The role that vitamin D plays in pulmonary function remains uncertain. Epidemiological studies reported mixed findings for serum 25-hydroxyvitamin D (25(OH)D)–pulmonary function association. We conducted the largest cross-sectional meta-analysis of the 25(OH)D–pulmonary function association to date, based on nine European ancestry (EA) cohorts (n 22 838) and five African ancestry (AA) cohorts (n 4290) in the Cohorts for Heart and Aging Research in Genomic Epidemiology Consortium. Data were analysed using linear models by cohort and ancestry. Effect modification by smoking status (current/former/never) was tested. Results were combined using fixed-effects meta-analysis. Mean serum 25(OH)D was 68 (sd 29) nmol/l for EA and 49 (sd 21) nmol/l for AA. For each 1 nmol/l higher 25(OH)D, forced expiratory volume in the 1st second (FEV1) was higher by 1·1 ml in EA (95 % CI 0·9, 1·3; P<0·0001) and 1·8 ml (95 % CI 1·1, 2·5; P<0·0001) in AA (Prace difference=0·06), and forced vital capacity (FVC) was higher by 1·3 ml in EA (95 % CI 1·0, 1·6; P<0·0001) and 1·5 ml (95 % CI 0·8, 2·3; P=0·0001) in AA (Prace difference=0·56). Among EA, the 25(OH)D–FVC association was stronger in smokers: per 1 nmol/l higher 25(OH)D, FVC was higher by 1·7 ml (95 % CI 1·1, 2·3) for current smokers and 1·7 ml (95 % CI 1·2, 2·1) for former smokers, compared with 0·8 ml (95 % CI 0·4, 1·2) for never smokers. In summary, the 25(OH)D associations with FEV1 and FVC were positive in both ancestries. In EA, a stronger association was observed for smokers compared with never smokers, which supports the importance of vitamin D in vulnerable populations.
OBJECTIVES/SPECIFIC AIMS: Patients with locally advanced pancreatic cancer typically have poor outcomes, with a median survival of ~16 months. Novel methods to improve local control are needed. Nab-paclitaxel (abraxane) has shown efficacy in pancreatic cancer and is FDA approved for metastatic disease in combination with gemcitabine. Nab-paclitaxel is also a promising radiosensitizer based on laboratory studies, but it has never been clinically tested with definitive radiotherapy for locally advanced disease. METHODS/STUDY POPULATION: We performed a phase 1 study using a 3+3 dose-escalation strategy to determine the safety and tolerability of dose escalated nab-paclitaxel with fractionated radiotherapy for patients with unresectable or borderline resectable pancreatic cancer. Following induction chemotherapy with 2 cycles of nab-paclitaxel and gemcitabine, patients were treated with weekly nab-paclitaxel and daily radiotherapy to a dose of 52.5 Gy in 25 fractions. Final dose-limiting toxicity (DLT) determination was performed at day 65 after the start of radiotherapy. RESULTS/ANTICIPATED RESULTS: Nine patients received nab-paclitaxel at a dose level of either 100 mg/m2 (n=3) or 125 mg/m2 (n=6). One DLT (grade 3 neuropathy) was observed in a patient who received 125 mg/m2 of nab-paclitaxel. Other grade 3 toxicities included fatigue (11%), anemia (11%), and neutropenia (11%). No grade 4 toxicities were observed. With a median follow-up of 8 months (range 5–28 months), median survival was 19 months and median progression-free survival was 10 months. Following chemoradiation, 3 patients underwent surgical resection, all with negative margins and limited tumor viability. Of the 3 patients, 2 initially had borderline resectable tumors and 1 had an unresectable tumor. Tumor (SMAD-4, Caveolin-1) and peripheral (circulating tumor cells and microvesicles) biomarkers were collected and are being analyzed. DISCUSSION/SIGNIFICANCE OF IMPACT: The combination of fractionated radiation and weekly nab-paclitaxel was safe and well tolerated. This regimen represents a potentially promising therapy for patients with unresectable and borderline resectable pancreatic cancer and warrants further investigation.
Effectiveness of the common soil fungus Sclerotinia sclerotiorum as a biological control for spotted knapweed and its effect on competitive interactions between spotted knapweed and bluebunch wheatgrass were evaluated in a growth chamber study using addition series methods. Total seeding densities ranged from 2000 to 60 000 seeds/m2. Mean bluebunch wheatgrass plant weight was 3.5 times greater than spotted knapweed weight per plant, respectively. Coefficient ratios estimating species interaction showed bluebunch wheatgrass density had a greater influence than spotted knapweed density on both bluebunch wheatgrass and spotted knapweed weights (2.11 and 0.51, respectively) when not under the influence of S. sclerotiorum. Niche differentiation ratios indicated a lack of resource partitioning between species (1.11). S. sclerotiorum reduced spotted knapweed density by 68 to 80% without reducing bluebunch wheatgrass density. Spotted knapweed weight per plant also was reduced by the addition of 5. sclerotiorum (1.4 to 1.2 mg) but there was not a corresponding increase in bluebunch wheatgrass weight. S. sclerotiorum decreased competition between spotted knapweed and bluebunch wheatgrass. This study provides evidence that establishment of bluebunch wheatgrass on spotted knapweed infested rangeland may be improved by combining S. sclerotiorum with high grass seeding rates.
