdermal muscle and was found in two very young fish only. Spores resembling those of the first two species were also found in one Haplochromis sp.

3. The spores of M. heterospora are roughly ellipsoidal but are very variable in shape, and range from 8·5–17·0 μ in length and 6·3–11·0 μ in breadth; the two anterior polar capsules measure from 2·0–9·5 by 1·5–3·5 μ. Those of M. brachyspora are flattened antero-posteriorly, measuring from 7·0–7·5 μ long and 12·0–13·5 μ broad; their polar capsules are from 2·5–3·8 by 2·3–2·5 μ in size. M. homeospora has ovoid spores with the anterior end narrowed, measuring from 13·5–17·0 by 8·5–11·0 μ; the polar capsules measure 4·0–6·0 by 2·0–4·0 μ.

4. It is suggested that M. heterospora, at least, is pathogenic and may play a considerable part in reducing the availability of fish as food in this part of Africa.

5. A check-list is given of all species of the genus Myxosoma.

This paper contains a revision of the morphology and classification of three species of holotrichous ciliates, Lembus pusillus, Cyclidium glaucoma and Uronema nigricans (=Uronema marina Dujardin, 1841=Cryptochilum nigricans Maupas, 1883=U. marina et U. marinum auctorum, pro parte), together with various observations on their biology. The revised data on these species are summarised in the diagnoses preceding their description.

The close resemblance between U. nigricans and the other species described has led to its transfer from the family Chiliferidae to the family Pleuronemidae.

Cyst formation in L. pusillus is described for the first time. The cysts, when fully formed, are provided with a double-layered wall, to which the body of the ciliate is more or less closely adherent. When preserved in dry condition, the cysts retain the same structure and are able to hatch after several months. Excystation is brought about by active rupture of the cyst-wall on the part of the ciliate. The primary cyst does not, however, afford adequate protection to the ciliate in a fluid medium. After remaining in the latter about three weeks, the animal secretes a cyst of the second order, in which it remains viable some time longer, but if left in the fluid, the ciliate ultimately dies, gradually breaking up into a number of granules.

In order to establish the relationship of the marine and fresh-water forms of the same species, experiments on their acclimatisation to the opposite environment were conducted. The fresh-water U. nigricans adapted itself to pure sea water within 12 days and developed for 12 months in this medium, in which it is still being maintained. The marine U. nigricans was gradually transferred to fresh water, in which it was kept for 60 days. Observations on the morphological changes undergone by this ciliate in the course of acclimatisation show that the fresh-water forms living in sea water gradually acquire the habitus of the marine ones, and that the latter undergo the reverse change in fresh water. The characters distinguishing the two strains of U. nigricans being insignificant and transitory, the two forms are regarded as constituting a single species.

The fresh-water L. pusillus required eight days for acclimatisation to sea water, in which it was maintained for 163 days without exhibiting any morphological changes.

Attempts to acclimatise the fresh-water C. glaucoma to pure sea water failed, but in 50 per cent, sea water it grew for 130 days. The morphological and biological differences between the fresh-water and marine strains of C. glaucoma are probably sufficient for them to be regarded as sub-species or races.

Experiments were conducted on the cultivation of these ciliates in faecal matter from normal and diseased human subjects, and varying widely in consistency. Tests on the influence of the reaction of the medium upon the growth of the ciliates showed that they prefer a slightly alkaline medium (pH 7·4–8·0), but are able to tolerate an increase of alkalinity up to pH 9·0–10·0, whilst the acidification of the medium beyond pH 5·0 inhibits their growth.

As regards the facility with which these ciliates grew in faecal cultures, Lembus occupied the first place, having developed in the majority (64 per cent.) of samples of pure (i.e. undiluted) faeces, and in all those slightly diluted with water. The second place belongs to Uronema, which grew in all the diluted specimens and in one undiluted, whereas Cyclidium was unable to grow in pure faeces, but developed in about half of the diluted samples. All these ciliates were successfully cultivated in suspensions of human and animal faeces.

The coprozoic habits of the ciliates in question are compared with the records for “Uronema caudatum,” comprising a number of ciliates described by some authors as human intestinal parasites. It is shown that the conditions under which these ciliates were observed indicate that they were coprozoic forms that had contaminated the material examined. Though described under the name “U. caudatum” these ciliates represent at least two distinct genera, those recorded by Martini being similar to Cyclidium or Uronema, the ones described by Yakimoff et al. probably representing Lembus pusillus.

The last-named ciliarte having been originally found in a sample of canine faeces, experiments were carried out with the view of ascertaining its origin. It was demonstrated that the ciliate was unable to withstand incubation at 37°C. for 24 hours, and, when fed to mice in the form of cysts, did not survive the passage through their intestine. These facts indicate that the faeces in which Lembus was found must have been contaminated by this ciliate from outside.

Two dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (2D SDS–PAGE) has been used to produce ‘fingerprint’ maps of the proteins from each of the 7 species of Eimeria which infect the chicken. All 7 species could be identified from their array of polypeptides but few differences were detected between strains of the same species. Alterations to the polypeptide array associated with the stage of sporulation of the oocysts were observed. lodination of sporozoites, 2D SDS–PAGE, autoradiography and immunoblotting techniques were combined to identify polypeptides with a surface moiety and those which were antigenic.

The effect of acquired immunity on the duration of Plasmodium falciparum infections is unclear, although this is an important term in models of malaria transmission. It is problematical to determine the duration of infections because of the difficulty of distinguishing persisting infections from new ones, and because parasite densities are often transiently below the limit of detection. We recently developed a dynamic model for infection incidence, clearance and detection of multiple genotype P. falciparum infections and fitted it to a panel dataset from a longitudinal study in Northern Ghana. We now extend this model to allow for seasonal and age variation in infection rates and also age dependence in clearance and in detectability of infections. These models indicate that there is seasonal variation in the infection rate, and age dependence in detectability. The best fitting models had no age dependence in infection or clearance rates, suggesting that acquired immunity mainly affects detectability.

In this paper, we aimed to explore the effects of the calpain inhibitor III (MDL28170) and to detect calpain-like molecules (CALPs) in epimastigote forms of Trypanosoma cruzi isolate Dm28c. MDL28170 at 70 μM promoted a powerful reduction in the growth rate after 48 h. The IC50 value was calculated to be 31·7 μM. This inhibitor promoted an increase in the cellular volume, but not cell lysis, resulting in a trypanostatic effect. T. cruzi CALPs presented a strong cross-reactivity with anti-Drosophila melanogaster calpain and anti-cytoskeleton-associated protein from Trypanosoma brucei antibodies, and labelling was found mainly intracellularly. Furthermore, an 80 kDa reactive protein was detected by Western blotting assays. No significant cross-reactivity was found with anti-human brain calpain antibody. The expression of CALPs was decreased in cells kept for long periods in axenic cultures in comparison to a strain recently isolated from mice, as well as in MDL28170-treated cells, the latter being paralleled by an increased expression of cruzipain. Different levels of CALPs expression were also detected in distinct phylogenetic lineages, like Y strain (lineage TCI), Dm28c (TCII) and INPA6147 strain (Z3 zymodeme). These results may contribute for the investigation of the functions of CALPs in trypanosomatids.