Two broad aims drive weed science research: improved management and improvedunderstanding of weed biology and ecology. In recent years, agriculturalweed research addressing these two aims has effectively split into separatesubdisciplines despite repeated calls for greater integration. Although someexcellent work is being done, agricultural weed research has developed avery high level of repetitiveness, a preponderance of purely descriptivestudies, and has failed to clearly articulate novel hypotheses linked toestablished bodies of ecological and evolutionary theory. In contrast,invasive plant research attracts a diverse cadre of nonweed scientists usinginvasions to explore broader and more integrated biological questionsgrounded in theory. We propose that although studies focused on weedmanagement remain vitally important, agricultural weed research wouldbenefit from deeper theoretical justification, a broader vision, andincreased collaboration across diverse disciplines. To initiate change inthis direction, we call for more emphasis on interdisciplinary training forweed scientists, and for focused workshops and working groups to developspecific areas of research and promote interactions among weed scientistsand with the wider scientific community.
We determined individual distances to a small number of rather round, quite regularly shaped planetary nebulae by combining their angular expansion in the plane of the sky with a spectroscopically measured expansion along the line of sight. For this goal, we combined up to three epochs of Hubble Space Telescope imaging data and determined the respective proper motions of rim and shell edges, and of other features as well. Ground-based radial velocities are assigned separately to the nebular rims and shells and used to determine individual distances, thereby assuming that the expansions in the line-of-sight and in the plane of sky are equal. We employed 1D radiation-hydrodynamics simulations of planetary nebulae evolution to correct for the difference between the spectroscopically measured expansion velocities of rim and shell and the expansion speeds of their respective shock fronts.
To assess the mean package size and manufacturer-recommended serving size of sweet beverages available in four high-income countries: Australia, Canada, the Netherlands and New Zealand.

The two largest supermarket chains of each country in 2012/2013.
For all IPS drinks, the mean package size was larger than the mean serving size (mean (sd)=412 (157) ml and 359 (159) ml, respectively). The mean (sd) package size of IPS drinks was significantly different for all countries (range: Australia=370 (149) ml to New Zealand=484 (191) ml; P<0·01). The mean (sd) package size of Dutch BPS drinks (1313 (323) ml) was significantly smaller compared with the other countries (New Zealand=1481 (595) ml, Australia=1542 (595) ml, Canada=1550 (434) ml; P<0·01). The mean (sd) serving size of BPS drinks was significantly different across all countries (range: Netherlands=216 (30) ml to Canada=248 (31) ml; P<0·00). New Zealand had the largest package and serving sizes of the countries assessed. In all countries, a large number of different serving sizes were used to provide information on the amount appropriate to consume in one sitting.
Conclusions
At this point there is substantial inconsistency in package sizes and manufacturer-recommended serving sizes of sweet beverages within and between four high-income countries, especially for IPS drinks. As consumers do factor serving size into their judgements of healthiness of a product, serving size regulations, preferably set by governments and global health organisations, would provide consistency and assist individuals in making healthier food choices.
Other than during the Civil War of 1945–1949, Taiwan has never been part of a Chinese state ruled by Han Chinese in Mainland China. With the arrival of the Dutch in 1624, Taiwan underwent a succession of six foreign colonial rulers: the Dutch (1624–1662), the Spanish (in northern Taiwan, 1626–1642), the Cheng family (1662–1683), the Manchus (1683–1895), the Japanese (1895–1945), and the Chinese Nationalists (1945–1988). Only with democratization, beginning in 1988, have the people of Taiwan been able to rule themselves. With democratization, the people of Taiwan have increasingly identified as Taiwanese, an identification that is explicitly not Chinese. China's racial appeals to Taiwan are scientifically inaccurate and have no place in the modern world. In order to gain international support, Taiwan's leaders and people need to avoid the Chinese framework of ‘independence’ versus ‘unification’. Rather, they should emphasize Taiwan's decolonialization, a process that Taiwan shares with much of the world.
Street criminals live in a dangerous world, but they cannot realistically rely on the criminal justice system to protect them from predation by fellow lawbreakers; they are on their own when it comes to dealing with crimes perpetrated against them and often use retaliation as a mechanism for deterring and responding to victimization. Although retaliation lies at the heart of much of the violence that plagues many inner-city neighborhoods across the United States, it has received scant attention from criminologists. As a result, the structure, process, and forms of retaliation in the real world setting of urban America remain poorly understood. Street Justice: Retaliation in the Criminal World, first published in 2006, explores the face of modern day retaliation from the perspective of currently active criminals who have experienced it first hand, as offenders, victims, or both.
High-content analytical methodologies are increasingly becoming integrated into the screening phase of the discovery pipeline for small molecule modulators of biological targets. The iterative analysis among targets with related functions or among a series of interacting targets can be replaced with multiplexing approaches whereby multiple targets are analyzed simultaneously in the same well against the same compound or even against a mixture of compounds in the same well. It has become practical to use the multiparameter optical capabilities of the flow cytometer for the discovery of active small molecules from compound libraries by high-throughput screening (HTS). In this chapter, we present the fundamental principles of flow cytometry–based multiplex methods, including assay design and data analysis, with specific examples of cell-based and bead-based assays and a discussion of the added value of multiplex data sets.
Overview
Over the past two decades, HTS methodologies (generally defined as testing ten thousand to one hundred thousand compounds per day, depending on the technology) have become an essential part of discovery science for novel drugs and biological probes within the pharmaceutical, biotech, and academic research communities [1–3]. Technologies that are inherently capable of multiparametric measurements, including microscopy and flow cytometry, are well suited to produce high-content, high-complexity data by obtaining multiple data points from each well in a microtiter plate or other high-density format. In multiplex applications, information content is further enriched by obtaining data for each parameter on multiple targets. The advantages, compared with single-point iterative assays, include a substantial savings in time and reagents and the ability to produce rich data sets under conditions that can be optimized for statistical rigor and data quality for multiple targets simultaneously. For example, multiplex analysis among a family of related targets, which often exhibit similar biochemical and pharmacologic properties, produces selectivity screens in a single well with inherent consistency. Furthermore, the conditions in each well are normalized across targets for any number of parameters, and counterscreens for artifacts can be rigorously examined. Table 17.1 is a comparison of characteristics between single-target and multiplexed assays that summarizes the advantages for each approach.
The development of novel organic polymer thin films is essential for the advancement of many emerging fields including organic electronics and biomedical coatings. In this study, the effect of synthesis conditions, namely radio frequency (rf) deposition power, on the material properties of polyterpenol thin films derived from nonsynthetic environmentally friendly monomer was investigated. At lower deposition powers, the polyterpenol films preserved more of the original monomer constituents, such as hydroxy functional groups; however, they were also softer and more hydrophilic compared to polymers fabricated at higher power. Enhanced monomer fragmentation and consequent reduction in the presence of the polar groups in the structure of the high-power samples reduced their optical band gap value from 2.95 eV for 10 W to 2.64 eV for 100 W. Regardless of deposition power, all samples were found to be optically transparent with smooth, defect-free, and homogenous surfaces.
Between November 1988 and January 1989, a total of 155 people in 36 reported outbreaks suffered gastrointestinal symptoms associated with eating Stilton cheese, produced from unpasteurized cow's milk in the English midlands. Symptoms were suggestive of a staphylococcal illness but extensive laboratory testing of cheeses implicated in several of the outbreaks failed to detect any pathogen, toxin or chemical. Control measures were implemented, and included a voluntary withdrawal of the implicated Stilton cheese from sale on 23 January 1989 and a subsequent decision to use pasteurized milk in production of the cheese.
An outbreak of Salmonella dublin infection occurred in England and Wales in October to December 1989. Forty-two people were affected, mainly adults, and most lived in south-east England. Microbiological and epidemiological investigations implicated an imported Irish soft unpasteurized cows' milk cheese as the vehicle of infection. A case-control study showed a statistically significant association between infection and consumption of the suspect cheese (p = 0·001). Salmonella dublin was subsequently isolated from cheeses obtained from the manufacturer's premises. Initial control measures included the withdrawal of the cheese from retail sale and a Food Hazard Warning to Environmental Health Departments, as well as a press release, from the Department of Health. Subsequently, a decision was taken by the manufacturer to pasteurize milk used in the production of cheese for the UK market and importation of the cheese resumed in June 1990.
This paper presents an operational semantics for the core of Scheme. Our specification improves over the denotational semantics from the Revised5 Report on Scheme specification in four ways. First, it covers a larger part of the language, specifically eval, quote, dynamic-wind, and the top level. Second, it models multiple values in a way that does not require changes to unrelated parts of the language. Third, it provides a faithful model of Scheme's undefined order of evaluation. Finally, we have implemented our specification in PLT Redex, a domain-specific language for writing operational semantics. The implementation allows others to experiment with our specification and allows us to build a specification test suite, which improves our confidence that our system is a faithful model of Scheme. In addition to a specification of Scheme, this paper contributes three novel modeling techniques for Felleisen Hieb-style rewriting semantics. All three techniques are applicable to a wider range of problems than modeling Scheme, and they combine seamlessly in our model, suggesting that they would scale to complete models of other languages.
Lee Teng-hui, president of Taiwan from 1988 to 2000, played an important role in launching the widespread nationalism in Taiwan today. Since the end of his presidency, Lee has pushed very hard for a separate Taiwan nation, but during his terms of office he moved very cautiously and only announced his “special state-to-state” relationship to describe Taiwan's relations with China after China repeatedly refused to negotiate. Although Lee's efforts at democratization have received widespread affirmation in Taiwan, his efforts in creating Taiwan nationalism have proven more controversial.
